ABSTRACT

OBJECTIVE@#To determine the expression level of linc-223 and miR-125a in the patients with adult acute leukemia (AL) and explore the relationship between the expression level and the occurrence, development, prognosis of leukemia.@*METHODS@#Bone marrow samples of 93 patients with AL treated in our hospital from January 2017 to September 2017 were enrolled, including 21 cases of acute lymphoblastic leukemia (ALL) and 72 cases of acute non-lymphocytic leukemia (ANLL). At the same time, bone marrow samples from 20 cases of non-malignant hematopathy patients in the same period were enrolled as control group. Real-time quantitative PCR (qRT-PCR) was used to test the expression level of linc-223 and miR-125a in bone marrow of 93 AL patients and the relationship between the level and the occurrence, development, prognosis of leukemia was analyzed.@*RESULTS@#There was no significant difference in the expression of linc-223 between AL patients (ANLL and ALL) and control group (P>0.05). Moreover, there was no significant correlation between linc-223 and PML-RARα gene or the remission rate of patients after treatment. The expression of miR-125a in ANLL patients was significantly lower than those in the control group (P0.05), and also for ALL and control group (P>0.05). In the newly treatment ANLL patients, the expression level of miR-125a showed negatively correlated with LDH level and the ratio of immature cells (r=-0.454, r=-0.400), but not with sex, degree of risk, peripheral blood leukocyte count, platelet count, hemoglobin content, WT1, CRP, etc. (P>0.05). There was a positive correlation between linc-223 and miR-125a in ANLL patients (r=0.296).@*CONCLUSION@#No abnormal expression of linc-223 was found in the bone marrow of AL patients, but miR-125a expression shows a low level and positively correlate with the expression level of linc-223 in ANLL, which is helpful for the diagnosis.

ABSTRACT

Objective To provide reference for further promoting the development ability of county-level public hospitals by analyzing the influencing factors of the development ability of county-level public hospitals in Jiangsu Province from 2014 to 2016.Methods Different development indicators of 32 county-level public hospitals in Jiangsu Province were collected,and data were analyzed by Excel2007,and SPSS22.0 was used to analyze the comprehensive development ability of hospitals.Results Scale development factor,human resources development factor and medical service development factor have impacts on the development capacity of county public hospitals in Jiangsu Province.Different hospitals have different scores.on three factors,and their development ability is identical.Conclusion The scale of the hospital,the staff development and the medical service are important to the development of the count-level public hospitals.

ABSTRACT

Objective To research the efficacy in the treatment of advanced esophageal cancer with Hongwu Mixture in combination with chemotherapy. Methods Forty-seveb advanced esophageal cancer patients were admitted, then were divided into two groups, one was the treatment group (24 cases, Hongwu Mixture combined with chemotherapy), and the other was the control group (23 cases, chemotherapy alone). After two cycles treatment, the clinical efficacy was observed. Results In the short team efficacy, the effective rate in the treatment group was 50.0% and the disease control rate was 87.5%, while these rates were 30.4% and 60.9% respectively in the control group. The difference of the effective rate between the two groups was no statistically significant (P > 0.05); But the difference of the disease control rate was statistically significant (P 0.05). Conclusion The treatment of Hongwu Mixture combined with chemotherapy in advanced esophageal cancer can improve the efficacy of chemotherapy and the quality of life. Moreover, it is safe and reliable without enhancing the toxic and side effects.

ABSTRACT

Previous studies have found that Musashi-2 (Msi-2) plays an essential role in regulating the gene expression of stem cell populations by inhibiting or activating the translation, and thus regulates stem cell function. Current research shows that Msi-2 involved in the regulation of a variety of malignant hematological diseases through different mechanisms; moreover abnormally expressed in a variety of malignant hematological diseases and involved in the occurrence, development of a variety of malignant hematological diseases. Further study on the role of malignant hematologic diseases will further clarify the pathogenesis of malignant hematologic diseases, and provide a new basis for the diagnosis and treatment of malignant hematological diseases. In this review, the structure and function of Msi-2 and its relationship with malignant hematologic diseases and its latest research progress are summarized.

ABSTRACT

Inflammasome is a group of polyprotein complexes located in the cytoplasm, its activation can induce the maturation and release of proinflammatory cytokines IL-1β and IL-18, and promote the early atherosclerosis. In the recent years, it is found that the inflammasome is activated in thrombotic deseases, moveover, the activated inflammasome and its activation induced cytokines promote the occurrence and development of thrombolic deseases, and show the unfavaourable effect on prognosis. With further exploration on the mechanisms of thrombotic diseases, the relationship between the inflammasome and thrombotic diseases increasingly become a hot spot of research. This review focuses on the action mechanisms of inflammasome in thrombotic diseases.

ABSTRACT

MicroRNAs (miRNAs) are a class of endogenous non-coding single-stranded small noncoding RNAs with the length of 20 to 23 nucleotides. MicroRNA-125 (miR-125) family, which is a highly conserved miRNA family, is consist of miR-125a, miR-125b-1 and miR-125b-2. Accumulating evidence demonstrated that miR-125 can be involved in various physiological and pathological processes in vivo. Importantly, it is closely related with the tumorigenesis and tumor development, including tumor cell proliferation, apoptosis, invasion and metastasis, metabolism and immune response. In malignant hematologic diseases, it is defined either as a oncogene, or as a tumor suppressor gene, even, closely related with the drug resistance in a variety of hematologic malignancies. MiR-125 is expected to become a new therapeutic target. Newly, the research of the relationship between miR-125 family and hematologic malignancies become increasing, including leukemia, lymphoma, multiple myeloma. In this review, the relationship between miR-125 family with malignant hematologic diseases and its latest research progress are summarized.

ABSTRACT

Long non-coding RNA (LncRNA) is defined as a class of transcripts more than 200 nucleotides in length and without the protein-coding function. It has been found for years, however, that little is known about the potential role of LncRNA in humans. But recent studies showed that LncRNA can regulate the coding-gene expression and participate in effects of human body. Accumulating evidence demonstrated that LncRNA are involved in cancer incidence, development and progression.With further exploration on the mechanisms of tumors, the relationship between the long non-coding RNA and hematological malignancies increasingly become a hot research. This review focuses on the mechanisms of LncRNA in hematological malignancies.

ABSTRACT

CRISPR/Cas genome editing technology is a newly developed powerful tool for genetic manipulation, which can be used to manipulate the genome at specific locations precisely, to restore the function of genetic defect cells, and to develop various disease models. In recentl years, with the advances of precise genome manipulation, CRISPR/Cas technology has been applied to many aspects of diseases research and becomes an unique tool to investigate gene function and discover new therapeutic targets for genetic diseases. Nowadays, CRISPR/Cas technology has been a hot research point in agriculture, graziery, biotechnology and medicine. This review focuses on the recent advances in CRISPR/Cas technology and its application in hematological diseases.

ABSTRACT

To evaluate lesion detection of MR1 in knee joint osteoarthritis in patients with symptoms of pain,the correlation between MRI findings and varying degrees of reported pain was assessed.Twenty-eight patients(31 knees)with osteoarthritis were recruited for this study.The degree of knee pain was assessed by VRS scores.The knees were evaluated by plain film radiograph utilizing Kellgren-Lawrence scores.Multiple MR sequences were performed on a 1.5T MR-system,including sagittal and coronal dual fast spin echo(TR/TE 3660/11/120 ms,slice thickness 5 mm),coronal spin echo T1-weighted(TR/TE 360/9 ms,slice thickness 5 mm),sagittal fat saturated 3D-spoiled gradient-recalled echo(TR/TE 50/6 ms; slice thickness 1.5 mm; flip angle 40°),and 3D steady-state free precession (TR/TE 6/2.2 ms; slice thickness 1.6 mm: flip angle 30°)pulse sequences for the purpose of detecting abnormities of cartilage,menisci,the anterior cruciate ligaments,bone marrow edema-like lesions,osteophytes,synovitis,and joint effusions.MR findings were compared with the degree of pain using Fisher exact test with P values less than 0.05 indicating a statistically significant difference.The results showed that,of the 31 knees evaluated,mild pain was reported in 11 and severe pain in the remainder.Kellgren-Lawrence scores of all 31 evaluated OA knees were as follows: grade 1 lesions(n=6),grade 2 lesions(n=14),grade 3 lesions(n=8),and grade 4 lesions(n=3).Articular cartilaginous defects were found in 37.1% of knees.Abnormalities of the menisci and anterior cruciate ligaments,bone marrow edema-like lesions,osteophytes,synovitis,and joint effusions were detected in 32.3%,38.7%,45.2%,100%,15.1% and 67.7% of knees,respectively.Of these variables,only the differences in prevalence of joint effusions were significantly different in the mild and severe pain groups(P=0.004).It is concluded that MRI evaluates the entire joint structure of the osteoarthritic knee,demonstrating abnormalities of the cartilage,menisci,and anterior cruciate ligaments as well as bone marrow edema-like lesions,osteophytes,synovitis,and joint effusions.The difference in pain grading between OA patients reporting mild and severe degrees of pain is related to the presence of joint effusion.

ABSTRACT

<p><b>BACKGROUND</b>Spinal hyperbaric ropivacaine may produce more predictable and reliable anesthesia than plain ropivacaine for cesarean section. The dose-response relation for spinal hyperbaric ropivacaine is undetermined. This double-blind, randomized, dose-response study determined the ED50 (50% effective dose) and ED95 (95% effective dose) of spinal hyperbaric ropivacaine for cesarean section anesthesia.</p><p><b>METHODS</b>Sixty parturients undergoing elective cesarean section delivery with use of combined spinal-epidural anesthesia were enrolled in this study. An epidural catheter was placed at the L1 approximately L2 vertebral interspace, then lumbar puncture was performed at the L3 approximately L4 vertebral interspace, and parturients were randomized to receive spinal hyperbaric ropivacaine in doses of 10.5 mg, 12 mg, 13.5 mg, or 15 mg in equal volumes of 3 ml. Sensory levels (pinprick) were assessed every 2.5 min until a T7 level was achieved and motor changes were assessed by modified Bromage Score. A dose was considered effective if an upper sensory level to pin prick of T7 or above was achieved and no intraoperative epidural supplement was required. ED50 and ED95 were determined with use of a logistic regression model.</p><p><b>RESULTS</b>ED50 (95% confidence interval) of spinal hyperbaric ropivacaine was determined to be 10.37 (5.23 approximately 11.59) mg and ED95 (95% confidence interval) to be 15.39 (13.81approximately 23.59) mg. The maximum sensory block levels and the duration of motor block and the rate of hypotension, but not onset of anesthesia, were significantly related to the ropivacaine dose.</p><p><b>CONCLUSION</b>The ED50 and ED95 of spinal hyperbaric ropivacaine for cesarean delivery under the conditions of this study were 10.37 mg and 15.39 mg, respectively. Ropivacaine is suitable for spinal anesthesia in cesarean delivery.</p>

ABSTRACT

One strain of Pseudoalteromonas sp. E18 was isolated from the sea mud of Ningbo, Zhejiang. It can produce indigo pigment. The morphological, cultural and biochemical characteristics of the bacterium were studied. The indigo pigment was also extracted. The results showed that the maximum absorption peak of the pigment was at 579nm. The pigment was stable to UV, Na_2SO_3,It was also stable at pH 3~9. The pigment was unstable to the sunlight and high concentration H_2O_2. Temperature of 60℃~80℃ could increase the hue while temperature higher than 90℃ could reduce the hue.
