ABSTRACT

ObjectiveTo analyze the epidemiological distribution and temporal trends of liver cancer incidence among Luzhou residents from 2016‒2022, and to provide a theoretical basis for improving liver cancer prevention and treatment strategies in Luzhou. MethodsData on liver cancer incidence among Luzhou residents from 2016 to 2022 were collected, and the incidence rate, age-specific incidence rate, and annual percentage change (APC) were calculated. A Joinpoint regression model was used to fit a time series segment to the monthly number of new cases in each district and county of Luzhou to explore the trend of liver cancer incidence rate. ResultsThe incidence rate of liver cancer in Luzhou increased from 22.96/105 in 2016 to 32.31/105 in 2022. The incidence rate of liver cancer in men was higher than that in women in both 2016 and 2022, and the incidence rate of liver cancer in men increased from 34.83/105 in 2016 to 47.95/105 in 2022, with an APC of 3.3%; the incidence rate of liver cancer in women increased from 10.50/105 in 2016 to 15.95/105 in 2022, with an APC of 3.0%, and the differences in the change trends were not statistically significant (P>0.05).The incidence of liver cancer was low in the age group of 0‒<40 years from 2016 to 2022 and increased with age; the incidence of liver cancer in the age group of 55 years and above was increasing at an average annual rate of 16.4%. ConclusionThe overall incidence of liver cancer in Luzhou is on the rise, and the incidence of liver cancer in men is higher than that in women. Middle-aged and elderly men are the key population for liver cancer prevention and treatment, and liver cancer prevention and treatment should be carried out in a targeted manner, taking into account regional development differences.

ABSTRACT

Objective:To evaluate the efficacy of levonorgestrel-releasing intrauterine system (LNG-IUS) (mirena) versus mifepristone therapy for adenomyosis.Methods:We retrived the Cochrane library, the Pubmed, EMbase, China National Knowledge Infrastructure (CNKI), WanFang Data, VIP, China Biomedical Literatue Database (CBM) and other databases to identify randomized controlled trials (RCTs) involving the comparison of levonorgestrel-releasing intrauterine system and mifepristone up to October 2018. The quality assessment and data extraction for RCTs were conducted by two reviewers independently. Then data were analyzed with Stata 11.0 software.Results:A total of 17 RCTs involving 1 772 patients were included. The results indicated that mirena was more effective than mifepristone [RR=1.25, 95% CI(1.19, 1.32), P<0.01]. The decrease in endometrial thickness [MD=-1.94, 95% CI(-2.19, -1.70), P<0.01], uterine volume [MD=-18.99, 95% CI(-19.90, -18.07), P<0.01] and the incidence of adverse reactions [RR=0.67, 95% CI(0.47, 0.95), P=0.026] in mirena group were significantly different compared with the mifepristone group. Also the dysmenorrhea score and menstrual blood volume of mirena group were lower than mifepristone group ( P<0.05). Conclusions:Current evidence shows that the efficacy and safety of mirena was better than mifepristone in the treatment for adenomyosis patients. Since the limitation of quantity and quality of included studies, large-scale high-quality RCTs are needed to verify the above conclusion.

ABSTRACT

Objective@#By comparing different repairing effects of different methods on articular cartilage defects in rabbit model, the ability of a new bioactive glass scaffold to repair cartilage defect was studied.@*Methods@#We prepared the PSC/CS bone cement by mixing the bioactive glass (BG) powder composed of 10.8%P2O5-54.2%SiO2-35%CaO (PSC), chitosan solution (CS) and calcium sulfate hemihydrate (CSH), then the bone cement was sized by a mold to form cylindrical scaffold. In vivo experiment, 18 male rabbits were divided into three groups randomly, including blank group, BG group and TGF-β1 group, which was added TGF-β1 into BG scaffold. Both knees of each rabbit were made cartilage defect for the same group, and no intervention was applied in B group, then implanted scaffolds into defects in both experimental groups. At 6 and 12 weeks after surgery, observed the macroscopic growth, histologic staining and collagen II immunohistochemistry (IHC), and the International Cartilage Repair Society (ICRS) and Wakitani score were used to analyze the experimental results quantitatively.@*Results@#At 6 weeks after surgery, there was no obvious difference between blank group and BG groups, but the macroscopic result of TGF-β1 group was better than the other two groups and its ICRS score 4.67±0.52 points was statistically higher than BG group 2.83±0.75 points (t=-2.817, P=0.015). As to the comparison of histologic staining and Wakitani socre among three groups, no statistical difference was observed (blank group=13.67±0.52, BG group=13.83±0.41, TGF-β1 group=13.33±1.03). At 12 weeks after surgery, there was still no obvious difference between blank and BG groups, while the results of macroscopic observation and ICRS score in TGF-β1 group were significantly higher than them 9.01±0.63 points (blank group vs TGF-β1 group: t=-2.289, P=0.022; BG group vs TGF-β1 group: t=-2.326, P=0.020). More importantly, much deeper positive staining were observed in TGF-β1 group, and the Wakitani score was higher than the other two groups (blank group=9.83±1.33, BG group=9.51±1.05, TGF-β1 group=6.50±1.38, blank group vs TGF-β1 group: t=-2.771, P=0.007; BG group vs TGF-β1 group: t=-2.756, P=0.006). By comparing the degree of histologic staining and Col II expression with normal cartilage, the regenerated tissue in TGF-β1 group was similar.@*Conclusion@#Single PSC/CS scaffold doesn’t possess excellent ability to repair cartilage defect. When TGF-β1 was added into PSC/CS bioactive glass, the scaffold was able to promote cartilage defect repair, and the regenerated tissue was similar to normal cartilage.

ABSTRACT

Objective To observe the clinical effect of Jiawei-Xiaoxianxiong decoction combined with conventional western medicine for the refractory angina pectoris with Qi stagnation and blood stasis syndrome and to explore its mechanism. Methods A total of 98 patients who met the inclusion criteria were divided into two groups according to the random number table method, 49 in each group. The control group was treated with conventional therapy. The study group was treated with Jiawei-Xiaoxianxiong decoction on the basis of the control group. Both groups were treated for 4 weeks. The symptom of angina pectoris was scored before and after treatment. The Serum level of cardiotrophin-1 (CT-1), cytochrome C (Cyt C), soluble intercellular adhesion molecule-1 (sICAM-1), von Willebrand Factor (vWF), MDA and SOD were measured by ELISA, and the clinical effect was evaluated. Results The total effective rate was 75.5% (37/49) in the study group and 63.3% (31/49) in the control group. There was significant difference between two groups (χ2=5.235, P=0.022). After treatment, the level of CT-1 (105.19 ± 12.04 ng/L vs. 158.25 ± 14.18 ng/L, t=19.967), Cyt C (0.53 ± 0.11 ng/L vs. 0.81 ± 0.15 ng/L, t=10.537) in the study group were significantly lower than those in the control group (P<0.01); the serum level of sICAM-1 (261.13 ± 38.40 μg/L vs. 415.55 ± 42.26 μg/L, t=18.930), vWF (105.22 ± 18.63 μg/L vs. 145.57 ± 21.18 μg/L, t=10.013), MDA (4.80 ± 0.59 mol/L vs. 6.21 ± 0.87 mol/L, t=9.389) in the study group were significantly lower than those of the control group (P<0.01); the SOD (12.07 ± 1.84 mol/L vs. 10.89 ± 1.65 mol/L, t=3.342) level in the study group was significantly higher than that of the control group (P<0.01). The number of attacks, duration, degree of pain, nature of pain and nitroglycerin dosage in the study group were significantly lower than those in the control group (t=9.711, 11.521, 8.818, 13.922, 17.791, respectively, all Ps<0.01). Conclusions The Jiawei-Xiaoxianxiong decoction combined with routine western medicine therapy can improve the symptoms of angina and clinical efficacy in patients with refractory angina pectoris of Qi stagnation and blood stasis syndrome. It is speculated that its mechanism may be related to anti-myocardial cell apoptosis, alleviating vascular endothelial injury and antioxidation.

ABSTRACT

Objective To evaluate systematically the efficacy of mirror therapy for improving the upper limb function, daily life and pain of stroke survivors. Methods The PubMed, EBSCO, Web of Science, Ovid, Co-chrane Library, Open Gray, China Biological Medicine, China National Knowledge Infrastructure, GoogleScholar, VIP and Wan Fang Data databases were searched for reports of randomized and controlled trials ( RCTs) of mirror therapy for improving the upper limb function of stroke survivors. Only papers published before December 2015 were surveyed. The relevant references of the primary studies were also searched. The literature screening, data extraction and assessments were conducted by two reviewers independently. The meta-analysis was performed using version 5.2 of the RevMan software. Results A total of 22 RCTs involving 823 patients were discovered, some not dealing with mirror therapy. Comparing the studies with and without visual feedback, there were significant differences in upper limb motor function as measured by the Fugl-Meyer, ARAT and Brunnstrom instruments. The average Barthel indexes and visual analogue scale were also significantly different. Conclusion Mirror therapy can improve upper limb function and ability in the activities of daily living, and also relieve pain among stroke survivors to some degree, though its long-term efficacy is still unclear. Large-sample, multi-center, high-quality RCTs are required to verify these conclusions.

ABSTRACT

Objective To systematically evaluate the consistency of sensitivity (SEN) and specificity (SPE) between positron-emission tomography (PET) /magnetic resonance imaging (MRI) and PET/CT in the diagnosis of lymphoma.Methods The databases,including PubMed,Cochrane Library,Chinese Biomedical Literature Database (CBM),CNKI,Wanfang and VIP,were retrieved till October 2016.The studies published the SEN and SPE of PET/MRI and PET/CT in the diagnosis of lymphoma were collected and selected based on inclusion and exclusion criteria by two reviewers independently.Then,data extraction and analysis were performed by two independent reviewers.Resnlts A total of 6 studies,including 221 cases of patients with lymphoma,wereincluded.Among the 6 studies,4 of them provided patients-based and site-based data (including one provided data about staging ofdisease),and 2 studies only provided site-based data (including one provided data about staging of disease).The results indicated thatboth PET/MRI and PET/CT can accurately detect the lesions in patients with lymphoma,and the locations and numbers of lesions found bythe two methods were almost the same.Additionally,PET/MRI and PET/CT had high SEN and SPE for detection of lesions in patientswith lymphoma.and had high consistency in the diagnosis of lymphoma.Condusion The SEN and SPE of PET/MRI were similar to those of PET/CT in the diagnosis of lymphoma.PET/MRI may become a significant method for diagnosing lymphoma.

Objective To verify the mediating effect of the moral identity on the relationship between sport moral disengagement and pro-social and anti-social behaviors in competition.Methods Two hundreds and sixty-seven athletes of group events(including 173 males,94 females,with an average age of 22.40,SD=2.75)were selected and investigated using the sport moral disengagement scale,moral identity scale and the scale of pro-social and anti-social behavior in competition.Results (1) The sport moral disengagement had significant influence on the athletes'pro-social and anti-social behaviors in competition.(2)The moral identity has significant positive influence on the athletes' pro-social behavior in competition,but significant negative influence on the athletes' anti-social behavior in competition.(3)The moral identity has significant mediating effect on the relationship between sport moral disengagement and pro-social behavior in competition.Conclusion The moral disengagement in sport has important influence on the athletes' pro-social and anti-social behavior in competition.The moral identity has a significant mediating effect on the relationship between sport moral disengagement and pro-social behaviors in competition.

Objective To construct the lncRNA-1700020I14Rik plasmid and detect its effect on the fibrosis of mice mesangial cell (MMC) cultured with high glucose medium.Methods RT-qPCR was used to measure the expression of 1700020I14Rik in MMC cultured with low glucose medium or high glucose.Total RNA was extracted from MMC and cDNA was got by RT-PCR.The whole fragment of lncRNA-1700020I14Rik amplified by PCR was constructed into plasmid pcDNA3.1(+) through PCR.Lipidosome 3000 was used to transfect the plasmid into the MMC cultured with high glucose medium and RT-qPCR was used to measure the expression level of 1700020I14Rik.Western blot was used to analyze the expression of fibronectin, collagen Ⅳ and TGF-β1.Results 1700020I14Rik was significantly down-regulated in MMC cultured with high glucose and it was significantly up-expressed in the MMC after transfecting with pcDNA3.1(+)-1700020I14Rik.The expressions of fibronectin, collagen Ⅳ and TGF-β1 were down-regulated by 1700020I14Rik.Conclusions The plasmidpcDNA3.1(+)-1700020I14Rik is able to effectively express the lncRNA-1700020I14Rik.Over-expression of 1700020I14Rik may protect mesangial cells from fibrosis conduced in high glucose medium.

ABSTRACT

Long non-coding RNA was initially identified as "noises" of gene transcriptions. However, with the developing researches of ENCODE, it was found that the long non-coding RNAs can regulate the genomic expressions in the form of RNAs in epigenetic, transcription, and post transcriptional levels, which is involved in the regulation of diverse cellular processes and has significant influences on occurrence and precaution of human diseases. This paper introduces functions and features of the long non-coding RNAs, and sums up the internal relation between long non-coding RNAs, diabetes and diabetic complications on the basis of existing researches. These advances can provide the basis for the further understanding of molecular medicine on occurrence and evolution of diabetes.

ABSTRACT

OBJECTIVE@#To determine the correlation between p16 gene CpG methylation sites in the promoter region and HPV16 infection in cervical squamous cell carcinoma in Xinjiang Uyghur women.@*METHODS@#MALDI-TOF MS was used quantitatively to analyze p16 gene promotor methylation status of CpG islands in 20 cervix squamous cell carcinomas and 20 corresponding non-cancerous tissues in Uyghur women. HPV16 infection was detected by polymerase chain reaction (PCR) in both groups.@*RESULTS@#Among the 16 CpG sites in the p16 gene promoter region, CpG1-2 and CpG 6 sites were different between the 2 groups, and the levels of CpG1-2 and CpG6 methylation sites in the cervical squamous cell carcinoma were higher than those in the control group. The presence of HPV16 infection was significantly different between the cervix squamous cell carcinoma tissue and non-cancerous tissues (P<0.05). There was no significant correlation between p16 gene CpG methylation sites and HPV16 infection of cervical squamous cell carcinoma in Uyghur women.@*CONCLUSION@#P16 gene CpG 1-2, CpG 6 hypermethylation and HPV16, which are independent of one another, play an important role in cervical squamous cell carcinogenesis in Uyghur women.

Via activating G-protein receptor transduction pathways,nitric oxide (NO),an important signal molecule,has a complex and diverse role which is closely related with activity and gene expression of nitric oxide synthetase (NOS).NO is involved in tumor related events such as cellular proliferation,migration,especially the angiogenic process,and it is closely related with the occurrence and progression of prostate cancer.NO donors and NOS inhibitors have anti-cancer and radio-chemotherapy enhancement roles.

ABSTRACT

Objective To produce an experimental information for the safety assessment of Vero cells during subculture. Methods Passage and freeze on Vero cells, and Vero cells in different passages in vitro and in vivo tumorigenicity were tested. The protein expression of different Vero cell passages was analyzed. Results Vero cells passaged to p270 and 14 cell banks were developed and stored for future evaluation. In vitro and in vivo tumorigenicity Lest results of Vero cells in different passages were negative. Conclusion Although the tumorigenicity test results in vitro and in vivo process were negative, the protein expression of more than p200 Vero cells were changed, which produced the experimental reference for the safety evaluation of the process during the Vero cell passage.

ABSTRACT

A serum method for evaluating therapeutic efficacy of acute leukemia has been built by magnetic beads-based biological mass spectrometric technology. After analysis of quality control sample, the reproduci-)bility) of within-run assay and between-run assay was obtained and consistent with the previous reports. This result) manifested that the method used for serum peptides analysis was stable and reliable. Serum samples from 30 patients with primary acute leukemia and those with complete remission after treatment were extracted by metal-chelated magnetic beads and detected by MALDI-TOF-MS. Peaks at the m/z range 1000-10000 were analyzed by FlexAnalysis 2.4 software and Biosun_MS software. Approximately 27 differential peaks that were of statistical significance(p<0.05) were obtained. Ten peaks were up-regulated) and 17 peaks were down-regulated) after complete remission, respectively. The diagnostic model based on these differential peaks achieved) 100% sensitivity and 90% specificity. The results indicated that the model based on magnetic beads extraction and MALDI-TOF-MS detection acquires high sensitivity and specificity. Further sequence identification of differential) peaks is useful for evaluation of therapeutic efficacy and mechanism research on acute leukemia.)

OBJECTIVE To study the aminoglycosides modifying enzyme genes and intⅠ gene in Stenotrophomonas maltophilia in Chinese Armed Police Forces General Hospital.METHODS The samples of 27 multi-resistant S.maltophilia were collected from inpatiens from Jan 2006 to Oct 2007 in this Hospital.The sensitivity of the isolates to 14 antibacterial agents was determined using a broth induction method.The aminoglycosides modifying enzyme genes and intⅠ 1 gene were detected by PCR.RESULTS The multi-drug resistance of S.maltophilia was a serious problem.In 27 strains of S.maltophilia,the positive ant(2″)-Ⅰ were in 5 strains(18.5%),aac(3)-Ⅱ in 3 strains(11.1%)and aac(6')-Ⅱ in 1 strain(3.7%).The positive intⅠ gene was found in 11 strains(29.6%).CONCLUSIONS Multi-resistant S.maltophilia resistant to aminoglycosides mainly due to the presence of aminoglycoside modifying enzymes ant(2″)-Ⅰ,aac(3)-Ⅱ and aac(6')-Ⅱ.The aminoglycoside modifying enzymes ant(3″)-Ⅰ and aac(6)-ⅠZ were not detected carrying IntⅠ would be the reason of S.maltophilia resistant to aminoglycosides.

ABSTRACT

In the present study, we constructed a prokaryotic expression vector containing SOX4 protein encoding sequences. The GST-SOX4 soluble protein was expressed in Escherichia coli DH5alpha and purified by glutathione sepharose-4B. The purified recombinant protein was used to immunize Balb/C mice and the monoclonal antibody against SOX4 was prepared by using hybridoma technique. The titer of the antibody was determined as 1 x 10(5) by indirect ELISA. The specificity of the antibody was verified by Western blotting analysis. The monoclonal antibody specifically recognized the overexpressed exogenous SOX4 protein as well as endogenous SOX4 protein. The expression level of SOX4 protein in different cell lines and mouse tissues was detected by using the antibody. Differential expression of the protein was demonstrated by Western blotting. The data indicated that the antibody was specific. The antibody can be used as an important tool for further exploration of the role of SOX4 in tumorigenesis.

ABSTRACT

Objective To investigate the effect of Akt on cardiomyocyte hypertrophy induced by hydrogen peroxide(H2O2).Methods The neonatal rat cardiomyocytes cultured in primary generation were treated with low concentrations of H2O2.The cardiomyocyte hypertrophy was evaluated by the determination of average cell volume and protein content.The effects of Akt inhibitor on cardiomyocyte hypertrophy induced by H2O2 was recorded.Western blot was performed to examine the phosphorylation of Akt induced by H2O2.ResultsH2O2 at 10 or 50 ?mol/L stimulated cardiomyocyte enlargement as measured by cell volume and the protein content per cell.The inhibitor of Akt inhibited the hypertrophic response of cardiomyoctes stimulated by H2O2.H2O2 increased the level of Akt phosphorylation in cardiomyocytes,while that is suppressed by the inhibitors of PI3K(phosphoinositide 3-kinase). Conclusion Akt signaling is involved in cardiomyocyte hypertrophy induced by H2O2.
