ABSTRACT

Abstract Germin-like proteins (GLPs) play an important role against various stresses. Vitis vinifera L. genome contains 7 GLPs; many of them are functionally unexplored. However, the computational analysis may provide important new insight into their function. Currently, physicochemical properties, subcellular localization, domain architectures, 3D structures, N-glycosylation & phosphorylation sites, and phylogeney of the VvGLPs were investigated using the latest computational tools. Their functions were predicted using the Search tool for the retrieval of interacting genes/proteins (STRING) and Blast2Go servers. Most of the VvGLPs were extracellular (43%) in nature but also showed periplasmic (29%), plasma membrane (14%), and mitochondrial- or chloroplast-specific (14%) expression. The functional analysis predicted unique enzymatic activities for these proteins including terpene synthase, isoprenoid synthase, lipoxygenase, phosphate permease, receptor kinase, and hydrolases generally mediated by Mn+ cation. VvGLPs showed similarity in the overall structure, shape, and position of the cupin domain. Functionally, VvGLPs control and regulate the production of secondary metabolites to cope with various stresses. Phylogenetically VvGLP1, -3, -4, -5, and VvGLP7 showed greater similarity due to duplication while VvGLP2 and VvGLP6 revealed a distant relationship. Promoter analysis revealed the presence of diverse cis-regulatory elements among which CAAT box, MYB, MYC, unnamed-4 were common to all of them. The analysis will help to utilize VvGLPs and their promoters in future food programs by developing resistant cultivars against various biotic (Erysiphe necator and in Powdery Mildew etc.) and abiotic (Salt, drought, heat, dehydration, etc.) stresses.


Resumo As proteínas do tipo germin (GLPs) desempenham um papel importante contra vários estresses. O genoma de Vitis vinifera L. contém 7 GLPs; muitos deles são funcionalmente inexplorados. No entanto, a análise computacional pode fornecer informações importantes sobre sua função. Atualmente, as propriedades físico-químicas, localização subcelular, arquitetura de domínio, estruturas 3D, sítios de N-glicosilação e fosforilação e estudos filogenéticos dos VvGLPs foram conduzidos usando as ferramentas computacionais mais recentes. Suas funções foram previstas usando a ferramenta Search para recuperação de genes/proteínas em interação (STRING) e servidores Blast2Go. ABSTRACT

The objective of the present study was to analyse the bioactive compounds of the leaves of Conocarpus lancifolius (C. lancifolius). The GC-MS analysis of the hot methanolic extract of the leaves (HMEL) of C. lancifolius exhibited the bioactive compounds such as 1-(3-Methoxy-2-nitrobenzyl) iso quinoline, morphin-4-ol-6,7-dione, 1-bromo N-methyl-, phytol, hexadecanoic acid, 2,3-dihydroxypropyl ester, 2,2’:4’,2”-terthiophene, ethyl iso-allocholate, caryophyllene oxide, campesterol, epiglobulol, cholestan-3-ol, 2-methylene-, (3á,5à)-, dasycarpidan-1-methanol, acetate (ester) and oleic acid, eicosyl ester. The FT-IR analysis of HMEL of C. lancifolius showed a unique peak at 3184, 2413, 1657 cm-¹ representing coumaric acid, chlorogenic acid and ferulic acid. The HMEL of C. lancifolius was actively inhibiting the proliferation of breast cancer cells MCF-7 ATCC at the concentration of 72.66 ± 8.21 µg/ml as IC50 value. The HMEL of C. lancifolius also revealed a good spectrum of activity against Gram-positive and Gram negative bacterial cultures screened in this work. The activity observed has shown more or less similar effects against screened bacteria. However, the magnitude of potentiality was significantly lesser compared to standard ciprofloxacin disc at p< 0.001 level (99% confidence intervals). Furthermore, the study demonstrating the bioactive compounds can be isolated from the leaves of C. lancifolius.


O objetivo do presente estudo foi analisar os compostos bioativos das folhas de Conocarpus lancifolius (C. lancifolius). A análise por GC-MS do extrato metanólico quente das folhas (HMEL) de C. lancifolius exibiu os compostos bioativos como 1- (3-Metoxi-2-nitrobenzil) isoquinolina, morfina-4-ol-6,7- diona, 1-bromo-N-metil-, fitol, ácido hexadecanoico, 2,3-di-hidroxipropil éster, 2,2 ‘: 4’, 2 ” - tertiofeno, isoalocolato de etil, óxido de cariofileno, campesterol, epiglobulol, colestano -3-ol, 2-metileno-, (3á, 5à) -, dasycarpidan-1-metanol, acetato (éster) e ácido oleico, éster eicosílico. A análise FT-IR de HMEL de C. lancifolius mostrou um pico único em 3184, 2413, 1657 cm-¹ representando ácido cumarico, ácido clorogênico e ácido ferúlico. O HMEL de C. lancifolius inibiu ativamente a proliferação de células de câncer de mama MCF-7 ATCC na concentração de 72,66 ± 8,21 µg/ml como valor de IC50. O HMEL de C. lancifolius também revelou bom espectro de atividade contra culturas de bactérias Gram-positivas e Gram-negativas rastreadas neste trabalho. A atividade observada mostrou efeitos mais ou menos semelhantes contra bactérias rastreadas. No entanto, a magnitude da potencialidade foi significativamente menor em comparação com o disco de ciprofloxacina padrão em nível de p < 0,001 (intervalos de confiança de 99%). Além disso, o estudo demonstrando os compostos bioativos pode ser isolado das folhas de C. lancifolius.

ABSTRACT

Seaweeds are a major marine resource that can be explored to develop novel pharmaceutical molecules. The present study showed the presence of unique bioactive components in the petroleum ether extract (PEE) and methanolic extract (ME) of Sargassum tenerrimum. The gas chromatography-mass spectrometry analysis suggested that the PEE of S. tenerrimum contained antibacterial biomolecules: hexadecanoic acid, methyl ester, 17-pentatriacontene, dasycarpidan-1-methanol, and acetate (ester). However, the ME of S. tenerrimum exhibited better antibacterial effect than the PEE due to the presence of the bioactive compounds 1,2-benzenedicarboxylic acid, diisooctyl ester, tetratetracontane, 1-docosene, 1,2-benzenediol, and benzoic acid. Thus, promising antibacterial molecules can be isolated from S. tenerrimum for better therapeutic use.


As algas marinhas são um importante recurso marinho que pode ser explorado para desenvolver novas moléculas farmacêuticas. O presente estudo mostrou a presença de componentes bioativos únicos no extrato etéreo de petróleo (PEE) e no extrato metanólico (ME) de Sargassum tenerrimum. A análise por cromatografia gasosa espectrometria de massa sugeriu que o PEE de S. tenerrimum continha biomoléculas antibacterianas: ácido hexadecanoico, éster metílico, 17-pentatriaconteno, dasycarpidan-1-metanol e acetato (éster). Entretanto, o ME de S. tenerrimum exibiu melhor efeito antibacteriano do que o PEE devido à presença dos compostos bioativos ácido 1,2-benzenodicarboxílico, éster diisooctil, tetratetracontano, 1-docosene, 1,2-benzoenodiol e ácido benzoico. Assim, moléculas antibacterianas promissoras podem ser isoladas de S. tenerrimum para melhor uso terapêutico.

ABSTRACT

Discharge of textile industrial effluent without proper treatment has become a severe hazard for the animal health and environment worldwide. Therefore, this study was designed to isolate azo dye-degrading bacteria from textile wastewater and evaluate their ability to biodegrade reactive dyes into non-toxic products. The potent bacterial strain which was isolated from textile wastewater was identified as Pseudomonas monteilii strain RZT1 on the basis of 16S rDNA sequence. The isolated bacterial strain exhibited good decolorization ability with yeast extract supplementation as cosubstrate in static conditions for Malachite Green dye. The optimal condition for the decolorization of Malachite Green dye by P. monteilii strain RZT1 were at pH 7.0 and 28癈. Decolorization rates of Malachite Green dye by P. monteilii strain RZT1 were varied with initial dye concentration as follow: 84.8%, 75.4%, 63.4% and 45.5% decolorization for 100ppm, 200ppm, 300ppm and 400ppm initial dye concentration respectively. We investigated the effects of dyes used in the textile industry on the seed germination of Five crops - Rice (Oryza sativa), Wheat (Triticum aestivum L.), Khesari (Lathyrus sativus), Mustard (Brassica nigra) and Bitter Melon (Momordica charantia). It was found that textile dye Malachite Green had negative effect on seed germination and seedling growth in test cultures. The harmful effects of dye on seed germination and early seedling growth parameters were augmented with increase of dye concentration. Interestingly, treatment of the Malachite Green dye with isolated bacteria reduced the adverse effects of that dye on seed germination and seedling growth. Thus, it indicated the potentiality of P. monteilii strain RZT1 for bioremediation of textile effluents into a non-toxic form for plants.

ABSTRACT

Due to rapid industrialization and market demand of vibrant textile products, the natural textile dyes have been replaced by the synthetic textile dyes. These synthetic dyes are released in environment with textile wastewater resulting in a major environmental pollution, especially in aquatic ecosystem. Hence, aquatic organisms like fish are highly vulnerable to the pollution caused by dyes of textile wastewater. This study was designed to evaluate the deleterious effects of Basic Red-18 (BR-18) dye on behavior, survivability, haematology and histology of Tilapia fish (Tilapia mossambica) and to minimize these deleterious effects of BR-18 dye by bioremediation with the novel bacteria isolated from textile wastewater. The isolated novel bacteria was identified as Mangrovibacter yixingensis strain AKS2 by 16s rRNA sequencing (Accession no. OM189530). The 30% and 70% mortality rates were observed in fish exposed to commercial BR-18 dye at concentrations of 100 and 200 ppm respectively. Interestingly, the mortality rate of fish was decreased significantly to 10% and 20% when fish were exposed to 100 and 200 ppm BR-18 dye respectively after bioremediation with M. yixingensis strain AKS2. Fish exposed to tap water and bioremediated BR-18 dye solution exhibited typical behavioral responses, whereas fish exposed to commercial BR-18 dye solution exhibited anomalous behavior. Fish subjected to commercial BR-18 dye solution displayed decreased RBC, Hb, but increased WBC levels, demonstrating the dye's haemotoxicity. Contrary, no remarkable haematological toxic effect was found when fish were exposed to bioremediated BR-18 dye indicating the non-toxic character of the bioremediated dye metabolites. Similarly, extensive histological abnormalities in the gill, liver, intestinal, stomach, and heart tissues were seen when fish was cultured in commercial BR-18 dye, but the abnormalities were less significant when fish were raised in bioremediated BR-18 dye. Altogether, it can be concluded that BR-18 dye are toxic to fish, but this toxicity can be minimized by bioremediation with M. yixingensis strain AKS2.

An experiment was conducted into the Horticulture Farm of Sher-e-Bangla Agricultural University, Dhaka from October 2019 to March 2020 this study the effect of macronutrients combination and plant spacing on growth and yield of black cumin (Nigella sativa L.). Black cumin variety BARI Kalozira-1 was used as planting material in this study. The experiment consisted of two factors: Factor-A: macro nutrient combinations (4 levels): T1= N0P0K0 kg ha-1 (control), T2= N90P50K40 kg ha-1, T3= N135P75K60 kg ha-1; Factor-B: plant spacing (3 levels): S1= 20 cm × 10 cm, S2= 20 cm × 15 cm and S3= 20 cm × 20 cm. The experiment was laid out in a randomized complete block design with factorial with three replications. Data on different growth, yield and yield contributing parameter of black cumin were recorded and significant variation was observed from different treatments. In case of nutrient combinations, the tallest plant (54.86 cm) was observed from T3 (N135P75K60 kg ha-1) treatment. The maximum primary branch plant-1 (8.62) and secondary branch plant-1 (12.18), flower plant-1 (22.20), capsules plant-1 (19.69) and 1000 seed weight (2.99 g) was observed from T2 (N90P50K40 kg ha-1) treatment. The highest seed yield ha-1 (1.18 t) was observed from T2 (N90P50K40 kg ha-1) treatment. In case of plant spacing the tallest plant (48.34 cm) was observed from S1 (20 cm × 10 cm) treatment. The maximum primary branch plant-1 (7.80) and secondary branch plant-1 (11.23) flower plant-1 (20.26), capsules plant-1 (18.81), 1000 seed weight (2.99 g) were observed from S3 (20 cm × 20 cm) treatment. The highest seed yield ha-1 (1.09 t) was recorded from S1 (20 cm × 10 cm) treatment. It can be concluded that, sowing of black cumin providing 90 kg N, 50 kg P and 40 kg K nutrient combination with 20 cm × 10 cm plant spacing was recorded to be more suitable practice for getting higher amount and quality of seed yield of black cumin.

ABSTRACT

The purpose of the study were to (i) determine the disaster coping strategy practiced by the affected people (ii) assess the socio-economic profile of the respondents, (iii) ascertain the contribution of selected characteristics of the people and (iv) identify the constraints of disaster coping strategy practiced by the affected people. The study was conducted at Gangachara Upazila of Rangpur District. Data were collected from a sample of 302 respondents, randomly selected from disaster affected population. Age, education, family size, farm size, disaster affected land, annual income, training received, extension media contact, knowledge on disaster coping strategy, environmental awareness, household assets, credit facilities, IGAs, water and sanitation condition, risk orientation, awareness about SSNP, perception of Climate Change (CC), perception of disasters and scope of work in vulnerable situation constituted the independent variables, while disaster coping strategy practiced was the dependent variable. For measuring the disaster coping strategy practices, a 4-point rating scale was used against 50 items taking 10 items from each of five components of human needs e.g. 1) food preservation, collection and management, 2) agricultural products protection, 3) maintaining social network, 4) safeguard of health and sanitation, and 5) protection of housing and shelter. Numerical values and scales were used to measure the personal attributes. Regression and path analysis were employed to determine the contribution among the variables. For exploring relationship between the two variables Pearson’s Product Moment Correlation (r) was used. The majority (77.8 per cent) of the respondents had regular coping strategies compared to 22.2 per cent were found to have occasional coping strategies in the study area. Among 19 independent variables 16 were found significant relationship with the dependent variable. Stepwise multiple regression revealed that six variables namely education (23 per cent), participation in IGAs (3.6 percent), awareness of SSNPs (2.9 percent), disaster affected land (1.2 percent), farm size (1.9 percent) and perception of CC (1 percent) were the important contributing variables which combined explained 33.6 of the total variation of practice of coping strategy. Path analysis indicated that disaster affected land (0.589) had the highest positive direct effect while farm size (0.643) had the highest positive indirect effect to the disaster coping strategy practice. The major constraints for practicing disaster coping strategy in the study area were ‘Lack of knowledge and skills of affected people‘, ‘Lack of relief materials during disaster‘, ‘Lack of technologies‘, ‘Weak weather forecasting‘, ‘Less motivation’, ‘Low sanitation & health coverage’ and ‘Lack of communication during disasters’,. To cope up with the challenges of the disasters, the people used reduction of food intake per meal, putting goods above flood level, keeping women and children in safer places, using boiled and tube well water, providing health care support to the sick family members, transfer important documents to the safer places, increasing level of homestead with soil, taking relief, credit, religious fasting, using mosque/temple as campaigning center, borrowing principal food from neighbor, use savings, migration, livestock sell, and social interconnectedness as the major coping strategies to survive the situation and improve their livelihood.

Background: Osteocalcin, has high affinity for calcium. In osteoporotic women, deficiency of calcium may lead to lowering of the formation of hydroxyapatite crystals. Thus, in the state of hypo mineralization, free osteocalcin available in the circulation. Therefore, present study was designed to evaluate significance of serum osteocalcin in diagnosis of osteoporosis, and relationship between Serum Osteocalcin and BMD (Bone mineral Density) in post-menopausal females with osteoporosis and without osteoporosis.Methods: One hundred and forty seven post-menopausal women between age 45 to 80 years attending the hospital OPD were studied. To be eligible for the study they had to have been postmenopausal for at least one year. The diagnosis of osteoporosis was made based on T-Scores (BMD) at the lumber spine (L1 to L4 and femaral neck) by DEXA (GE lunar Densitometer). Serum osteocalcin level was estimated by LIAISON osteocalcin assay. Patients with chronic conditions affecting skeletal health and patients on drugs affecting the skeleton were excluded from the study.Results: Serum osteocalcin level in post-menopausal female without osteoporosis was 9.87±1.04ng/ml, while post-menopausal female with osteoporosis had 22.62±2.25ng/ml suggesting significant increase in bone marker level in osteoporotic females (p<0.05.) Correlation study between BMD and osteocalcin showed strong Negative Correlation (r=-0.77, p<0.05).Conclusions: Serum osteocalcin can be considered as a specific marker of osteoblast function as its levels have been shown to correlate with bone formation rates. Thus, serum osteocalcin can be used for diagnosis and monitoring of response to therapy and this may be the better predictor than BMD.

Aim: The objective of this study was to understand the possible fate of 4-NP through the molecular mechanism and to identify potential enzymes involved in 4-NP biodegradation by Rhodococcus sp. strain BUPNP1 Methodology: Biodegradation of 4-NP was detected spectrophotometrically at 400 nm and also confirmed by TLC and HPLC. Comparative study of proteomes was performed by 2-D gel electrophoresis followed by peptide mass fingerprinting and bioinformatic analysis to identify and/ or predict the possible functions of over-expressed proteins in 4-NP treated cells of BUNP1. Results: Utilization of 4-nitrophenol and its hydrolysis intermediate 4-nitrocatechol (4-NC) and 1,2,4-Benzenetriol as sole carbon source indicated the presence of genomic information encoding the enzyme necessary for the operation of 4-nitrophenol degradation pathway in the strain BUPNP1. It could transform 4-NP into 4-NC by monooxygenase whose major activity was detected during initial stage of degradation. The 4-NC further depleted in the medium to release nitrite ions. In order to investigate the molecular changes occurring during degradation, a comparative study of proteome profiles was carried out where; 4-nitrophenol treated cells were compared against cells grown on glucose as control. The comparative study indicated expression of several protein spots under 4-nitrophenol treated condition. Interpretation: This study showed the potential of BUPNP1 strain belonging to genus Rhodococcus towards induced expression of some unique proteins which might have possible role in 4-NP biodegradation process

ABSTRACT

A reproducible stability-indicating Reverse Phase-HPLC technique for the quantification of enzalutamide in in pharmaceuticals was developed and validated. Chromatography was achieved on Inertsil-ODS-C18 (250mm×4.6 mm) 5µmanalytical column with acetonitrile: methanol: water in 40:30:30% v/v proportion as mobile phase and flow rate of 1 ml/min. Enzalutamide was detected at 237 nm UV-wavelength maximum. In the present work mobile phase used as a diluent. Developed technique was validated over 20-150 µg/ml linear concentration range for enzalutamide. This method established with linearity coefficient value of 0.99 and the percentage recovery was found to be 99.3%. This method was proven with LOD and LOQ values of 0.53 µg/mL and 1.61µg/ml respectively. The drug was degraded in acid and alkaline conditions and the percentage degradation values were 3.10 % and 4.54 % respectively. There was no degradation of drug when exposed to neutral, UV, thermal, sun-light and oxidative conditions.Drug was undergoing degradation when exposed to acid and alkaline conditions. The developed technique was useful in the routine quantitation of enzalutamide.

Objective: To develop an innovative, rapid, simple, cost-effective, stability indicating reverse phase-high performance liquid chromatography (RP-HPLC) method for simultaneous estimation of ledipasvir (LP) and sofosbuvir (SB) in combination pill dosage form. Methods: The method was developed using C8 column, 250 mm x 4.6 mm, 5 mm using mobile section comprising of 0.1% (v/v) orthophosphoric acid buffer at pH 2.2 and acetonitrile in the ratio of 45:55 that was pumped through the column at a flow rate of 0.8 ml/min. Temperature was maintained at 30 °C, the effluents were monitored at 260 nm with the help of usage of PDA detector. Results: The retention time of LP and SB were found to be 2.246 min and 3.502 min. The approach was found to be linear with the variety of 9-36 µg/ml and 40-240 μg/ml for LP and SB respectively, the assay of estimated compounds were found to be 99.65% and 99.73% w/v for LP and SB respectively. Conclusion: The pressured samples changed into analyzed and this proposed a technique turned into determined to be particular and stability indicating as no interfering peaks of decay compound and excipients were observed. Hence, the approach was easy and economical that may be efficiently applied for simultaneous estimation of both LP and SB in bulk and combination tablet system.

The popliteal artery is the branches off from the femoral artery. It is located in the knee towards the back of the legs. The clinicians and radiologists dealing with the popliteal area use description of branching pattern of the popliteal artery, in which it first gives rise to anterior tibial artery followed by the tibial peroneal trunk, which then terminates by dividing into the posterior tibial and peroneal arteries. As popliteal artery is one of the most common sites for aneurysm formation, the knowledge of the normal popliteal artery diameter is essential to determine the relevance of popliteal dilatations. Objective: Considering the clinical importance of diameter of popliteal artery, this study was undertaken to note the internal diameter of popliteal artery. Subjects and Methods: The present study was conducted on 50 specimen embedded in 10% formalin from the department of Anatomy of tertiary care hospital & medical college. Cadavers which had gross pathological deformities in its lower limbs were excluded from the study. All the cadavers were adult males and females. Results: This study states right popliteal arteries are slightly larger (4.9 ± 0.7 mm) as compare to left popliteal artery (4.8± 0.5 mm). This study also reconfirm popliteal arteries of men are slightly larger than that of female (4.9 ± 0.7 mm), (4.6± 0.9 mm) and left popliteal artery (4.8± 0.5 mm), (4.6± 0.7 mm) respectively. On the other hand terminal part its diameter is slightly larger in male (4.5 ± 0.6 mm) as compare to female (4.3 ± 0.5 mm). This study also shows terminal end of left popliteal arteries (3.8 ± 0.7 mm) are smaller than right popliteal arteries (4.3 ± 0.5 mm). Conclusion: The present study was done in a small cadaveric sample. In our prospective different live and cadaveric study will be helpful for vascular surgeons, radiologists. The variation in the measurement of our study compare to other study may be due to the difference in live and cadaveric spacemen or due to the method of measurement. Therefore, we suggest before making any decision study on much larger sample should be consider
