ABSTRACT

Objective:To investigate clinical characteristics of pediatric psoriasis based on the information systems from two children′s hospitals.Methods:Clinical data on outpatients confirmly diagnosed with pediatric psoriasis were collected from information systems of Beijing Children′s Hospital affiliated to Capital Medical University and Children′s Hospital of Chongqing Medical University from January 1, 2015 to December 31, 2019, and a clinical and epidemiological investigation was conducted. Statistical analysis was carried out by using t test and chi-square test. Results:A total of 5 235 children with psoriasis were included, with the ratio of male to female being 1∶1.08. Their age at the clinic visit ( M [ Q1, Q3]) was 8.37 (6.48, 10.50) years, and the school-age children were the most common population; their age at onset was 7.57 (5.37, 9.82) years. Among the 5 235 children with psoriasis, there were 3 195 (60.82%) with psoriasis vulgaris, 281 (5.37%) with pustular psoriasis, 19 (0.36%) with erythrodermic psoriasis, and 1 (0.02%) with psoriatic arthritis. The trunk (87.76%, 1 097/1 250) was most frequently affected, followed by the limbs (87.68%, 1 096/1 250) , the scalp (62.56%, 782/1 250) , and the face and neck (35.76%, 477/1 250) . Among the 5 235 patients, 4 319 (82.50%) received topical treatments, 177 (3.38%) received systemic treatments, and 832 (15.89%) were treated with antibiotics. Among 3 497 children who received initial treatment regimens, the disease could be controlled in 3 423 (97.88%) without change in treatment regimens, while treatment regimens needed to be adjusted in 2.12%. Conclusions:In the two children′s hospitals, most children with psoriasis developed this condition and visited the clinic at school age, and the predominant clinical type was psoriasis vulgaris. Most skin lesions were extensive, and commonly occurred on the trunk and limbs. Scalp involvement was not uncommon. The condition could be controlled by topical treatments in most children with psoriasis, while a few patients needed systemic treatments.

ABSTRACT

Children with atopic dermatitis often have a variety of psychological and behavioral problems, such as attention-deficit/hyperactivity disorder, depression, autism, and even suicidal tendencies. The mechanism of comorbidity between atopic dermatitis and psychological and behavioral problems is still unclear. Reasonable and effective treatment of atopic dermatitis, as well as correct identification of and early intervention with related psychological and behavioral problems can effectively reduce the occurrence of psychological and behavioral problems, and improve the prognosis of children with atopic dermatitis.

ABSTRACT

Mast cells (MC),a major participant in inflammation and allergy,can initiate an early defense response to foreign invaders.Besides expressing high-affinity IgE receptor,MC also expresse a large number of G protein-coupled receptors (GPCRs).Various studies have shown that cationic micromolecules represented by substance P and many peptidergic drugs can induce MC degranulation through a novel receptor,Mas-related G protein-coupled receptor X2 (MrgprX2).MrgprX2 plays an important role in allergy,itching and pseudo-allergic drug reactions.It will help explain some clinical difficulties which can not be explained by classical IgE-dependent activation of MC,and provides potential therapeutic targets for MC-mediated diseases.

ABSTRACT

Objective: To analyze the prognostic factors of non-Hodgkin's lymphoma(NHL)and to investigate the prognostic value of peripheral blood absolute lymphocyte count(ALC)at admission for patients with NHL. Methods: Clinical features and follow-up data of 108 patients with pathologically confirmed NHL seen in our hospital between January 2000 and January 2008 were reviewed.SPSS14.0 package was used for statistical analysis.Kaplan-Meier was applied to assess the survival probability.All parameters statistically significant concluded by univariate analysis were then computed as co-variates for multivariate analysis with Cox regression model. Results: The ratio of males to females was approximately 1.5:1.The median age of patients was 48 years.Before treatment.the Ann Arbor clinical classification showed that 61.1% of the cases were of stage Ⅰ and Ⅱ.Approximately 93%of the patients had ECOG performance status(PS)score of 0-1 and 19.2%of the cases had elevated serum lactate dehydrogenase(LDH).According to intemational prognosis index score.80.6%of the patients were in a low risk group.At admission,35.2%of the cases had ALC≤1×10~9/L.Hemoglobin (Hb)≤110g/L and B symptoms were seen in 29.6%and 26.9%of the patients.The mean Hb was 129.2±17.5g/L in cases with ALC>1×10~9/L(n=70)and 98.1±20.6g/L in cases with ALC≤1×10~9/L(n=38),with a statistically significant difference between the two groups(P<0.05).With a median follow-up duration of 2 years,the median overall survival(OS)time was 2.3 years for all patients.The 2-year and 5-year OS rates were 73.2%and 39.6%,respectively.ALC≤1×10~9/L,Hb≤110g/L,B symptoms and intemational prognostic index(IPI)≥2 were statistically significant unfavorable prognostic factors for NHL revealed by univariate analysis.Multivariate analysis showed that ALC≤1×10~9/L,B symptoms and IPI ≥2 were statistically significant unfavorable prognostic factors for NHL. Conclusion: ALC and B symptoms may be prognostic factors independent of IPI for NHL.Evaluation of the prognosis with IPI,ALC,and B symptoms is of clinical value for individualized therapy of NHL patients.

ABSTRACT

Objective To study the changes of gene expression profiling in HL-60 cells treated by Chan'su, and the possible mechanisms of Chan'su-induced apoptosis in HL-60 cells. Methods Apoptosis was determined by morphology and agarose gel electrophoresis of DNA fragmentation. The changes of gene expression profiling were detected with DNA microarray. Results 1%(v/v) Chan'su could obviously induce HL-60 cells apoptosis. In the tested 1152 genes, mRNA expression levels of 9 genes changed prior to apoptosis in Chan'su-treated HL-60 cells. Of the 9 genes, 8 gene mRNA levels were up-regulated and 1 gene mRNA level was down-regulated. Conclusion Apoptosis in HL-60 cells treated by Chan'su was involved in many genes. Bcl-2 down-regulation and IL-8 up-regulation were closely correlated with the Chan'su-induced HL-60 cells apoptosis.

ABSTRACT

To investigate the effects of granulocyte macrophage colony stimulating factor (GM-CSF) which was secreted from the fresh leukemic cells of patients with acute myeloid leukemia (AML) and recombinant human GM-CSF(rhGM-CSF) on the apoptosis of the primary incubative AML cells,the level of GM-CSF was measured by means of radioimmunoassay,the DNA ladder was detected by using DNA electrophoresis,and the percentage of apoptotic cells,percentage of expression and the mean fluorescence intensity(MFI) of bcl-2 were respectively assayed by means of flow cytomtric analysis.After the fresh leukemic cells from 5 cases were incubated for 24 hours,GM-CSF in 3 of 5 cases were seen,their percentage of apoptotic cells was less than the other 2 cases,and the MFI of bcl-2 in the 3 of 5 cases was obviously enhanced.After the fresh leukemic cells were treated with cytarabine (Ara-C) 5?g/ml in combination with rhGM-CSF 5?g/ml for 24 hours,the percentage of apoptotic cells was more than the cells treated with Ara-C alone and it upward to (56 4?7 4)%(p
