ABSTRACT

Abstract Vegetation cover may show diversity and composition patterns of the soil invertebrate community, as a function of litter quantity and quality in a specific habitat. The objective of this work was to characterize the distribution of edaphic fauna in different monocultures. The study was carried out at Chapada Grande farm in Regeneração, PI. Four monoculture areas were chosen: no-tillage soybean, eucalyptus, pasture, and a preserved native cerrado forest. Soil fauna was collected in a dry and wet period by pitfall traps containing 4% formaldehyde. The edaphic fauna was evaluated by the number of individuals per trap per day, average richness and richness, Shannon diversity index and Pielou uniformity index. Data were submitted to analysis of variance and multivariate Principal Component Analysis (PCA). The highest number of individuals per day trap and of average richness were registered in the pasture, eucalyptus and forest areas in both periods, while soybean showed lower values with predominance of Coleoptera and Formicidae groups. The pasture and forest areas showed of higher Shannon index values in the two evaluated seasons, probably due to higher contributions organic residues in the soil that favors the shelter, feeding and reproduction conditions. Regarding the Pielou index, the soybean system showed higher values in this variable. The Aranae, Coleoptera, Formicidae and Diptera groups predominated in the humid period, while Coleoptera and Formicidae predominated in the dry period. Systems that generate greater accumulation of residues harbor a greater diversity of invertebrates of the edaphic fauna. Seasonality had an effect on all variables analyzed and the wet period showed more expressive values.


Resumo A cobertura vegetal pode mostrar padrões de diversidade e composição da comunidade de invertebrados do solo, em função da quantidade e qualidade da serapilheira em determinado habitat específico. O objetivo deste trabalho foi caracterizar a distribuição da fauna edáfica em diferentes monocultivos. O estudo foi realizado na fazenda Chapada Grande no município de Regeneração, PI. Foram escolhidas quatro áreas de monocultivos: soja em sistema de plantio direto, eucalipto, pastagem, além de uma mata nativa de cerrado preservada. Foram realizadas coletas da fauna do solo, em período seco e úmido, por meio de armadilhas do tipo pitfall contendo 4% de formol. A fauna edáfica foi avaliada pelo número de indivíduos por armadilha por dia, riqueza e riqueza média, índice de diversidade de Shannon e índice de uniformidade de Pielou. Os dados foram submetidos à análise de variância e à análise multivariada de Componentes Principais (ACP). O maior número de indivíduos por armadilha dia e de riqueza média foram registrados nas áreas pastagem, eucalipto e mata nos dois períodos, enquanto que a soja mostrou valores inferiores com predomínio dos grupos Coleoptera e Formicidae. As áreas de pastagem e mata mostraram maiores valores de índice de Shannon nas duas épocas avaliadas provavelmente em função de maiores aportes de resíduos orgânicos no solo que favorece as condições de abrigo, alimentação e reprodução. Em relação ao índice de Pielou o sistema com soja mostrou maiores valores nessa variável. Os grupos Araneae, Coleoptera, Formicidae e Diptera predominaram no período úmido, enquanto Coleoptera e Formicidae se destacaram no período seco. Sistemas que geram maior acúmulo de resíduos abrigam uma maior diversidade de invertebrados da fauna edáfica. A sazonalidade apresentou efeito sobre todas as variáveis analisadas sendo que o período úmido mostrou valores mais expressivos.

This study verified the effects of respiratory muscle training (RMT) on hemodynamics, heart rate (HR) variability, and muscle morphology in rats with streptozotocin-induced diabetes mellitus (DM). Thirty-six male Wistar rats were randomized into 4 groups and 34 completed the study: i) sham-sedentary (Sham-ST; n=9); ii) sham-RMT (Sham-RMT; n=9); iii) DM-sedentary (DM-ST; n=8); and iv) DM-RMT (DM-RMT; n=8). Hemodynamics were assessed by central cannulation, and R-R intervals were measured by electrocardiogram. In addition, the effects of RMT on the cross-sectional area of the diaphragm, anterior tibial, and soleus muscles were analyzed. The induction of DM by streptozotocin resulted in weight loss, hyperglycemia, reduced blood pressure, and attenuated left ventricular contraction and relaxation (P<0.05). We also observed a decrease in root mean square of successive differences between adjacent RR intervals (RMSSD) index and in the cross-sectional area of the muscles assessed, specifically the diaphragm, soleus, and anterior tibial muscles in diabetic rats (P<0.05). Interestingly, RMT led to an increase in RMSSD in rats with DM (P<0.05). The induction of DM produced profound deleterious changes in the diaphragmatic and peripheral muscles, as well as impairments in cardiovascular hemodynamics and autonomic control. Nevertheless, RMT may beneficially attenuate autonomic changes and improve parasympathetic modulation.

Abstract The vegetation physionomy and cover can show patterns of diversity and composition of the edaphic community, depending on the quantity and quality of litter in a specific habitat. The objective of this study was to evaluate the effect of the vegetation gradient formed by Graminoid Field (GRF), Cerrado Sensu Stricto (CSS), Cerradão (CRD) and Semideciduous Seasonal Forest (SSF) on density, diversity and composition of the edaphic fauna community in two seasons of the year, in the Sete Cidades National Park (Piauí state). For fauna sampling, a total of eight pitfall traps, distanced 10 m, were placed in each area in the central part of each system, where they remained for seven days. In the wet period, there was a tendency to increase the number of individuals as a function of the complexity of the vegetation formation, with the inverse occurring in the dry period. It was verified an environmental variation of the climatic factors temperature and humidity according to the vegetal formation, contributing to a heterogeneous distribution of the fauna. The GRF formation presented a significantly lower value of average richness only in the dry period. Regarding the variables of diversity and uniformity, they did not show drastic variations in relation to the vegetation gradient studied. The dominant groups in the vegetation gradient were Formicidae, Coleoptera, Aranae, Acari and Collembola, with reduction of the number of Coleoptera in the dry season. Principal component analysis (PCA) revealed greater differences in the composition of the communities between the vegetation formations for the rainy season. At this time, the formations SSF and CRD were associated to a greater diversity of invertebrates than CSS and GRF, demonstrating the influence of the vegetation complexity on the soil fauna community.


Resumo A fisionomia e cobertura vegetal podem mostrar padrões de diversidade e composição da comunidade edáfica, em função da quantidade e qualidade da serapilheira em determinado habitat específico. O presente trabalho teve por objetivo avaliar o efeito do gradiente vegetacional formado por Campo Graminóide (CGR), Cerrado Sensu Stricto (CSS), Cerradão (CRD) e Floresta Estacional Semidecídua (FES) sobre a densidade, diversidade e composição da comunidade da fauna edáfica em duas épocas do ano do Parque Nacional de Sete Cidades (PI). Para amostragem da fauna foram colocadas um total de oito armadilhas do tipo Pit-fall, distanciadas por 10 m, em cada área na parte central de cada sistema, onde permaneceram por sete dias. No período úmido houve uma tendência a aumento do número de indivíduos conforme a complexidade da formação vegetal e o inverso ocorreu no período seco. Verificou-se uma variação ambiental dos fatores climáticos temperatura e umidade conforme a formação vegetal contribuindo para uma distribuição heterogênea da fauna. Apenas no período seco a formação CGR apresentou valor significativamente menor de riqueza média. Em relação às variáveis de diversidade e uniformidade não apresentaram variações drásticas em relação ao gradiente vegetacional estudado. Os grupos dominantes no gradiente vegetacional foram Formicidae, Coleoptera, Aranae, Acari e Collembola, com redução do número de Coleoptera na época seca. Pela análise de componentes principais (ACP) constatou-se maiores diferenças na composição das comunidades entre as formações vegetais para o período chuvoso. Nesta época, as formações vegetais os estádios FES e CRD estiveram associados a uma maior diversidade de invertebrados do que CSS e CGR, demonstrando influência da complexidade vegetacional sobre a comunidade da fauna do solo.

Abstract The aim of this study was to measure soil microbial biomass and soil surface fauna in undisturbed and disturbed Cerrado sensu stricto (Css) from Sete Cidades National Park, Northeast Brazil. The following sites were sampled under Cerrado sensu stricto (Css) at the park: undisturbed and disturbed Css (slash-and-burn agricultural practices). Total organic and microbial biomass C were higher in undisturbed than in disturbed sites in both seasons. However, microbial biomass C was higher in the wet than in the dry season. Soil respiration did not vary among sites but was higher in the wet than in the dry season. The densities of Araneae, Coleoptera, and Orthoptera were higher in the undisturbed site, whereas the densities of Formicidae were higher in the disturbed site. Non-metric multidimensional scaling analysis separated undisturbed from disturbed sites according to soil biological properties. Disturbance by agricultural practices, such as slash-and-burn, probably resulted in the deterioration of the biological properties of soil under native Cerrado sensu stricto in the Sete Cidades National Park.


Resumo O objetivo do estudo foi avaliar as propriedades biológicas do solo em área de Cerrado sensu stricto preservadas e não preservadas do Parque Nacional de Sete Cidades, Nordeste do Brasil. Os seguintes sites sob Cerrado sensu stricto (Css) foram avaliados: preservado (UND) e não preservado (DIS). O C orgânico total e da biomassa microbiana foram maiores na área preservada do que na não preservada em ambas estações seca e chuvosa. Entretanto, o C da biomassa microbiana foi maior na estação chuvosa do que na seca. A respiração do solo não variou entre as áreas, mas foi maior na estação chuvosa. As densidades de Araneae, Coleoptera e Ortoptera foram maiores na área preservada, enquanto as densidades de Formicidae foram maiores na área não preservada. A analise de escalonamento não-métrico separou a área preservada da não-preservada de acordo com as propriedades biológicas. Os distúrbios no solo pelas atividades agrícolas, como corte-e-queima, resultaram na deterioração das propriedades biológicas do Cerrado nativo no Parque Nacional de Sete Cidades.

ABSTRACT

Os efeitos do uso de minerais complexados durante o pré-parto sobre a ocorrência de retenção de placenta foram avaliados em 135 vacas Holandesas de dois ou mais partos: grupo mineral iônico (69 animais) e grupo mineral complexado (66 animais). Em 55 desses animais foram também avaliadas as concentrações séricas da imunoglobulina G (IgG), Zn, Cu e a qualidade do colostro. O experimento foi realizado em delineamento inteiramente ao acaso, em arranjo em parcelas subdivididas. As concentrações séricas de IgG e dos microminerais foram avaliadas por análise de variância, sendo utilizados, respectivamente, os testes de Duncan e Fisher. A taxa de erro α admitida foi de 7%. Não foram observadas diferenças entre os grupos para ocorrência de retenção de placenta, qualidade do colostro, concentrações séricas de Zn e IgG (P>0,07), sendo observada diferença para a concentração de Cu (P<0,07). As concentrações de IgG foram diferentes nas semanas pré-parto avaliadas (P<0,07).


The effects of the use of complex minerals on the occurrence of retained placenta during pre-partum were valued on 135 Holstein cows from two or more deliveries. The animals were divided in two groups: ionic mineral (69 animals) and complexed mineral (66 animals). In 55 of these animals serum concentrations of imunoglobulin G (IgG), Zn and Cu and colostrum quality were also evaluated. The experiment was conducted in complete randomized split-plot design, serum IgG and trace minerals were evaluated by analysis of variance and used, respectively, Duncan's test and Fisher. The α error rate of 7% was accepted. There were no differences between groups for the occurrence of retained placenta, colostrum quality and serum concentrations of Zn and IgG (P>0.07), a difference was observed for Cu (P<0.07) concentrations. The IgG concentrations were different on the weeks pre partum evaluated (P <0.07).

ABSTRACT

Alpha-thalassemia is the most common inherited disorder of hemoglobin synthesis. Genomic deletions involving the alpha-globin gene cluster on chromosome 16p13.3 are the most frequent molecular causes of the disease. Although common deletions can be detected by a single multiplex gap-PCR, the rare and novel deletions depend on more laborious techniques for their identification. The multiplex ligation-dependent probe amplification (MLPA) technique has recently been used for this purpose and was successfully used in the present study to detect the molecular alterations responsible for the alpha-thalassemic phenotypes in 8 unrelated individuals (3 males and 5 females; age, 4 months to 30 years) in whom the molecular basis of the disease could not be determined by conventional methods. A total of 44 probe pairs were used for MLPA, covering approximately 800 kb from the telomere to the MSLN gene in the 16p13.3 region. Eight deletions were detected. Four of these varied in size from 240 to 720 kb and affected a large region including the entire alpha-globin gene cluster and its upstream regulatory element (alpha-MRE), while the other four varied in size from 0.4 to 100 kb and were limited to a region containing this element. This study is the first in Brazil to use the MLPA method to determine the molecular basis of alpha-thalassemia. The variety of rearrangements identified highlights the need to investigate all cases presenting microcytosis and hypochromia, but without iron deficiency or elevated hemoglobin A2 levels and suggests that these rearrangements may be more frequent in our population than previously estimated.

ABSTRACT

The present study was designed to evaluate the time course changes in peripheral markers of oxidative stress in a chronic HgCl2 intoxication model. Twenty male adult Wistar rats were treated subcutaneously daily for 30 days and divided into two groups of 10 animals each: Hg, which received HgCl2 (0.16 mg kg-1 day-1), and control, receiving the same volume of saline solution. Blood was collected at the first, second and fourth weeks of Hg administration to evaluate lipid peroxidation (LPO), total radical trapping antioxidant potential (TRAP), and superoxide dismutase (Cu,Zn-SOD), glutathione peroxidase (GPx), glutathione-S-transferase (GST), and catalase (CAT). HgCl2 administration induced a rise (by 26 percent) in LPO compared to control (143 ± 10 cps/mg hemoglobin) in the second week and no difference was found at the end of the treatment. At that time, GST and GPx were higher (14 and 24 percent, respectively) in the Hg group, and Cu,Zn-SOD was lower (54 percent) compared to control. At the end of the treatment, Cu,Zn-SOD and CAT were higher (43 and 10 percent, respectively) in the Hg group compared to control (4.6 ± 0.3 U/mg protein; 37 ± 0.9 pmol/mg protein, respectively). TRAP was lower (69 percent) in the first week compared to control (43.8 ± 1.9 mM Trolox). These data provide evidence that HgCl2 administration is accompanied by systemic oxidative damage in the initial phase of the process, which leads to adaptive changes in the antioxidant reserve, thus decreasing the oxidative injury at the end of 30 days of HgCl2 administration. These results suggest that a preventive treatment with antioxidants would help to avoid oxidative damage in subjects with chronic intoxication.

Patients with sickle-cell anemia submitted to frequent blood transfusions are at risk of contamination with hepatitis C virus (HCV). Determination of HCV RNA and genotype characterization are parameters that are relevant for the treatment of the viral infection. The objective of the present study was to determine the frequency of HCV infection and the positivity for HCV RNA and to identify the HCV genotype in patients with sickle-cell anemia with a history of blood transfusion who had been treated at the Hospital of the HEMOPE Foundation. Sera from 291 patients were tested for anti-HCV antibodies by ELISA 3.0 and RIBA 3.0 Chiron and for the presence of HCV RNA by RT-PCR. HCV genotyping was performed in 19 serum samples. Forty-one of 291 patients (14.1 percent) were anti-HCV positive by ELISA and RIBA. Both univariate and multivariate analysis showed a greater risk of anti-HCV positivity in those who had started a transfusion regime before 1992 and received more than 10 units of blood. Thirty-four of the anti-HCV-positive patients (34/41, 82.9 percent) were also HCV RNA positive. Univariate analysis, used to compare HCV RNA-negative and -positive patients, did not indicate a higher risk of HCV RNA positivity for any of the variables evaluated. The genotypes identified were 1b (63 percent), 1a (21 percent) and 3a (16 percent). A high prevalence of HCV infection was observed in our patients with sickle-cell anemia (14.1 percent) compared to the population in general (3 percent). In the literature, the frequency of HCV infection in sickle-cell anemia ranges from 2 to 30 percent. The serological screening for anti-HCV at blood banks after 1992 has contributed to a better control of the dissemination of HCV infection. Because of the predominance of genotype 1, these patients belong to a group requiring special treatment, with a probable indication of new therapeutic options against HCV

ABSTRACT

The purpose of the present study was to examine myocardial antioxidant and oxidative stress changes in male and female rats in the presence of physiological sex hormone concentrations and after castration. Twenty-four 9-week-old Wistar rats were divided into four groups of 6 animals each: 1) sham-operated females, 2) castrated females, 3) sham-operated males, and 4) castrated males. When testosterone and estrogen levels were measured by radioimmunoassay, significant differences were observed between the castrated and control groups (both males and females), demonstrating the success of castration. Progesterone and catalase levels did not change in any group. Control male rats had higher levels of glutathione peroxidase (50 percent) and lower levels of superoxide dismutase (SOD, 14 percent) than females. Control females presented increased levels of SOD as compared to the other groups. After castration, SOD activity decreased by 29 percent in the female group and by 14 percent in the male group as compared to their respective controls. Lipid peroxidation (LPO) was assessed to evaluate oxidative damage to cardiac membranes by two different methods, i.e., TBARS and chemiluminescence. LPO was higher in male controls compared to female controls when evaluated by both methods, TBARS (360 percent) and chemiluminescence (46 percent). Castration induced a 200 percent increase in myocardial damage in females as determined by TBARS and a 20 percent increase as determined by chemiluminescence. In males, castration did not change LPO levels. These data suggest that estrogen may have an antioxidant role in heart muscle, while testosterone does not
