ABSTRACT

Aim To investigate the effect of RORα antagonist T0901317 promoting EMT (epithelial-mesenchymal transition) of human gastric cancer MGC803 cells by RORα/β-catenin signal. Methods Cell proliferation was detected by MTT. Cell migration and invasion were detected by cell scratch and Transwell assay respectively. RORα/β-catenin signaling molecules were detected by Western blot and immunofluorescence. RORα binding to β-catenin protein was detected by immunoprecipitation. Results MTT assay showed that the proliferation ability of T0901317 cells increased in a time-dependent manner compared with MGC803 cells (P < 0. 05). Cell scratches and Transwell experiments showed that the migration and invasion ability of T0901317 cells were significantly enhanced compared with MGC803 cells (P < 0. 05). Western blot analysis showed that RORα protein was significantly down-regulated after T0901317 compared with untreated group (P < 0. 05), and total β-catenin protein and nuclear β-catenin in MGC803 cells were up-regulated after T0901317 (P < 0. 05). Compared with the control group, RORα protein binding to β-catenin protein significantly decreased after T0901317 treatment (P < 0. 05). Compared with MGC803 cells treated with T0901317, the long spindle cells increased and the heteromorphism was more obvious. T0901317 significantly up-regulated the expression of Rac1, TGFβ1 and Vimentin in MGC803 cells (P < 0. 05), and inhibited the expression of E-cadherin (P < 0. 05). Conclusion T0901317 can promote the proliferation, migration, invasion and EMT in human gastric cancer MGC803 cells by RORα/β-catenin signal.

ABSTRACT

Objective:To evaluate the emergence and prevalence of drug-resistant strains among HIV-1-infected individuals who had failed antiretroviral therapy (ART) in Suqian.Methods:A total of 561 HIV-1-infected patients who had received ART for six months and above in Suqian in 2019 were recruited. Whole blood samples were collected from them with EDTA anticoagulation tubes. Viral loads (VLs) were tested for evaluating the outcomes of ART. HIV-1 subtypes and drug-resistant genotypes were detected if VL in blood samples was ≥1 000 IU/ml for further analyzing the emergence and prevalence of resistant strains.Results:ART successfully suppressed the replication of HIV-1 in 91.1% of the 561 patients (VLs≤1 000 IU/ml). Forty-six HIV-1 pol gene sequences were obtained through RT-PCR from 50 patients experiencing failed ART. Phylogenetic analysis showed that the predominant subtype was CRF07_BC (32.6%), and some new recombinant subtypes (CRF67_01B, CRF79_0107 and CRF87_cpx) were also detected. Drug-resistant strains mainly caused by K65R (30.8%), M184V/I (30.8%), K103N (19.2%), Y181C (23.1%) and G190A/S (23.1%) mutations were isolated form 56.5% of the patients failed to respond to ART. ART failure in the cases (43.5%) negative for drug-resistant strains might result from non-adherence. Conclusions:CRF07_BC was the predominant subtype of HIV-1 circulating in Suqian and various new recombinant subtypes were emerged. Drug resistance might be the main cause of ART failure. It was very necessary to enhance the adherence education during the course of ART for suppressing the replication of HIV-1 and preventing from the emergence of drug-resistant strains.

ABSTRACT

Hemodynamic situation is an important factor of recurrence of postoperative carotid artery aneurysm. In order to investigate the hemodynamic factors of postoperative carotid artery aneurysm affect carotid artery aneurysm recurrence, we established a 3D finite element carotid artery aneurysm for the preoperative and postoperative periods using the three-dimensional reconstruction techniques. And then we measured the hemodynamic factors of carotid artery aneurysm of preoperative and postoperative by the finite element method. The carotid artery aneurysm model has an accurate and realistic shape; the pressure of the recurrence of aneurysm was reduced significantly after surgery,wall shear stress increased significantly at residual neck, and blood flow velocity increased significantly, which will increase the risk of recurrence. The hemodynamic analysis provides a reference for development of aneurysm clinical treatment programs and prevention of recurrence.

ABSTRACT

@#ObjectiveTo analysis the clinical characteristics and the high risk factors of children with cerebral palsy.MethodsClinical data of 100 children with cerebral palsy were retrospectively analyzed.Results51% of children were born with asphyxia, 42% were born prematurely, 39% were low birth body weight. 85.7% of children involved had mental retardation, 12% had hearing loss and 7% had visual impairment. 88% of children involved had abnormal cranial CT results and 46.1% had abnormal EEG. 50.0% of the children had abnormal brain stem auditory evoked potentials.ConclusionAsphyxia, prematurely birth and low birth body weight are high risk factors of cerebral palsy. Most of the children with cerebral palsy in this group are mental retarded.

@#ObjectiveTo investigate the efficacy of acupuncture point injection for fine motor functions in children with cerebral palsy.Methods57 children with cerebral palsy were treated by acupuncture point injection combined with occupational therapy. The efficacy was evaluated by the Peabody Developmental Motor Scale-Fine Motor (PDMS-FM).ResultsThe fine motor quotient (FMQ) of 34 children (59.7%) elevated. The raw scores of grasping subtest (Gr) elevated in 45 children (78.9%) and the raw scores of visual-motor integration (VI) elevated in 53 children (93.0%).ConclusionThe efficacy of acupuncture point injection combined with occupational therapy is significant in children with cerebral palsy.
