ABSTRACT

Objective:To investigate the analytical performance verification protocols and performance specifications of CD34+cell enumeration by flow cytometry for clinical laboratories. Methods:According to international guidelines and National Health Standard of China, we designed the performance verification protocols of CD34 +cell enumeration (including percent count and absolute count) by flow cytometry. Four quality assessment materials, three leukapheresis products and three samples of peripheral blood were selected to verify the precision, linearity, carryover, trueness and accuracy of FACSCanto Ⅱ measurement system, and the assessment criterion was set according to the detection technologies of clinical laboratories. Results:The CVs of intra-run precision of percent count and absolute count were 2.5% to 8.9% and 3.0% to 9.0%; the CVs of inter-run precision were 2.8% to 10.5% and 3.8% to 9.9%, respectively. The slopes of linearity regression equation of low range (3.6/μl to 123.6/μl) and high range (113.2/μl to 1196.3/μl) were 0.993 2 and 0.965 2, and R2 were 0.999 6 and 0.993 9, and the biases were -8.67% to 0.22%. The carryover of percent and absolute count were 0.07% and 0.00%. When percent count≤0.2% or absolute count≤20/μl, the absolute biases of trueness were in the range of ±0.006% or ±0.5/μl, and the absolute biases of accuracy were in the range of ±0.02% or ±0.9/μl; when percent count>0.2% or absolute count>20/μl, the relative biases of trueness were in the range of ±5.65%, and the relative biases of accuracy were in the range of ±8.19%. The verification results met the assessment criterion set in this study. Conclusions:The performance verification protocols and assessment criterion formulated in this study not only conform to the recommendations of domestic and foreign guidelines, but also conform to state of the detection technologies of native clinical laboratories, which can be taken as a reference of performance verification for clinical laboratories.

ABSTRACT

Objective To compare the effects of hip abductor fatigue on postural stability and neuromuscular control in different gender groups during single-leg side-jump landing. Methods Twenty male and twenty female adults were required to execute single-leg side-jump landing before and after hip abductor fatigue protocol. The center of pressure (COP), ground reaction force (GRF), lower extremity kinematics, joint moment, muscle activity were compared. Results For both males and females after fatigue, the maximum displacements and average velocities of COP decreased, and the peak angles of hip abduction and ankle eversion increased, and the peak torques of ankle inversion increased. At 200 ms pre-landing, the activiation of rectus femoris, biceps femoris, tibialis anterior, peroneus longus were lower in male than that in female. At 200 ms post-landing, the activiation of biceps femoris in male was lower than that in female after fatigue. Conclusions The postural stability in frontal plane decreased after hip abductor fatigue. The frontal plane control of hip joint and ankle joint was weakened, which might increase the risk of joint injury. There are differences in postural strategies for different genders, indicating that gender difference in the mechanism of lower limb joint injury is worth of further investigation.

ABSTRACT

Objective:The purpose of current study was to evaluate the interplay effects in intensity-modulated proton therapy (IMPT) for lung cancer and compare the results of different Iso-energy layer repainting techniques in the mitigation of interplay effects. Methods:Eight patients with lung cancer who underwent 4DCT were retrospectively selected. A robust CTV-based IMPT plan was generated for each based on commercial TPS, considering patient setup errors ±5 mm, range uncertainties ±3.5%, and CTV time structure motion in 4DCT image. Monte Carlo dose engines were used for all IMPT plans in the final dose calculation. The 4D static dose (4DSD) and 4D dynamic dose (4DDD) were calculated using a hybrid deformable algorithm and simulated proton delivery system for interplay effects. An index[ΔI(ROI, DVH)] was developed to quantitatively evaluate the interplay effects. We applied Iso-energy layer repainting techniques with different numbers of repainting (3, 4, 5, 6, 7) to the robust IMPT plans and evaluated the difference in the mitigation of interplay effects based on the ΔI(ROI, DVH) index. Results:Due to interplay effects, the mean values of target coverage, conformity and homogeneity index reduced by 13.7%, 12.7% and 24.6%, respectively. The mean values of lung V 5Gy and V 20Gy improved by 0.8%, 3.4% and 2.6%. Compared to the IMPT plans without layer repainting, Multiple iso-energy layers repainting techniques improved the mean values of CTV coverage by 4.5%, 3.8%, 3.8%, 3.6% and 5.7%, respectively. The average values of lung V 20Gy reduced by 1.5%, 1.8%, 1.7%, 1.6% and 1.9%, respectively. Conclusions:In the robust CTV-based IMPT plans, the interplay effects degraded the target dose distribution but were mitigated using iso-energy layer repainting techniques. We recommended to use the layer repainting technique according to the characteristics of the patient.

ABSTRACT

Objective:To study the accuracy and positioning error of image guidance during the treatment of body stereotactic radiotherapy (SBRT) in patients with non-small cell lung cancer (NSCLC) by cone beam CT (CBCT) of VERO system. Methods:The clinical data of 80 NSCLC patients from January 2017 to December 2018 in Lishui People′s Hospital of Zhejiang Province were retrospectively analyzed. The maximum density projection and average density projection were reconstructed by four dimensional cone beam CT (4DCBCT), and internal target volume (ITV) was obtained by mapping the tumor profile. The translational and rotating tumor positioning errors were determined by comparing the manual ITV registration with the motion-fuzzy tumor on the CBCT. At the same time, correction was completed by the robot positioning bed and circular rotation. After correction, the CBCT was verified to evaluate the residual error. Results:The initial set average three dimensional vector displacement was significantly greater than the six dimensional automatic calibration displacement: (6.8 ± 2.1) mm vs. (1.5 ± 0.5) mm, and there was statistical difference ( P<0.01); 94.0% (425/452) of the rotation error was ≤ 3°. If there was no image guidance, the stereo positioning based on ExacTrac required 6.6 mm in the left-right direction, 9.8 mm in the up-down direction, and 6.5 mm in the front-back direction. After six dimensional correction of residual errors, the safety margins in three directions were reduced to 3.0, 3.2, and 3.1 mm, respectively. Conclusions:Online image guidance using ITV-CBCT matching technology and the automatic six dimensional correction of the VERO system can reduce the positioning error in SBRT treatment of NSCLC patients and improve the accuracy of tumor positioning.

ABSTRACT

Objective@#In clinical, cephalic index is the most common method to evaluate scaphocephaly. However, cephalic index alone is not sufficient to describe the nuances of skull shape. The authors hypothesize that the location of euryon could be a valuable and practical index as the supplement of cephalic index to better describe the skull.@*Methods@#This is a retrospective study. CT scans of 17 scaphocephaly patients and 17 normal controls were included. Eight of 17 scaphocephaly patients have 6 months postoperative CT scan. The preoperative location of euryon of scaphocephaly patients was compared to that of the controls and postoperative. The horizontal location of euryon was defined as the ratio of the distance, from the most prominent point of forehead to euryon, to anteroposterior cranial length. The vertical location of euryon was defined as the ratio of the vertical distance, from the nasion to euryon, to cranial height.@*Results@#Both the horizontal and vertical locations of euryon of preoperative scaphocephaly patients were significantly decreased compared to normals (46.50%±4.02% vs. 50.39%±2.21%, t=3.49, P=0.0014; 35.78%±8.62% vs. 52.6%±8.08%, t=5.86, P<0.001). Both the postoperative horizontal vertical locations of euryon were significantly increased, when compared to preoperative (47.25%±4.23% vs. 44.02%±3.72%, t=2.42, P=0.045; 42.39%±9.06% vs. 35.18%±5.36%, t=2.554, P=0.0379).@*Conclusions@#The location of euryon could be a practical and valuable index to describe the nuances of skull shape.

ABSTRACT

Objective@#To investigate the effect and mechanism of LncRNA ANRIL on the radiosensitivity of HCT116 cells line and nude mouse transplant tumors.@*Methods@#The expression of LncRNA ANRIL in colorectal cancer cells was detected by qPCR. The negative control siRNA, ANRIL siRNA, miR-NC mimic, miR-195 mimic, miR-NC inhibitor and miR-195 inhibitor were transfected into HCT116 cells, and marked as negative control group, silencing ANRIL group, overexpressing miR-NC group, overexpressing miR-195 group, inhibiting miR-NC group and inhibiting miR-195 group, and the HCT116 cells without any treatment were marked as the blank control group. The clone formation assay was used to detect radiosensitivity of colorectal cancer cells, flow cytometry was used to detect apoptosis. The web site, StarBase, was used to predict the downstream miRNAs of ANRIL and dual luciferase reporter gene assay was used to further verify. Subcutaneous tumor transplantation assay was used to detect the effect of ANRIL on the growth of colorectal cancer cells after irradiation.@*Results@#After irradiation with 2, 4, 6 and 8 Gy, the cell survival fraction of silencing ANRIL group was significantly decreased when compared with that of negative control group (P<0.05), and the radiosensitivity ratio was 1.52. The apoptosis rate of the silencing ANRIL+ 4 Gy group was significantly higher than that of the negative control+ 4 Gy group ((27.86±2.78)% vs. (12.06±1.46)%, P<0.05). The results of the experiment on nude mouse transplant tumors showed that the tumor volume in the negative control group was lower than that of the silent ANRIL group on days 13, 16, 19, 22 and 25 ((234±66) mm3, (273±63) mm3, (296±72) mm3, (321±85) mm3 and (403±94) mm3 vs. (357±79) mm3, (485±124) mm3, (617±143) mm3, (764±174) mm3 and (985±221) mm3P<0.05). MiR-195 is a target gene of ANRIL, and inhibition of miR-195 can reverse the inhibitory effect of silencing ANRIL on radiosensitivity, apoptosis and xenografts of HCT116 cells.@*Conclusions@#LncRNA ANRIL regulates the radiosensitivity of colorectal cancer cells by miR-195, which may provide a new sensitizing target for clinical colorectal cancer radiotherapy.

Objective@#To assess the association of single nucleotide polymorphisms (SNPs) in SLCO1B3 gene with prognosis of breast cancer (BC) patients treated with neoadjuvant chemotherapy of TA regimen (taxane and antharcycline drugs).@*Methods@#439 female BC patients were recruited and treated with neoadjuvant chemotherapy of TA regimen. A blood sample (2 ml) of peripheral blood was collected from each patient before chemotherapy. Tagging SNPs (tag-SNPs) were selected. We investigated the association of tag-SNPs with prognosis, by Sequenom Mass ARRAY system platform, characterizing tag-SNPs. The hazard ratio (HR) and 95% confidence interval (CI) for progression or death were calculated by multivariable-adjusted Cox regression model.@*Results@#Seven tag-SNPs (rs11045689, rs200104106, rs3764006, rs3834935, rs4149117, rs7305323 and rs73241801) were selected for study. Compared with individuals carrying the rs11045689 GG genotype, individuals carrying rs11045689 AA genotype performed worse PFS and OS, with the HR (95% CI) for progression being 1.39 (1.11~1.75) and the HR (95% CI) for death being 1.38 (1.04~1.83). Compared with individuals carrying the rs73241801 CC genotype, individuals carrying rs73241801 TT genotype performed better OS (P=0.041), with the HR (95% CI) for death being 0.65 (0.44~0.94). The number of risk allele was significantly associated with PFS (P=0.012) and OS (P=0.017) of BC patients by accumulation analysis. Compared with individuals carrying one or less than one risk allele, individuals carrying four risk alleles performed worse PFS and OS, with the HR (95% CI) for progression being 1.37 (1.09~1.72) and the HR (95% CI) for death being 1.36 (1.02~1.81).@*Conclusion@#The variations of rs11045689 and rs73241801 in SLCO1B3 gene were significantly associated with prognosis of BC patients treated with neoadjuvant chemotherapy of TA regimen, which might serve as biomarkers for predicting prognosis of BC patients treated with neoadjuvant chemotherapy.

ABSTRACT

The present study was aimed to investigate the underlying mechanisms of the protective effect of proanthocyanidin (Pro) against hypoxia/reoxygenation (H/R) injury in H9C2 cells with a focus on Janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) signaling pathway. H9C2 cells were randomly assigned to 5 groups, including the control group (Con), the H/R-injured group (H/R), the Pro-treated group (H/R+Pro), the JAK2 siRNA-treated group (H/R+Pro+JAK2 siRNA) and the JAK2 siRNA control group (H/R+JAK2 siRNA). The cells were pretreated with Pro (40 µmol/L) for 8 h before 2 h of hypoxia and 4 h of reoxygenation. Cellular viability and apoptosis rate were detected by MTT and TUNEL methods, and superoxide generation was measured. JAK2/STAT3 signaling, oxidative stress markers and endoplasmic reticulum stress markers were also detected by Western blot. We found that Pro treatment significantly improved cellular viability and reduced apoptosis rate in H/R-treated H9C2 cells. In addition, Pro treatment significantly up-regulated the phosphorylation levels of JAK2 and STAT3, down-regulated the superoxide generation, gp91, glucose-regulated protein 78 (GRP78), CCAAT/enhancer binding protein homologous protein (CHOP) and caspase-12 expression. However, these protective effects of Pro were all attenuated by JAK2 siRNA administration. Taken together, we demonstrated that Pro protects H9C2 cells against H/R-induced oxidative stress and endoplasmic reticulum stress injury via JAK2/STAT3 signaling pathway.

ABSTRACT

Objective To prepare and evaluate the reference materials for plasma von Willebrand Factor antigen testing with fresh frozen plasma. Methods The candidates were prepared by low temperature centrifugation in 5 different concentration levels. The homogeneity and stability of the preparation was evaluated according to the ISO Guide35 and CNAS-GL03.The comparability between STAGO and IL system was evaluated according to the WS/T 356-2011.Then the preparations were characterized by six laboratories with the Secondary Coagulation Standard established by NIBSC(SSCLOT4).Results Homogeneity evaluation of the preparation showed that there was no statistically significant difference between the groups (P >0.05),the F values of factor analysis of variance were 0.317~0.844,the uncertainty range was 1.01% ~2.06%.A linear regression based on stability evaluation indicated that the linear trend (within 24 weeks)was insignificant (P >0.05). The uncertainty range of long-term (within 24 weeks)stability was 0.79% ~ 1.20%.The results of the preparations on STAGO and IL system were comparable. The certificated values of the candidates were range from 12.2% to 138.9% with uncertainties were 0.06%~0.09%,respectively.The range of combined standard uncertainty was 0.03% ~ 0.16% while the expanded uncertainty was 2.2%~6.7%.Conclusion The reference materials for von Willebrand Factor antigen testing were stable and homogenous with comparability between STAGO and IL.The method of characterization was accurate and reliable.

ABSTRACT

To explore the treatment effects of HydroCoil Embolic System [HES] on large- and med sized intracranial aneurysms. Fifty cases of intracranial aneurysm patients were retrospectively analyzed, in which 27 an cases were treated with HES [n=27] and ordinary coils [n=23], respectively. All the patients were followed up for two years. The 27 cases [54%, 27/50] treated with HES include 23 cases of densely packed occlusion [23/50] and 4 cases of subtotal occlusion [8%, 4/50]. The 23 cases [46%, 23/50] treated with ordinary include 15 cases of densely packed occlusion [30%, 15/50] and 8 cases of subtotal occlusion [16%, 8/5. HydroCoil Embolic System [HES] may increase the ratio of densely packed occlusion prevent the recurrence of large- and medium-sized intracranial aneurysms

ABSTRACT

Objective To evaluate the dosimetric characteristics of hippocampal formation (HF) in the intensity-modulated radiotherapy (IMRT) for nasopharyngeal carcinoma (NPC).Methods Fifty-nine NPC patients underwent IMRT.Simultaneous integrated boost technology was used to determine the doses for the target areas. The dose ranges of the HF were collected by dose-volume histogram. The influence of T stage on the exposure doses ( Dmax,Dmean,D20,V10,V20,V30,and V40 ) were compared. Results The maximum dose for the HF (Dmax) ranged from 11.1 to 78.2 Gy(F =24.2,P <0.05) and the Dmean ranged from 3.2 to 44.6 Gy ( F =16.3,P < 0.05 ).The Dmax and Dmean of the T1-2 stage patients were (40.8 ±9.4) Gy and ( 12.5 ±5.1 ) Gy,respectively, both significantly lower than those of the T3-4 stage patients [ (58.6± 14.8) Gy,(20.9± 9.3 ) Gy].The mean exposed volume of the T4 stage patients was significantly larger than that of the T1 and T2 stages patients. Conclusions In the IMRT of NPC,the HF receives rather high irradiation dose.T stage is the main factor influencing the dose, especially T3 and T4 stages deserve serious attention.
