ABSTRACT

AIM: To explore the promoting effect of 2-APB on skin wound healing in mice and its potential mechanism. METHODS: KM mice were divided into 5 groups: control group, DMSO group, low (50 mg/L), medium (100 mg/L) and high (200 mg/L) concentration 2-APB group. On the back of each mouse's skin use a circular punch about 1 cm on both sides of the midline of the spine to make a skin wound with a diameter of 10 mm and as deep as the fascia. The control group was only wrapped with gauze and no drugs were applied; the DMSO group was applied 1 g DMSO/Vaseline ointment per day; in the 2-APB group, apply 1 g of 2-APB/Vaseline ointment at a corresponding concentration every day. Pictures were taken the next day to observe the healing, and the material was taken on the 21st day, HE staining was used to observe the pathological morphology of the wound and western blot to detect TRPM7, TGF-β, collagen-I and IL-1β expression. RESULTS: Compared with the control group and the DMSO group, different concentrations of 2-APB could significantly promote skin wound healing in mice (P<0.01), but there was no significant difference in wound healing rate between the DMSO group and the control group group. The results of HE staining showed that, compared with the control group group and the DMSO group, 2-APB could increase the collagen content of the wound and the thickness of the dermis (P<0.01), but there was no significant difference between the DMSO group and the control group group. At the same time, 2-APB could also significantly increase the expression of TGF-β and Col-I on the wound, and inhibit the expression of TRPM7 and IL-1β. CONCLUSION: Different concentrations of 2-APB (50, 100 and 200 mg/L) can promote skin wound healing, and its mechanism may be related to the inhibition of TRPM7.

ABSTRACT

Objective:To investigate the therapeutic effect of Postpartum Zhuyu capsule combined with norethisterone in the treatment of menorrhagia and its influence on sex hormone.Methods:From January 2016 to December 2018, 120 patients with irregular menstruation admitted to the First People's Hospital of Jiande were divided into observation group (60 cases) and control group (60 cases) according to the random digital table method.The control group was treated with norethisterone, while the observation group was treated with Postpartum Zhuyu capsule combined with norethisterone.Both two groups began to use drugs on the 5th day of menstruation, and the course of treatment was three menstrual cycles.The therapeutic effect, PBAC, Pittsburgh sleep quality index (PSQI), sex hormone were compared before and after treatment.Results:The total effective rate of the observation group (95.00%) was higher than that of the control group (75.00%) (χ 2=9.412, P<0.05). After treatment, the scores of PBAC [(72.34±10.38)points] and PSQI[(3.65±1.20)points] in the observation group were lower than those in the control group [(87.82±8.90)points and (6.98±1.45)points] ( t=8.770, 13.705, all P<0.05). The serum E 2 level [(221.28±19.87)ng/L] in the observation group after treatment was higher than that in the control group [(135.42±24.71)ng/L], while FSH [(12.39±2.87)IU/L] and LH [(14.68±2.37)IU/L] were lower than those in the control group [(19.83 ± 2.32)IU/L and (39.94±3.24)IU/L], the differences were statistically significant( t=20.975, 15.616, 48.742, all P<0.05). Conclusion:Postpartum Zhuyu capsule combined with norethisterone is effective in the treatment of menorrhagia.It can improve the serum levels of sex hormone, and improve the quality of life of patients.

ABSTRACT

Objective:To investigate the effect of estradiol valerate combined with dydrogesterone on prevention of uterine adhesions after abortion and its influence on sex hormone level.Methods:From January 2018 to October 2019, 142 patients who underwent artificial abortion in the First People's Hospital of Jiande were divided into observation group(71 cases) and control group(71 cases) according to the random digital table mathod.The control group was given postoperative treatment to promote uterine contraction and congestion, and the observation group was given estradiol valerate tablets combined with dydrogesterone tablets on the basis of the control group.The course of treatment in both two groups was 3 months.The bleeding time, bleeding volume and menstrual rehydration time, menstrual volume after operation, the change of serum sex hormone level preoperation and postoperative 3 months, and the occurrence of intrauterine adhesions at 12 months after operation were compared between the two groups.Results:The bleeding time[(4.35±0.97)d] and menstrual recovery time[(24.83±1.43)d] in the observation group were shorter than those in the control group[(5.67±1.14)d and (28.79±1.78)d], and the bleeding amount[(42.54±8.97)mL]in the observation group was less than that in the control group[(53.67±6.81)mL], the differences were statistically significant between the two groups( t=7.431, 14.614, 8.327, all P<0.05). The patients with normal amount of menstruation in the observation group (94.37%) was more than that in the control group(81.69%)(χ 2=5.413, P<0.05). The serum E2 level[(167.39±27.84)ng/L] in the observation group was higher than that in the control group[(108.92±21.01)ng/L], while FSH[(17.83±3.27)IU/L] and LH[(28.98±7.63)IU/L] in the observation group were lower than those in the control group[(26.54±4.58)IU/L and (49.83±13.54)IU/L], the differences were statistically significant between the two groups( t=14.126, 13.042, 11.304, all P<0.05). The incidence of intrauterine adhesions in the observation group(2.82%) was lower than that in the control group(14.08%)(χ 2=5.826, P<0.05). Conclusion:Estradiol valerate combined with dydrogesterone has good effect on the prevention of postoperative uterine adhesions, which can effectively reduce the amount of postoperative bleeding, reduce the amount of postoperative uterine adhesions, improve the amount of menstruation and improve the level of sexual hormones of patients.It is worthy of clinical reference.

ABSTRACT

Promoter-trap strategy for enriching targeted colonies has been usually used to elevate the gene targeting efficiency in somatic cells. Knocking out Prnp in animals by gene targeting can render them resistant to Prion diseases. We constructed a bovine Prnp promoter-less targeting vector BoPrPneo, then transfected the linearized vector into the bovine fetal fibroblasts BFF through electroporation. After selecting in cell culture medium with 250 microg/mL G418, we obtained 99 drug-resistant cell colonies, 4 of them were positive for targeted events after PCR screening, and the targeted colonies were further confirmed by sequencing and Southern blotting. This suggests that one allele of Prnp has been successfully knocked out in bovine fetal fibroblasts. This research supplies a simple, safe and effective method to targeting bovine Prnp.
