ABSTRACT

WRKY transcription factors are novel transcriptional regulatory factors, which play an important role in regulating plant development, metabolism and other physiological processes. In this study, a new Dendrobium officinale WRKY transcription factor, designated as DoWRKY1 was cloned by using RT-PCR and RACE (GenBank Accession No. KF953910). Bioinformatic analysis demonstrated that, the full-length cDNA of DoWRKY1 was 1,704 bp. And DoWRKY1 contained a 1,629 bp open reading frame (ORF) that encoding a peptide of 542 amino acid residues. The putative DoWRKY1 protein contained two conserved WRKY domains and it belonged to the group I WRKY family protein. Yeast one-hybrid experiment showed that DoWRKY1 had transcriptional activation ability in yeast, and it could activate the expression of downstream report genes (His3 and Ade2). Semi-quantitative RT-PCR experiment showed that DoWRKY1 expressed in roots, stems, leaves and protocorm-like bodies. Real-time qRT-PCR proved that DoWRKY1 could be induced by methyl jasmonate (MeJA) and chitosan (Chitosan), and the expression level of this gene can reach the expression peak at 2 h and 1 h, respectively. These results are useful for further determination of the regulation function of this gene in secondary metabolism of D. officinale.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expressions of CDX2 and β-catenin and their correlation in acute myeloid leukemia (AML).</p><p><b>METHODS</b>Real-time PCR was used to examine the expressions of CDX2 and β-catenin mRNA in 92 de novo AML and 30 non-malignant patients (control), and Western blot was employed to detect CDX2 and β-catenin protein expressions in 26 de novo AML and 8 non-malignant patients.</p><p><b>RESULTS</b>The positivity rates of CDX2 mRNA and protein expressions in AML group were 84.78% and 76.92%, respectively, but no CDX2 mRNA or protein expression were detected in control group (P<0.01). The expressions of β-catenin mRNA and protein were detected in all the patients in both groups but its expressions in AML group were significantly higher (P<0.01). The expressions of CDX2 and β-catenin mRNA were significantly correlated with WBC counts and LDH levels (P<0.01). The expression of CDX2 mRNA was not detected and the expression of β-catenin mRNA was decreased to normal level in AML patients with complete remission, while the expressions of CDX2 and β-catenin mRNA were increased again in those with disease relapse. There were significantly positive correlations between CDX2 and β-catenin expressions at both mRNA and protein levels (P<0.01).</p><p><b>CONCLUSION</b>Up-regulation of CDX2 gene and activation of Wnt/β-catenin pathway co-exist in AML patients and the expressions of CDX2 and β-catenin are positively correlated.</p>

ABSTRACT

Objective To investigate the expressions of CDX2 and β-catenin in acute lymphoblastic leukemia (ALL) and their correlation. Methods Real-time PCR was used to determine the expressions of CDX2 and β-catenin mRNA in 43 de novo ALL and 30 non-malignant patients (used as control). Results The positivity rate of CDX2 mRNA expression in ALL group was 93%, but CDX2 mRNA expression couldn′t be detected in the control group (P<0.01). The mRNA expression of β-catenin could be detected in patients in both two groups, butβ-catenin mRNA expression in the ALL group was significantly higher than in the control group (P < 0.01). And mRNA expressions of CDX2 andβ-catenin were significantly correlated with WBC counts and LDH level (P<0.01). When the ALL patients acquired complete remission (CR), the mRNA expressions of CDX2 and β-catenin were significantly decreased compared with their newly diagnosed status , while disease-relapsed the mRNA expressions of CDX2 andβ-catenin were increased again. There was significantly positive correlation between CDX2 and β-catenin mRNA expressions (r = 0.835, P = 0.000). Conclusion Up-regulation of CDX2 and activation of Wnt/β-catenin pathway coexist in the ALL patients and the mRNA expressions of CDX2 and β-catenin are positively correlated.
