ABSTRACT

We conducted purification of filamentous temperature-sensitive protein Z of Streptococcus suis serotype 2 (S.suis 2) and measured its GTPase activity. The ftsz gene in the genome of the Chinese 05ZYH33 strain of S.suis 2 was successfully amplified using PCR,and then the ftsz gene was cloned into prokaryotic expression plasmid pET28a,and the recombinant plasmid pET28a-ftsz was transformed into E.coli BL21.After induction by IPTG,the isolated FtsZ protein was analyzed with SDS-PAGE.Then the recombinant protein was purified by Ni2+-NTA affinity chromatography. The rabbit serum was harvested after immunization with recombinant FtsZ protein, and was analyzed by indirect ELISA and Western blotting. The GTPase activity of FtsZ was measured with the malachite green method. Results showed that successfully constructed recombinant plasmid pET28a-ftsz and the recombinant protein with high purity was obtained;Western blot result indicated that FtsZ could react with the His-tag antibody and the rabbit serum;the polyclonal antibody titer of the rabbit serum reached 1 ∶ 13 107 200;FtsZ have GTPase activity. We successfully prepared S.suis 2 recombinant protein FtsZ having GTPase activity and high titer antiserum would be useful for the further study of S.suis 2 cell division mechanism.

ABSTRACT

Objective This study was to investigate the relation of nitric oxide synthase with cell proliferation and apoptosis in hepatocellular carcinoma (HCC). Methods The expression and localization of inducible nitric oxide synthase (iNOS) and PCNA in 21 patients with HCC were studied by immunohistochemistry. Apoptotic cell in tissue section was detected by TUNEL (terminal deoxynucleotidyl transferase [TdT] mediated desoxyuridinetriphosphate [dUTP] nick end labeling). Results The iNOS protein was markedly expressed in the liver tissue of hepatic cirrhosis or chronic liver hepatitis, which was close to tumor edge. Meanwhile it was weakly or undetectable in the liver tissue localized beyond 1.5 cm from the tumor edge. The iNOS protein was markedly expressed at the invasive tumor edge and HCC cells that invased into stroma, but it was weakly or undetectable at the most tumor core. The expression of iNOS was correlated with the proliferative activity of cells, and the number of apoptotic cells was correlated to the number of PCNA positive cells ( P

ABSTRACT

ObjectiveTo study the surgical treatment of pancreatic carcinoma. Methods 69 cases of pancreatic carcinoma in recent five years were studied retrospectively. Twenty four out of 51 cases underwent surgical resection. Whipple′s procedure was performed in 16 cases, extended regional pancreatectomy and autograft intestinal transplantation in 2 cases, combined organ resection in 6 cases of pancreatic body or pancreatic tail carcinoma. 27 cases of unresectable pancreatic carcinoma received internal drainage. Results Wound dehiscence and digestive bleeding developed in one each respectively. Pancreatic fistula occurred in one undergoing extended regional pancreatectomy plus autograft intestinal transplantation.ConclusionIn this series, 47% of the patients received surgical resection.Extended regional pancreatectomy plus intestinal autograft transplantation is feasible in surgical technique.

ABSTRACT

Objective To explore factors influencing the prognosis of hepatocellular carcinoma (HCC) patients after hepatectomy. Methods From May 1994 to January 1998, 189 patients who underwent hepatectomy for HCC were enrolled for reviewing their clinical characteristics, treatment and prognosis. Twenty-two parameters contributing to long-term survival rate (SR) and disease-free SR were analysed. Results The 3,5-year cumulative SR of the whole group was 63%,45%, respectively. The 3,5-year SR and disease-free SR in the curative resection (CR) group ( n =162) were 67%,47% and 45%,26% respectively. Results showed that the way by which a tumor was found, tumor size, portal tumor thrombi, satellite nodule, TNM stage, cirrhosis type, recurrent and treatment, blood transfusion, differentiation grade, and CR were risk factors by individual variable analysis( P =0.0000~0.0034); A multivariable analysis showed that CR, tumor size, tumor finding mode and reoperation were significant factors associating with prognosis( P =0.0000~0.0024). Blood transfusion and type of cirrhosis were closely correlated with tumor-free survival ( P =0.0001). Conclusions Curative resection, tumor size, reoperation for recurrence were important factors for recurrence by multivariate analysis. The severity of concomittant liver cirrhosis and perioperative blood transfusion were closely correlated with postoperative tumor free survival.

ABSTRACT

Objective To investigate the safety and feasibility of laparoscopic hepatic left lateral lobectomy. Methods Five cases of liver cancer, 4 cases of hepatic hemangioma, and 2 cases with suspected liver tumors were included in this series. Tumors were all located at left lateral lobe, with a diameter less than 10?cm. Tumors were resected by laparoscopy under a combination of pneumoperitoneum and non-pneumoperitoneum, with hand assistance through a mini-incision. Several manoeures were used for the dissection of liver tissue, including microwave coagulation, ultrasonic dissection, tainium clips clamping, rotating suction, endogia ligation. ResultThe procedure was successful in all cases with the operation time ranging from 1.5?h to 6?h, and bleeding was 50?ml to 500?ml. The hospitalization time were from 5?d to 15?d. ConclusionLaparoscopic liver resection is safe and feasible for the left liver lateral lobe tumor when the diameter is less than 10?cm, and not involving surrounding organs.
