ABSTRACT

Hydroxytyrosol as one of natural anti-oxidants,extracted from the fruits and leaves of Olea europaea,is a natural polyphenol compound in the form of esters. Recently,considerable studies showed that hydroxytyrosol demonstrated intrinsic biological activity for metabolic syndromes, cardiovascular- and neurodegenerative-related diseases,and it was revealed to play the roles in the anti-activities of cancerous,inflammatory as well as depressing issues. In addition,hydroxytyrosol is an oleophilic and hydrophilic compound with high bioavailability and low cellular cytotoxicity. It could be absorbed by various tissues and could easily passe through blood brain barrier. Therefore,hydroxytyrosol was introduced as one of the key subjects targeted by innovative drug development. However,it has a short half-life in vivo and non-tissue specific,which lead to its limitation in clinical application, so further in-depth studies are still needed. The authors had a literature review of hydroxytyrosol,and summarized the basic properties of its pharmacokinetic,pharmacological effects and molecular mechanisms. This article mainly focused on it’s pharmacological activity and the mechanism involved in treating damages induced by the oxidative stress,in alleviating cardiovascular diseases and in inhibition of neurodegenerative diseases. In this article, its anti-inflammatory,anti-tumor,anti-depressant effects,other biological activities,and pharmacokinetics were also briefly reviewed. The authors put forward some personal thoughts on its future research direction,hoping to provide ideas and inspirations for the vast number of researchers,and provide references for its further development,research and application.

ABSTRACT

Objective:To investigate the effects of recombinant human interluekin-1 receptor antogonist (rhIL-1Ra) on the cartilage repair in rat temporomandibular joint(TMJ) with osteoarthritis(OA).Methods:Collagenase-Ⅱ was injected into bilateral TMJs of 24 adult rats for the induction of bilateral TMJOA,1 week after injection,5μg rhIL-1Ra(diluted in 0.05 ml normal saline) was injected into each right TMJ and the left joint received the same amount of normal saline injection as the control.12 animals were sacrificed at 2and 4 weeks after the first injection respectively.HE staining,immunnohistochemical method and RT-PCR examination were conducted.Mankins scere was used to evaluate the TMJOA degree.1 adult SD rat was used as healthy control,and sacrificed at 2 weeks of the experiment.Results:The TMJs of both sides showed typical OA-related cartilage degradation 2 week after IL-1Ra treatment,the Mankin~ score of the IL-1Ra treated and control joints was 1.33±0.52 and 2.00±6.63 (P>0.05),4 week after treatment that was 3.00± 0.63 and 6.50 ± 0.84 (P<0.05),respectively.Lower expression of ADAMTS-4 and ADAMTS-5 was observed in the treated joints than in the controls (P<0.05).Conclusion:Intra-articular injection of IL-1Ra into TMJ can alleviate the cartilage lesion,the mechanism may lie in the inhibition of the expression of ADAMTS-4 and ADAMTS-5.

ABSTRACT

Mandibular symphysis fracture combined with anteromedial dislocated condyle fracture is commom in clinical,but mandibular symphysis fracture associated with superolateral dislocation of the mandibular condyle is rare,which is often misdiagnosed or completely over-looked.Malpractice can lead to ankylosis and other sequelae.This article reviews 10 patients with mandibular symphysis fracture associated with superolateral dislocation of the mandibular condyle,discusses the causative mechanism,diagnostic features and clinical management according to literature data.

ABSTRACT

Objective:To observe the effect of NGF on the expression of BMP-2 in rabbit model and explore the molecular mechanism of NGF which might promote the healing of mandibular fracture with nerve injury. Method:The 48 New-Zealand white rabbits were randomly divided into the experimental group (mandibular fracture+to cut off the nerve bundle+NGF by GS), the control group (mandibular fracture+to cut off the nerve bundle+NS by GS), the blank group (mandibular fracture+to cut off the nerve bundle) and the full-set group (mandibular fracture+retains the nerve bundle). After 2 weeks, 4 weeks, 6 weeks and 8 weeks, 3 rabbits were sacrificed in each group for HE staining and RT-PCR, respectively. Result:HE staining showed the osteogenesis phenomenon: the experimental group was clearer than control group, the full-set group was clearer than the blank group and the control group is similarly to the blank group. RT-PCR results revealed that there was a statistically significance in the early stage. The expression of BMP-2 peaked in 2 weeks and decreased later with time. Conclusion:The local application of NGF can prompt BMP-2 expression in the early stages of the mandibular fracture with partial nerve injury healing and this may be one of the molecular mechanisms.

ABSTRACT

OBJECTIVE@#To investigate the expressions of transforming growth factor beta1 (TGF-beta1) and bone morphogenetic protein-2 (BMP-2) in human mandible fracture callus and their quantity changes in the process of healing.@*METHOD@#Thirty callus samples from the fractured mandible bone stumps were collected during operation, and two callus samples were collected from the angle-square jaw patients as controls. The expressions of TGF-beta1 and BMP-2 were test by the immunohistochemistry technic-SABC-staining in different periods of human fractured mandibular callus and in osseous tissue of normal angle of mandible.@*RESULT@#The TGF-beta1 and BMP-2 were expressed in callus of different periods but not in normal bone tissue. The expression of TGF-beta1 increased slowly during the first three weeks after fracture and reached its maximum in the third week, and then weakened gradually. The expression of BMP-2 increased gradually during the first two weeks after fracture and reached its maximum in the second week, then the expression weakened gradually.@*CONCLUSION@#(1) BMP-2 may be one of the factors promoting the repair of fracture. (2) TGF-beta1 could be another signal pathway in repairment of fracture. (3) There could exist some synergistic effects between TGF-beta1 and BMP-2 in the process of fracture healing.

ABSTRACT

Objective To study the possibility of ultr asonic examination in evaluation of cleft palate repair. Methods The length, open-close movement of soft palate and velopharyngeal insufficiency were observed by ultrasonography in submaxillary region pre-operation and post-operation in 7 patients. Results After operation- the length of soft palate was longer than that before operation, the mean extended length was 13.7- mm; the movement of soft palate was increased; the velopharyngeal insufficiency was improved. Conclusions The observation of soft palate structure pre-operation and post-operation by ultrasonic examination is an objective index for evaluating the cleft palate repair.
