ABSTRACT

Objective To compare classical and molecular typing methods for Streptococcus pneumoniae,in order to provide fasty and accurate method for classification.Methods Selected the 150 strains of Streptococcus pneumoniae which isolated from the First People’s Hospital of Zhaoqing from October 2013 to February 2015 for the research objects,using the classi-cal quellung classification method and molecular typing of multiple PCR method for classification.Compared the result and the rate of the classification,used the non parametric test of two independent samples for statistical analysis several higer rate of serotypes,and calculated the total coincidence rate,sensitivity and specificity.Results The typing rate of Streptococ-cus pneumonia capsular was 53.3%,and the main serum group were 19(32.7%),6(7.3%)and 23(4%)respectively.The serotype of 19F(23.3%),19A(2.7%),6C(2%)and 23F(2.7%)respectively by single factor serum.The typing rate of the multiple PCR method was 76.0%,and the main serum group were 19(41.3%),6(10.7%)and 23(10.7%).The serotype were mainly 19F(32.7%),19A(8.0%),6(10.7%)and 23F(9.3%).Used the 2 Independent Sample Test for statistical a-nalysis,compared with the serotypes 19F,19A and 23F(P>0.05).There was no statistically significant difference in the distribution of the types of the two methods.The total coincidence rate was 74.6%.To verify the classic quellung method by multiple PCR method,sensitivity was 70.2%,and specificity was 100%.That showed the capsular swelling method was less sensitivity than the molecular typing method.Conclusion Compared with the traditional serological typing techniques,mo-lecular typing technique have the advantages of fast,accurate,sensitive and high resolution,and become the classification method which can be rely on.

ABSTRACT

Aim To detect the possible ameliorative effects of APS on the airway inflammation and whether the effects are associated with inhibiting the NF-κB/MAPK signaling pathway in ovalbumin( OVA)-induced asthmatic rats. Methods Asthma was induced by OVA sensitization and challenge. The asthmatic rats in the APS group were treated with APS. Pulmonary in-flammation was assessed with hematoxylin and eosin staining and inflammatory cell counting in BALF. Ul-trastructural changes of type I pneumocyte were ob-served by electron microscopy. Levels of NF-κB p65, p-NF-κB p65, p-IκBα, ERK1/2, p-ERK1/2, JNK, p-JNK, p38 MAPK, p-p38 MAPK, IL-1β, IL-4, IL-5 , IL-6 and IL-13 were measured using ELISA to as-sess the activity of NF-κB/ MAPK signaling pathway. Results Compared to the normal group, OVA in-duced significantly pulmonary inflammation and ultra-structural changes of type I pneumocyte in the asthma group. Besides, OVA increased the activity of the NF-κB/MAPK signaling pathway and promoted the produc-tion of inflammatory cytokines IL-1β, IL-4 , IL-5 , IL-6 and IL-13 in the asthma group. If compared to the asthma group, APS markedly attenuated the pulmonary inflammation and type I pneumocyte damage, as well as inhibited the activity of the NF-κB/MAPK signaling pathway and decreased the production of inflammatory cytokines in the APS group. Conclusion The amelio-rative effects of APS on airway inflammation might be associated with the inhibition of the activity of the NF-κB/MAPK signaling pathway.

ABSTRACT

Objective To establish a serological diagnostic method about NSCLC by investigating the expres-sion of 3 tumor markers of the sulfate glycosaminoglyean fragments(SGF), TSGF and CEA in NSCLC patients' serum. Methods To assay the level of SGF,TSGF and CEA in 50 NSCLC patients with latex particleagglutiation test,colori-metry of biochemistry and CLIA, respectively. Compared the clinical value of different examination combination way in NSCLC diagnosis value. Results There was high difference value between the Ⅰ - Ⅱ stage and the Ⅲ time,the Ⅲ time and the Ⅳ time about the level of SGF(P <0.05) in NSCLC patients. The specificity between SGF,TSGF and CEA each single item and the SGF and TSGF combination examination were not significant(P > 0.05). The semitivity between the SGF and TSGF combination and the SGF,TSGF and CEA combination was not significant(P >0.05), its specificity had significant difference(P <0.05). Conclusions There are higher expression about the 3 tumor mark-ers in NSCLC patients' serum. In the different combination examination, SGF and TSGF combined has great clinical value than other combinations to diagnosis NSCLC.

Methods The serum levels of HCMV-DNA and HBV-DNA were determined by real-time fluorescent ration PCR, and the HCMV-IgG, IgM level were determined by ELISA in 162 serums from hepatitis patients HBsAg reported the masculine. Results The masculine rate of HCMV-DNA and HCMV-IgM, and the mean vahe of HCMV-DNA ration were much consilient: controls<hepatitis B sufferer<hepatocirrhosis<liver cancer. The masculine rate of HCMV-DNA and HCMV-IgM in hepatitis B HBV-DNA positive patients was higher in hepatitis B HBV-DNA negative patients(P<0.01). ABSTRACT

OBJECTIVE:To investigate the effects of the antihypertensive protein from Pheretima on blood pressure, angiotensinⅡ and angiotensin Ⅱ AT1 receptor in spontaneously hypertensive rats (SHR).METHODS: The dynamic change of blood pressure in SHR after singel intravenous injection of antihypertensive protein from Pheretima was observed. After intervention with antihypertensive protein from earthworm for 28 d,the levels of blood pressure, angiotensinⅡ and expression of angiotensin Ⅱ AT1 receptor in SHR were detected. RESULTS: Either single intravenous injecion or multiple dosing of the antihypertensive protein from Pheretima could significantly reduce the blood pressure in SHR (P0.05).The expression of angiotensin Ⅱ AT1 receptor in kidney of SHR model increased significanlty compared the normal control (wistar rats), but decreased after the intervention of the antihypertensive protein from Pheretima.CONCLUSION: The antihypertensive protein from Pheretima has antihypertensive effect on SHR. The mechanism may be related to the reduction of angiotensinⅡ level and lowering of the expression of a angiotensin Ⅱ AT1 receptor in kidney.
