ABSTRACT

Objective:The cluster analysis method was applied to evaluate the scientific research performance evaluation index system of departments in an obstetrics and gynecology hospital, and analyze the weaknesses of scientific research work of various departments, to provide a basis for the improvement of scientific research strength of departments.Methods:On the basis of the scientific research performance evaluation index system of departments in the obstetrics and gynecology hospital, the indicators and weights of the system were optimized through expert consultation, and the scientific research values from 2019 to 2021 were brought into the optimized scientific research performance evaluation index system to calculate the scientific research scores of each department, and then the cluster analysis method was applied to evaluate the index system.Results:Before and after the optimization of the scientific research performance evaluation index system of departments, the conformity with the discipline classification was 76.00% and 96.67% respectively ( P=0.039). In total of 30 departments were clustered into 4 categories: excellent (7), good (7), medium (4), and concerned (12). The average score of the total scientific research performance evaluation indicators of the 4 categories of departments was 15.022. The highest average score was for papers and monographs, and the lowest was for awards. Conclusions:This study applied the cluster analysis method to evaluate the scientific research performance evaluation index system of departments in the obstetrics and gynecology hospital and replaced quantitative indicators with quality indicators. It will optimize and improve the hospital hierarchical management methods, provide data support and classified guidance for the scientific research development of different categories of departments, and provide a reference basis for hospitals to formulate scientific research management policies such as achievement transformation, award, industry standard guidelines, etc.

ABSTRACT

Objective:To understand the cognition of scientific research integrity of the authors of scientific and technological papers in a grade A tertiary hospital, the necessity and relevant needs of carrying out scientific research integrity construction, and provide a reference for improving the construction of relevant scientific research integrity system in the hospital.Methods:A questionnaire was used to survey 746 people who had published scientific and technological papers in a grade A tertiary hospital from 2016 to 2021, and the influencing factors of scientific research integrity cognition and construction were analyzed via a generalized linear model.Results:The overall awareness rate of scientific research integrity cognition of scientific papers was 76.72%, and the overall necessity rate of scientific research integrity construction was 77.75%. Generalized linear model analysis showed that other degrees refer to a doctor's degree, associate senior title, intermediate title, refer to junior title, or no title were negatively correlated with the cognition of scientific research integrity ( P<0.05), other ages refer to ≤30 years old, middle-level and above positions refer to no position, postgraduate mentor refer to non-postgraduate mentor, senior title refer to junior title or no title, postgraduate, nursing, administrative and research personnel refer to medical technology and pharmacy personnel, and presiding projects refer to no presiding project experience were positively related to scientific research integrity cognition ( P<0.05). Bachelor′s degree and no degree refer to doctor′s degree, associate senior and intermediate titles refer to junior title or no title were negatively related to the necessity of scientific research integrity construction ( P<0.05), middle level and above positions refer to no position, postgraduate, nursing and research personnel refer to medical technology and pharmacy personnel, were positively related to the necessity of scientific research integrity construction ( P<0.05). Behaviors with less than 60% cognition of scientific research integrity included the ethical problems of artificial intelligence, the ethical problems of stem cells, the problems of human genetic resources, and the definition of unauthorized human living drug and medical technology experiments. Conclusions:The hospital will take publicity and education as the basis, system norms as the criteria, daily supervision as the starting point, evaluation system as the core, carry forward the spirit of scientists, improve and implement the construction of the hospital's scientific research integrity system and supervision system, strengthen the integrity management of the whole process of scientific research activities, and guide medical personnel to practice excellent scientific research style and style of study.

ABSTRACT

Objective:To explore the competence characters of hospital young scientists and build a competency model.Methods:The model characters were screened by literature review, behavioral event interview, expert consultation and questionnaire survey. The competency characters were extracted by exploratory factor analysis.Results:The competency characters model includes 5 factors and 34 characters: comprehensive accomplishment, practical ability, personal trait, professional skill and knowledge quality. The Cronbach's α coefficient of the model was 0.980 and the Cronbach's α coefficient of the 5 factors ranged from 0.832 to 0.964, with a split-half reliability of 0.922. The content validity index of the scale was 0.977, and the content validity index of each entry ranged from 0.857 to 1.000. The value of KMO was 0.944.Conclusions:The competency model of hospital young scientists has high stability, and provides reference for young scientists' ability and quality, which will provide the basis for optimizing the training system and training strategy of hospital scientists.

ABSTRACT

Objective:Delphi method and weighted average method are used to establish the evaluation index system of basic and clinical scientific research projects respectively, to provide support for screening high-quality projects, to avoid the disadvantages of " one size fits all" , and promote the transformation of project management from quantity and restatement to quality and results, in order to release the innovative vitality of talents and produce more high-quality results.Methods:Using the methods of literature review and expert interview, form the questionnaire of evaluation indicators for the conclusion of basic and clinical scientific research projects, then carry out two rounds of expert consultation with Delphi method, evaluate the authority of experts with authority coefficient, form the second round of questionnaire after analysis and discussion according to the results from the first round of expert consultation, screen and determine the evaluation indicators again in the same way. After two rounds of discussion, the evaluation index system of basic and clinical scientific research projects was developed, and the weighted average method and product method were used to calculate the weight and combination weight.Results:After two rounds of consultation, the evaluation index system for the conclusion of basic scientific research projects was finally established, with 2 first-class indicators, 12 second-class indicators and 33 third-class indicators, and the evaluation index system for the conclusion of clinical scientific research projects had 2 first-class indicators, 10 second-class indicators and 33 third-class indicators. In the second round, the authority coefficient of basic and clinical experts was 0.8, indicating that the authority of experts in this study was high. 29 basic and clinical projects concluded from 2018 to 2020 were selected to verify the index system.Conclusions:The index system formed in this study will be used to classify and evaluate the conclusion evaluation of basic scientific research projects and clinical scientific research projects. In the next step, the weight of the index system will be adjusted to make it more realistic and highlight high-quality achievements, guide the project leader to pay more attention to quality and results, and conduct follow-up evaluation at different stages.

ABSTRACT

Abnormal activation and secretion of pancreatic enzymes in pancreatic acinar cells is one of the important pathogeneses of acute pancreatitis (AP) and can directly damage the pancreatic tissue to accelerate disease progression and induce severe AP. At present, the drugs inhibiting the abnormal activation and secretion of pancreatic enzymes tend to have an unsatisfactory effect in clinical practice, and therefore, it is of great importance to search for new therapeutic targets. This article summarizes the pathological events of abnormal activation and secretion of pancreatic enzymes (cytoplasmic calcium overload, colocalization of lysosomes and zymogen granules, organelle injury, obstructed apical secretion of trypsin, and increased basal secretion of trypsin), collects the molecular mechanisms of related events, and discusses the role of abnormal activation and secretion of pancreatic enzymes in the early stage of AP, so as to provide ideas for the development of targeted drugs in the future.

ABSTRACT

As a prenatal testing for chromosomal abnormalities, non-invasive prenatal testing (NIPT) has been integrated into prenatal healthcare service. NIPT has shown a high sensitivity and specificity for screening fetal trisomies 13, 18 and 21, and has attained excellent clinical results. With the propagation of the NIPT screening, international organizations have issued guidelines and comments for its clinical utility with regular updating. China has also developed guidelines for NIPT in 2016. NIPT guidelines in various countries have provided valuable guidance for its target diseases and suitable patient groups, but there has been few research data on its clinical application for special groups of patients. Based on the guidelines and comments of various professional bodies and published data on the clinical utility of NIPT, in addition with consideration of the conditions in China, clinical utility of NIPT for particular groups of pregnant women, including those with advanced maternal age, obesity, twin pregnancy and fetal ultrasonographic anomalies, are reviewed. The value of genetic counseling for NIPT is also emphasized, which is critical for the clinical application of NIPT.

ABSTRACT

OBJECTIVE@#To evaluate the efficacy of non-invasive prenatal testing (NIPT) in the prenatal screening and its role in the system of prenatal diagnosis.@*METHODS@#A total of 22 649 singleton pregnant women who were registered and finally delivered or had induced labor at Beijing Obstetrics and Gynecology Hospital of Capital Medical University were enrolled. The routes of prenatal screening were analyzed to evaluate the efficacy of prenatal screening. Meanwhile, 9268 pregnant women who underwent invasive prenatal diagnosis procedure were enrolled. The indications and results of prenatal diagnosis were analyzed to evaluate the effectiveness of prenatal screening.@*RESULTS@#60.24% of singleton pregnant women have opted for Down syndrome screening, and their age was mainly under 35. The proportion of women opted for NIPT was 34.74%, and were mainly between 35 and 39. The overall diagnostic rate of trisomy 21, 18 and 13 trisomy for those with high risk by NIPT was 0.89%, which yielded a positive predictive value of 75.71%. For those with moderate risk by serum screening, 0.30% was predicted with a high risk by NIPT. Among women undergoing prenatal diagnosis, 63.04% and 21.22% had the indication of advanced age or high risk by serum screening, and the positive predictive values were 5.1% and 5.13%, respectively. By contrast, 2.30% of women undergoing prenatal diagnosis had a high risk by NIPT, which yielded a positive predictive value of 54.46%.@*CONCLUSION@#With the change of the age composition of pregnant women and increase in the complexity of pregnancy in China, to build a prenatal screening system based on NIPT will be helpful to improve the efficiency of the current system of prenatal screening and diagnosis.

ABSTRACT

Objective:Data envelopment analysis (DEA) was applied to evaluate the evaluation index system of scientific research performance of hospital departments.Methods:Based on the three-level index assignment of the constructed evaluation index system of scientific research performance of hospital departments, the relevant data of departments are calculated. Scientific research projects, talent team and scientific research platform are taken as input indexes, papers, awards, patents, published monographs/papers, professional standards and guidelines, as well as achievements transformation are taken as output indexes, and departments are taken as evaluation units. Data envelopment analysis software is used to evaluate departments Results of scientific research performance evaluation, redundant input and insufficient output.Results:The average comprehensive technical efficiency of 25 departments is 0.404 8, the average pure technical efficiency is 0.837 1, and the average scale efficiency is 0.428 3. Among them, 7 departments are generally efficiency, 17 departments obtained pure technical efficiency, 7 departments obtained scale efficiency.Conclusions:The analysis results can provide a reference for the scientific research development of the department. On the basis of building a reasonable talent echelon and assure the sufficient scientific research time, the hospital further optimizes and improves the scientific research performance evaluation index system and hierarchical management method of the Department.

ABSTRACT

Objective:To analyze the current situation of scientific research in hospitals in China based on the indicators and results of Science and Technology Evaluation Metrics in Chinese hospitals, and propose possible countermeasures for scientific research development.Methods:The external and internal environment of scientific research in a Grade-A tertiary obstetrics and gynecology hospital was discussed through the results of Science and Technology Evaluation Metrics and situation analysis.Results:The external opportunity of the hospital is that the national and municipal government lead the development direction of science and technology, the hospital leaders attach great importance to the development direction of the hospital and have a clear direction for the development of the hospital; the challenge is that the new generation of information technology puts forward the requirements of cross integration of scientific research work, the flow of high-level talents is accelerated, and there is a significant gap with the similar hospitals in the field of obstetrics and gynecology. The internal advantages are embodied in the following points: the advantages of specialties are obvious, the output of scientific research results has increased year by year after the implementation of the scientific research management of hospital departments, the training and introduction of talents are equally important; the disadvantages are reflected in the following points: lack of basic research and scientific research platforms, lack of high-end scientific and clinical research talents, insufficient academic influence, lack of national-level scientific research projects, insufficient high-level scientific research results, and low achievement conversion rate.Conclusions:The hospital should improve performance assessment, build research platform and achievement transformation platform, strengthen interdisciplinary integration and personnel training, and promote the fine management of high-level achievements, so as to improve the scientific and technological output, academic influence and scientific and technological conditions of the hospital.

ABSTRACT

Objective:This article aims to sort out the current situation and problems of horizontal scientific research projects in a tertiary hospital, so as to improve the management of horizontal scientific research projects, and to mobilize the enthusiasm of researchers to undertake horizontal scientific research projects, and give full play to their characteristics of interdisciplinary integration, transfer and transformation of scientific and technological achievements.Methods:A total number of 129 horizontal projects undertaken by Beijing Obstetrics and Gynecology Hospital, Capital Medical University from 2010 to 2019 were reviewed.The comparison of management policies and risk prevention of horizontal scientific research projects before and after 2017 was also analyzed.The existing problems were analyzed and relevant management suggestions were put forward.Results:From 2017 to 2019 the hospital undertook 18.7 horizontal research projects per year, which was 1.8 times of that 2010 to 2016.The hospital strengthened the cooperation with foundations, companies and foreign countries, carried out substantive scientific research, standardized the budget, leased or borrowed equipment, and built the joint laboratories by medical research and enterprise, and carried out risk prevention in human genetic resources and biological security.At present, there are still some problems, such as the lack of relevant policy basis for management, the management of project approval and conclusion is not in place, the conversion rate of achievements is low, and the horizontal scientific research projects of platform construction are need to be constructed.Conclusions:Our hospital will improve the relevant system of horizontal scientific research projects management, incorporate the horizontal scientific research projects into the position title evaluation, performance evaluation and reward, build various platforms through multiple channels, promote the interdisciplinary integration, and give full play to the characteristics of horizontal scientific research projects in promoting the transfer and transformation of scientific and technological achievements, so as to promote the discipline construction and scientific and technological innovation of the hospital, and accelerate the transformation of the hospital into innovative and research-oriented hospital.

ABSTRACT

Acute pancreatitis in pregnancy is one of the rare and serious complications in pregnancy and has sudden onset, rapid disease progression, and difficulty in diagnosis, and thus it greatly threatens the health of mother and infant. Accurate judgement of the etiology of acute pancreatitis in pregnancy and confirmed diagnosis as early as possible are the key to successful clinical treatment. This article summarizes the recent advances in the early diagnosis of this disease from the aspects of etiology of acute pancreatitis in pregnancy, methods for early diagnosis and examination, and differential diagnosis of this disease from other diseases.

ABSTRACT

OBJECTIVE:To provide reference for improving safe drug use in pregnant women. METHODS :Retrieved from as domestic and foreign literature retrieval platforms as CNKI and Web of Science ,the literatures about drug use in pregnancy period were collected and analyzed by using the literature analysis function provided by the retrieval platform and CiteSpace 5.5R2 software. The collected data of annual volume ,author,organization,keyword co-occurrence and literature co-citation analysis of domestic and foreign literatures are visualized ,and the research focus was analyzed. RESULTS :A total of 38 946 foreign literatures and 698 domestic literatures were included ,and domestic literatures were less than foreign literatures ;the literature volume on pregnancy drug use were increasing year by year at abroad,but the volume of domestic literatures fluctuates greatly and increased slowly. Foreign institutions and scholars worked closely together ,and built a close cooperation network ,especially in the United States. However ,the author team of domestic research was relatively scattered as a whole ,and the cooperation between researchers was less ,so a large cooperation team had not yet been formed. The most frequently cited literatures in the world were randomized controlled studies and large-scale cohort studies,the keyword “Risk”appears most frequently ,indicating that it focuses on the effects of pregnancy medication on pregnancy outcomes and offspring. The effects of antiepileptic drugs ,antidepressants and opioids used in pregnancy on pregnancy outcomes and child development were currently a research hotspot abroad. However ,there was a lack of large-scale sample and high-quality clinical research in China ,high-frequently keywords in domestic literatures indude “fetus”“hypertension in pregnancy ”etc.,which focused on the treatment of combined diseases in pregnancy and paid little attention to the long-term effects of medication in pregnancy. CONCLUSIONS:More and more attention had been paid to the study of drug use in pregnancy period. Domestic research needs to seize the international cooperation communication ,learns from the excellent experience of foreign research team ,and establishes domestic specific pregnancy medication research team. Relevant departments should introduce measures as soon as possible ,support medical institutions and drug production and developmententerprises to do a good job in the registration of pregnancy medication,establish the database of pregnancy medication inChina as soon as possible ,and improve the medication safety of pregnant patients.

ABSTRACT

Objective:To establish a scientific research performance evaluation index system for departments by applying Delphi method and analytic hierarchy process.Methods:A questionnaire composed of 10 first-level indicators, 36 second-level indicators and 95 third-level indicators was developed according to literature review and expert interview. Then two rounds of expert consultation were conducted by adopting Delphi method, the authority of experts was evaluated by the authority coefficient. Based on the analysis and discussion of the findings of the first round of expert consultation, an updated version questionnaire was developed, the evaluation indicators were re-selected and determined by the same way. After two rounds of discussions, the scientific research performance evaluation index system of the hospital department was finally developed. Finally, the analytic hierarchy process and product method were used to calculate the weights and combined weights, respectively.Results:After two rounds of consultation, 9 first-level indicators, 28 second-level indicators and 67 third-level indicators were finally established in the scientific research performance evaluation index system of hospital departments. The expert authority is 0.79, which shows that this research has a higher authority. From the standpoint of significance test, the P values are all less than 0.01, indicating that the consulting experts meet the requirements for the consistency of the index scores, and the results are reliable. Conclusions:An effective index system for scientific research performance evaluation of departments has been established in this research, however, it still needs further validation in practice.. In the future, it will need to be combined with grading standards and methods, and subjective assessment by experts to test the scientificity and feasibility of the evaluation index system to assure continuing improvement.

ABSTRACT

Objective:To understand the cognition of scientific research integrity of applicants of the National Natural Science Foundation of China (NSFC) and the necessity of developing scientific research integrity construction.Methods:Questionnaire survey was used to collect information from 121 applicants who had applied for the NSFC from 2016 to 2020, collected data were statistically analyzed.Results:The cognitive average score of scientific research integrity knowledge of NSFC applicants was (287.03±49.45) points, and the overall awareness rate was 80.44%. The average score of the necessity of scientific research integrity construction work was (20.71±3.59) points, and the overall necessity rate 87.27%; educational background, position title, and experience as principle investigator are factors that affect the perception of integrity, while educational background, position, job category, and research management experience of a project are factors that affect the necessity of scientific research integrity construction. Behaviors with less than 70% of scientific research integrity behaviors include stem cell ethical issues, genetic technology ethical issues, human genetic resources issues, conditions for secondary publication, and use of official cards for settlement.Conclusions:The hospital should strengthen the scientific research integrity knowledge training of NSFC and other project applicants. On the basis of giving full play to the supervision role of relying units, the hospital will improve the relevant system of scientific research integrity, improve the construction of scientific research integrity at the same time, carry out classified evaluation, and put an end to scientific research misconduct from the source.

Objective:To lay a foundation for the continuous improvement of the birth cohort management in the future, and provides references for other hospitals in establishing the birth cohort through sharing the practice, experience and reflection of the establishment and management of the birth cohort of 500, 000 pregnant women.Methods:The present situation and data of the establishment and management of the birth cohort of 500, 000 pregnant women were summarized to describing the practice, experience and reflection.Results:The specific work regarding the establishment and management of the cohort include personnel investment, standards development, implementation process and quality control. Three phases of the management experiences were summarized which including explorative process implementation; standardize project implementation, and quality improvement along with the fast development of the birth cohort. Generally, the attention of leadership was one of the most important factors for the establishment and management of birth cohort in each research site.Conclusions:It is important that comprehensive considerations of factors including the personnel, standards, practical management process, quality control and the attention of leadership plays important role in improving the quality and efficiency of birth cohort establishment and management.

ABSTRACT

Acute pancreatitis (AP) is an exocrine inflammatory disease of the pancreas, the pathophysiological mechanism of AP remains unclear. Autophagy is an important pathway of metabolic degradation in cells, and autophagy impairment is closely associated with the development and progression of many diseases. Recent studies have found that the process of autophagy is abnormal in AP, with impaired fusion of autophagosome and lysosome and a significant reduction in the degradation function of autolysosome. Abnormal activation of pancreatic enzymes and inflammatory response is the key event in the initiation of AP, and abnormal autophagy is closely associated with these two events. The study of the upstream and downstream mechanisms of autophagy dysfunction in AP may help to find new molecular targets and strategies for the treatment of AP.

Objective@#To investigate the clinicopathological characteristics and diagnosis of ovarian Brenner tumors.@*Methods@#Forty-seven cases of ovarian Brenner tumors were enrolled from January 2012 to May 2018 at Beijing Obstetrics and Gynecology Hospital, Capital Medical University. Clinical data, imaging examination, histopathological characteristics and immunohistochemical phenotype were analyzed.@*Results@#The age of the patients ranged from 30-73 years and the mean age was 55 years. Thirty-nine patients (83.0%) were postmenopausal. Forty cases (85.1%) of the Brenner tumors were benign, five (10.6%) borderline and two (4.3%) malignant. Usual tumor markers of ovarian carcinoma, including CA199 and CA125 were normal or mild elevated in the 47 cases. Imaging before surgery was not specific to Brenner tumors. Microscopically, benign Brenner tumors were composed of nests of bland, transitional-type cells within a fibromatous stroma. In our 5 cases of borderline Brenner tumors, mildly atypical transitional-type cells were projected into the cyst lumens and lack of stromal invasion. In 2 cases of malignant Brenner tumors, different degrees of nuclear atypial transitional-type cells exhibited stromal invasion. Immunohistochemical stains for CK7, GATA3, p63 and CK5/6 were positive in all cases. Ki-67 was less than 5% in Brenner tumors, and up to 20%-30% in malignant Brenner tumors.@*Conclusion@#Brenner tumors are mostly seen in postmenopausal patients and are usually benign. Imaging examination and usual ovarian tumor markers do not provide diagnostic value. Diagnosis and classification of Brenner tumors depend on histopathological evaluation.

ABSTRACT

Objective@#To investigate the value of short tandem repeat (STR) genotyping in the diagnostic workup of molar and non-molar gestations with correlation of histological characteristics.@*Methods@#Six hundred and fifty-six cases were selected based on clinically suspected hydropic abortion and/or molar pregnancy from July 2015 to September 2017 at Beijing Obstetrics and Gynecology Hospital. DNA was extracted from dissected chorionic villi and paired maternal endometrial FFPE tissue samples by Simplex OUP™ FFPE DNA Tissue Kit. STR genotyping was performed by PowerPlex 16 HS system.@*Results@#DNA genotyping was informative in 649 of 656 cases, leading to identification of 215 hydatidiform mole gestations and 434 non-molar gestations. Most of non-molar gestations (375 cases, 86.4%) were diploid hydropic abortion. Various trisomy syndromes were found (53 cases, 12.2%), including trisomy 2, 3, 4, 7, 8, 13, 16 and 21. Only 2(0.5%) digynic triploid gestations were detected. Moreover, 4 cases (0.9%) of uniparental disomies (homologous or heterologous) were found. There were 196 cases with histologic diagnostic suspicious of hydatidiform moles were accurate sub-classified. Among them, 59 cases hydatidiform moles were under-diagnosed as diploid hydropic abortions, and 28 cases diploid hydropic abortions were over-diagnosed as hydatidiform moles.Compared with partial moles(PHM), there were no specific histomorphological features between the various types of non-molar gestations and partial moles for definitive diagnostic separation. There was no significant difference in the expression of p57kip2 among PHM, trisomy and diploid hydropic abortions group (P=0.247).@*Conclusions@#STR genotyping can distinguish non-molar gestations from early hydatidiform moles, and efficiently avoid misdiagnosis based only on histological evaluation. Therefore, using STR genotyping, not only can the overdiagnosis of non-molar pregnancy be avoided, but also individualized management can be offered to patients including monitoring of serum hCG.

ABSTRACT

Objective To observe the protective mechanism of angiotensin-converting enzyme (ACE) 2 on lipopolysaccharide (LPS)-induced hepatocyte inflammation by inhibiting P38 mitogen activated protein kinase (MAPK)/activator protein (AP)-1 pathway.Methods Rat liver BRL cells after immortalized culture were randomly divided into five kinds of groups:control group,LPS (10 μg/ mL) group,LPS + recombinant(r) ACE2 (5,10,20 ng/mL rACE2 for 30 min before cells stimulated with LPS) groups,LPS+ACE2 inhibitor MLN-4760 (10-7,10-6,105 mmol/L MLN-4760 for 30 min before cells stimulated with LPS) groups,LPS + rACE2 (20 ng/mL rACE2 for 30 min before cells stimulated with LPS) + P38MAPK inhibitor SB203580 (10-5 mmol/L SB203580 for 30 min before cells stimulated with LPS) groups.The changes in protein levels of ACE2,P38MAPK,p-P38MAPK and AP-1 were detected by western blot after LPS exposure for 6,12 and 24 hours,and the mRNA expressions of P38MAPK,AP-1 and tumor necrosis factor-α were quantified by real-time RT-PCR.Results Compared with control group,the protein levels of ACE2,P38MAPK and AP-1 were up-regulated in LPS-induced hepatic cells in a time-dependent manner,peaking at 12 h after LPS stimulation (all P<0.05).Compared with LPS group,the mRNA expressions ofAP-l,P38MAPK,p-P38MAPK and tumor necrosis factor-oα decreased significantly in rACE2 group (all P<0.05).The dose of 20 ng/mL rACE2 had the best inhibitory effects on the mRNA expression of AP-1 (0.12±0.002 vs.0.04±0.005,P<0.01),P38MAPK (0.17±0.02 vs.0.02±0.002,P<0.01) and p-P38MAPK(0.29±0.01 vs.0.02±0.01,P<0.01)compared with LPS group.The mRNA expressions of AP-1,P38MAPK and p-P38MAPK increased in MLN-4760 group in a concentration dependent manner (all P<0.05).Furthermore,the inhibitory effects of rACE2 on AP-1 and tumor necrosis factor-α levels were cancelled by SB203580.Conclusion The rACE2 can alleviate the LPS-induced hepatocyte injury by down regulating the P38MAPK/AP-1 signaling pathway.
