ABSTRACT

Objective:To establish the best processing conditions of fried with earth for Atractylodes macrocephala.Methods:The contents of water extract , ethanol extract and atractylenolide Ⅲwere used as the evaluation indices , the temperature of pot room , pro-cessing time and the amount of terra flava usta were chosen as the influencing factors , and the processing conditions of fried with earth for Atractylodes macrocephala was optimized by L9(34) orthogonal design.Results:The best conditions of fried with earth processing technology for Atractylodes macrocephala were as follows:the pot temperature of 220℃-250℃, the processing time of 6min and the a-mount of terra flava usta of 30%.Conclusion: The processing technology optimized by orthogonal design can ensure the quality of products .

ABSTRACT

Objective To explore an UPLC method for simultaneous content determination of the five nucleosides (cytidine, uridine, adenine, thymidine and adenosine) in Hedyotis diffusa and its adulterants; To compare the content differences.Methods The analysis was performed on a BEH C18 column (2.1 mm×50 mm, 1.7 μm) by UPLC eluted with acetonitrile and water in gradient mode. The flow rate was 0.5 mL/min; the detection wavelength was set at 254 nm; the column temperature was set at 30℃.Results Five nucleosides have good linear relationship, precision, stability, and repeatability according to the requirements of the methodology determination. The recoveries were among 98.7%–101.5%. Five nucleosides in Hedyotis diffusa and its adulterants from different areas were determined by the UPLC method.Conclusion The method is certified to be simple, rapid, accurate and reliable, which can be used for the determination of nucleosides in Hedyotis diffusa and its adulterants.

ABSTRACT

Objective:To develop an HPLC determination method for ferulic acid and caffeic acid in Atractylodes macrocephala Koidz fromdifferentorigins.Methods:ThecolumnwasAgilent-C18(150mm×4.6mm,5μm),themobilephasewas0.2% acetonitrile-aceto-nitrile(70 ∶30),the UV detection was performed at 320 nm,the flow rate was 1. 0 ml·min-1, and the drift tube temperature was set at 35℃. Results:The calibration curve was linear within the range of 0. 040-0. 500μg for ferulic acid(r=0. 999 8)and 0. 060-0. 750 μg for caffeic acid(r=0.999 8). The average recovery of ferulic acid and caffeic acid was 98.10%(RSD =1.08%)and 98.66%(RSD =0. 61%)(n=6), respectively. Conclusion: The method is simple and accurate. It is suitable for the determination of ferulic acid and caffeic acid in Atractylodes macrocephala Koidz.

ABSTRACT

Objective To establish an HPLC method for concentration determination of emodin in plasma and colon tissues after gavage withHuchang Qingdu Pellets;To explore pharmacokinetic properties and colon tissue distribution ofHuchang Qingdu Pellets in rats.Methods Wistar rats were randomly divided into Pellets group and ordinary capsule group. Blood was taken after gavage withHuchang QingduPellets or capsule content in given time points (1, 2, 3, 4, 5, 6, 8, 12, 24 h) from rat venous sinus and analyzed. Rats were put to death, and gastric and colon tissues were isolated and extracted for analysis. Chromatographic conditions:Phenomenex C18 column (4.6 mm × 150 mm, 10μm), the mobile phase of methanol-0.1% acetic acid solution (80∶20), flow rate of 1 mL/min, the detection wavelength of 254 nm.Results Emodin was in good linear range of 0.102-20.40μg/mL. The regression equations were:plasma solutionA=76 825C + 3567.8,r=0.999 4 (n=6);intestinal tissue solutionA=75 931C + 3384.2,r=0.999 0 (n=6). Day-to-day precision and within-day precision were less than 4%. The recovery rate was more than 90%. Pharmacokinetic parameters of T1/2 and Tmax of emodin in rat blood and intestinal tissues increased compared with the ordinary capsule group;Cmax and AUC0-24 were obviously improved compared with the ordinary capsule group.ConclusionHuchang Qingdu Pellets do have controlled release and colon targeting effect.
