ABSTRACT

Objective To detect the expression of a disintegrin and metalloproteinase with thrombospondin motifs 4 (ADAMTS4),Versican,and Aggrecan in hearts of myocardial infarction rats.Methods Male Wistar rats were chosen to make myocardial infarction model.Fifteen rats were randomly assigned into the myocardial infarction group and five rats into the sham-operation group.Rats were killed 3 d after surgery and the left ventricles were taken to produce frozen section.ADAMTS4,Versican,and Aggrecan protein expressions were detected by imnunohistochemistry method.Results ADAMTS4 expressed in myocardial cells at marginal zone of the infarcted area.ADAMTS4 expressed weakly while Versican expressed strongly in capillary endothelial cells of marginal zone of the infarcted area.Aggrecan showed no expression in the cardiac tissue and vascular endothelium of both myocardial infarction group and sham-operation group.Conclusions ADAMTS4 is involved in the inflammation after myocardial infarction.Degradation of ADAMTS4 to Versican exists in the vessels after myocardial infarction.Aggrecan is not involved in the pathophysiological process of myocardial infarction.

ABSTRACT

BACKGROUND:An animal model of myocardial infarction is of great significance for the study of the pathogenesis, pathophysiology and evaluation of treatment methods of human coronary artery disease. Many researchers aim to improve survival rate and the success rate of model induction. OBJECTIVE:To improve the method of making a rat model of myocardial infarction. METHODS:One hundred Wistar rats were divided into model group (n=80) and sham-operated group (n=20). The rats were intraperitoneal y anesthetized with 3%sodium pentobarbital. Tracheostomy was operated. An automatic breathing apparatus was utilized for assisted breathing. A left anterior thoracotomy was performed at the fourth intercostal space. The left anterior descending coronary artery was ligated. Sham-operated rats underwent similar surgery but without coronary artery ligation. The thoracic cavity was closed layer by layer. The breathing apparatus was removed and tracheal intubation was also removed. To prevent suffocation, the trachea and the neck incision were not closed. RESULTS AND CONCLUSION:Heart morphology and hematoxylin-eosin staining demonstrated that the success rate of establishing myocardial infarction model was 98.6%. The survival rate of model group at 3 weeks after surgery was 88.75%. Total y nine rats died, including seven dead rats during operation and within 24 hours after operation, and two dead rats in later stage. The survival rate of the sham-operated group was 100%. We modified the method of establishing a rat model of myocardial infarction and improved the operation success rate and survival rate of rats, such as anesthesia method, longitudinal incision during tracheotomy, without closing the trachea after operation, and the method of opening and closing the chest.

ABSTRACT

Objective To compare the incidence of MACE and predictors in patients with unprotected left main coronary artery disease (ULMCAD) after percutaneous coronary intervention (PCI) or coronary artery bypass grafting (CABG). To establish a risk stratification to guide treatment and prognosis of patients with ULMCAD who underwent PCI. Methods The incidence of MACE in 201 patients with ULMCAD who underwent PCI or CABG was compared, retrospectively. The predictors of MACE of the two groups were obtained by logistic regression analyses. The predictors of MACE of the PCI group were assigned and stratified. The validity of the risk stratification on the prediction of MACE was verified in a new group of 126 patients with ULMCAD who underwent PCI. Results The incidence of MACE of PCI and CABG group was 16% and 11.9%, respectively. Logistic regression analyses showed that LVEF, diabetes mellitus and Syntax score were related to MACE in PCI group, while age, LVEF and renal function insufficiency were related to MACE in CABG group. The verification results showed a good predictive value of the risk stratification based on LVEF, diabetes mellitus and Syntax score on the incidence of MACE. Conclusions PCI treatment in patients with ULMCAD is feasible and has a good long-term outcome. The risk stratification in accordance with LVEF, diabetes mellitus and Syntax score has good predictive value on the incidence of MACE.

ABSTRACT

Objective Reverse transcription-polymerase chain reaction (RT-PCR) is widely used in gene expression analysis.Selection of proper housekeeping gene is very important.Rat model of myocardial infarction is a major animal model of myocardial infarction.Few reports are found about the selection of rat housekeeping gene after myocardial infarction.This study was aimed to explore the stability of the housekeeping genes expression in the rat model of myocardial infarction.Methods Myocardial infarction models were made using the anterior descending coronary artery ligation method.Combining with practical work,analysis and selection were made of four widely used standard housekeeping genes:glyceraldehydes-3-phosphate dehydrogenase (GAPDH),ribosomal protein L13A (RPL13A),beta-actin (ACTB) and acidic ribosomal phosphoprotein P0 (ARBP).The expressions in the rat heart were compared using real-time fluorescent RT-PCR and analysis was carried out to figure out which was the most suitable housekeeping gene for study of gene expression in rats after myocardial infarction using GeNorm program.Results The M values of RPL13A,GAPDH,ARBP and ACTB gene were 0.812,0.721,0.812 and 1.2 respectively.Conclusion GAPDH and ARBP are the most stable genes for the rat myocardial infarction model.
