ABSTRACT

ObjectiveProteomics was used to investigate the protein differences between porcine cardiac blood(PCB) and porcine blood(PB) from Menghe medical school and to compare the effects of both on the microglial inflammation of Salviae Miltiorrhizae Radix et Rhizoma(DS). MethodNanoliquid chromatography-quadrupole orbitrap mass spectrometry(nLC-MS/MS) and bioinformatics were utilized to compare the proteomic differences of PCB and PB in simulated gastrointestinal digestion. Furthermore, Western blot was used to verify the contents of some shared proteins and differential proteins identified in PCB and PB. In addition, BV2 neuroinflammation model constructed by corticosterone(CORT) and lipopolysaccharide(LPS) was applied to detect the intervention effects of PCB and PB on the levels of tumor necrosis factor(TNF)-α and interleukin(IL)-6 in BV2 inflammatory cells of DS. ResultA total of 69 common proteins and 68 differential proteins were identified in PCB and PB, among which the common proteins included transferrin(Tf) with brain-targeting effect, and the differential proteins in the two were 41 and 27, respectively. Western blot validation showed that the difference in the content of the same protein Tf between PCB and PB was not statistically significant, while the difference in the contents of the specific proteins of creatine kinase M and heart-type fatty acid binding protein(H-FABP) were statistically significant(P<0.05). Moreover, in vitro experimental studies revealed that compared with the same concentration of DS group, in addition to the 100 mg·L-1 PB-DS group, PCB-DS and PB-DS groups could significantly inhibit the levels of TNF-α and IL-6 in BV2 inflammatory cells(P<0.05, P<0.01), and PCB-DS group had more significant anti-inflammatory effect than PB-DS group with the same concentration(P<0.05, P<0.01). ConclusionBoth of PCB and PB can enhance the inhibitory effect of DS on the release of inflammatory factors, thus playing a neuroprotective role, and PCB promotes DS inhibition more significantly, which may be due to the existence of the two involved in energy metabolism-related differential proteins, which can lay a foundation for revealing the scientific connotation of the processing of PCB-DS and PB-DS.

ABSTRACT

ObjectiveTo evaluate the methodological quality of systematic review / Meta analysis (SR/MA) of intervention randomized controlled trial (RCT) published in the Sichuan Mental Health. MethodsThe literature databases such as Wanfang Data, China National Knowledge Infrastructure (CNKI), VIP Database for Chinese Technical Periodical (VIP) and Chinese Biomedical Literature Database (CBM) were searched for the SRs/MAs of intervention RCTs published in the Sichuan Mental Health from the initial issue to the issue published on 31 June 2021. Then the methodological quality of eligible SRs/MAs were assessed using A Measurement Tool to Assess systematic Reviews 2 (AMSTAR 2). ResultsThe literature search yielded 24 full-text articles, and the mean AMSTAR 2 score of the included SRs/MAs was (5.21±3.63) with a range from 1 to 11. The total AMSTAR 2 score for SRs/MAs showed difference in terms of the publication date prior to or later than the publication of AMSTAR-2 tool (t=-5.499), number of authors ≤ 2 or ≥ 3 (t=-6.736), with or without funding support (t=3.329) and author unit nature (F=7.827), with statistical significance (P<0.01). All selected studies had deficiencies on explicit statement of a priori design and registrations, list of excluded studies and reasons for exclusion, sources of funding for the research in systematic review, potential conflicts of interest statement, and funding information of the systematic review. ConclusionThe methodological quality of SRs/MAs of intervention RCTs published in the Sichuan Mental Health varies widely, After the release of AMSTAR 2, the methodological quality has improved, but the report still needs to be further standardized to provide high-quality evidence-based evidence.

ABSTRACT

Objective To study the effects of endoplasmic reticulum stress on liver damage in young rats fed with high-fat diet. Methods We divided 48 male weaned young rats randomly into high-fat diet group and control group, which were separately fed with high-fat diet and normal diet. After feeding 8,12 and 1 6 weeks, the body weight and visceral fat of the rats were measured. The serum liver function was measured. The morphology of livers was observed by HE and transmission electron microscopy. The mRNA expressions of ATF6 and GRP78 in hepatocytes were measured with RT-PCR.Results ① The body weight and visceral fat weight of rats in high-fat diet group increased compared with those in control group (P 0.05);alanine aminotransferase at week 1 6 was increased significantly compared with that in controls (P < 0.05 ).③ Liver cells in high-fat diet group had steatosis at week 8 and the steatosis became more serious between week 12 and week 1 6.④ In high-fat diet group at week 8 there were a large number of lipid droplets in the cytoplasm, and the cell structure was close to that of normal cells;rough endoplasmic reticulum was nearly normal and the ribosome was visible. At week 12 and week 1 6,besides a large number of lipid droplets,we could also see that some substances with line-like structure deposited in rough endoplasmic reticulum pool.⑤ The expressions of ATF6 and GRP78 mRNA in hepatocytes in high-fat group at weeks 8, 12 and 1 6 were significantly increased compared with those in control group (P <0.05). Conclusion High-fat diet in infants can cause visceral fat accumulation, fatty degeneration of hepatocytes and liver injury.ATF6-mediated endoplasmic reticulum may be closely related to the liver injury which results from highfat diet.

ABSTRACT

Sugarcane bagasse modified by polyethylenimine (PEI) and glutaraldehyde (GA) was used as a carrier to immobilize Clostridium acetobutylicum XY16 in the process of butanol production. The effects of chemically modified sugarcane bagasse on batch and repeat-batch fermentations were investigated. Batch fermentation was conducted with an addition of 10 g/L modified sugarcane bagasse and 60 g/L glucose, resulting in a high solvent concentration of 21.67 g/L and productivity of 0.60 g/(L x h) with the treatment of 4 g/L PEI and 1 g/L GA. Compared to the fermentations by free cells and immobilized cells on unmodified sugarcane bagasse, the productivity increased 130.8% and 66.7%, respectively. The fibrous-bed bioreactor also maintained a stable butanol production during repeat-batch fermentations, achieving a maximum productivity of 0.83 g/(L x h) with a high yield of 0.42 g/g.

ABSTRACT

Objective To report the clinical effect of microsurgical salpingostomy Methods Follow up 116 patients who used microsurgical techniques and analyzed its relevant factors, such as age, sterilization time limit, the length of the reconstructed fallopian tube, microsurgical techniques, and the site of salpingitis. Results Pursuit 116 cases, the re-pregnant rate was 86.2%(110/116). Conclusions The success of microsurgical salpingostomy depend on several factors including the length of the reconstructed fallopian tube, microsurgical techniques and the site of salpingitis.

ABSTRACT

The hospital the authors work with succeeded in achieving three sublimations of the idea of hospital management control in the course of hospital management control innovation, viz. sublimations from one-sided development concept to scientific development concept, from people-oriented management of human resources to ability-oriented management of human resources, and from emphasizing comprehensive strength to stressing developmental ability in hospital accreditation.
