ABSTRACT

Depressive disorder ranks as a major bur-den of disease worldwide,yet the current antidepressant medications are limited by frequent non-responsiveness and significant side effects.The lateral septum(LS)is thought to control of depression,however,the cellular and circuit substrates are largely unknown.Here,we identified a subpopulation of LS GABAergic adenosine A2A receptors(A2AR)-positive neurons mediating depres-sive symptoms via direct projects to the lateral habenula(LHb)and the dorsomedial hypothalamus(DMH).Activa-tion of A2AR in the LS augmented the spiking frequency of A2AR-positive neurons leading to a decreased activation of surrounding neurons and the bi-directional manipula-tion of LS-A2AR activity demonstrated that LS-A2ARs are necessary and sufficient to trigger depressive pheno-types.Thus,the optogenetic modulation(stimulation or inhibition)of LS-A2AR-positive neuronal activity or LS-A2AR-positive neurons projection terminals to the LHb or DMH,phenocopied depressive behaviors.Moreover,A2AR are upregulated in the LS in two male mouse mod-els of repeated stress-induced depression.This identifica-tion that aberrantly increased A2AR signaling in the LS is a critical upstream regulator of repeated stress-induced depressive-like behaviors provides a neurophysiological and circuit-based justification of the antidepressant poten-tial of A2AR antagonists,prompting their clinical transla-tion.

Abstract Croton argyrophylloides Muell. Arg., from the Euphorbiaceae family, popularly known as marmeleiro prateado or sacatinga, is a plant from the Caatinga biome commonly found in Brazils northeastern region. The present study aimed to evaluate the antioxidant activity of the species. The phytochemical study was performed through qualitative analysis of chemical constituents and quantitative determination of the total phenol content through the Folin-Ciocalteu test. The qualitative and quantitative antioxidant tests were performed using the DPPH method (2.2 diphenyl-1-picryl hydrazil) and ferric reducing antioxidant power (FRAP). The minimum inhibitory concentration (MIC) was determined by microdilution in 96-well plates. The ethanolic extract of the leaves of C. argyrophylloides manifested antioxidant action in the quantitative DPPH test with a significant bioactivity of 84.70 AAO% in 500 µg/mL, with an EC50 of 236.79. The content of total phenolic compounds was 946.06 mg of gallic acid equivalents/g of sample, and total flavonoids was 58.11 mg of quercetin equivalents/g of sample, the result obtained for FRAP was 15294.44 µM Trolox/g of sample and ABTS was 718 M Trolox of sample. The prospecting of the chemical constituents of the leaves of C. argyrophylloides revealed the presence of the main compounds that manifests the antioxidant activity and it was proven by the DPPH method that there is antioxidant activity in the analyzed sample, in addition to demonstrating a significant content of phenolic compounds and total flavonoid content in the species, which corroborates the antioxidant activity of the plant sample. Arg., pertencente à família Euphorbiaceae, conhecida popularmente como marmeleiro prateado e sacatinga, é um vegetal do bioma caatinga comumente encontrado no Nordeste do Brasil. O presente trabalho teve como objetivo avaliar a atividade antioxidante da espécie. O estudo fitoquímico foi realizado por meio de análise qualitativa dos constituintes químicos e determinação quantitativa do teor de fenóis totais pelo teste de Folin-Ciocalteu. ABSTRACT

Agroforestry systems are examples of soil exploration that is closer to the natural form of the forest, with intercropping of several species within an area, thus being sustainable alternatives. Therefore, the objective of this work was to evaluate the weed community in agroforestry systems in two ecosystems: solid ground and floodplain in the state of Pará. Such collections were made in eight areas, with about one hectare each, cultivated in agroforestry systems, four of which are on dry land and four on floodplains, in the rural area of Cametá-PA. Four plots of one square meter per area were randomly sampled, where the species were identified, counted, and taken to weigh to evaluate the weed community. Among the evaluated environments, the species that stood out the most in the solid ground environment was Kyllinga brevifolia, leading most of the evaluated indices. In the lowland ecosystem, the Brachiaria species purpuracens were the most relevant.

ABSTRACT

Este estudo teve como objetivo avaliar biomarcadores de mastite após terapia da secagem com plasma rico em plaquetas (PRP), associado ou não a antibiótico. Trinta e seis quartos mamários foram utilizados em três tratamentos (T): T1 - antibiótico (ATB), T2 - PRP e T3 - PRP + ATB. Amostras de leite foram coletadas antes da secagem (A1), no parto (D0) e 14, 30 e 60 dias pós-parto (D14, D30 e D60), para determinar contagem de células somáticas (CCS) e amiloide A (AA). O delineamento foi inteiramente ao acaso, com arranjo em parcelas subdivididas (SigmaPlot®). Dados de CCS foram transformados (log 10). As médias foram comparadas utilizando-se testes de Tukey ou Holm-Sidak (P<0,05). A CCS em A1 foi elevada em todos os grupos (P>0,05). No D30, CCS foi maior em T2 (P<0,05), igualando-se no D60. Não houve diferença na AA entre Ts em qualquer dia de coleta (P>0,05). Houve diferença nos momentos de coleta (P<0,05), A1 maior que D14 e D30. Houve uma correlação positiva fraca com CCS (0,280). Os tratamentos foram semelhantes em manter a saúde da glândula mamária na lactação subsequente. O PRP intramamário pode ser usado para terapia de vaca seca em casos de mastite subclínica.(AU)


This study aimed to evaluate biomarkers of mastitis after autologous platelet rich plasma (PRP) dry cow therapy, associated or not with antibiotic, compared to conventional treatment. Thirty-six mammary quarters were used in three treatments (T): T1 - antibiotic, T2 - PRP and T3 - PRP + ATB. Milk samples were collected before drying (A1), on calving (D0) and at 14, 30 and 60 days postpartum (D14, D30 and D60), for Somatic Cell Count (CCS) and amyloid dosage A (AA). The design was completely randomized with arrangement in subdivided plots (SigmaPlot ®). Data from CCS were transformed in log10. Means were compared using the Tukey or Holm-Sidak tests, at a 95% confidence level (P<0.05). CCS in A1 was elevated in allgroups (P>0.05). On D30 CCS was higher in T2 (P<0.05), matching D60. There was no difference in AA among T on any day of collection (P>0.05). There was difference in collection moments (P<0.05), A1 different from D14 and D30. There was a weak positive correlation with CCS (0.280). Three treatments were similar in maintaining the health of the mammary gland at subsequent lactation. PRP intramammary can be used for dry cow therapy in subclinical mastitis.(AU)

ABSTRACT

This trial aimed to evaluate the effects of thyme essential oils (EO) on rumen parameters, nutrient digestibility and nitrogen balance in wethers fed with high-concentrate diet. Twenty rumen-cannulated wethers were blocked according to body weight (BW= 64.0±2.1kg), and received one of the following treatments: 25mg of monensin/kg of dry matter (DM; MON) or doses of thyme EO (1.25, 2.50 or 3.75g/kg of DM). The diet was composed of 90% concentrate. Thyme EO was composed mainly by thymol (46.6% of DM) and p-cymene (38.9% of DM). The nutrient intake and apparent digestibility were similar among treatments. The inclusion of 3.75g of thyme EO tended (P= 0.07) to increase butyrate compared to MON and 1.25OE and wethers fed with 1.25g of thyme EO tended (P= 0.07) to decrease ruminal pH on the 14th day compared to MON. The treatments did not affect acetate:propionate ratio, total short chain fatty acids (SCFA) and nitrogen retention. Results from this study suggest that adding thyme EO to high-concentrate diets may be used as an alternative to monensin as feed additive in feedlot lambs.(AU)


O objetivo do estudo foi avaliar os efeitos do óleo essencial (OE) de tomilho nos parâmetros ruminais, na digestibilidade e no balanço de nitrogênio em borregos alimentados com elevado teor de concentrado. Vinte borregos providos de cânulas ruminais foram blocados de acordo com o peso corporal (PC=64,0±2,1kg) e receberam um dos tratamentos: 25mg de monensina/kg de matéria seca (MS; MON) ou doses de OE de tomilho (1,25; 2,50 ou 3,75g/kg de MS). A dieta foi composta por 90% de concentrado. A composição do OE de tomilho foi principalmente timol (46,6% da MS) e p-cimeno (38,9% da MS). A ingestão e a digestibilidade dos nutrientes foram semelhantes entre os tratamentos. A inclusão de 3,75g de OE de tomilho tendeu (P=0,07) a aumentar o butirato em relação aos tratamentos MON e 1,25OE. Os borregos alimentados com 1,25g de OE tenderam (P=0,07) a apresentar menor pH ruminal no 14º dia comparado a MON. No entanto, os tratamentos não afetaram a relação acetato:propionato, concentração total de ácidos graxos de cadeia curta e retenção de nitrogênio. Os resultados sugerem que a adição de OE de tomilho em dietas com elevado teor de concentrado pode ser uma alternativa à monensina como aditivo alimentar para cordeiros em confinamento.(AU)

ABSTRACT

The effect of the systemic absorption of 0.1% diclofenac sodium (DS) eyedrop was compared to that of 0.5% ketorolac tromethamine (KT) in female New Zealand white rabbits treated on both eyes three times a day for 90 days. The rabbits were divided in three groups of six animals (n= 18): KT group, DS group, and control (Co) group, in which saline (0.9% NaCl) solution was instilled. Water and food consumption were measured daily, clinical examination was performed weekly, and blood samples were collected every 30 days for laboratory examination. The plasma was analyzed for the presence of KT and DS by solid-phase extraction (SPE) associated with mass spectrometry (MS). Systemic absorption of these drugs was confirmed by SPE-MS, allowing their separation and identification in the plasma. At the end of the treatment, the animals were euthanized and necropsied, and no macroscopic or microscopic changes were found. This observation confirmed the laboratory results, which were within normal reference standards for the species. According to the results obtained, it can be concluded that treatment with eyedrops containing KT and DS for 90 days in healthy rabbits does not cause adverse systemic effects.(AU)


Comparou-se o efeito da absorção sistêmica do colírio de diclofenaco de sódio 0,1% (DS) em relação ao de cetorolaco de trometamina 0,5% (CT) em coelhas da raça Nova Zelândia, tratadas nos dois olhos, três vezes ao dia, por 90 dias. As coelhas foram separadas em três grupos de seis animais (n=18): grupo CT, grupo DS e grupo controle (Co), no qual foi instilada solução fisiológica (NaCl 0,9%). Os consumos de água e ração foram mensurados diariamente, os exames clínicos foram realizados semanalmente e o sangue foi coletado a cada 30 dias para realização de exames laboratoriais. O plasma foi analisado para detectar a presença de CT e DS por extração em fase sólida (SPE) associada à espectrometria de massas (MS). A absorção sistêmica desses fármacos foi confirmada por SPE-MS, permitindo sua separação e identificação no plasma. Ao final do tratamento, os animais foram eutanasiados e necropsiados, e não foram encontradas alterações macroscópicas ou microscópicas. Essa observação confirmou os resultados laboratoriais, que estavam dentro dos padrões de referência para a espécie. De acordo com os resultados obtidos, pode-se concluir que o tratamento com colírio contendo KT e DS, por 90 dias, em coelhos saudáveis, não causa efeitos adversos sistêmicos.(AU)

Background: Neonatal infection is a serious public health problem. The aim of this study was to assess the influence of the antenatal care on the risk of early-onset neonatal healthcare associated infection in two Brazilian maternities. Methods: Cohort study - Newborns admitted at two public neonatal intensive care units from 2008 to 2009 were included in the study. Data on antenatal and perinatal variables were collected from maternal prenatal cards and medical charts. Newborns were actively surveyed for early-onset neonatal healthcare associated infection, defined as a neonatal infection diagnosed within 48 h after birth. Multiple logistic regression was used to assess variables independently associated with early-onset neonatal healthcare associated infection. Results: 561 neonate-mother pairs were included in the study. Early-onset neonatal health-care associated infection was diagnosed in 283 neonates (51%), an incidence rate of 43.5/1000 live births. Neonates whose mothers had less then six antenatal visits were under risk significantly higher for early-onset neonatal healthcare associated infection (OR = 1.69, 95% CI = 1.11-2.57), after adjusting for birth weight, membranes ruptured for >18 h, maternal complications during delivery, maternal infection at admission, and hospital where patients received care. Conclusions: The risk of neonatal early-onset neonatal healthcare associated infection was significantly associated with insufficient number of antenatal care visits. Further studies assessing the quality of antenatal care and targeting its improvement are warranted. .

ABSTRACT

RESUMO Piper hispidum é uma espécie pioneira pertencente à família Piperaceae, com importância na medicina popular e na obtenção de óleo essencial. Assim como outras espécies da família, possui poucas informações sobre técnicas de cultivo. O objetivo foi avaliar a propagação via estaquia de Piper hispidum em função do tipo de substrato e estaca em Manaus, Amazonas, Brasil. O experimento foi realizado na Embrapa Amazônia Ocidental. O delineamento utilizado foi inteiramente casualizado em esquema fatorial 3 (estacas) x 5 (substratos), com três repetições de 12 estacas, sendo as estacas (apical, mediana e basal) e os substratos (areia lavada, substrato comercial, solo + esterco de aves, solo + casca de guaraná e fibra de coco). Foram avaliadas: enraizamento (%), número de brotações, comprimento da maior brotação (cm), número de folhas, comprimento da maior raiz (cm), massa seca da raiz (g) e massa seca das brotações (g). Foi realizada análise de variância pelo teste F a 5% de probabilidade e para as médias foi realizado o teste Tukey ao nível de 5% de probabilidade. Recomenda-se utilizar estacas apicais e basais, nesta ordem. Ocorreu interação entre os fatores substrato e tipos de estaca somente para variável número de brotações. Os substratos areia lavada e substrato comercial são indicados para maiores porcentagens de enraizamento com 81,56% e 81,33%, respectivamente. O enraizamento foi superior nas estacas apicais (85,67%) e basais (74,47%). Porém, para esta espécie os substratos solo + esterco de aves e/ou solo + casca de guaraná foram mais indicados quando o objetivo é obter estacas de qualidade.


ABSTRACT Piper hispidum is a pioneer species belonging to the family Piperaceae, with relevance in popular medicine and in obtaining essential oil. As other species of this family, there is little information about cultivation techniques. The aim of this work was to evaluate the propagation through cutting from the Piper hispidum according to the type of substrate and cutting technique in Manaus, Amazonas, Brazil. The trial experiment was conducted at the Embrapa Western Amazon. The design was completely randomized factorial 3 x 5, with three replications of 12 cuttings with types of cuttings (apical, median and basal) and substrates (washed sand, commercial substrate, soil + poultry manure, soil + guarana shell and coconut fiber). After 60 days, the following characteristics were evaluated: rooting (%), number of shoots, length of the largest sprouting (cm), number of leaves, length of the longest root (cm), root dry weight (g) and dry weight of shoots (g). An analysis of variance was performed by the F test at 5% probability and for the averages` comparison the Tukey test was done at 5% level of probability. It is recommended to employ apical and basal cuttings, respectively. There was interaction between the factors and substrate types of cuttings only for the variable number of sprouts. These two substrates, washed sand and commercial substrate, are suggested for higher percentages of rooting with 81,56% and 81,33 %, respectively. The rooting was higher in the apical cuttings (85,67 %) and basal ones (74,47 % ). However, for this species, the substrates soil + poultry manure and/or soil + guaraná shell were most indicated when the goal was to obtain high-quality cuttings.

ABSTRACT

New approaches are needed to quickly indicate possible contamination of UHT milk, among them the technique of ATP-Bioluminescence. Therefore, the aim of this study was to compare the results of culture methods with the results of ATP-Bioluminescence technique of 102 UHT whole milk samples incubated at 48, 72, and 168 hours. UHT milk samples were analyzed for the presence of mesophilic and psychrotrophic aerobic microorganisms using Plate Count Agar (PCA), Brain-Heart Infusion (BHI) media and PetrifilmTM Aerobic Count (AC) plates. The ATP-Bioluminescence technique was applied through the Microbial Luminescent Screening (MLS) system. Significant correlations were found between counts of aerobic mesophilic microorganisms on PCA, PetrifilmTM AC, BHI and results of ATP bioluminescence technique (P≤0.05). The ATP-Bioluminescence technique had higher correlation with counting method in PCA than BHI media. At lower pass/fail limits of Relative Light Units (60, 50, 45 and 40 RLU), the number of samples identified as positive increased and statistically agreed with aerobic mesophilic microorganism counts (P>0.05). For the dairy industry, the ATP-Bioluminescence technique may become an important tool that assists the official methods to quickly monitor the microbiological quality of UHT milk though this will likely require a threshold below 150 RLU...


Novos métodos são necessários para detectar de forma rápida a contaminação do leite UAT, entre eles, a técnica de ATP-Bioluminescência. Portanto, objetivou-se comparar os resultados de métodos de cultura tradicionais com os resultados de ATP-Bioluminescência de 102 amostras de leite UAT integral incubadas por 48, 72 e 168 horas. Os leites UAT foram analisados quanto à presença de micro-organismos aeróbicos mesófilos e psicrotróficos usando os ágares PCA, BHI e placas PetrifilmTM AC. A técnica de ATP-Bioluminescência foi aplicada por meio do sistema Microbial Luminescent Screening (MLS). Significantes correlações foram obtidas entre as contagens de micro-organismos mesófilos aeróbios em PCA, PetrifilmTM AC, BHI e os resultados da técnica de ATP-Bioluminescência (p<0,05). A técnica de ATP-Bioluminescência tem maior correlação com o método de contagem em meio PCA que BHI. Quando valores limites de Unidades Relativas de Luz (60, 50, 45 e 40 RLU) foram menores, o número de amostras identificadas como positivas aumentou e concordou estatisticamente com a contagem de micro-organismos mesófilos aeróbios (P>0,05). Para as indústrias de laticínios, a técnica de ATP-Bioluminescência pode se tornar uma ferramenta auxiliar aos métodos oficiais para o monitoramento rápido da qualidade microbiológica do leite UAT, desde que sejam utilizados limites abaixo de 150 RLU...

Avaliou-se a presença de resíduos antimicrobianos (ceftiofur, estreptomicinas, quinolonas, tetraciclinas, tianfenicol e tilosina) e anti-helmínticos (benzimidazóis, aminobenzimidazóis, levamisol, avermectinas, tiabendazóis, moxidectina e triclabendazóis) em, respectivamente, 70 e 83 amostras de leite cru provenientes de quatro mesorregiões (Triângulo Mineiro/Alto Paranaíba, Central Mineira, Oeste de Minas e Metropolitana de Belo Horizonte) do estado de Minas Gerais. Este estudo demonstrou a presença de quinolonas (2,86por cento), estreptomicinas (2,86por cento) e tetraciclinas (11,43por cento) nas amostras de leite cru analisadas. Ademais, observou-se expressiva porcentagem de amostras de leite cru positivas para os anti-helmínticos amino- benzimidazóis (55,42por cento), levamisol (53,57por cento), avermectinas (60,24por cento), tiabendazóis (67.47por cento), moxidectina (73,49por cento) e triclabendazóis (45,78por cento), e em menor porcentagem os benzimidazóis (6,02por cento). Desta forma, os resultados do presente estudo indicam que os resíduos de antimicrobianos e anti-helmínticos no leite em Minas Gerais devem ser constantemente monitorados pelas autoridades competentes com intuito de oferecer aos consumidores um alimento sem riscos à saúde humana...

ABSTRACT

O potencial probiótico in vitro de 12 amostras de bactérias ácido-láticas (11 Lactobacillus spp. e uma Weissella paramesenteroides), isoladas de queijo-de-minas artesanal da Serra da Canastra, foi investigado. Essas amostras foram caracterizadas in vitro quanto à susceptibilidade a antimicrobianos, antagonismo contra microrganismos de referência patogênicos e não patogênicos e sensibilidade a pH gástrico e sais biliares. Apenas L. rhamnosus B4, W. paramesenteroides C10 e L. rhamnosus D1 apresentaram resistência a menor número de antimicrobianos (ceftazidima, oxacilina, estreptomicina e vancomicina). Todas as amostras apresentaram atividade antagonista frente a todas as bactérias patogênicas testadas e não inibiram as bactérias não patogênicas. Todas as amostras foram resistentes ao pH gástrico (2.0). Alguns microrganismos mostraram pouca inibição do crescimento em presença de sais biliares, enquanto outros foram moderadamente ou altamente inibidos. L. rhamnosus B4, W. paramesenteroides C10 e L.rhamnosus D1 apresentaram o melhor potencial probiótico de acordo com os testes in vitro realizados.


The in vitro probiotic potential of twelve samples of lactic acid bacteria (eleven Lactobacillus spp. and one Weissella paramesenteroides) isolated from Minas artisanal cheese from Serra da Canastra was evaluated for their antimicrobial susceptibility, antagonistic activity against pathogenic and lactic acid bacteria, as well as for their tolerance to gastric pH and biliary salts. Only L. rhamnosus B4, W. paramesenteroides C10, and L. rhamnosus D1 showed resistance to tested antimicrobials (ceftazidime, oxacillin, streptomycin, and vancomycin). All samples showed antagonism against all pathogenic bacteria tested and did not inhibit the growth of nonpathogenic samples. Regarding gastric pH tolerance, all samples showed low inhibition at pH 2. The tolerance to biliary salts varied from low to great among tested samples. L. rhamnosus B4, W. paramesenteroides C10, and L. rhamnosus D1 showed the best probiotic potential revealed by their in vitro test results.

The potential of in vitro probiotic Lactobacillus spp. was evaluated in fermented milks marketed in Belo Horizonte, MG, Brazil. Of the samples analyzed, 86.7% had at least 10(6) CFU/mL of Lactobacillus spp., complying with the Brazilian quality standards for fermented milks. Furthermore, 56.7% had minimum count ranging from 10(8) to 10(9) CFU/mL, which is in accordance with legal parameters. The remaining 43.3% would not be able to satisfactorily guarantee benefits to consumers. The amount of Lactobacillus spp. varied between batches of products, which may indicate failures in monitoring during manufacture, transport or storage. All strains of Lactobacillus spp. showed some inhibitory activity against the indicator microorganisms, being more pronounced against pathogenic microorganisms than against non-pathogenic (P<0.05). Samples of Lactobacillus spp. showed different profiles of antimicrobial susceptibility, with an occurrence of cases of multidrug resistance. All strains tested showed sensitivity to bile salts (0.3%) and resistance to gastric pH (2.0). Lactobacillus spp. of commercial fermented milks should be present in higher amounts in some brands, be resistant to bile salts and have no multiple resistance to antimicrobials.


O potencial probiótico in vitro de Lactobacillus spp. foi avaliado em leites fermentados comercializados em Belo Horizonte, MG, Brasil. Das amostras analisadas, 86,7% apresentaram quantidade mínima de 106UFC/mL de Lactobacillus spp., enquadrando-se no padrão brasileiro de qualidade de leites fermentados. Além disso, 56,7% apresentaram quantidade mínima de 108a 109UFC/mL, estando em conformidade com os parâmetros legais vigentes. As demais 43,3% não estariam aptas a garantir satisfatoriamente efeitos benéficos aos consumidores. Observou-se variação na quantidade de Lactobacillus spp. entre lotes dos produtos, o que pode indicar falhas no monitoramento durante a fabricação, transporte ou estocagem destes. Todas as amostras de Lactobacillus spp. demonstraram alguma atividade inibitória frente aos microrganismos indicadores, sendo mais intensa contra microrganismos patogênicos que não patogênicos (P<0,05). As amostras de Lactobacillus spp. apresentaram diferentes perfis de susceptibilidade aos antimicrobianos, ocorrendo casos de multirresistência. Todas as amostras testadas apresentaram sensibilidade in vitro a sais biliares (0,3%) e resistência in vitro ao pH gástrico (2,0). Lactobacillus spp. dos leites fermentados testados deveriam estar presentes em quantidades mais elevadas em algumas marcas comerciais, possuir resistência aos sais biliares e não apresentar resistência múltipla aos antimicrobianos.

ABSTRACT

Embora métodos tradicionais sejam utilizados na avaliação microbiológica de produtos UAT, metodologias rápidas, baseadas em ATP-Bioluminescência, têm sido desenvolvidas. Os resultados da aplicação dessa técnica em 54 amostras de bebida láctea UAT achocolatada e 12 de creme de leite UAT foram comparados com os resultados de métodos microbiológicos, utilizando-se diferentes meios de cultura e tempos de incubação das referidas amostras. A técnica de ATP-Bioluminescência foi aplicada por meio do sistema MLS, e os resultados foram expressos em unidades relativas de luz (RLU). Em todos os tempos de incubação - 48, 72 e 168 horas - , as amostras apresentaram contagens baixas de microrganismos mesófilos e psicrotróficos aeróbios quando analisadas em meio PCA, BHI, PetrifilmTM AC e por ATP-Bioluminescência (<150 RLU), demonstrando alta especificidade da técnica. Apenas uma amostra de creme de leite UAT apresentou contagem de mesófilos aeróbios acima do padrão estabelecido pela legislação brasileira (<100 UFC/mL) quando analisada em meio PCA (260 UFC/mL) e PetrifilmTM AC (108 UFC/mL), no tempo de 168 horas. Essa alta contagem de microrganismos mesófilos aeróbios também foi detectada pela técnica de ATP-Bioluminescência (416 RLU). Os resultados da técnica de ATP-Bioluminescência foram iguais aos resultados em meio PCA, BHI e PetrifilmTM AC.


Although traditional methods are used for the microbiological evaluation of UHT products, rapid methodologies based on ATP-Bioluminescence have been developed. The results of applying this technique in 54 samples of chocolate UHT milk drink and 12 of UHT milk cream were compared with the results of microbiological methods, using different culture media and incubation times for the referred samples. The ATP-Bioluminescence technique was applied through the MLS system and the results were expressed as relative light units (RLU). In all incubation times - 48, 72, and 168 hours - , the samples showed lower counts of mesophilic and psychrotrophic aerobic microorganisms when analyzed using PCA, BHI, PetrifilmTM AC and ATP-Bioluminescence (<150RLU), demonstrating the technique's high specificity. Only one sample of UHT milk cream showed a mesophilic aerobic count above the standard established by Brazilian legislation (<100CFU/mL) when analyzed in PCA (260 CFU/mL) and PetrifilmTM AC (108CFU/mL) at 168 hours. This high count of aerobic mesophilic microorganisms was also detected by the ATP-Bioluminescence (416 RLU) technique. The results of the ATP-Bioluminescence technique were equal to the results in PCA, BHI and PetrifilmTM AC.

ABSTRACT

Amostras de queijo de minas artesanal foram coletadas em 18 queijarias localizadas em propriedades rurais da região da Serra da Canastra, Minas Gerais, com o objetivo de avaliar a influência da altitude sobre a população de bactérias acidolácticas. As queijarias estavam distribuídas nas altitudes de 600 a 900m, 900 a 1000m e mais de 1000m. Observaram-se populações mais elevadas de bactérias acidolácticas nas amostras de queijo da altitude de 600 a 900m. Lactobacillus rhamnosus, Lactobacillus casei e Lactobacillus plantarum foram os principais microrganismos isolados e identificados por PCR ARDRA 16S-23S rDNA, além de Enterococcus spp., Lactococcus spp. e outras espécies de Lactobacillus. Sugere-se que estas espécies estejam adaptadas ao ambiente de produção do queijo de minas artesanal produzido na região, o que resultaria em características sensoriais próprias do produto.


Samples of minas artisanal cheese were collected in 18 small-scale producer properties located in the rural region of Serra da Canastra, Minas Gerais state, aiming to evaluate the influence of three altitudes, from 600 to 900m, 900 to 1000m, and higher than 1000m, on the lactic acid bacteria (LAB) population. High populations of LAB were observed in the cheese samples, mainly in the lowest altitude. Lactobacillus rhamnosus, Lactobacillus casei, and Lactobacillus plantarum were the major LAB isolated from the cheese samples and identified according to PCR ARDRA 16S-23S rDNA. Enterococcus spp., Lactococcus spp., and other species of Lactobacillus genus were also found. It is suggested that these microorganisms are adapted to the production environment of the minas artisanal cheese which result in the unique sensorial properties of the product.

ABSTRACT

Com o objetivo de modificar a escala de coma de Glasgow pediátrica para utilização na medicina veterinária, foram utilizados 30 cães adultos com alterações neurológicas passíveis de avaliação da consciência. A escala modificada para cães foi aplicada em três momentos com intervalos de 48 horas entre eles, resultando em 90 eventos diversos. A escala foi aplicada para avaliação da abertura ocular (AO), da melhor resposta associada à vocalização (MRV) e da melhor resposta motora (MRM). Com a análise fatorial para os indicadores AO, MRV e MRM, obtiveram-se valores iniciais de 2,482, 0,302 e 0,215, respectivamente. A variância foi de 82,7 por cento, 10,1 por cento e 7,2 por cento, respectivamente, e cumulativa de 82,7 por cento, 92,8 por cento e 100 por cento, respectivamente. A extração esperada do principal fator, AO, foi de 2,482 com variância de 82,7 por cento e cumulativa de 82,7 por cento. O resultado da avaliação da consciência dos cães foi normal em 10 por cento dos animais, alteração leve em 20 por cento, moderada em 45 por cento e grave em 25 por cento, com escores de Glasgow iguais a 15, entre 13 e 14, entre 9 e 12 e entre 8 e 3, respectivamente. A escala de coma de Glasgow pediátrica modificada para cães é ferramenta segura para avaliação da consciência de cães adultos.


The pediatric Glasgow coma scale was modified for use in veterinary medicine. Thirty adult dogs with neurological alterations susceptible to evaluation of the conscience were studied. The modified scale for dogs was applied at three moments with intervals of 48 hours, resulting in 90 events. The scale was applied for evaluation of the ocular opening (OO), the best answer associated to vocalization (BAV), and the best motor answer (BMA). With the factorial analysis for the indicators OO, BAV, and BMA the initial values of 2.482, 0.302, and 0.215, respectively, were obtained. The variance was of 82.7 percent, 10.1 percent, and 7.2 percent, respectively, and cumulative of 82.7 percent, 92.8 percent, and 100 percent, respectively. The expected extraction of the main factor, OO, was 2.482 with variance of 82.7 percent and cumulative of 82.7 percent. The evaluation result of the conscience was normal in 10 percent of the animals, light alteration in 20 percent, moderate in 45 percent, and severe in 25 percent, with scores of Glasgow equal to 15, from 13 to 14, nine to 12, and eight to three, respectively. The pediatric Glasgow coma scale modified for dogs was a safe tool for evaluation of the conscience of adult dogs.

The gut barrier monitors and protects the gastrointestinal tract from challenges such as microorganisms, toxins and proteins that could act as antigens. There is evidence that gut barrier dysfunction may act as a primary disease mechanism in intestinal disorders. The aim of the present study was to evaluate the barrier function towards sugars after the appropriate treatment of celiac disease and Crohn's disease patients and compare the results with those obtained with healthy subjects. Fifteen healthy volunteers, 22 celiac disease patients after 1 year of a gluten-free diet, and 31 Crohn's disease patients in remission were submitted to an intestinal permeability test with 6.0 g lactulose and 3.0 g mannitol. Six-hour urinary lactulose excretion in Crohn's disease patients was significantly higher than in both celiac disease patients (0.42 vs 0.15 percent) and healthy controls (0.42 vs 0.07 percent). Urinary lactulose excretion was significantly higher in celiac disease patients than in healthy controls (0.15 vs 0.07 percent). Urinary mannitol excretion in Crohn's disease patients was the same as healthy controls (21 vs 21 percent) and these values were significantly higher than in celiac disease patients (10.9 percent). The lactulose/mannitol ratio was significantly higher in Crohn's disease patients in comparison to celiac disease patients (0.021 vs 0.013) and healthy controls (0.021 vs 0.003) and this ratio was also significantly higher in celiac disease patients compared to healthy controls (0.013 vs 0.003). In spite of treatment, differences in sugar permeability were observed in both disease groups. These differences in the behavior of the sugar probes probably reflect different mechanisms for the alterations of intestinal permeability.

ABSTRACT

Estudaram-se aspectos clínicos e hematológicos em eqüinos submetidos a um programa de controle estratégico de A. cajennense. Os tratamentos carrapaticidas foram realizados a cada sete dias e divididos em dois módulos, o primeiro com início em abril de 2004, e o segundo com início em julho do mesmo ano, utilizando-se a base química piretróide - cipermetrina na concentração de 0,015 por cento. Além do acompanhamento clínico dos animais, foram realizados hemogramas completos antes e após o programa. As dosagens bioquímicas de bilirrubinas, gama-glutamiltransferase (GGT), aspartato aminotransferase (AST), creatina kinase (CK), proteína total, albumina e globulinas, foram realizadas antes, durante e ao final do programa. Os resultados demonstraram que houve uma melhora no quadro hematológico dos animais após o programa de controle. Os tratamentos carrapaticidas, na forma em foram aplicados, não provocaram alterações desfavoráveis nos parâmetros clínicos e hematológicos dos eqüinos. Tais informações podem ser consideradas na busca de alternativas viáveis e seguras para o controle dessa espécie de carrapato.


Clinical and hematological parameters were studied in equines submitted to a strategic control program of Amblyomma cajennense. The acaricide treatments were carried to each seven days and divided in two batteries, the first one began in April 2004 and the second in July 2004. A pyrethroid chemical base - 0.015 percent cypermethrin was used. Clinical examinations of the animals and complete hemograms were carried before and after the control program of the tick. Seric dosages of bilirrubins, gamma-glutamiltransferase (GGT), aspartate aminotransferase (AST), creatin kinase (CK), total protein, albumin, and globulins, were carried before and throughout the experiment. The results showed an improvement in the hematological parameters of the animals after the end of control program. The acaricide treatments did not cause undesirable alterations of the clinical and hematological parameters studied. Such information can be considered as viable and safe alternatives for the control of this tick.
