ABSTRACT

AIM: To explore the effect of spinal anesthesia on ventricular arrhythmia and involved mechanisms in myocardial ischemia-reperfusion (MIR) rats. METHODS: The rat MIR model was made by occlusion the left anterior descending coronary artery for 30 minutes and reperfusion for 45 minutes. Bupivacaine (0.05 mL / 100 g, 1 mg / kg) was injected slowly via intrathecal for spinal anesthesia. The electromyelogram at T2 thoracic spinal cord was recorded. Ventricular arrhythmias, cardiac function, myocardial damage were assessed by electrocardiography, echocardiography and TTC or HE staining. RESULTS: MIR reduced left ventricular short-axis shortening (LVFS) and left ventricular ejection fraction (LVEF), caused myocardial histological damage and ventricular arrhythmias, promoted spinal electrical discharge frequency and amplitude in T2 dorsal horn. Spinal injection of bupivacaine could significantly reduce spinal cord electrical activities and eliminate MIR-induced arrhythmias. Moreover, bupivacaine also significantly improved MIR-induced myocardial histological damage and cardiac function inhibition. CONCLUSION: Spinal anesthesia can reduce ventricular arrhythmias induced by MIR. The mechanism may be related to the effect of abolishing spinal nerve excitability.

ABSTRACT

Background@#Due to the unavailability of an effective vaccine or antiviral drug against the African swine fever virus (ASFV), rapid diagnosis methods are needed to prevent highly contagious African swine fever. @*Objectives@#The objective of this study was to establish the ladder-shape melting temperature isothermal amplification (LMTIA) assay for the detection of ASFV. @*Methods@#LMTIA primers were designed with the p72 gene of ASFV as the target, and plasmid pUC57 was used to clone the gene. The LMTIA reaction system was optimized with the plasmid as the positive control, and the performance of the LMTIA assay was compared with that of the commercial real-time polymerase chain reaction (PCR) kit in terms of sensitivity and detection rate using 200 serum samples. @*Results@#Our results showed that the LMTIA assay could detect the 10 4 dilution of DNA extracted from the positive reference serum sample, which was the same as that of the commercial real-time PCR kit. The coincidence rate between the two assays was 100%. @*Conclusions@#The LMTIA assay had high sensitivity, good detection, and simple operation. Thus, it is suitable for facilitating preliminary and cost-effective surveillance for the prevention and control of ASFV.

ABSTRACT

AIM: To investigate the effect of sakubatril valsartan combined with dagliflozin in the treatment of patients with HFrEF and the effect on serum cardiac troponin I (CTnl) and B-type brain natriuretic peptide (BNP) levels. METHODS: Seventy patients with HFrEF admitted to our hospital from January 2020 to October 2021 were selected and divided, using random number table method, into control group (35 cases, conventional treatment + sakubatril valsartan) and observation group (35 cases, conventional treatment + sakubatril valsartan + dagliflozin). The treatment effect, myocardial markers (serum cTnl, BNP), exercise capacity (6 min walking experiment), myocardial remodeling-related indexes [(left ventricular end-diastolic diameter (LVEDd), left ventricular end-systolic internal diameter (LVESD), left ventricular ejection fraction (LVEF)] and adverse effects were compared between the two groups. RESULTS: The total effective rate of treatment in the observation group was lower than that in the control group (P 0.05); the changing trends of serum BNP and cTnl expressions were the same in the two groups at 3 months and 6 months of treatment, and the serum BNP and cTnl expressions of patients in the observation group were lower than those in the control group (P 0.05). CONCLUSION: The combination of sakubatril valsartan and dagliflozin is effective for patients with HFrEF, and can effectively regulate serum cTnl and BNP levels with low adverse reactions.

ABSTRACT

Firstly, this article discussed the significance of medical ethics enacted by Nuremberg Code from three aspects. Thereafter, it introduced the fundamental principles of Nuremberg Code in regulating the aspect of human experiment, which included that the subjects should voluntarily agree to the experiment and informed con-sent, the experiment should be guided by scientific ideas, beneficial to society and harmless to subjects. Finally, it pointed out the basic norms of Nuremberg Code in the aspect of human experiment that the researchers must tell subjects the relevant information about the strictly assessed risk and danger, the experiments should carry on based on the standardized animal experiments, and subjects should have the freedom to decide whether they want to quit the experiments.

ABSTRACT

Objective To observe the changes of serum cystatin C(Cys C) in patients with coronary heart disease(CHD) and normal kidney function ,and evaluate the predictive value of Cys C concentration on coronary arterial lesions .Methods Serum levels of Cys C were detected in 316 patients with coronary heart disease and normal renal function .The relationship between serum 1evels of Cys C and coronary heart disease was evaluated from three aspects :the number of diseased vessels ,the severity of diseased ves‐sels and the CHD Gensini scores .Results The Cys C level in CHD group was significantly higher than that of non‐CHD group [(1 .24 ± 0 .32)mg/L vs .(1 .12 ± 0 .27)mg/L ,P<0 .01] .Serum Cys C was significantly higher in the multi‐vessel disease group than that of the single‐vessel disease group[(1 .31 ± 0 .31)mg/L vs .(1 .20 ± 0 .32)mg/L ,P<0 .01] .Serum Cys C was significantly higher in the severe coronary artery stenosis group than in the mild stenosis group [(1 .29 ± 0 .36)mg/L vs .(1 .16 ± 0 .23)mg/L , P<0 .01] .Cys C was positively correlated with the Gensini scores of coronary arterial lesion (r=0 .195 ,P<0 .01) .Conclusion Cys C levels increased in patients with coronary heart disease and normal renal function .Cys C was positively correlated with the Gensini scores of coronary arterial lesion .Serum Cys C levels has certain predictive value on coronary arterial lesions in patients with CHD .

ABSTRACT

AIM: To observe the local expression of nuclear factor-?B(NF-?B) in spontaneously hypertensive rat(SHR) kidney and effects of AT1 receptor contagonist valsartan.METHODS: 16 SHRs were randomly divided into two groups: SHR control group and valsartan group.Another 8 WKY rats act as normal control group.Systolic blood pressure(SBP) of SHR was measured at the beginning and the end of 2,4,6 and 8 weeks of intervention treatment.Radioimmunoassay was used to determine the activities of rennin and angiotensin II(AngII).The renal tissue NF-?B protein expression was detected by immunobiochemistry.RESULTS: SBP of SHR was remarkably decreased after valsartan intervention.However,the rennin activities and AngII level in plasma increased in valsartan group.In the renal tissue of SHR,there was remarkably increased in expression of NF-?B protein.Valsartan could significantly reduced NF-?B expression.CONCLUSION: Valsartan can depress NF-?B renal expression in protein level and might benefit hypotension renal function.

ABSTRACT

Aim To research the changes of myocardial endoxin level in rats with myocardial ischemia reperfusion (MIR) and the protevtive effects of anti digoxin antiserum, an endoxin specific antagonist, on MIR injury. Methods Myocardial ischemia reperfusion injury models were obtained by ligating left anterior descending coronary artery 30 min followed by 45 min reperfusion. Sprauge Dawley rats were randomly divided into seven groups each with 10 rats. There were sham group, MIR group, normal saline group, verapamil group, low dose anti digoxin antiserum group, middle dose anti digoxin antiserum group, and high dose anti digoxin antiserum group. After reperfusion of left ventricular myocardium, sample of ischemia were processed immediately. Myocardial endoxin levels, Na +, K + ATPase activities, and intramitochondrial Ca 2+ contents were measured. The myocardial morphology were observed. Results Myocardial endoxin levels were significantly increased; Na +, K + ATPase activities were remarkably decreased; intramitochondrial Ca 2+ contents were remarkably raised. Meanwhile, myocardial morphology injury were remarkable in light microscope and electric microscope. Middle and high dose of anti digoxin antiserum intervention, myocardial endoxin levels were remarkably decreased; Na +, K + ATPase activities were drastically increased; intramitochondrial Ca 2+ declined. The myocardial histological morphology was significantly improved. Conclusion Antidigoxin antiserum, an endoxin antagonist, had protective effect against MIR. The mechanism maybe related to antagonizing endoxin, restoring energy metabolism, attenuating intracellular Ca 2+ overload.

ABSTRACT

Aim To investigate effects and the mechanism of endoxin special antagonist anti-digoxin antiserum on heart function in myocardial anoxia-reoxygenation injury in rats. Methods The isolated Langendorff perfused rat heart model was established. Sixty Sprague Dawley(SD) rats were randomly divided intosix groups and each group had 10 rats: control group, anoxia-reoxygenation group, verapamil group, low, middle, high dose anti-digoxin antiserum groups. ECG, HR, LVDP and ?dp/dtmax were continuously recorded. The endoxin levels and intramitochondrial Ca2+ contents in myocardial tissues and nitric oxide (NO) contents in coronary artery fluence were measured after reoxygenation. Structures of mitochondrial and endothelial cells were observed by microscope. Results The anoxia-reoxygenation group showed a remarkable increase in endoxin level and intramitochondrial Ca2+ content, an obvious decrease NO content, an obvious injury of mitochondrial and endothelial cell, an obvious inhibition of heart function. Middle, high dose of anti-digoxin antiserum group could remarkably decrease endoxin level and intramitochondrial Ca2+ content; increase NO content; obviously relieve the injury of mitochondrial and endothelial cells; remarkably improve the discovery of heart function. Conclusion Anti-digoxin antiserum could inhibit the failure of heart function induced by myocardial anoxia-reoxygenation injury. Its mechanism may be related to antagonize endoxin, relieve mitochondrial Ca2+ overload, increase NO contents, and protect the function of mitochondrial and endothelial cells.
