ABSTRACT

@#Objective    To explore the effect of expression of miRNA-21 on bone marrow mesenchymal stem cells (BMSCs). Methods    In this study, flow cytometry was used to identify the surface-associated antigens of BMSCs. The 10 μmol/L 5-azacytidine was used to induce BMSCs to differentiate to cardiomyocyte-like cells. Immunofluorescence was used to detect the expression of troponin I (cTnI). The samples were assigned to 3 groups: a blank group, a miRNA-21 mimic group, and a negative control (NC) group. The proliferation of BMSCs was detected by methyl thiazolylte-trazolium (MTT), the apoptosis of BMSCs was analyzed by flow cytometry. Western-blotting was used to identify the expression of cTnI and myod in the BMSCs. Results    The proliferation of BMSCs was increased, because of the over expression of miRNA-21. But the apoptotic rate of the BMSCs was slower in the miRNA-21 group, on account of the expression of miRNA-21 was higher than that in the NC group and the CK group. The expression of cTnI in the miRNA-21 group was higher than that in the NC group or the CK group. Conclusion    The results suggest that the up-regulation of miRNA-21 enhances proliferation of BMSCs, reduces the apoptosis of BMSCs. miRNA-21 promotes the differentiation of BMSCs, which may pave the way for the treatment directed toward restoring miRNA-21 function for myocardial ischemia.

ABSTRACT

Objective: To investigate the effects of progranulin on migration and invasion of human colon cancer cells

Study Design: An experimental study

Place and Duration of Study: Department of Histology and Embryology, College of Basic Medical Sciences, Jilin University, Changchun, China, from October 2015 to July 2017

Methodology: Cell transfection was used to obtain progranulin knocked down/overexpressing cells. MTT and soft agar assay were applied to investigate the cell growth. Migration and invasion were detected using transwell assay

Results: Tumor growth, migration and invasion decreased in progranulin knocked down cells and increased in progranulin overexpressing cells. The decrease was recovered when exogenous progranulin was added. Progranulin-induced migration and invasion were inhibited by neamine

Conclusion: Progranulin has a direct effect on tumor growth, migration, and invasion of human colon cancer cells. It may serve as a potential therapeutic target for human colon cancer

ABSTRACT

The teaching methods were explored to improve the quality of medical genetic teaching for foreign students according to the common problems during the teaching process. The negative effects of communication barriers in medical genetic teaching could be reduced by interactive teaching or problem-based learning in groups,in which the ability to resolve problems by themselves could be improved. In order to improve the teaching systematicness and teaching quality, the teaching contents in class should be from simple to deep, covering genetic laws, pathogenesis, diagnosis and control measure of genetic diseases. From the perspective of practical application and combining with the construction of self-de-signed teaching textbook and cases, the quality of medical genetic teaching ultimately could be further improved.

ABSTRACT

Objective To investigate the anti-mutation of Chinese medicinal herb Herba Leonuri and its effect on T lymphocyte proliferation in spleen. Methods The micronucleus test of mouse bone marrow cell(MNT) :thirty mice were divided into six groups(n= 5),negative control(NS),cyclophosphamide group(CP 3.0 mg?kg-1),Herba Leonuri antimutagenesis groups(Herba Leonuri with dosages of 1.0,2.0,4.0,8.0 g?kg-1+CP30 mg?kg-1).The improved method was used to detect the micronuclei frequency.Lymphocyte transformation test:twenty-four mice were divided into four groups(n=6),saline control,CP control(30 mg?kg-1),Herba Leonuri(2.0 g?kg-1),Herba Leonuri +CP(2.0 g?kg-1 Herba Leonuri +CP 30 mg?kg-1).MTT assay was used to calculate the stimulation index(SI).Results The micronuclei frequencies in Herba Leonur 2.0,4.0,8.0 g?kg-1 groups were lower than that in CP group(P

ABSTRACT

Objective To study the effect of anti-mutation of Chinese medicinal herb Poly gala tenui folia willd on T lymphocyte proliferation in spleen. Methods The micronucleus test of mouse bone marrow cell (MNT): thirty mice were divided into six groups (n = 5) , negative control (NS) , cyclophosphamide group (CP 3. 0 mg ? kg-1) , Poly gala tenui folia willd antimutagenesis groups (Polygala tenui folia willd with dosage of 0.5, 1.0. 2.0, 4.0 g ? kg-1 + CP 30 mg ? kg-1 ). The improved method was used to detect the micro-nuclei frequency. Lymphocyte transformation test: twenty-four mice were divided into four groups (n = 6), saline control , CP control (3.0 mg ? kg-1), Polygala tenui folia willd (2.0 g ? kg-1), Polygala tenui folia willd + CP (2. 0 g ? kg-1 Polygala tenuifolia willd + CP 30 mg ? kg- ) group. MTT assay was used to calculate the stimulation index (SI). Results The micro-nuclei frequency was significant difference between Poly gala tenui folia willd antimutagenesis groups and CP groups (P
