ABSTRACT

Objective To investigate exogenous ceramide-induced apoptosis in colon carcinoma LoVo cells.Methods LoVo cells were pretreated for 3 h with or without the presence of phorbol 12-myristate 13-acetate (PMA), a potent protein kinase C (PKC) stimulator, later treated with C2-, C6-ceramide or C2-dihydroceramide, and then passed through gel electrophoresis, Heochest 33342 fluorescence staining and flow cytometry with Annexin V/PI staining. The treated LoVo cells were observed for biochemical and morphologic changes. Results Treatment with C2- or C6-ceramide in the range of indicated concentrations (10~25 μmol/L) for 12 to 24 hours resulted in apoptosis in LoVo cells, whereas C2-dihydroceramide, which is similar to C2-ceramide in configuration but lacks the trans double bond at C4-C5 of the sphingoid base backbone, did not induce the apoptosis at the same or even higher concentrations, indicating that the ceramide-induced apoptosis was stereospecific. Moreover, the exogenous ceramide-induced apoptosis in LoVo cells was inhibited in part by PMA. Conclusion Ceramide takes part in the process of apoptotic signal transduction in LoVo cells. PKC may be one of downstream target molecules acted by ceramide.

ABSTRACT

Objective To study on the clinical application and value of double-balloon video en teroscopy in diagnosing small inlestinal bleeding. Methods Fifty-four cases with suspected small intestinal bleeding were subjected to double-balloon video enteroscopy, via the mouth and /or anus in 21, 20 and 13 cases respectively, the procedure was performed under X-ray monitoring. Results The positive rate of en-doscopy was 90. 7% , the findings were isolated or multi small intestinal ulcer 11 cases, Crhon' s disease 7 cases; chronic nonspecific inflammation 6 cases, entero-mesenchymoma 6 cases; high differentiated adeno-carcinoma 3 cases; polyps 2 cases, lymphoma 1 case, stero-pro-nematodiasis 2 cases, ancylostomiasis 2 cases, vascular deformity 2 cases ( 1 with active hemorrhage) , Michael diverticulosis 2 case, iliac polydivertic ulosis 1 case, ulcerative colonitis 1 case, duodenal stasis I case, duodenal ulcer 2 cases and essentially normal 5 cases. Complications related to the procedure never occurred. Conclusions The main causes of small intestinal bleeding are benign ulcers and tumor, as well as chronic inflammation. Parasitosis is the fourth cause. Diverticulosis and vascular deformity are the rare cause. But Michael diverticulosis is an important cause for the children with small intestinal bleeding., Double-balloon video enleroscopy is the most valuable method in diagnosing small intestinal diseases.

ABSTRACT

Objective To evaluate diagnostic value of pit pattern analysis on detection of early colorectal carcinoma. Methods 4176 patients were examined with colonoscopy and had the mucosal lesions stained with 0.4% indigo carmine, and part of them observed with magnifying endoscope and stereomicroscope, then compared the mucosal crypt patterns (the pit patterns Kudo classification) with pathologic diagnosis. Results There were 955 protruded and flat lesions on the large intestine mucosa in 752 patients, and among them there are 14 early cancers, 209 advanced cancers, 76Ⅱa、Ⅱb、Ⅱc、Ⅱa+Ⅱc lesions. We also found 43 laterally spreading tumors (LST) ranging from 16 to 110 mm in diameter, 2 for pit Ⅱ,18 for pit Ⅲ L, 19 for pit Ⅳ, 1 for pit Ⅴ A, 1 for Ⅴ N. The pit pattern of the most non neoplastic lesions was type Ⅰ or Ⅱ, which is about 85.4% (303/355), and the type of the adenomas was type Ⅲ or Ⅳ, about 86.0% (504/586). All the invasive carcinomas'pit patterns were type Ⅴ and there were 8 for type Ⅴ (2 Ⅴ A, 6Ⅴ N) among 14 early carcinomas. Conclusion Pit pattern analysis is a very important tool to determine the nature of lesions, which helps to decide the kinds of later therapeutic intervention.

ABSTRACT

Objective: To strengthen the immunogenicity of hepatocarcinoma cells and activate immnological cells recognizing and killing the tumor cells.Methods:The murine MHC-I gene H-2Kb which can express immunologial rejection antigens was transfected into human hepatocarcinoma cells HepG2 by liposome DNA mediated gene gene transfer.The transfection of H-2Kb gene were detected by molecular hybridization techniues.The exogeous antigens expressed on the membrane of trans- fected tumor cells were detected with ABC immunohistochemical method and flow cytometer. [3H] release assays were used to detect the recognizing and killing effects of lymphocytes to HepG2 cells transferred with murine H-2Kb gene. The nude mice ex- periment was used to further verify CTL cells killing active.Results:Southern blot hybridization showed that the H-2Kb gene was integrated into the chromosome of HepG2 cells. The RNA dot blot hybridization showed that there was transcription of H-2Kb DNA in the transfected tumor cells.ABC immunohistochemical method and flow cytometer detection showed that the murine H-2Kb antigens were expressed on the membrane of HepG2 cells. [3H] release assays showed that the cytotoalcyty to HepG2 cells fected with H-2Kb gene was obviously higher than that to control cells.The results demonstrated that the growth of hepatocarcino- ma cells which were transferred with H-2Kb gene was obviously inhibited.Conclusion:The murine MHC-I gene H-2Kb could be transferred into the human hepatocinoma cells and expressed on the membrane of transferred cells.The HepG2 cells transferred with H-2Kb gene could induce human effective lymphocytes to recognize and kill these transferred tumor cells.

ABSTRACT

AIM: To explore the regulatory effect of bifidobacteria adolescence on NF-?B in murine peritoneal macrophages(PMs).METHODS: Macrophages were collected and were divided into tow groups,normal control group and bifidobacterium stimulation groups.The cells were fixed at 60 min after stimulated with bifidobacterium at 10~6,10~7, 10~8 and 10~9 cells/L,or fixed respectively at 15,30,60,120 and 180 min after stimulated with bifidobacterium at 10~8 cells/L,then the total protein and nucleoprotein were extracted.The activities of NF-?B and I?B? were measured with the methods of MSA and Western blotting.RESULTS: NF-?B activity markedly increased and reached the peak at 0.5 h after stimulation,while I?B? markedly decreased at same time.CONCLUSION: NF-?B activity is markedly activated by bifidobacterium.NF-?B pathway participates in the regulation of peritoneal macrophage in this process.

ABSTRACT

AIM: To deplore the adjustion of whole peptidoglycan of bifidobacterium bifidum to functions of macrophages.METHODS:Levels of interleukin 6, interleukin 12 produced by peritoneal macrophages of nude mice was detected by ELISA method, and the content of nitric oxide (NO) was detected by using Griess reagent, after whole peptidoglycan were injected into nude mice peritoneally. RESULTS:The content of interleukin 6, interleukin 12 and NO secreted from peritoneal macrophages of nude mice in the whole peptidoglycan injection group was 732.54?190.30(pg/mL)、816.37?96.40(pg/mL) and 48.90?6.51(?mol/L) respectively.It was 303.78?171.75(pg/mL), 510.27?123.46(pg/mL) and 30.67?12.83(?mol/L) respectively in the control group. ABSTRACT

Objective To observe the influences of combined therapy of ulinastatin and thymosin ?1 on the immune state in the treatment of sepsis and multiple organ failure(MODS),and to evaluate the therapeutic rationality.Methods 46 patients with sepsis and 18 patients with MODS were randomly divided into two groups.The patients in control group were treated with classical SSC therapy,those in treatment group were treated with the combined ulinastatin and thymosin ?1 therapy.The contents of IL-6 and IL-10 were detected with ELISA at the beginning of and after the treatment.Results After treatment,the concentrations of IL-6 and IL-10 declined significantly in treatment group,while no apparent changes were observed in the control group.There existed a significant difference between the two groups(P

ABSTRACT

4219 subjects were screened for colorectal cancer by fiberoptic sigmoidoscopy.5 subjects(0.12%)werc found to have cancers: 2 Dukes A, 2 Dukes B, and 1 Dukes C. One or more polyps in 160 subjects (3.79%) were detected, of which 86 were adenomas and 3 severe dysplasia. All the polyps were removed by means of endoscopic polypectomy.For selected screening in small population,fiberoptic sigmoi-doscopy is recommended. As for mass screening, it is advisable to run fecal occult blood test (FOBT) first. All persons with positive FOBT or negative FOBT but belonging to high-risk category should be advised to have fiberoptic sigmoidoscopy. Special diet is unnecessary prior to sigmoidoscopy. Glycerol enema is recomended for bowel preparation.

ABSTRACT

Biopsies of ileal mucosa of 38 healthy Chinese were obtained through colonofibroscope. The observations under SEM showed that the major type of villi of the ileal mucosa are finger-shaped; a small number tongue-shaped, ridge-shaped villi are in rare cases. The branched or fused villi were not found in our study.
