ABSTRACT

ABSTRACT Purpose: To explore the therapeutic effects of orthokeratology lens combined with 0.01% atropine eye drops on juvenile myopia. Methods: A total of 340 patients with juvenile myopia (340 eyes) treated from 2018 to December 2020 were divided into the control group (170 cases with 170 eyes, orthokeratology lens) and observation group (170 cases with 170 eyes, orthokeratology lens combined with 0.01% atropine eye drops). The best-corrected distance visual acuity, best-corrected near visual acuity, diopter, axial length, amplitude of accommodation, bright pupil diameter, dark pupil diameter, tear-film lipid layer thickness, and tear break-up time were measured before treatment and after 1 year of treatment. The incidence of adverse reactions was observed. Results: Compared with the values before treatment, the spherical equivalent degree was significantly improved by 0.22 (0.06, 0.55) D and 0.40 (0.15, 0.72) D in the observation and control groups after the treatment, respectively (p<0.01). After the treatment, the axial length was significantly increased by (0.15 ± 0.12) mm and (0.24 ± 0.11) mm in the observation and control groups, respectively, (p<0.01). After the treatment, the amplitude of accommodation significantly declined in the observation group and was lower than that in the control group, whereas both bright and dark pupil diameters significantly increase and were larger than those in the control group (p<0.01). After the treatment, the tear-film lipid layer thickness and tear break-up time significantly declined in the two groups (p<0.01). Conclusions: Orthokeratology lens combined with 0.01% atropine eye drops can synergistically enhance the control effect on juvenile myopia with high safety.


RESUMO Objetivo: Explorar os efeitos terapêuticos das lentes de ortoceratologia combinados com colírio atropina 0,01% em miopia juvenil. Métodos: Um total de 340 pacientes com miopia juvenil (340 olhos) tratados entre 2018 e Dezembro de 2020 foram divididos em Grupo Controle (170 casos com 170 olhos, lentes de ortoceratologia) e Grupo Observação (170 casos com 170 olhos, lentes de ortoceratologia combinadas com colírio atropina 0,01%). A acuidade visual melhor corrigida para longe, acuidade visual melhor corrigida para perto, dioptria, comprimento axial, amplitude de acomodação, diâmetro da pupila brilhante, diâmetro da pupila escura, espessura da camada lipídica da película lacrimal e tempo de ruptura do rasgo foram medidos antes do tratamento e 1 ano depois. A incidência de reações adversas foi observada. Resultados: Antes do tratamento, o grau esférico equivalente foi significativamente melhorado em 0,22 (0,06, 0,55) D e 0,40 (0,15, 0,72) D respectivamente no Grupo Observação e no Grupo Controle após o tratamento (p<0,01). Após tratamento, o comprimento axial foi significativamente aumentado em (0,15 ± 0,12) mm e (0,24 ± 0,11) mm respectivamente nos Grupos Observação e controle (p<0,01), enquanto, no grupo de observação, a amplitude de acomodação diminuiu significativamente e foi inferior a do Grupo Controle, e o diâmetro da pupila brilhante e o diâmetro da pupila escura aumentaram significativamente e foram maiores do que os do Grupo Controle (p<0,01). A espessura da camada lipídica da película lacrimal e o tempo de ruptura do rasgo diminuíram significativamente nos dois grupos (p<0,01) após o tratamento. Conclusões: As lentes de ortoceratologia combinadas com colírio atropina 0,01% podem melhorar significativamente o efeito controle em miopia juvenil com elevada segurança.

ABSTRACT

Simiao Wan (SMW) is a traditional Chinese formula, including Atractylodis Rhizoma, Achyranthis Bidentatae Radix, Phellodendri Chinensis Cortex and Coicis Semen at the ratio of 1:1:2:2. It can be used to the treatment of diabetes. However, its bioactive compounds and underlying mechanism are unclear. This study aimed to screen the antilipolytic fraction from SMW and investigate its therapeutic mechanisms on hepatic insulin resistance. Different fractions of SMW were prepared by membrane separation combined with macroporous resin and their antilipolytic activities were screened in fasted mice. The effects of 60% ethanol elution (ESMW) on lipolysis were investigated in 3T3-L1 adipocytes stimulated by palmitic acid (PA) and high fat diet (HFD)-fed mice. In our study, ESMW is the bioactive fraction responsible for the antilipolytic activity of SMW and 13 compounds were characterized from ESMW by UHPLC-QTOF-MS/MS. ESMW suppressed protein kinase A (PKA)-hormone-sensitive lipase (HSL) related lipolysis and increased AMP-activated protein kinase (AMPK) phosphorylation in PA challenged 3T3-L1 adipocytes. AMPKα knockdown abolished the inhibitory effects of ESMW on IL-6 and HSL pSer-660, revealing that the antilipolytic and anti-inflammatory activities of ESMW are AMPK dependent. Furthermore, ESMW ameliorated insulin resistance and suppressed lipolysis in HFD-fed mice. It inhibited diacylglycerol accumulation in the liver and inhibited hepatic gluconeogenesis. Conditional medium collected from ESMW-treated 3T3-L1 cells ameliorated insulin action on hepatic gluconeogenesis in liver cells, demonstrating the antilipolytic activity contributed to ESMW beneficial effects on hepatic glucose production. In conclusion, ESMW, as the antilipolytic fraction of SMW, inhibited PKA-HSL related lipolysis by activating AMPK, thus inhibiting diacylglycerol (DAG) accumulation in the liver and thereby improving insulin resistance and hepatic gluconeogenesis.

ABSTRACT

For local treatment of ulcerative colitis, a new azoreductase driven prodrug CDDO-AZO from bardoxolone methyl (CDDO-Me) and 5-aminosalicylate (5-ASA) was designed, synthesized and biologically evaluated. It is proposed that orally administrated CDDO-AZO is stable before reaching the colon, while it can also be triggered by the presence of azoreductase in the colon to fragment into CDDO-Me and 5-ASA, generating potent anti-colitis effects. Superior to olsalazine (OLS, a clinically used drug for ulcerative colitis) and CDDO-Me plus 5-ASA, CDDO-AZO significantly attenuated inflammatory colitis symptoms in DSS-induced chronic colitis mice, which suggested that CDDO-AZO may be a promising anti-ulcerative colitis agent.

ABSTRACT

At present, the incidence and mortality of gastric cancer in China are 10.26% and 12.45%, ranking the second and third places, respectively, in the incidence and mortality of malignant tumors in China. Cancer often goes through three stages: precancerous lesions, carcinoma in situ, and invasive carcinoma. It is of great significance to advance cancer prevention and control to the stage of precancerous lesions. The popularization of digestive endoscopy-assisted diagnosis and treatment has enabled the timely diagnosis and treatment of early gastric cancer and severe dysplasia. However, the endoscopic mucosal resection is not suitable for most precancerous lesions of gastric cancer (PLGC), and the effective drugs are not available. The long-term clinical dynamic monitoring has imposed considerable physical and mental burdens on patients. Gastrointestinal microenvironment is a dynamic balance system composed of gastrointestinal flora, chemical barrier, mechanical barrier, immune barrier, and gastrointestinal nerves. The imbalance of gastric microenvironment has been proved to be the key mechanism of PLGC. According to traditional Chinese medicine (TCM), PLGC is a result of long-term interaction between deficient healthy Qi and excessive pathogens. In syndrome differentiation and treatment, PLGC is often believed to be developed from chronic gastritis. Besides, the inflammation-cancer transformation model put forward by Correa and the evolution of its TCM pathogenesis are also considered. Guided by the basic treatment principle of reinforcing healthy Qi to eliminate pathogenic factors, we determined the basic therapeutic methods as follows: invigorating spleen, clearing heat, and resolving dampness. At the same time, such methods as soothing liver and regulating Qi, resolving phlegm and dissipating mass, activating blood and resolving stasis, clearing heat and removing toxin, and tonifying deficiency can be combined based on the results of syndrome differentiation. After discussing the correlation between the imbalance of gastric microenvironment and PLGC and summarizing TCM intervention methods and mechanisms against PLGC from the perspective of gastric microenvironment regulation, this paper believed that TCM improved the gastric microenvironment by regulating the disorder of gastric flora, eliminating the gastric mucosal inflammation, and relieving the abnormal immune response, thereby preventing and controlling the PLGC.

ABSTRACT

Objective The influenza monitoring results in Huzhou city were analyzed to understand the characteristics of influenza pandemic and to provide a scientific basis for prevention and control of influenza. Methods The influenza-like cases(ILI)were collected from two sentinel hospitals in Huzhou according to the protocol of The National Influenza Surveillance Program. Pharynx swab samples of the ILI were taken randomly for the test of influenza virus. Results Among 5 322 788 patients who went to the sentinel hospitals in Huzhou from 2011 to 2017, 185 010 were ILI. The percentage of ILI was 3.48% and there was a slowly rising trend in ILI% of month from 2011 to 2017(trend χ2 = 2 970.93, P < 0.001). Winter /spring and summer seasons marked the peaks in influenza epidemics. Patients were mostly children under 14 years old(accounting for 89.1%). In a total of 11 490 throat swabs collected from ILI, 1 975 were tested to be positive for influenza virus(positive rate 17.19%). These included A type H3N2, 1 008; A type H1N1, 350; B type, 614(160 Victoria type, 238 Yamagata, 216 unclarified B type), and 3 mixed type. The correlation between positive rate of influenza nucleic acid test and ILI% is statistically significant(r = 0.40, P < 0.001). Conclusion The incidence peaks of influenza in Huzhou are mainly in the winter /spring and summer periods. The influenza is mainly occurs in young people under 14 years old and the main type virus is A(H3N2). The prevalence of influenza B and H1N1 occurs at intervals. There is a positive correlation between influenza activity and ILI%.

Objective:The TIFY gene family will be identified and characterized from the whole genome level in Cannabis sativa,which will lay the foundation for gene function study on TIFY family genes and their regulation mechanism on the biosynthesis of cannabinoids and other secondary metabolites. Method:Using the existing genomic data of cannabis,the CsTIFY genes were identified through bioinformatics analysis tools such as NCBI,PlantTFDB,MEME and TBtools etc.,and physicochemical properties,phylogenetic trees,gene structures,chromosome locations and gene expression patterns were analyzed and visualized. Result:Fourteen TIFY family genes(CsTIFY1-CsTIFY14) were identified in Cannabis sativa,which belong to four subfamilies:TIFY,JAZ,ZML,and PPD. The CsTIFYs are composed of 365-1 369 bp nucleotides encoding 118-442 amino acid residues,and their isoelectric points are 4.64-9.96. The 14 CsTIFYs are unevenly distributed on 8 chromosomes,and their proteins are all located in the nucleus. The promoter of CsTIFYs contain multiple abiotic stress responsive cis-acting elements,which indicated that CsTIFYs might involved in the regulation of different abiotic stresses. Transcriptome profiling revealed that CsTIFYs expressed differently in female flowers of 10 differently cannabis varieties,or in flowers,bracts,stems,and leaves of the same variety. Conclusion:Fourteen TIFY family genes were characterized from the whole genome level in C. sativa,and their phylogenetic evolutions and gene expression patterns were analyzed,indicating that CsTIFYs may play important regulatory roles in JA signal transduction,abiotic stress and cannabinoid biosynthesis. This study will provide valuable reference for gene function study of the TIFY family genes in cannabis and cannabis breeding.

ABSTRACT

OBJECTIVE@#To explore the efficacy of ultrasound-guided needle-knife with precise three-dimensional stereotactic localization of points for stenosing tenosynovitis of flexor tendon (trigger finger).@*METHODS@#A total of 74 patients were randomly divided into an observation group and a control group, 37 cases in each group. The patients in the observation group were treated with ultrasound-guided intrathecal injection and releasing method of needle-knife, while the patients in the control group were treated with ultrasound-guided intrathecal injection. The self-made 9-score scale of trigger finger was recorded before treatment, immediately after treatment, 1 month and 3 months after treatment; the curative effect of the two groups was evaluated.@*RESULTS@#The results of self-made 9-score scale in the observation group immediately after treatment, 1 month and 3 months after treatment were lower than that before treatment (all <0.01); the scores in the observation group were lower than those in the control group at each time point after treatment (all <0.01). The excellent and good rate immediately after treatment was 100.0% (37/37) in the observation group, which was superior to 8.1% (3/37) in the control group (<0.05); the cured rates in the observation group were 100.0% (37/37) 1 month after treatment and 97.3% (36/37) 3 months after treatment, which were superior to 13.5% (5/37) and 10.8% (4/37) in the control group, respectively (<0.05).@*CONCLUSION@#The needle-knife with three-dimensional stereotaxic location of point could significantly improve the symptoms of trigger finger, with superior immediate and long-term efficacy.

BACKGROUND@#With increasing media coverage of food safety incidents, such as that of clenbuterol residues in pork, food safety has become a major public health concern in China. Rapidly developing online markets attract increasing numbers of Chinese consumers to purchase food on the Internet. However, the quality and safety of food sold online are uncertain and are less reported on.@*OBJECTIVE@#This research aimed to systematically study the quality and safety of chilled pork from wet markets, supermarkets, and online markets in China.@*RESULTS@#The chilled pork samples from online markets were fresher than those from wet markets and supermarkets based on the surface redness (a* value). Chilled pork contained high levels of nutritional elements, especially the magnesium and phosphorus levels in samples from online markets. The levels of heavy metal element residues and veterinary drug residues in all chilled pork samples were within the standards limits. In addition, huge differences existed in the quality and freshness of the chilled pork samples from online markets according to principal component analysis (PCA).@*CONCLUSIONS@#Most chilled pork sold in Chinese markets was qualified and safe. It is necessary to establish an effective online market supervision system for chilled pork.

ABSTRACT

Known as "the king of the five grains",tartary buckwheat (Fagopyrum tataricum) is a medicinal and edible plant with both nutritional and healthcare functions. It contains rich flavonoids, such as rutin,with balanced amino acid composition and multiple effects, like blood fat-lowering,blood giucose-lowering,blood pressure-lowering,anti-oxidation,anti-aging,anti-cancer,anti-cancer,and microcirculation-improving. Transcription factors play important roles in plant growth and development by regulating gene expressions. MYB family is one of the largest transcription factor families in plant,contains the MYB domain,and can be divided into four subfamilies:1R-MYB,R2R3-MYB,3R-MYB,4R-MYB. This family plays various roles in plant growth,plant development and flavonoid biosynthesis of secondary metabolism. In this study,the reported MYB transcription factors in F. tataricum were summarized and systemically clustered,and their interrelationships were defined to provide references for further exploring and cloning MYB transcription factors in F. tataricum. In addition,this study reviewed their regulatory functions of MYB transcription factors in flavonoid biosynthesis pathway,plant hormones pathways and other abiotic stress pathways,and made a conclusion and advances about the future research in F. tataricum. Therefore,this study will provide valuable scientific references for the functional studies of MYB family transcription factors in F. tataricum and its molecular breeding for high-quality varieties.

ABSTRACT

Cardiovascular disease (CVD) is the most common cause of death in patients with non-alcoholic fatty liver disease (NAFLD). New therapeutic strategies which have the potential for slowing down the evolution of NAFLD and reducing CVD-related mortality are urgently needed. Statins are well recognized in the treatment of dyslipidemia, but their use in the treatment of NAFLD is limited due to the safety concerns. Ilexgenin A (IA) is one of the main bioactive compounds in 'Shan-lv-cha', an herbal tea commonly used in China. In the present study, we investigated the possible synergistic therapeutic effects of IA and simvastatin (SV) on NAFLD. IA or SV showed beneficial effects on the rats with NAFLD by lowering the liver weight, liver index and plasma levels of alanine aminotransferase and aspartate aminotransferase, regulating abnormal metabolism of lipids and ameliorating steatosis in liver. IA significantly enhanced the hypolipidemic and anti-inflammation effects of SV. Furthermore, a sensitive, accurate, convenient and reproducible LC-MS method was developed to investigate the effects of IA on the pharmacokinetics of SV. No significant changes were observed in pharmacokinetic parameters of SV and simvastatin hydroxy acid in the IA plus SV co-treated group in comparison with those in the group treated with SV alone. The mRNA levels and activity of CYP3A1 were not altered by IA. In conclusion, the results obtained from the present study should be helpful for further clinical application of SV and IA alone or in combination.

ABSTRACT

BACKGROUND: Due to its strong catalytic activity and selective catalytic oxidation, gold-silver bimetallic nanoparticles have potential applications in biomedicine, drug delivery and electrochemical analysis.OBJECTIVE: To prepare the gold-silver alloy nanoparticles using mitochondria in animal cells as templates. METHODS: Separating mitochondria from the fish liver: 1 mL of mitochondrial mother liquor was added into 1 mL of alkaline HAuCl4solution (10 mmol/L) and shaken; then, 1 mL of AgNO3solution (10 mmol/L) was added, and the mixture was reacted in an electromagnetic stirrer for 20-30 hours until the color of the reaction solution was changed from colorless to purple, which indicated that gold and silver alloy nanoparticles were successfully obtained; finally, the characterization of the nanoparticles was analyzed. (1) Cytotoxicity test: gold-silver alloy nanoparticles atdifferent concentrations (0, 25, 50, 75, 100, 125, 150 mg/L) were added into gastric cancer cells. After 48 hours of culture, the proliferation of cells (absorbance value) was detected by MTT assay. (2) Stability evaluation: 1 mL of gold-silver alloy nanoparticles at different concentrations (0, 25, 50, 75, 100, 125, 150 mg/L) were placed into glass reaction flasks and then, 0.2 mL of ultrapure water was added in portions following by shaking. After addition of the ultra-pure water, ultraviolet-visible spectrum was used to analyze the characterization of the solution. RESULTS AND CONCLUSION: The gold-silver alloy nanoparticles were spherically distributed and confirmed to have an alloy structure with the average particle size of 20-30 nm, and presented to have the advantages of simple structure, good crystallinity, and active groups such as hydroxyl and carbonyl groups on the surface. MTT results showed that the gold-silver alloy nanoparticles at the mass concentration of < 150 mg/L had no obvious cytotoxicity. The characteristic absorption peak with the largest intensity was positively correlated with the concentration of gold-silver alloy nanoparticles.To conclude,the gold-silver alloy nanoparticles have good cytocompatibility and stability in vitro.

Aim To establish human U87-MG glioma model in nude mice brain and to observe the characteristics of the tumor growth. Methods Human U87-MG glioma cells were cultured in vitro. 5 μL of cell suspension containing 3.0 ×1010·L-1, 4.0×1010·L-1and 5.0×1010·L-1respectively was inocula-ted into the right caudate nucleus of 18 male nude mice brain un-der the guidance of stereotaxic apparatus, separately, whereas another 6 nude mice as the control group, were inoculated into the same volume of Hanks solution. The moving and survival state of rats with gliomas were observed. The examinations of the tumors formation, volumes, metastasis and histopathology were performed and the obtained brain samples were stained with HE and immunohistochemistry. Results All the tested rats of dif-ferent inoculation doses developed brain tumors without extracra-nial metastasis. The mean survival time of three groups was (46.50 ± 3.27) d,(38.50 ± 3.28) d and (30.67 ± 3.51) d,respectively. The tumors showed the similar morphological fea-tures and immunophenotype to human glioma. There was positive expression of GFAP and S-100 in the tumors. Conclusions The orthotopic implantation model of human U87-MG glioma, by in-oculating quantitative U87-MG cells stereotaxically into the brains of the nude mice, is successfully established with 100 yield of intracranial tumor and no extracranial growth extension. It resembles the histopathological and morphological features of human glioma,which can be used as a reliable animal model for the study of the tumorigenesis, pathogenesis, biological charac-teristics and therapy of glioma.

ABSTRACT

Objective To analyze the situation of malaria epidemic and the course of prevention and control,and to summarize and promote the experience of malaria elimination. Methods The data of malaria prevention measures of different stages in Huzhou City from 1950 to 2015 were analyzed retrospectively. Results Huzhou City went through three serious malaria epidemics during the past 60 years:1954 to 1955,1962 to 1963 and early 1970. The highest incidence rate was 13 981. 76 / 1 00,000 in 1963. Since standard treatments(including anti - relapse treatment)were carried out and anti - mosquito facilities had improved,the average annual incidence rate in the 70 s declined gradually. From 1980 to 1989,Huzhou City set up 15 - 30 longitudinal monitoring stations which covered 70% villages and towns. In 1989,the City passed the fundamental evaluation of malaria eradication. The average annual incidence rate from 1990 to 2009 was 0. 37 / 1 000,000,and 48. 95% were imported cases. After 2010,Huzhou City had no local malaria case,and all cases reported were imported from 2010 to 2015. Conclusion A remarkable effects of the targeted control measures have been made at different stages in Huzhou City. In order to consolidate the achievements,we should strengthen the monitoring of floating population,timely and effective treatment of imported cases.

ABSTRACT

Objective·To explore the correlation of etomidate dose and bispectral index (BIS) in children with congenital heart disease shunting from left to right during anesthesia induction. Methods·After general anesthesia induction to 24 children with congenital heart disease shunting from left to right, sevoflurane inhaling was stopped in order to make the BIS value gradually return to 80. Intravenous injection of etomidate was sustained by 0.3 mg/(kg·min) until the BIS value reached 40. The heart rate (HR), arterial blood pressure (ABP) and central venous pressure (CVP) of the patients were recorded when BIS value was 80, 70, 60, 50,and 40, respectively.The correlation of etomidate dose and BIS value in the children during anesthesia induction was analyzed. Results·There were no significant hemodynamic changes when the BIS value were from 80 to 40. The amount of etomidate was highly negatively correlated with the BIS value (r=-0.769, P<0.01), the average dose of etomidate in children under 2 years of age was significantly higher than that in children over 2 years old (P=0.034). Conclusion·The etomidate dose is highly negatively correlated with the BIS value. Etomidate has less influence on hemodynamic parameters of children with congenital heart disease shunting from left to right. The BIS value may have relationship with patients'age.

ABSTRACT

Objective:To study the in silico cloning and bioinformatics of chalcone synthase gene from Ribes americanum. Meth-ods:Ribes nigrum chalcone synthase sequence and Ribes americanum were cloned by retrieving the EST database as the querying probe, which were assembled by CAP3 sequence assembly program, and the bioinformatics database and related software were used to predict the structure and perform the function analysis. Results:Bioinformatical analysis showed chalcone synthase gene encoded 1423 bp and contained a 1173bp ORF, the protein was with 390 amino acid, which was a hydrophilic protein located in cytoplasm including three transmembrane regions with the secondary structure composed of alpha helix. Conclusion:The study is helpful to the further ex-planation for the molecular function mechanism of chalcone synthase gene from Ribes americanum.

ABSTRACT

Previously,hand-surgeons made great advancements and pioneering surgeries such as thumb reconstruction,flap transplantation surgery.Under the leadership of professor Yu-dong GU,we put forward several surgical strategies to treat the brachial plexus injury including the widely-used contralateral cervical 7 (CC7) nerve root transfer surgery.Now our department is one of the leading centers in treating peripheral nerve injury disease worldwide.During the past 10 years,we focused on the brain plasticity research in brachial plexus injury patients and revealed the reorganization pattern of sensorimotor cortex after the CC7 nerve root transfer surgery.Our study indicated the potential of one hemisphere in controlling bilateral upper limb and we further applied this conclusion in treating hemiplegic paralysis patients.In the past 10 years,we applied basic research findings in clinical practice and expanded the indication of CC7 nerve transfer surgery.By making innovations we keep making progress.
