ABSTRACT

Sepsis-induced uncontrolled systemic inflammatory response syndrome (SIRS) is a critical cause of multiple organ failure. Acute kidney injury (AKI) is one of the most serious complications associated with an extremely high mortality rate in SIRS, and it lacked simple, safe, and effective treatment strategies. Leontopodium leontopodioides (Willd.) Beauv (LLB) is commonly used in traditional Chinese medicine for the treatment of acute and chronic nephritis. However, it remains unclear whether lipopolysaccharide (LPS) affects LPS-induced AKI. To identify the molecular mechanisms of LLB in LPS-induced HK-2 cells and mice, LLB was prepared by extraction with 70% methanol, while a lipopolysaccharide (LPS)-induced HK-2 cell model and an AKI model were established in this study. Renal histopathology staining was performed to observe the morphology changes. The cell supernatant and kidney tissues were collected for determining the levels of inflammatory factors and protein expression by ELISA, immunofluorescence, and Western blot. The results indicated that LLB significantly reduced the expression of IL-6 and TNF-α in LPS-induced HK-2 cells, as well as the secretion of IL-6, TNF-α, and IL-1β in the supernatant. The same results were observed in LPS-induced AKI serum. Further studies revealed that LLB remarkably improved oxidative stress and apoptosis based on the content of MDA, SOD, and CAT in serum and TUNEL staining results. Notably, LLB significantly reduced the mortality due to LPS infection. Renal histopathology staining results supported these results. Furthermore, immunofluorescence and Western blot results confirmed that LLB significantly reduced the expression of the protein related to the NF-κB signaling pathway and NLRP3, ASC, and Caspase-1 which were significantly increased through LPS stimulation. These findings clearly demonstrated the potential use of LLB in the treatment of AKI and the crucial role of the NF-κB/NLRP3 pathway in the process through which LLB attenuates AKI induced by LPS.

ABSTRACT

BACKGROUND: Some clinical randomized controlled trials are trying to answer the clinical effects of SuperPATH approach and traditional approach in hip replacement, but the conclusions of present studies are different. OBJECTIVE: To compare the clinical outcomes between SuperPATH approach and traditional approach in hip arthroplasty. METHODS: The randomized controlled trials about SuperPATH approach and traditional approach in hip arthroplasty published before July 2017 were searched in the PubMed, EMbase, EBSCO, Cochrane Library, and CNKI. Some published references were hand-searched. Two reviewers independently screened the literatures, extracted the data and evaluated methodological quality. Meta-analysis was conducted by Revman 5.3 software. RESULTS AND CONCLUSION: Eight randomized controlled trials (involving 483 cases) were involved in this analysis. The SuperPATH approach group consisted of 228 cases; the traditional approach group consisted of 255 cases. Meta-analysis results demonstrated that compared with traditional approach group, operation time was longer; incision was shorter; hip joint score was higher at postoperatively 3 months; Visual Analogue Scale scores were lower at postoperatively 1, 3 and 7 days; intraoperative blood loss and postoperative drainage volume were less in the SuperPATH approach group. No significant difference was found in postoperative complications, postoperative anteversion angle, and postoperative abduction angle, and hip score at postoperatively 1 and 6 months. These findings suggested that the total short-term effects in SuperPATH approach was better than that of traditional approach in hip arthroplasty. Long-term clinical outcomes require further investigation.

The pathogenesis is complex, and disease locations are related to the lung, spleen, kidney, heart, and liver. Based on the relationship between lung and chronic glomerulonephritis, this article discussed the feasibility, common treatment and prescription of chronic glomerulonephritis from the lung, which could provide a theoretical basis for the treatment of chronic glomerulonephritis in clinic.

ABSTRACT

This study aims to isolate and identify the bacteria from the surface of Musca domestica adults in different livestock farms.The adults of Musca domestica were collected with sweep nets,then the surface of adult flies were washed with sterile water,and the eluted bacteria cultured with LB culture medium.The DNA of the bacteria isolated was extracted,16SrDNA fragment was amplified through general PCR,and the 16SrDNA sequence was blasted with bacteria sequence in NC-BI.Results showed that the number of Musca domestica adults and the bacteria from their surface had significant different in different livestock farms.The population density of Musca domestica adults had linear correlation with the number of bacteria from Musca domestica adults and the number of bacteria genus.Among the bacteria isolated from Musca domestica adults,Staphylococcus,Klebsiella,Escherichia and Serratia were the most common genus in the livestock farms.Most of the bacteria isolated from livestock farm house flies were opportunistic human bacterial pathogen,and the risk of bacterial pathogens spread to people was existed.The population density of house flies reflected the management level of different livestock farms.The house flies could mechanical transmit opportunistic human bacterial pathogen from livestock residue to human.So the house flies control and the monitoring of bacteria on body surface of house flies carried should be strengthened.

ABSTRACT

AIM:To observe the anti-inflammatory effect of triptolide (TP) on lipopolysaccharide (LPS)-in-duced uveitis (LIU). METHODS:BALB/c mice (n=24) were randomly divided into 3 groups:blank control group (treated with normal saline), model group (treated with normal saline) and triptolide treatment group (treated with 0.05% triptolide eye drops),with 8 mice in each group. The mice in model group and triptolide treatment group were in-travitreally injected with LPS after 30 d of drug treatment. The anterior chamber was assessed by slit-lamp examination at different time points after modeling. Ocular tissues were collected for histological examination. The protein levels of inter-cellular adhesion molecule-1 (ICAM-1),interleukin-1β (IL-1β) and monocyte chemotactic protein-1 (MCP-1) in iridial and retinal samples were tested by ELISA. RESULTS:Compared with model group,the inflammatory reaction and clinical score were obviously decreased in triptolide treatment group at 24 h after modeling (P<0.01). The histopathological ob-servation indicated that infiltrating inflammatory cells were dramatically reduced in the anterior and posterior segments by triptolide eye drops. In addition, the expression levels of ICAM-1, IL-1β and MCP-1 were significantly decreased in the ocular tissues by treatment with triptolide (P<0.05). CONCLUSION:Triptolide prevents the injury of LIU by down-regulating inflammatory cytokines,and may be a new immune therapy for uveitis.

ABSTRACT

Objective To study the accuracy of preoperative planning and postoperative satisfaction of prosthesis posi?tion by applying the personal surgical cutting guide (PSCG) to total knee arthroscopy (TKA) based on 3D printing technique. Meth?ods In this randomized study, 20 patients were selected from August 2014 to October 2015, who were randomly divided into two groups. In PSCG group, 4 males and 6 females were enrolled, with a mean age of 70.9±6.69 (from 59 to 78) years. In con?ventional instrumentation group, 2 males and 8 females were enrolled, with a mean age of 69.9 ± 6.35 (from 58 to 77) years. The conventional instrumentation group was treated with conventional TKA, while the PSCG group was treated with personal surgical cutting guides. Then the knee valgus angle of patients, the angle between prosthesis components on the coronal and the sagittal plane of two groups were evaluated. Subsequently the correlation of the actual osteotomy amount, the valgus angle, caster angle, external rotation angle between intra? and preoperative planning were compared in the PSCG group. Results Compared with the ideal value of each angle, the mean deviation of the hip?knee?ankle angle 0.77°±0.51°, frontal femoral compo?nent angle 0.37° ± 0.53° and frontal tibial component angle 0.11° ± 0.24° showed statistically significant difference between two groups (P0.05). The actual osteotomy amount, the valgus angle, caster angle, external rotation angle corre?lated well between intra?and preoperative planning (t=-2.547, 3.864, 0.537,-0.040,-1.290, P>0.05). Conclusion TKA assist?ed by PSCG can make lower extremity alignment and accuracy of prosthesis implantation more accurate compared with convention?al TKA, especially in hip?knee?ankle angle, frontal femoral component angle and frontal tibial component angle.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the quantitative relationship between the reduction of distortion product otoacoustic emission (DPOAE) and the percentage of outer hair cell loss.</p><p><b>METHODS</b>Coadministration of cisplatin (0.2 mg/kg) and ethacrynic acid (40 mg/kg) were used to establish a cochlear lesion model in chinchillas. DPOAE was measured before and 1 week, 2 weeks, and 3 weeks later respectively after cisplatin and ethacrynic acid treatment. Animals were terminated 3 weeks after the treatment. Cochlear surface preparations were performed, and the cochlear hair cells were counted through entire length of the cochlea. The correlation between DPOAE reduction and outer hair cell missing was analyzed using Pearson correlation analysis.</p><p><b>RESULTS</b>Cisplatin and ethacrynic acid treatment induced cochlear hair cell lesion that the outer hair cell loss in the cochlea developed in a stereotypic pattern; damage began in the base of the cochlea and progressed towards the apex. Reduction of DPOAE was relatively consistent with outer hair cells loss. On the average, 1% outer hair cells loss may result in 0.24 dB reduction in DPOAE levels. Pearson analysis showed a positive correlation between the reduction in DPOAE and missing of outer hair cells (r = 0.796, P < 0.05).</p><p><b>CONCLUSIONS</b>It may be helpful to evaluate missing percentage of outer hair cells from reduction in DPOAE levels.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate if caspase pathway was involved in streptomycin-induced cell apoptosis in cochlear hair cells.</p><p><b>METHODS</b>F344 rats at postnatal day 3 or 4 were used for the study in cochlear organotypic cultures. The cochlear basilar membrane was micro-dissected out and cultured overnight, and then treated with 1 mmol/L streptomycin for 24 hours. Before the termination, the activity of caspase-8, 9 or 6 were detected with FAM-peptide-FMK labeled caspase-8, 9 or 6, respectively. The stereocilia and cuticular plate of hair cells were stained with TRITC conjugated phalloidin, and the nuclei were stained with Topro-3 DNA probe. The specimens were observed and photographed under confocal fluorescent microscope.</p><p><b>RESULTS</b>Streptomycin with 1 mmol/L causes about 80% cochlear hair cells missing in the basal turn and 10% hair cell loss in the apex. After streptomycin treatment, the nuclear shrinkage and fragmentation were found in most cochlear hair cells, and the caspase-8, caspase-9 and caspase-6 were greatly activated.</p><p><b>CONCLUSIONS</b>Apoptosis is involved in the cochlear hair cells death induced by Streptomycin in vitro. The caspase activities in upstream and downstream are maybe the major apoptotic pathway.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the ototoxic effects of streptomycin in vestibular organotypic cultures.</p><p><b>METHODS</b>F344 rats with age at postnatal day three or four were used for this study. The maculae of saccule and utricle were routinely dissected out and cultured with serum-free medium containing various dose of streptomycin for 24 hours. The ciliary turf of vestibular hair cells was stained with fluorescent phalloidin, and its nucleus was stained with to pro-3 DNA probe. The vestibular hair cells were quantitatively counted and photographed under confocal fluorescent microscope.</p><p><b>RESULTS</b>Morphological feature of vestibular hair cells were good in normal control cultures. However, the density of hair cells was decreased in evidence with increase of streptomycin sulfate concentrations. Twenty-four hours after streptomycin cultures, 0.25 mmol/L streptomycin caused a 10% hair cell missing, 50% hair cell loss from 1 mmol/L streptomycin treatment, and more than 75% hair cells gone post-3 mmol/L streptomycin cultures. After streptomycin treatment, the nuclear shrinkage and fragmentation were found in vestibular hair cells, whereas the vestibular supporting cells were normal.</p><p><b>CONCLUSION</b>Streptomycin induced-vestibular hair cells lesion was in a dose dependent manner with nuclear shrinkage and fragmentation. This may suggest that streptomycin leads vestibular hair cell die through apoptosis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To discuss the value of one stage flap or musculocutaneous flap to repair the soft tissue defect in Pilon fracture.</p><p><b>METHODS</b>Twelve cases with Pilon fracture included 9 male and 3 female with an average age of 39.2 years ranging from 21 to 61 years. All fractures were type III according to Rüedi and Allgöwer. According to Gustilo sysytem, III a were in 4 cases, III b were in 8. A thorough debridement was made before internal fixation. After the internal fixation implanted, the tourniquet was released and a thorough debridement was made again. Be sure of the wound cleansed of all dead and foreign material, the wound was covered with local flap, musculocutaneous or gastrocnemius flaps depending on the size and localization of wound.</p><p><b>RESULTS</b>All the patients were followed up from 6 to 36 months, 18 months in average. The function of ankle was assessed according Mazur system. The results were excellent in 6 cases, good in 3, fair in 2 and poor in 1. No infection or necrosis happened on flaps. Although necrosis happened in the wound margin of two patients, they all healled up by conservative methods.</p><p><b>CONCLUSION</b>Primary closure of soft tissue defect in Pilon fracture using flap or musculocutaneous flap have ability to shorten the treating time and recover the function of ankle. It is important to have a thorough debridement of the dead tissue and free bone.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate duration tuning in the inferior colliculus (IC) of guinea pigs and the role of GABA-mediated inhibition on this tuning.</p><p><b>METHODS</b>Totally 23 healthy albino guinea pigs of either sex were employed in this study. After anesthesia, spikes of neurons in inferior colliculus were recorded using five-barrel glass-pipettes. The characteristic frequency was determined by recording iso-intensity response curves at moderate intensity level (40-70 dB SPL) and duration tuning was measured with signals of fixed intensity and varied durations. GABA-A receptor antagonist, bicuculline, was applied to neurons by means of in vivo micro-iontophoresis through one channel in the five-barrel glass-pipettes.</p><p><b>RESULTS</b>IC neurons of guinea pigs, especially for those who showed sustained temporal response pattern, showed stronger duration tuning in their transient response peak to signal onset. Among 207 neurons recorded, totally 93 neurons were found to show clear duration selectivity. The duration selectivity was eliminated or turned to be weaker in most of the neurons in which the effect of bicuculline was observed successfully.</p><p><b>CONCLUSIONS</b>Unlike what was reported in bats, duration selectivity may be a transient process for most of IC neurons in guinea pigs. Duration selectively of IC neurons in the guinea pig was also largely dependent on the GABAergic inhibition.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study anatomic and computed tomographic measurements of Chinese mandibular genial tubercles and to evaluate the correlations between them.</p><p><b>METHODS</b>The axial images were taken by spiral CT in 40 adult human skulls with 1 mm thick section from infraorbital margin to menton. Sagittal plane reconstruction was produced through mandibular central line. Then the height and width of superior genial tubercles, the distance between menton and inferior margin of genial tubercles, the distance between mandibular incisor apex and superior margin of superior genial tubercles, the thickness of mandible were measured respectively. Thereafter anatomic measurements were taken by the same methods as computed tomographic images. The measured value were showed as means +/- standard deviation, then paired-t test and correlation analysis was made by SPSS 10.0.</p><p><b>RESULTS</b>The genioglossus almost origins from superior genial tubercles, geniohyoideus from inferior genial tubercles. The height of superior genial tubercles which were measured by anatomy and spiral CT were (5.82 +/- 0.71) mm and (6.17 +/- 0.71) mm respectively. The width of superior genial tubercles were (6.98 +/- 1.35) mm and (7.01 +/- 1.13) mm. The distance between menton and inferior margin of superior genial tubercles were (11.08 +/- 2.05) mm and (10.41 +/- 1.55) mm. The distance between mandibular incisor apex and superior margin of superior genial tubercles for male were (15.57 +/- 1.82) mm and (14.34 +/- 2.06)mm, and for female were (9.36 +/- 2.79) mm (8.78 +/- 2.53) mm. The thickness of mandibles at genial tubercles were (11.95 +/- 1.59) mm and (12.19 +/- 1.64) mm. The distance from menton to superior margin of superior genial tubercles were (16.1 +/- 2.30) mm and (15.73 +/- 2.12) mm. The correlations between anatomic measurements and spiral CT measurements of the above mentioned parameters were significant except for height of superior genial tubercles (r = 0.59 - 0.92).</p><p><b>CONCLUSION</b>The anatomic and spiral CT measurements of genial tubercles appear to have significant correlations. Preoperative spiral CT measurements of genial tubercles could be help for the design of osteotomy in genioglossus advancement.</p>
