ABSTRACT

OBJECTIVE@#To investigate the effects of Gastrodiae rhizoma, a dried root of Gastrodia elata Blume, on proliferation and differentiation of human NSCs derived from embryonic stem cells.@*METHODS@#A 70% ethanol extract of Gastrodiae rhizoma (EEGR) was estimated with 4-hydroxybenzyl alcohol as a representative constituent by HPLC.@*RESULTS@#MTT assay showed that the treatment with EEGR increased the viability of NSCs in growth media. Compared to control, EEGR increased the number of dendrites and denritic spines extended from a differentiated NSC. Whereas EEGR decreased the mRNA expression of Nestin, it increased that of Tuj1 and MAP2 in NSCs grown in differentiation media. Immunocytochemical analysis using confocal microscopy also revealed the increased expression of MAP2 in dendrites of EEGR-treated NSCs. Furthermore, EEGR decreased mRNA expression of Sox2 in NSCs grown even in growth media.@*CONCLUSIONS@#In conclusion, our study demonstrates for the first time that EEGR induced proliferation and neuronal differentiation of NSCs, suggesting its potential benefits on NSC-based therapies and neuroregeneration in various neurodegenerative diseases and brain injuries.

ABSTRACT

This study was performed to observe the sequential charge of the morpholcgic characteristics in experimentally induced herpes simplex keratitis. Duration and morphology of corneal lesion following infection of rabbit cornea with the Kos strain of HSV-I were followed by a daily slit-lamp examination. Three types of virus inoculation methods were used such as scratching, deepithelialization, and intrastromal injection. Herpetic corneal lesions appeared 24 hours after inoculation with punctate and dendritic figures. They persisted up to 14 or 15 days. The characteristic finding in punctate herpetic keratitis was grouped, round-shaped, punctate lesion. When scratching method was emplyed, the most remarkable finding was the discontinuity of the lesion occurred along the scratching wound at relatively regular intervals. There was no difference in lesional morphology and duration between three inoculation methods.

ABSTRACT

In diagnostic methods of herpes simplex keratitis are included clinical appearance, cytology, viral culture, immunofluorescent technique, and identification with electron microscopy. We studied isolation rate of virus with viral culture method from 44 eyes that was clinically diagnosed as herpes simplex keratitis. Herpes simplex virus was isolated from 24 eyes among 44 eyes(56.8%). The result of viral culture showed a high positive isolation rates in superficial keratitis(72.0%) and relatively low rates in stromal keratitis(36.8%). The positive isolation rate was high in cases of short duration after the onset of herpes simplex keratitis. It is suggested that virus isolation with viral culture method is a relatively sensitive, simple, and highly specific method for the diagnosis of herpes simplex keratitis.

ABSTRACT

The cultured cells were derived from rabbit corneal stroma by explant technique following microdissection and serial passage. The ultrastructural features of fourteenth-passage keratocytes were examined with both scanning and transmission electron microscope. The cells can be divided into activated, intermediate and old cell according to the differences in electron density and surface microvillous pattern. The morphologic characteristics of the cultured keratocytes partially resemble those shown in corneal keratocytes in vivo.
