ABSTRACT

Objective To discuss the ultrasonic features and diagnostic value of high frequency ultrasound in the diagnosis of the injury of triangular fibrous cartilage complex (TFCC) of wrist joint.Methods The ultrasonic features of 32 patients with pathology confirmed unilateral TFCC injury were retrospectively analyzed.The preoperative MRI and ultrasound data were complete.The difference between the two methods in diagnosis of TFCC injury were contrasted.Results The major ultrasonic features of the injure of TFCC included form swelled,structures disorganized,inhomogeneous echoes,irregular echoless in damaged place.The flow signals could be displayed from the tear opening edge of the injured TFCC by CDFI.Among 32 cases of TFCC injury,there were 26 cases (26/32,81.25%) diagnosed by ultrasound and 27 cases (27/32,84.38%) diagnosed by MRI.There was no significant difference in the detection rate of the two methods for TFCC injury (x2 =0.143,P=0.706).Conclusion High frequency ultrasound can be used to diagnose the injury of TFCC accurately,and can provide imaging evidence simply and rapidly for clinical therapy.

ABSTRACT

Objective To evaluate the differential diagnosis value of shear wave elastography for Hashimoto's disease with benign or malignant thyroid nodules.Methods Fifty four patients of Hashimoto's disease confirmed by biopsy or pathology were enrolled.Based on benign or malignant of nodules,patients were divided into benign nodules group and malignant nodules group.And the benign group were divided into true nodules and sham nodules.Young's modulus values were measured and compared among different groups.The ROC curve was drawn to evaluate the diagnostic efficency.Results There were tatol 162 nodules in 54 cases with Hashimoto's disease,including 131 of benign nodules (93 of true nodules,38 of sham nodules) and 31 of malignant nodules.The values of Young's modulus for benign,malignant,ture nodules and sham nodules were (33.04±10.95)kPa,(66.15±19.17)kPa,(35.95±9.74)kPa and (30.13±11.87)kPa.For mean values of Young's modulus,there was significantly statistical difference between benign and malignant nodules (t=12.91,P<0.01).There was significant statistical difference of the Young's modulus among sham nodule group,true nodule group and malignant nodule group (F 36.20,P<0.01).Compared with the other two groups,the value of Young's modulus in malignant nodules was significantly higher than that in the other two groups (both P<0.01).Taking 43.65 kPa as the diagnostic point,the sensitivity,specificity,positive likelihood ratio,and negative likelihood ratio of Youngs modulus in diagnosis of benign and malignat nodules were 90.63%,81.25%,4.83,and 0.12.Conclusion Shear wave elastography technique can quantitative analysis the Hashimoto's disease and complicated nodules,which is helpful to determine the essence of the nodules.

ABSTRACT

Objective To evaluate the effect of ultrasound elastography and radiography in the assessment of hepatic fibrosis in chronic hepatitis B patients. Methods Seventy patients with chronic hepatitis B were en-rolled in our hospital from August 2014 to August 2016. Liver ultrasonography was performed in all patients. HAVTT1 and HAVTT2 were measured by visual method. SWV was measured by ultrasonic elasticity imaging. Re-sults With the increase of liver fibrosis,SWV test also increased,while HAVTT gradually shortened.The correct rates of HAVTT1 and HAVTT2 and SWV in the diagnosis of liver fibrosis were 72.36%,76.84% and 89.78%.The diagnostic accuracy of SWV was higher than that of HAVTT1 and HAVTT2,the difference was statistically sig-nificant(P<0.05).Conclusion Acoustic contrast measurement of HAVTT1 and HAVTT2 can reflect the chang-es of liver microcirculation,which is helpful to distinguish liver cirrhosis,fibrosis and no fibrosis. The measure-ment of SWV can reflect the difference of texture hardness of liver,which can be used as a kind of judgment on the degree and effect of hepatic fibrosis new method.

ABSTRACT

Objective To study the expressions of osteopontin (OPN),caspase-3 and mt-P53 proteins, and their relationship in gliomas. Methods Seventy gliomas specimens of patients (glioma group) were selected, and 10 samples of non-glioma brain tissue were used as control group. The SP method was used to detect the positive rates of protein expressions of OPN, caspase-3 and mt-P53 between two groups. The relationship between protein expressions of OPN, caspase-3 and mt-P53 in gliomas and grade of gliomas were detected by Western blot assay. Spearman rank correlation was compared between the positive expression of OPN, caspase-3 and rate mt-P53. Results The positive expression rates of OPN and mt-P53 were significantly higher in glioma group (64.29%and 60%) than those of control group (no positive expression), but the positive expression rate of caspase-3 was significantly lower than that of control group (47.14%vs. 90%, P0.05). The higher the WHO classification, the higher the positive expression rates of OPN and mt-P53 (P<0.001), and the lower the positive expression rate of caspase-3 (P<0.001). With the increased level of glioma grade, OPN and mt-P53 protein levels were increased, but caspase-3 protein expression level was decreased. There was a negatively correlation between OPN and the positive expression of caspase-3, but there was a positive correlation between OPN and the expression of mt-P53 (rs=-0.720 and 0.722, P<0.05). There was a negative correlation between caspase-3 and mt-P53 expressions (rs=-0.556, P<0.05). Conclusion The higher the WHO classification, the higher the positive expression rates of OPN and mt-P53, while the lower the positive expression rate of caspase-3. The study reveals that OPN, caspase-3 and mt-P53 expressions are associated with the occurrence and the progress of gliomas. The combined detection of them can contribute to the judgment of biological behavior of gliomas.

Objective:Peptides were designed on the basis of high conservative regions of amino acid sequences and structures of the SEs,three-dimension structure of P72 was constructed.Methods: Bioinformatics analysis softwares such as Vector NTI 10.3, InsightII 2000,Discovery Studio 1.7 were used to analyse and predict the space structure of P72.Results:three-dimensional domains of the peptide P72 from SEA, SEB and SEC were quite similar, Peptide P72 was far away from TCRVβchain and MHC class II molecule.Conclusion:The inhibitory activity of peptide P72 may not due to binding to MHCⅡ and TCRVβchain.The exact mechanism of inhibitory activity of P72 should be explored.

ABSTRACT

Objective This research is aimed to investigate the efficacy and toxicity of icotinib for lung adenocarcinoma pa-tients with poor performance status and unknown EGFR gene status .Methods A total of 27 lung adenocarcinom patients with poor Eastern Cooperative Oncology Group-Performance status(ECOG-PS) and unknown EGFR gene status referred to Chongqing Canc-er Institute from August 2012 to August 2014 were analyzed .Icotinib (125 mg) was orally administered three times per day .Asess the efficacy and adverse reaction ,calculate survival rates .Results Among the 27 patients ,the objective response rate(ORR) and disease control rates(DCR) were 29 .6% and 81 .5% ,respectively .The median progression free survival time was 6 .0 months .A to-tal of 70 .4% of patients had an significant improvment in ECOG-PS scores ,following icotinib treatment (Z= - 2 .157 ,P= 0 .031) . Fatigue ,anorexia and diarrhea were the most frequent adverse reaction ,which defined as grade 1 to 2 rashes .Conclusion Lung ade-nocarcinoma patients with poor performance status and unknown EGFR gene status may benefit from icotinib therapy ,and patients were tolerated well .

ABSTRACT

Objective To analyze the clinical characteristics of malignant tumor complicating herpes zoster.Methods Fifty-seven cases of malignant tumor complicating herpes zoster in our hospital from September 2003 to November 2014 were performed the retrospective analysis on the anti-tumor treatment history and the clinical characteristics of herpes zoster.Results Most of the cases were over 60 years old.Herpes zoster had the high incidence rate within 2 years of operation and radiochemotherapy.The on-set sites of herpes zoster were most commonly in the chest and back.Conclusion The factors of advanced age,surgery,radiochemo-therapy,poor nutritional status,etc.aggravate or promote the occurrence of herpes zoster in the patients with malignant tumor.

ABSTRACT

Objective:To observe the effect of the proteininhibitor of activated STAT 3 (PIAS3) on the proliferation and apopto-sis of U251 glioma cells after PIAS3 expression was inhibited by RNAi. Methods: Three RNAi expression vectorstargeting PIAS3 were constructed and transfected into CHG-5 cells by liposomein vitro. The most efficient RNAi vector was subsequently selected by examiningthe mRNA expressions of PIAS3 in the transfected cells by semi-quantitativeRT-PCR. The selected RNAi vector was then transfected into U251 cells. After 48h of transfection, the mRNA and protein expressions of PIAS3 in glioma cellswere examined by semi-quantitative RT-PCR and western blot. Apoptosis wasobserved by flow cytometry using a double-staining method with FITC-con-jugatedannexin V and PI. Flow cytometry was also applied in cell cycle assay. Results:RNAidownregulated the mRNA (P<0.01) and protein (P<0.01) expressionsof PIAS3 in transfected cells.RNAi promoted the resistance of U251 cells to apoptosisand subsequently al-tered the cell cycle. A high percentage of G2 phaseand a low percentage of Sphase were observed in U251 cells. Conclusion:The down-regulation of PIAS3arrested U251 cells in the G2 phase andinduced the resistance of U251 cells to apoptosis.

ABSTRACT

Objective To explore the feasibility of enhanced green fluorescent protein (EGFP) transfection into the joint synovial tissues of rheumatoid arthritis (RA) rats by ultrasound-mediated microbubble destruction.Methods Twenty-eight normal rats were established the RA rat model,four rats were control group,twenty-four rats were categorized into four experimental groups:EGFP,ultrasound +EGFP,microbubbles + EGFP,and ultrasound + microbubbles + EGFP.The last group was irradiated with ultrasound for 10 min after the mixture consisting of 300 μl Sono Vue and 10 μg EGFP was injected into the joint cavity.The rats were sacrificed after 3 days and the joint synovial tissues were collected for EGFP observation under fluorescence microscopy and quantitative analysis by real-time polymerase chain reaction (RT-PCR).Results Comparing with control group,EGFP expression was observed in the rat joint synovial tissues from all groups.However,a strong EGFP expression was observed in the ultrasound + microbubbles +EGFP group.EGFP expression had no statistically significant difference (the P values were 0.89,0.93,and 0.82,respectively,P > 0.05) in the EGFP,ultrasound + EGFP and microbubbles + EGFP groups.However,EGFP expression in the EGFP,ultrasound + EGFP,microbubbles + EGFP groups significantly differed (all P values were <0.01) from that in the ultrasound + microbubbles + EGFP group.Conclusions Ultrasound-mediated microbubble destruction can improve EGFP transfection efficiency into the joint synovial tissues of RA rats.

ABSTRACT

Objective To observe the bioeffects on the normal rats synovial with diagnostic ultrasound combined with different doses contrast medium. Methods Twenty normal cleaning level Wister rats were divided into 5 groups (each with 4 rats and 8 knees): ultrasound+100 μl contrast medium, ultrasound+200 μl contrast medium, ultrasound+300 μl contrast medium, ultrasound+500 μl contrast medium, a simple ultrasound irradiation (control group). Each group was irradiated with diagnostic ultrasound for 10 min. Rats were sacrificed after 1 week and joints synovial tissues were observed with pathological HE staining. Results In group ultrasound+100 μl contrast medium, ultrasound+200 μl contrast medium, ultrasound+300 μl contrast medium and control group, no significant damage in synovial tissues was found. In group ultrasound+500 μl contrast medium, synovial cells swelling, showing round, oval or polygonal, synovial tissues hyperplasia, arranged disorder, capillary proliferation, some muscle fiber structure dissolved fracture, inflammatory cell infiltration in the surrounding tissue were found. Conclusion Ultrasound combined with contrast medium can significantly enhance the bioeffects of synovial tissues, and the more the contrast medium doses, the more serious injury occurs in synovial tissues.

@#Objective To use the multi-slice CT(MSCT)cerebral perfusion imaging and CT angiography(CTA)in the clinical diagnosis of transient ischemic attack(TIA).Methods 20 cases with transient ischemic attack within 48 h were examined with 16-slice CT perfusion imaging(CTPI)and CTA in head and neck.The interesting regional cerebral blood flow(rCBF),cerebral blood volume(rCBV)and the time to peak(rTP)were analyzed.Results The rTP of affected side delay compared with that of the healthy(t=3.04,P=0.008).CTA showed the responsible vascular stenosis or occlusion in varying degrees.There was not significant correlationship between rTP and vascular stenosis(r=0.32,P=0.17).Conclusion Cerebral perfusion imaging and head and neck CTA can provide valuable information for diagnosis and treatment of TIA.

ABSTRACT

Objective: To construct a liver targeting gene transfer vector using hepatitis B virus envelope particles. Methods: Hepatitis B viruses were obtained from the supernatant of HepG 2.2.15 cells by a PEG8000 system and were inactivated by ?-propiolactone to prepare hepatitis B virus envelope. The hepatitis B virus envelope was used to pack 5.3 kb pIRES_2-EGFP to assess their packing ability. Subsequently, the products were studied with ELISA, PCR, SDS-PAGE, and electron microscopy. Finally, the product was used to transfect HepG2 cells and the green fluorescent protein (GFP) expression was observed under a fluorescent microscope. The rate of GFP positive cells was determined by flow cytometer.Results: The acquired hepatitis B virus envelope retained the surface protein HBsAg+pre S_1+pre S_2, but with no virus DNA. The prepared envelope had high packing ability for GFP and the packed GFP had a high transfection rate in HepG2 cell. Conclusion: Hepatitis B virus envelope has been successfully obtained from the supernatant of HepG 2.2.15 cells with a PEG8000 system and ?-propiolactone.

ABSTRACT

<p><b>BACKGROUND</b>Endostar™ (rh-endostatin, YH-16) is a new recombinant human endostatin developed by Medgenn Bioengineering Co. Ltd., Yantai, Shandong, P.R.China. Pre-clinical study indicated that YH-16 could inhibit tumor endothelial cell proliferation, angiogenesis and tumor growth. Phase I and phase II studies revealed that YH-16 was effective as single agent with good tolerance in clinical use.The current study was to compare the response rate , median ti me to progression (TTP) ,clinical benefit andsafety in patients with advanced non-small cell lung cancer ( NSCLC) , who were treated with YH-16 plus vi-norelbine and cisplatin (NP) or placebo plus NP.</p><p><b>METHODS</b>Four hundred and ninety-three histologically or cy-tologically confirmed stage IIIB and IV NSCLC patients , withlife expectancy > 3 months and ECOG perform-ance status 0-2 , were enrolledin a randomized ,double-blind ,placebo-controlled , multicenter trial ,either trialgroup : NP plus YH-16 (vinorelbine 25 mg/m² on day 1 and day 5 ,cisplatin 30mg/m² on days 2 to 4 , YH-167.5mg/m² on days 1 to 14) or control group : NP plus placebo (vinorelbine 25 mg/m² on day 1 and day 5 ,cis-platin 30 mg/m² on days 2 to 4 ,0.9% sodium-chloride 3 .75 ml on days 1 to 14) every 3 weeks for 2-6 cycles .The trial endpoints included response rate ,clinical benefit rate ,time to progression,quality of life and safety .</p><p><b>RESULTS</b>Of 486 assessable patients , overall response rate was 35.4% in trial group and 19.5% in controlgroup (P=0 .0003) . The median TTP was 6 .3 months and 3 .6 months for trial group and control group respectively (P < 0 .001) . The clinical benefit rate was 73 .3 %in trial group and 64.0% in control group (P=0 .035) .In untreated patients of trial group and control group ,the response rate was 40 .0% and 23.9%(P=0 .003) ,the clinical benefit rate was 76 .5 % and 65 .0 % (P=0 .023) ,the median TTP was 6 .6 and 3 .7months (P=0 .0000) ,respectively .In pretreated patients of trial group and control group ,the response ratewas 23.9% and 8.5%(P=0 .034) ,the clinical benefit rate was 65.2% and 61.7%(P=0 .68) ,the median TTP was 5 .7 and 3 .2 months (P=0 .0002) ,respectively . The relief rate of clinical symptoms in trial groupwas higher than that of those in control group ,but no significance existed (P > 0 .05) . The score of quality oflife in trial group was significantly higher than that in control group (P=0 .0155) after treatment . There were no significant differences in incidence of hematologic and non-hematologic toxicity , moderate and severe sideeffects betweentrial group and control group .</p><p><b>CONCLUSIONS</b>The addition of YH-16 to NP regimen results in significantly and clinically meaningful improvement in response rate , median time to tumor progression,and clinical benefit rate compared with NP alone in advanced NSCLC patients . YH-16 in combination with chemotherapy shows a synergic activity and a favorable toxic profile in advanced cancer patients .</p>

Objective To evaluate the effects of 0.3 kb human alpha-fetoprotein (AFP) promotor on the expression of enhanced green fluorescent protein (EGFP) in human hepatoma cells. Methods By constructing the primer with special enzyme site, human AFP promoter was selectively amplified from human gene bank and eukaryon-expressed pAF-EGFP and pAF-DTA with AFP promotor and EGFP and with AFP promotor and DTA, respectively. pAF-EGFP was transfected into HepG2, SMMC-7721, and NIH3T3. ABSTRACT

To study the effects of MRF4 transfection on differentiation and expression of myogenic regulatory factors of human rhabdomyosarcoma RD cells, the plasmid-MRF4 cDNA was transfected into cultured rhabdomyosarcoma RD cells with lipofectin method. The myogenic regulatory factors MRF4 and MyoD mRNA were measured with in situ hybridization and the expressions of myosin heavy chain(MHC) and a-actin in the cells were assayed with immunocytochemical method. The cell growth and morphology were observed at the same time. It was found that the morphology of differentiation increased and the growth was suppressed in RD cells after transfection. The expression of MHC and a-actin were significantly increased in RD cells after transfection, while the expressions of MRF4 and MyoD mRNA were up-regulated. It is suggested that transfection of MRF4 can induce differentiation of RD cells and up-regulate the expression of MyoD.

Background and purpose:The reason for hepatitis B virus (HBV) with hepatocyte specificity is PreS1 enchased on the hepatitis B virus envelope (HBVE). So HBVE may have a potential application in liver targeting gene transfer. In this study, we investigated whether HBVE has the ability to target liver cancer cells. Methods:HBVE was obtained from the supernatant of Hep G 2.2.15 cells through PEG8000 system and ?-propiolactone method. The pIRS2-EGFP was packed with HBVE and resulted in the product HBVE-GFP. HBVE-GFP was transfected into HepG2, A549, HeLa and FB cells. The green fluorescent protein (GFP) expression was observed under a fluorescent microscope. The rate of GFP positive cells was determined by flow cytometer.Results:The GFP could be observed in the four groups, but the HepG2 group had a higher fluorescent intensity than the other 3 groups. Objective To construct an expression vector containing Ewing's sarcoma EWS FLI 1 specific binding sequence and diphtheria toxin A chain (DTA) gene sequence to investigate the killing effect of DTA on cultured Ewing's sarcoma cells so as to provide proof for the exploration of new methods for gene therapy for cancer. Methods Expression vector (pS2 DTA) containing EWS FLI 1 specific binding sequence and DTA gene sequence was constructed by replacing the luciferase gene in pS2 with DTA gene at the points of Nco Ⅰ and Xba Ⅰ by molecular technique. After transfection of pS2 DTA into Ewing's sarcoma cells and the control cells, DTA expression and its killing effect on cells were detected. Results pS2 DTA was highly expressed in Ewing's sarcoma cell line, and the killing effect of DTA was much higher than that in the control cells. Conclusion pS2 DTA has selective killing effect on Ewing's sarcoma cells and can inhibit the growth of Ewing's sarcoma cells. So pS2-DTA has potential value in the treatment of Ewing's sarcoma.

After electrical stimulation of the phrenic nerve,the function of the diaphragm was assessed by measurement of the transdiaphragmatic pressure (PDi),establishment of the Pdi-sti-mulating frequency (F) curve,and analysis of the power spectrum on the diaphragmatic elec-tromyogram (EMGdi) at spontaneous breathing.Hypercapnia of the dogs was induced by forcing the animals to inhale a mixture of CO2 and air.The findings were as follows:(1) After acute hypercapnia,PDi rapidly and progressively decreased.The power spectrum of EMGdi was also reduced.The PDi-F curve was shifted to right.These findings suggest that the function of the diaphragm was severely damaged.(2) The decrease of pH value was the important cause of diaphragmatic fatigue.(3) The diaphragm in a static condition could reduce the damage of hypercapnia.(4) Cedi-lanid exerted certain effects on the prevention of diaphragmatic fatigue created by acute hypercapnia.
