ABSTRACT

The present study evaluated the volume and function of the left atrium by two-dimensional echocardiographic feature-tracking imaging (2D-FTI) and Simpson's monoplanar modeling in dogs with asymptomatic degenerative mitral valve disease (DMVD). The study consisted of 80 dogs that were divided into the following three groups: Group 1, 21 dogs (A); Group 2, 30 dogs (B1) and Group 3, 29 dogs (B2). The variable strain (contraction phase) was significantly lower in Group 3 than in Group 1 (12.92±4.54 x 16.69±5.74, p=0.014), and significant differences in the contraction strain index (CSI) were observed between all of the groups that were evaluated (1 = 46.82±8.10, 2 = 39.88±8.03, 3 = 35.25±5.64, p<0.0001). The atrial diastolic volume index (AdVi) that was measured by 2D-FTI was significantly higher in Group 3 than in Group 1 (1.31±0.95 x 0.96±0.31, p=0.038), and the atrial cardiac index (ACI) was also higher in Group 3 than in Group 1 (102.38±80.18 x 78.19±33.38, p=0.030). Atrial function was assessed by Simpson's monoplanar method, which demonstrated an increase in the left atrial systolic volume, while the contractile function decreased with an increasing disease severity (Group 1 0.21±0.06; Group 2 0.25±0.06; Group 3 0.32±0.08, p<0.0001). The intraobserver and interobserver assessments showed low to moderate variability; most of the values for the coefficient of variation for the variables that were analysed with each method were below 25%. Thus, DMVD was determined to cause an alteration in atrial function, especially in the contraction phase, and even in asymptomatic animals, and the methods of 2D-FTI echocardiography and Simpson's monoplanar evaluation are sensitive and early methods for the detection of left atrial dysfunction.(AU)


O presente estudo avaliou o volume e a função atrial esquerda obtidos por meio da ecocardiografia bidimensional feature tracking (2D-FTI) e pelo método monoplanar de Simpson em cães saudáveis e cães com DMVD assintomáticos. Foram avaliados 80 cães distribuídos em três grupos: Grupo 1, 21 cães (classe A); Grupo 2, 30 cães (classe B1) e Grupo 3, 29 cães (classe B2). A variável strain (fase de contração) foi significativamente menor no Grupo 3 que no Grupo 1 (12,92±4,54 x 16,69±5,74, p=0,014) e para a variável índice de strain de contração (CSI), houve diferença estatística entre todos os grupos avaliados (1 = 46,82±8,10; 2 = 39,88±8,03; 3 = 35,25±5,64, p<0,0001). O índice de volume diastólico atrial (iVdA) mensurado por meio do 2D-FTI foi significativamente maior no Grupo 3 que no Grupo 1 (1,31±0,95 x 0,96±0,31, p=0,038), assim como para o índice cardíaco atrial (iCA) também foi maior no Grupo 3 (102,38±80,18 x 78,19±33,38, p=0,030). A função atrial avaliada pelo método monoplanar de Simpson demonstrou um aumento do volume atrial esquerdo e do volume sistólico do átrio esquerdo, enquanto que a função contrátil diminuiu com o aumento da gravidade da doença (Grupo 1 0,21±0,06; Grupo 2 0,25±0,06; Grupo 3 0,32±0,08; p<0,0001). A avaliação intraobservador e interobservador, demonstrou variabilidade baixa a moderada, uma vez que a maioria dos valores de coeficiente de variação se concentraram abaixo de 25% para as variáveis analisadas em ambos os métodos. Dessa forma, conclui-se que a DMVD causa alteração na função atrial, principalmente na fase de contração, mesmo em animais assintomáticos e que a ecocardiografia 2D-FTI e o método monoplanar de Simpson são métodos sensíveis e precoces na detecção da disfunção atrial esquerda.(AU)

ABSTRACT

The objective of this study was to evaluate the adherence to and invasion of HeLa cells by Campylobacter spp. strains (total n=63) isolated from chickens (n=4), dogs (n=4), non-human primates (n=16), pigs (n=9), calf feces (n=18), and bovine genital tracts (n=12). Thirty-two strains adhered to and 13 invaded HeLa cells. Invasive strains included 1 of 4 dog isolates, 4 of 16 non-human primate isolates (2 C. jejuni and 2 C. coli), 1 of 9 C. coli strains isolated from pigs, and 7 of 18 C. fetus subsp. fetus isolated from calf feces. Only 25% of chicken and dog isolates and 23% of pig isolates were able to adhere to HeLa cells, a property of 65% of strains obtained from calf feces and 83% of bovine genital tract-isolated strains. The adherent phenotype was observed in 5 of 19, 6 of 15, and 21 of 29 strains of C. jejuni, C. coli, and C. fetus subsp. fetus, respectively, whereas 3 of 19, 3 of 15, and 7 of 29 strains were additionally able to invade HeLa cells, respectively. C. jejuni, C. coli, and C. fetus subsp. fetus strains isolated from animal feces are able to adhere and invade HeLa cells, whereas C. fetus subsp. fetus strains isolated from the bovine genital tract were not invasive in HeLa cells. The present study showed that C. jejuni isolated from primates and dogs, C. coli isolated from non-human primates and pigs, and C. fetus subsp. fetus isolated from calf feces have the ability to adhere to and to invade HeLa cells. Moreover, the lack of invasive ability by C. fetus subsp. fetus strains isolated from the bovine genital tract could be important in the pathogenesis of the genital tract diseases caused by this bacterium.(AU)


O objetivo deste estudo foi avaliar a adesão e invasão de células HeLa por amotras de Campylobacter spp. (total n=63) isoladas de frangos (n=4), cães (n=4), primatas não-humanos (n=16), porcos (n=9), fezes de bezerros (n=18), e trato genital de bovinos (n=12). Trinta e duas amostras foram capazes de aderir e 13 invadiram células HeLa. As amostras invasivas incluíram 1 de 4 isolados de cão, 4 de 16 isolados de primatas não-humano (2 C. jejuni e 2 C. coli), 1 de 9 C. coli isoladas de porcos e 7 de 18 C. fetus subsp. fetus isoladas de fezes de bezerros. Apenas 25% dos isolados de frango e de cão e 23% dos isolados de suínos foram capazes de aderir a células HeLa, propriedade exibida por 65% das cepas obtidas a partir de fezes de bezerros e por 83% das amostras isoladas de trato genital bovino. O fenótipo aderente foi observado em 5 de 19, 6 de 15 e 21 de 29 amostras de C. jejuni, C. coli e C. fetus subsp. fetus, respectivamente, enquanto que 3 de 19, 3 de 15 e 7 de 29 amostras foram adicionalmente capazes de invadir as células HeLa, respectivamente. Amostras de C. jejuni, C. coli e C. fetus subsp. fetus isoladas de fezes de animais foram capazes de aderir e invadir as células HeLa, enquanto amostras de C. fetus subsp. fetus isoladas a partir de amostras de trato genital bovino não foram invasivas, em células HeLa. O presente estudo mostrou que amostras de C. jejuni isoladas de primatas não-humanos e cães, C. coli isoladas de primatas não-humanos e porcos, e C. fetus subsp. fetus isolados a partir de fezes de bezerros foram capazes de aderir e invadir células HeLa. Além disso, a falta de capacidade invasiva de amostras de C. fetus subsp. fetus isoladas de trato genital bovino pode ser importante na patogênese das doenças das vias genitais causadas por esta bactéria.(AU)

ABSTRACT

The aim of this study was to evaluate the growth of the B. abortus reference strains and field isolates on media containing different inhibitor agents. Reference strains were seeded on tryptose agar containing: i-erythritol (1.0 mg/mL), fuchsin (20 μg/mL and 80 μg/mL), thionin (2.5 μg/mL and 10 μg/mL), rifampicin (200 μg/mL) and safranin O (200 μg/mL). Field isolates were tested only on media containing i-erythritol, rifampicin and thionin. Furthermore, each suspension was also inoculated on tryptose agar incubated in air, to test its ability to grow without CO2. Sensitivity to fuchsin was similar among reference strains evaluated. Growth of S19, 544 and 2308 but not RB51 were inhibited on media containing rifampicin. Medium with safranin O showed no inhibition for RB51, 544 and 2308, but it partially inhibited the S19 growth as well as medium containing i-erythritol. Treatment/control growth ratio for 2308 on tryptose agar containing thionin (2.5 μg/mL) was approximatelly 1.0, whereas S19 and RB51 showed 0.85 and 0.89 ratios, respectively. Growth of 544, S19 and RB51 but not 2308 was completely inhibited on medium with thionin (10 μg/mL). All field strains grew on medium containing i-erythritol, but were completelly inhibited by rifampicin. With exception of A1 (B. abortus biovar 3) all field isolates grew on medium with thionin, although some strains showed a treatment/control growth ratio of 0.75–0.80 (10 μg/mL). These results showed that tryptose agar with thionin, i-erythritol or rifampicin could be useful for differentiating vaccine, challenge and field strains of B. abortus.

ABSTRACT

Foram identificadas diferenças no perfil protéico das amostras de Sponselee, Norma e Hardjoprajitno, com bandas protéicas entre 175, 47 kDA e 12,10 kDa. A amostra Sponselee foi a que apresentou maior número de bandas (12), seguida da Norma que apresentou 11 bandas e de Hardjoprajitno que apresentou 9 bandas. Todas as bandas observadas na amostra Sponselee possuíam correspondentes nas outras duas amostras. A amostra Norma não apresentou uma banda em torno de 35,77 kDa e a Hardjoprajitno não apresentou bandas em torno de 89,59 kDa, 35,77 kDa e 12,10 kDa. O reconhecimento dessas proteínas por soros hiperimunes contra cada uma das amostras também mostrou diferenças, sendo que o maior número de proteínas reconhecido em todas as amostras por todos os soros se encontrou entre 35,83 kDa e 29,19 kDa. Os soros contra bovino na amostra Norma só reconheceu proteínas de baixa massa molecular nas amostras Norma (6,80 kDa) e Hardjoprajitno (6,80 kDa e 5,30 kDa). Soro bovino contra a amostra Hardjoprajitno reconheceu uma proteína de 44,33 kDa todas às amostras e proteínas de 4,22 kDa nas amostras Sponselee e Norma e de 10,49 kDa e 6,16 kDa na Hardjoprajitno. As diferentes proteínas identificadas poderiam se constituir em alvos específicos para o desenvolvimento de testes diagnósticos e vacinas contra leptospirose bovina.


Differences in the protein profile of Leptospira sp. strains Sponselee, Norma and Hardjoprajitno were observed, with bands ranging from 175.47 kDa to 12.10 kDa. Strain Sponselee presented a 12-band profile, while strain Norma showed 11 and strain Hardjoprajitno showed 9 bands in the profile. All bands observed in Sponselee strain profile could match bands in the other two strains. Strain Norma lacks a band at 35.77 kDa and strain Hardjoprajitno lacks the bands at 89.59 kDa, 35.77 kDa and 12.10 kDa. The recognition profile from hyperimmune sera was also different for the studied serovar Hadjo strains. The majority of recognized proteins was in the range of 35.83 kDa to 29.19 kDa. Cattle sera against strain Norma only recognized low molecular mass proteins in strains Norma (6.80 kDa) and Hardjoprajitno (6.80 kDa and 5.30 kDa). Bovine sera against strain Hardjoprajitno recognized a 44.33 kDa protein in all studied strains and proteins of 4.22 kDa in strains Sponselee and Norma and of 10.49 kDa and 6.16 kDa in strain Hadjoprajitno. The different identified proteins could become specific targets to the development of diagnostic tests and vaccines against bovine leptospirosis.
