ABSTRACT

ObjectiveTo form a questionnaire of high reliability and validity on mate preferences of postgraduates in military academy. MethodsIn December 2020, full-time postgraduates of a military academy were randomly selected as research objects, and their mate preferences were investigated via a self-compiled open-ended questionnaire. The item screening and expert consultation were carried out to compile an initial questionnaire on mate preferences of the postgraduates in military academy, and the questionnaire survey was conducted on 89 postgraduates in the military academy. Then the item analysis and exploratory factor analysis were performed to test the questionnaire, and the reliability and validity of the questionnaire were tested by calculating Pearson correlation coefficient, Cronbach's α coefficients and split-half coefficient. ResultsThe questionnaire on mate preferences of postgraduates in military academy has been formed, which consisting of 4 factors (intrinsic quality, personal ability, external conditions and quality of life) and 25 items was formed. The correlation coefficient between the total score of the questionnaire and the score of each factor ranged from 0.739 to 0.864, Cronbach's α coefficient of total score and each factor ranged from 0.723 to 0.926, and the split-half coefficient was between 0.682 and 0.899. ConclusionThe compiled questionnaire on mate preferences of postgraduates in military academy has good credibility and validity, and can be used as a tool to collect the data of psychological marriage and love on the mate preferences of postgraduates in military academy.

ABSTRACT

Objective: To explore the clinical value of ultrasound-guided radiofrequency ablation in the treatment of retroperitoneal tumors. Methods: The clinical data of 13 patients with retroperitoneal tumors treated with ultrasound-guided radiofrequency ablation in the First Affiliated Hospital of Zhengzhou University from January 2018 to January 2020 were analyzed retrospectively. The ablation effect was evaluated and the postoperative complications were observed. The changes of tumor volume before and after radiofrequency ablation were compared. Results: The symptoms of pain and dyspepsia were significantly improved after radiofrequency ablation, and the hospital stay was (9.2±2.9) days. The tumor was ablated completely in 10 cases, tumor residual in 1 case and tumor metastasis in 2 cases. One patient had postoperative duodenal perforation complicated with intra-abdominal infection, and no serious complications occurred in other patients. There were 20 lesions in 13 patients. The maximum diameter of 20 lesions before operation and 1, 3, 6 months after operation were (39.5±15.9) mm, (30.6±4.9)mm, (15.6±7.7) mm and (9.9±3.1) mm, respectively, the maximum diameters of 1, 3 and 6 months after operation were smaller than that before operation (P<0.05). Conclusion: Ultrasound-guided radiofrequency ablation is a real-time, accurate, safe and effective minimally invasive treatment with few complications, and has a high clinical value for retroperitoneal tumors.

ABSTRACT

Esophageal cancer is a digestive tract malignancy with high morbidity and mortality and mainly occurs in males. The 5-year survival rate is lower than 20%. In China, the morbidity and mortality of esophageal cancer rank the first in the world, seriously threatening national health. The pathogenesis of esophageal cancer is diverse, which is generally considered as the consequence of environmental-genetic-gene interaction. In addition to genetic factors and regional characteristics, gene mutation, RNA interference, DNA damage repair, tumor microenvironment, dietary habit, chronic adverse stimulation, and inflammatory reaction are all involved in the occurrence and development of esophageal cancer. However, there is no unified and accurate conclusion. Clarifying the exact pathogenesis of esophageal cancer is of great significance for its early screening, diagnosis, prevention, treatment, and prognosis. Surgery, radiotherapy, and chemotherapy are the three effective methods for the treatment of esophageal cancer. However, due to the atypical early symptoms, most patients have missed the best operation period when diagnosed, resulting in poor clinical prognosis. Moreover, radiotherapy and chemotherapy will cause side effects such as loss of appetite, low immune function, esophagitis, pneumonia, and malnutrition, which is not conducive to the prognosis and treatment maintenance of patients. With definite efficacies on esophageal cancer, traditional Chinese medicine (TCM), which is flexible and diverse in the treatment, can primarily or alternatively be involved in the treatment of esophageal cancer. TCM can eliminate postoperative complications and postoperative infections and relieve adverse gastrointestinal reactions, weakened immune function, and organ damage caused by radiotherapy and chemotherapy. It can enhance clinical efficacy and improve the quality of life of patients. Therefore, it is necessary to systematically summarize the clear pathogenesis or risk factors of esophageal cancer and review the clinical characteristics of TCM in the prevention and treatment of esophageal cancer to facilitate the early screening, diagnosis, and treatment of esophageal cancer and promote the application of TCM in the prevention and treatment of esophageal cancer and related adverse reactions.

ABSTRACT

ObjectiveTo explore the efficacy of Interpersonal and Social Rhythm Therapy (IPSRT) on first-episode adolescent depressive disorder, so as to provide references for the treatment. MethodsA total of 310 first-episode adolescents with depressive disorder who met the International Classification of Diseases, tenth edition (ICD-10) were included as the research objects. They were randomly divided into research group (n=155) and control group (n=155). The research group received IPSRT, while the control group received conventional psychological intervention. Before and 2 weeks after the intervention, all participants were asked to complete Hamilton Anxiety Scale (HAMA), Hamilton Depression Scale-24 item (HAMD-24) and Interpersonal Competence Questionnaire (ICQ). The sleep quality was evaluated by portable monitoring devices. ResultsAfter intervention, compared with the control group, HAMA score [(5.92±1.04) vs. (10.23±1.09), t=-10.310, P<0.01], time to fall asleep [(0.26±0.09)h vs. (1.29±0.78)h, t=-4.701, P<0.01] and time in bed[(10.19±0.99)h vs. (11.30±1.38)h, t=-2.353, P=0.027] in research group were significantly lower. In research group, active social interaction [(20.23±1.59) vs. (17.38±1.33), t=4.959, P<0.01], appropriate rejection [(14.77±1.17) vs. (11.77±1.48), t=5.740, P<0.01], self-disclosure [(20.92±1.66) vs. (16.15±1.46), t=7.780, P<0.01] and affective support [(21.00±1.53) vs. (18.85±1.99), t=3.092, P<0.01] of ICQ were significantly higher than those in control group. ConclusionThe implementation of IPSRT in first-episode adolescents with depressive disorder may help to improve sleep quality, adjust inappropriate psychological state and improve interpersonal skills.

ABSTRACT

Endoplasmic reticulum (ER) is an important organelle responsible for protein, steroid, lipid and carbohydrate synthesis and calcium-dependent signal transduction in eukaryotic cells. ER homeostasis is essential for normal cell function. ER homeostasis imbalance can induce ER stress (ERS), which participates in the occurrence and development of diseases of the digestive system, respiratory system, circulatory system, nervous system, reproductive system, and endocrine system, and affects body health. Among various diseases, cancers seriously endanger people′s health due to its high mortality rate, disability rate, and recurrence rate. Due to the survival characteristics of unlimited proliferation, tumor cells are often exposed to various internal and external stimuli such as hypoxia, ischemia, excessive proliferation, and starvation, which destroy intracellular protein balance and induce ERS to some extent for survival. ERS plays a major role in various tumors and has dual functions in the survival of tumor cells: promoting the survival of tumor cells by activating a series of adaptive responses, while inducing ERS-related apoptosis pathways, so as to promote tumor cell death and inhibit tumor growth and invasion. As multiple functions of ERS in tumors are reported, many scholars have tried to intervene in the progress of tumors from the perspective of ERS. The therapeutic effect of traditional Chinese medicine (TCM) on tumors has been widely recognized. TCM can participate in the regulation of tumors from many aspects, including ERS, chemoradiotherapy resistance, gastrointestinal adverse reactions caused by chemotherapy, postoperative recurrence and metastasis. Since there are few reports on the antitumor effect of TCM from the perspective of ERS, this paper expounds the influence of ERS on tumorigenesis and development and the progress of TCM intervention in tumor through ERS, in order to provide a new direction for tumor treatment.

ABSTRACT

Nanocrystal drugs have many advantages, such as no carrier materials, easy industrialization, diversified dosage forms, and can significantly improve the solubility and bioavailability of insoluble drugs, so many drugs have been on the market. The traditional nanocrystal preparation technology has the problems of low preparation efficiency and process limitation of the smallest achievable particle size. With the progress of pharmaceutical preparation technology, the preparation technology of nanocrystal drugs is constantly improving, and new preparation technologies are constantly emerging. The emergence of new technologies has greatly shortened the process time and makes it possible to prepare nanocrystal drugs with smaller particle diameters. In this paper, the preparation technologies of nanocrystal drugs, especially the new preparation technologies such as high gravity controlled precipitation, microfluidic reaction technology and various combination technologies, are reviewed from three aspects: "Top-down" technology, "Bottom-up" technology and combination technology. This article also prospects the development of new preparation technologies, hoping to provide reference for the related research of nano-preparations.

Due to the increasing incidence of central nervous system diseases,especially the increasing incidence and mortality of stroke,brain-targeted drug delivery has attached more and more attention. Nasal administration,as one of the ways of brain-targeted administration,can effectively make the drug delivered to the brain in a targeted way after by passing the blood-brain barrier,providing a new idea for the treatment of central nervous system diseases. Therefore,it is a promising administration way. In recent years,the treatment of encephalopathy by nasal administration of traditional Chinese medicine has become a hot topic in the research of traditional Chinese medicine. Ischemic stroke is one of the most important diseases endangering human health. Nasal administration has a history of thousands of years in treatment of stroke. Modern medical research has proved that there is a subtle connection between the nasal cavity and the brain,and the complex and ingenious structure of the nasal cavity provides the possibility for drugs delivery to the brain through the nose. Drug administration through nasal cavity has obvious advantages in treatment of central nervous system diseases represented by ischemic stroke. Nasal administration is characterized by non-invasion,low infection,rapid absorption and brain targeting. The author will expound the theoretical basis of brain targeting of nasal administration from the aspects of anatomy and physiology,and summarize the transport pathway of drugs through the nose into the brain,the in vitro and in vivo experimental research basis of the " nose-brain"pathway,and the clinical nasal administration of traditional Chinese medicine to prevent cerebral ischemia. It provides a reference for better research of drugs to prevent and treat cerebral ischemia injury through the " nose-brain"pathway and lays a foundation for further research of the " nose-brain" pathway.

ABSTRACT

Acute cellular rejection (ACR) remains a major concern after liver transplantation. Predicting and monitoring acute rejection by non-invasive methods are very important for guiding the use of immunosuppressive drugs. Many studies have shown that exosomes and their contents are potential biomarkers for various liver diseases. Here, we identify and validate the role of exosomes and galectin-9 in ACR after liver transplantation. Exosomes were isolated from three sets of paired patients, with and without ACR, and the proteins within the exosomes were isolated and identified. Candidate proteins were then validated using a tissue microarray containing resected liver samples from 73 ACR and 63 non-rejection patients. Finally, protein expression and clinical manifestations were included in Kaplan-Meier survival and Cox regression analyses. Circulating exosomes were isolated from ACR and non-rejection patients and characterized using transmission electron microscopy and western blotting for CD63/CD81. Western blotting experiments revealed higher levels of galectin-9 protein in circulating exosomes from ACR recipients. Immunohistochemical analysis of the tissue microarray showed that the expression of galectin-9 in resected liver was significantly higher in the ACR group than in the non-rejection group (P<0.05). Higher levels of galectin-9 expression in resected livers were associated with poorer prognosis (P<0.05). Exosome-derived galectin-9 may be a novel predictor of rejection and prognosis after liver transplantation.

ABSTRACT

Objective: To investigate the effect of dodder total flavone on polycystic ovary syndrome (PCOS) rat models induced by dehydroepiandrosterone (DHEA) combined with human chorionic gonadotropin (HCG). Method: Except the blank group, the remaining rats were injected with DHEA 0.06 mg·g-1 in the morning on the nucha and 1.5 U HCG in the afternoon for 21 consecutive days. On the 16th day after the modeling, the vaginal smear was performed to monitor the estrus cycle. Sixty rats with successful modeling were selected and divided into model group, dacin-35 group, and high, middle and low-dose dodder total flavonoids groups, with 10 rats in each group. On the day of grouping, drugs were given respectively to the drug treatment groups, and the blank group and the model group were given equal volume solvent. The drugs were given continuously for 21 days. Blood was collected from abdominal aorta 2 h later after the final administration, serum levels of testosterone (T), estrogen (E2), luteinizing hormone (LH) and follicle stimulating hormone(FSH) were measured; rats were put to death, and the ovaries at the same part of each rat were fixed in 10% formalin solution. htoxylin eosin (HE) staining was performed, and the morphological changes in the ovaries were observed by light microscopy; the same part of the ovary was taken, and androgen receptor (AR), B-cell lymphoma-2(Bcl-2) and Bcl-2-associated X protein(Bax) were measured in the ovary by immunohistochemistry to observe the expressions of relevant proteins in the ovary; the hypothalamus and pituitary were taken, and the expressions of gonadotropin-releasing hormone (GnRH) in hypothalamus and gonadorelin releasing hormone receptor (GnRHR) in pituitary were detected by immunohistochemistry. Result: PCOS rat model was successfully replicated. Serum levels of T, E2 and LH were significantly reduced, and FSH level of PCOS was significantly increased in each drug treatment group (PPPPPPConclusion: The PCOS rat model was successfully replicated. Total flavonoids of dodder may play a protective role in PCOS model rats by regulating the secretion of androgen, inhibiting the expression of ovarian apoptotic protein and impacting the hypothalamic-pituitary-ovary axis pathway.

ABSTRACT

Decoction pieces are the main body of traditional Chinese medicine (TCM) clinical medication. With the modernization of TCM medication, TCM formula granules have gradually developed and begun to take shape. Although there are processing forms of TCM injection and oral liquid, they only account for a small part of TCM market and are mainly taken orally. However, when oral administration of lipid soluble ingredients is conducted, the dissolution is limited and the absorption is relatively small. In addition, it has side effects of stimulating gastrointestinal tract, resulting in low tolerance of patients. Although there are topical preparations made by water extraction and alcohol extraction, most of them are crude preparations with random dosage, which results in the waste of lipid soluble components to a certain extent. Most of the lipid soluble components of TCM have good biological activities, such as schisandrin b, tanshinone, methyl chlorogenic acid, etc., but there is no suitable medicinal form. To make the best use of "medicine", it is necessary to systematically study the lipid soluble components of TCM. On the basis of fairly mature extraction, separation and collection processes, a medicinal form with definite pharmacological effect and beneficial to its absorption is developed based on lipid soluble components. TCM essential oil has anti-aging, anti-dementia, anti-oxidation and other activities, which has been widely used in clinical, cosmeceuticals, health products and other fields. However, the effect of single essential oil is limited. Compatibility can not only enhance the efficacy and reduce toxicity, but also expand the application range of essential oil. Therefore, formula essential oil can be used as a new form of TCM, and mainly for external use, supplemented by internal use, to make up for the disadvantages of crude preparations and random dosing for external use of TCM, as well as the disadvantages of orally absorbed less and stimulated gastrointestinal tract, to meet the needs of clinical medication, faster, more accurate, better and stronger play the effect of lipid soluble components.

ABSTRACT

Objective: To analyze the medication rules of traditional Chinese medicine (TCM) in treating type 2 diabetes mellitus (T2DM),and explore the mechanism of action.Method: The articlesabout TCM treatment for T2DM in CNKI and Wanfang Med Online were retrieved.The data was entered into Excel 2013 to set up a database,and the factor analysis,association rules and cluster analysis were used to analyze core medication combinations by SPSS Statistics 19.0 and SPSS Modeler 14.1.kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis was performed using BATMAN-TCM to reveal the mechanism of action.Result: A total of 92 prescriptions were included,the high frequency medicines were Astragali Radix.The major properties of TCM were cold and plain.The major flavors of TCM were sweet and bitter.The major meridian tropism of TCM was livermeridian.These high frequency medicines were deficiency-nourishing herbs,and blood circulation-invigorating and stasis-eliminating herbs.Totally 6 core medication combinations were mined through association rules.Seven common factors were extracted from factor analysis.Clustering analysis was used to get 6 kinds of herbs.KEGG pathway analysis revealed 24 biological pathways and 6 diseases relating to T2DM.Conclusion: The most commonly used Astragali Radix for treating T2DM.In clinical,it can be combined with Poria,Salviae Miltiorrhizae Radix et Rhizoma and Rehmanniae Radix for treating T2DM.At the same time,it can be combined with such herbs as deficiency-nourishing herbs,and blood circulation-invigorating and stasis-eliminating herbs.The mechanism of action of high-frequency herbs may be related to the targets of exophosphatase/phosphodiesterase (PDEs),adenosine A1 receptor gene 1(ADORA1),adenosine A2A receptor gene (ADORA2A),neuropetide second receptor (NPY2R),and endothelial carbon monoxide synthase (NOS3) by interfering with PI3K/Akt,starch and sucrose metabolism signaling pathways.

ABSTRACT

Polycystic ovary syndrome (PCOS) is a common gynecological reproductive endocrine and metabolic disorder disease. The clinical manifestations of PCOS are diversified, with and no ovulation, insulin resistance, hyperinsulinemia and polycystic ovaries as the main pathological and physiological changes, which can lead to non-ovulation infertility and ovulatory dysfunction infertility, cause complications, such as amenorrhea, diabetes, lipid metabolism and glucose metabolism disorder, and endanger the long-term health of the body. Traditional Chinese medicine (TCM) does not have the name of this disease, and classifies PCOS to the category of "amenorrhea", "infertility" and "abdominal mass kidney" according to its clinical manifestations. Compared with western medicine, TCM has a unique view in treating the disease, and believes that the disorders of liver, kidney and spleen functions jointly affect the occurrence and development of the disease. According to the literature review, kidney-deficiency syndrome is the main syndrome type of this disease, the kidney-tonifying method has achieved a good effect in clinic. Besides, the drug use is flexible and diversified, and both primary and secondary symptoms are taken into consideration. However, in the clinical application process, the application of kidney-tonifying method in PCOS has been affected by the thinking mode of "kidney-tonifying medicine for male diseases" to some extent. In view of this, the therapeutic effect of kidney-tonifying drugs in the treatment of PCOS is preliminarily demonstrated from several aspects of the relationship between kidney deficiency and reproduction, pathogenesis of PCOS, clinical research and experimental research of PCOS, in the expectation of providing theoretical and practical supports for kidney-tonifying therapy for PCOS and other gynecological diseases.

ABSTRACT

Objective: To investigate the effect of dodder total flavone on polycystic ovary syndrome (PCOS) rat models induced by letrozole. Method: Except the blank group, the other rats were given letrozole 1 mg·kg-1 for 21 consecutive days to replicate PCOS animal model. On the 16th day of the modeling, the estrous cycle was detected by vaginal smear, and rat with persistent keratinization of vaginal epithelial cells were selected as the PCOS model rat. The model rats were randomly divided into model group, Dacin-35 group, and high, middle, low-dose dodder total flavonoids groups. The corresponding drugs were given for 21 consecutive days. At the end of the administration, materials were collected to calculate ovary index, and enzyme-linked immunosorbent assay(ELISA) was used to measure estrogen (E2), testosterone (T), gonadotropin-releasing hormone (GnRH), follicle stimulating hormone (FSH) and luteinizing hormone (LH) levels of serum. The right ovary of rats was stained with haematoxylin-eosin (HE), and the pathological changes were observed by optical microscope. Androgen receptor(AR) expressions in hypothalamus, pituitary and ovary were detected by mmunohistochemistry. Result: Compared with the blank group, serum T, GnRH and LH levels, ovarian index and LH/FSH ratio were significantly increased, while FSH and E2 levels were significantly decreased (PPP2, T, GnRH and LH levels, ovarian index and LH/FSH ratio were significantly decreased in high, middle and low-dose dodder total flavonoids groups (P2 level was significantly increased (PP2 level in PCOS model rats was obviously increased in low-dose dodder total flavonoids group (PPConclusion: Dodder total flavonoids may play a protective role in PCOS model rats by regulating the secretion of estrogen and androgen and affecting the hypothalamic-pituitary-ovary axis pathway.

ABSTRACT

Objective To establish a Real-time quantitative PCR method (qPCR) for the detection of diatom UPA barcoding genes and evaluate its application in the drowning diagnosis. Methods The homologous sequences of diatoms UPA gene was obtained by Blast from GeneBank, based on which the universal primers for diatoms were designed. DNA were extracted from 2 common human symbiotic bacteria (Escherichia coli, Bifidobacterium longum), 3 species of planktonic bacteria, 15 species of planktonic algae, tissue samples (lung, liver and kidney) from human cadavers (28 drowning victims, 1 victims by non-drowning in the water, 3 victims deaths on land) in 32 cases. The specificity, sensitivity and repeatability of the designed primers were tested. The positive rates of diatoms detection in the drowning cases were calculated. The results of the real-time quantitative method were evaluated comparatively by Microwave Digestion-Vacuum Filtration-Automated Scanning Electron Microscopy (MD-VF-Auto SEM) and PCR-Capillary Electrophoresis (PCR-CE). Results The results showed that the primers UPA99 had strong specificity for the diatomaceae (Synedra radians, Navicula sp., Melosira varians, Cyclotella sp. and Nitzschia sp.) DNA. The melting curve of the amplified product was smooth; the peak was narrow; the melting temperature was (87±1)℃. The sensitivity of qPCR method was 1.56×10-5ng/μL with the detection range of 1.56×102ng/mL~1.56×10-5ng/μL, in contrast with the PCR-CE method (1.56×10-3ng/μL). This real-time PCR method showed high repeatability and stability with the coefficient of variation less than 2%. The detection rate of lung, liver and kidney was 89.3%, 71.4% and 64.3% respectively. Conclusion The established qPCR method, based on the universal primers designed for diatom UPA gene, has high specificity, high sensitivity and good repeatability. With a promising prospect for application, qPCR is suitable for drowning diagnosis.

ABSTRACT

Objective To induce a novel temperate bacteriophage from staphylococcus haemolyticus strain SA98,observe the morphology and size,complete the whole genome sequencing,analyse the structure of genome and evolutionary relationship.Methods The mitomycin C was used to induce the temperate phage from staphylococcushaemolyticus strain SA98,the induced phage was observed by transmission electron microscopy after be concentrated and purified.The genome DNA was extracted and high through sequenced.The feature of whole genome and evolutionary relationship was analyzed.Results A temperate phage IME-SA4 was successfully induced from staphylococcus haemolyticus strain SA98.Transmission electron microscopy analysis indicated that IME-SA4 had an isometric head and a non-contractile long tail.The whole genome of IME-SA4 was long as 41 843 bp,and the whole genome blast result indicated IME-SA4 shared only 13% homology with most related strain phiRS7.Conclusions A novel staphylococcus haemolyticus temperate phage with low homology with other staphylococcusphages was successfully induced from staphylococcus haemolyticus strain SA98.The research of its morphology,whole genome sequencing and comparative genome analysis make the foundation for further study of staphylococcus phages' properties and practical application.

ABSTRACT

Patients with hip osteoarthritis or other hip joint diseases suffer from pain, limited joint function and psychological disorder, which are responsible for difficulties in sexual life. Total hip arthroplasty can effectively relieve the pain of hip joint and improve the range of joint motion, thus improving the frequency and performance of sexual life of patients. Good quality of sexual life is related to the postoperative satisfaction of patients. However, patients can hardly get the correct guidance on this. After total hip arthroplasty, male patients are earlier to recover sexual life than female patients. The time of the most patients to recover sexual life is 1-3 months after surgery, and the safe position of sexual life is stand position. Sugerons should make positive and correct guidance for patients and their sexual partners about safe resumption of sexual life after total hip arthroplasty.

ABSTRACT

<p><b>OBJECTIVE</b>To study the distribution of single nucleotide polymorphisms (SNP) of arginine-vasopressin (AVP) gene rs66818855 and rs1078152 in Chinese Guangxi healthy population in comparison with that in different ethnic populations.</p><p><b>METHDOS</b>Polymerase chain reaction-single base extension (PCR-SBE) and DNA sequencing were used to detect the allele and genotype frequencies of AVP gene among 303 Chinese healthy individuals in Guangxi, China, and the results were compared with the reported frequencies in 4 other populations (HapMap-CEU, HapMap-YRI, HapMap-JPT, and HapMap-HCB) from Human Genome Project group (HapMap) data.</p><p><b>RESULTS</b>We found significant AVP gene polymorphisms in this Guangxi healthy population. The frequencies of allele and genotype of AVP gene rs66818855 and rs1078152 polymorphisms in this Guangxi population differed significantly from those in HapMap-CEU population (P<0.01), and allele frequencies of AVP gene rs66818855 polymorphism differed significantly from those in HapMap-YRI populations (P<0.05).</p><p><b>CONCLUSION</b>The distribution pattern of AVP gene polymorphisms in this Guangxi population is significantly different from that in other ethnic populations, which might account for the difference in the morbidity of AVP-related disease among different ethnic groups and may have important indications in the study of population genetics and anthropology.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the race- and gender-specific distribution characteristics of rs1891385A/C and rs10975519C/T polymorphism of interleukin-33 (IL-33) gene in Zhuang and Han populations.</p><p><b>METHODS</b>The polymorphisms of rs1891385A/C and rs10975519C/T of IL-33 gene in 283 subjects from Guangxi Zhuang Autonomous Region were analyzed with single base extension (PCR-SEB) and DNA sequencing to analyze the differences in their distribution frequencies between genders and between Zhuang and Han populations.</p><p><b>RESULTS</b>Three genotypes (AA, AC and CC) were found in rs1891385A/C with frequencies of 64.3%, 32.5% and 3.2%, respectively. The genotype and allele frequencies of rs1891385A/C in this Guangxi population showed no significant difference between Zhuang and Han subpopulations and between genders (P>0.05), but differed significantly from those in European and African black populations (P<0.01). Three genotypes (CC, CT and TT) were identified in rs10975519C/T with frequencies of 34.3%, 53.0%, and 12.7%, respectively, showing no significant ethnic or gender-specific differences in this population (P>0.05). The genotype frequency of rs10975519C/T in this population differed significantly from those in the European and Japanese populations (P<0.01), but the allele frequencies only showed significant differences from those in the European population (P<0.01).</p><p><b>CONCLUSION</b>rs1891385A/C and rs10975519C/T polymorphisms of IL-33 gene show a race-specific difference.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the diagnostic value of R-banding technique (RT), dual-color fluorescence in situ hybridization (D-FISH) and quantitative real-time PCR (RT-PCR) for acute promyelocytic leukemia.</p><p><b>METHODS</b>The cytogenetic characteristics and PML/RARα fusion gene in 340 patients with suspectable APL were analyzed by using 3 detection methods. MICM (morphology, immunology, cytogenetic and molecular biology) was used as diagnostic standard of APL, and the diagnostic value of RT, D-FISH and RT-PCR was evaluated by comparing the detection results of RT, D-FISH and RT-PCR as well as their combination.</p><p><b>RESULTS</b>For the diagnosis of APL, the sensitivity of RT, D-FISH and RT-PCR was 81.3% (78/96), 95.0% (91/96) and 96.9% (93/96) respectively. RT failed to detect 18 cases, the results of D-FISH showed 5 cases with false positive and 2 cases with false negative, the RT-PCR showed 4 cases with false positive, 3 cases with false negative. The sensitivity and specificity of combined detection of 3 methods were 99.97% and 100% respectively.</p><p><b>CONCLUSION</b>The 3 detection methods alone all have certain defects for diagnosis of APL, but their combined detection is helpful to improve the definitive diagnostic rate and can decrease misdiagnosis rate and missed diagnostic rate.</p>
