ABSTRACT

Objective:To explore the application effect of multimodal quantitative rehabilitation exercise in patients with ankylosing spondylitis and to provide reference for patients′ rehabilitation exercise.Methods:The quasi-experimental study method was used to select 78 patients with ankylosing spondylitis admitted to Rheumatology and Immunology Department of First Affiliated Hospital of Xinjiang Medical University from February 2021 to February 2022 as the research objects. The 39 patients admitted from February 2021 to August 2021 as the control group, and 39 patients admitted from September 2021 to February 2022 as the experimental group. The control group adopted conventional rehabilitation exercise program, and the experimental group adopted multimodal quantitative rehabilitation exercise program. The Bath ankylosing spondylitis disease activity index, Bath ankylosing spondylitis function index, inflammatory factors after 6 months of intervention were compared between the two groups.Results:The Bath ankylosing spondylitis disease activity index in the experimental group after 6 months of intervention was (2.35 ± 0.81) points, and that in the control group was (3.47 ± 1.04) points, with a statistically significant difference ( t = 4.02, P<0.05). The Bath ankylosing spondylitis disease activity index in the two groups were analyzed by repeated measurement variance. The differences of time effect, inter group effect and interaction effect were statistically significant ( Fgroup = 11.27, Ftime = 62.05, Finteraction = 5.47, all P<0.05). The Bath ankylosing spondylitis function index in the experimental group after 6 months of intervention was (2.11 ± 0.32) points, and that in the control group was (3.07 ± 0.58) points, with a statistically significant difference ( t = 4.03, P<0.05). The Bath ankylosing spondylitis function index in the two groups were analyzed by repeated measurement variance. The differences in time effect, inter group effect and interaction effect were statistically significant ( Fgroup = 21.44, Ftime = 42.25, Finteraction = 16.67, all P<0.05). After 6 months of intervention, C-reactive protein, interleukin-6, transforming growth factor β, tumor necrosis factor-α were (43.15 ± 2.21) mg/L, (3.28 ± 0.85) mg/L, (41.67 ± 9.04) ng/L, (176.63 ± 20.15) ng/L respectively in the experimental group, and (50.12 ± 1.67) mg/L, (5.27 ± 0.68) mg/L, (48.65 ± 8.96) ng/L, (194.56 ± 19.45) ng/L respectively in the control group. There was a statistically significant difference in the content of inflammatory factors between the two groups ( t values were 2.05-4.45, all P<0.05). Conclusions:Multimodal quantitative rehabilitation exercise can improve the physiological function of ankylosing spondylitis patients′spine, reduce the disease activity of patients, and promote the rehabilitation of patients.

ABSTRACT

Objective:To investigate the effects of complement C3a receptor(C3aR) and receptor for advanced glycation end product(RAGE) on bone metabolism in APP/PS1 mice model of Alzheimer′s disease.Methods:Alzheimer′s disease model APP/PS1 mice was hybridized with C3aR knockout mice(C3aR -/-), RAGE knockout mice(RAGE -/-) to generate APP/PS1-C3aR -/- and APP/PS1-RAGE -/-, respectively. In vivo, micro computed tomography(Micro-CT) scan, bone tissue osteocalcin immunohistochemical staining, tartrate-resistant acid phosphatase(TRAP) staining and Goldner′s trichrome staining were used to understand the variabilities of bone metabolism between different genotypes of mice; In vitro, bone marrow-derived osteoblast and osteoclast induction cultures were used to understand the effects of C3aR and RAGE on osteoblast and osteoclast differentiation. Results:In in vivo experiments, APP/PS1-C3aR -/- and APP/PS1-RAGE -/- mice showed increased bone mass, increased bone formation, decreased bone resorption, and increased osteoid compared to APP/PS1 mice( P<0.05). In in vitro experiments, bone marrow mesenchymal stem cells(BMSCs) derived from APP/PS1-C3aR -/- and APP/PS1-RAGE -/- mice showed enhanced osteoblast differentiation and elevated expression levels of alkaline phosphatase(ALP) and runt-related transcription factor 2(RUNX2), diminished osteoclast differentiation, and reduced positive TRAP staining( P<0.05). Conclusions:Both C3aR and RAGE are involved in regulating the process of APP/PS1 bone metabolism. Knockout of C3aR and RAGE can improve osteoporosis in APP/PS1, providing a new target for the clinical treatment of Alzheimer′s disease combined with osteoporosis.

ABSTRACT

OBJECTIVE:To study the effects of gin seng root extract on autophagy ,proliferation and cytokine of splenic lymphocyte of mice ,and to study its mechanism. METHODS :The splenic lymphocyte of mice were divided into blank control group,positive control group (concanavalin A ,5 μg/mL),different concentration of ginseng root extract groups (32,125,500 μg/mL,by lyophilized powder ). After 48 h of culture with the corresponding medicine ,acridine orange staining method was used to detect autophagy of splenic lymphocyte ;CCK-8 assay was used to detect the cell proliferation ;ELISA assay was used to determine the levels of IL- 4,IL-6 and TNF-α in cell culture;Western blotting method was used to detect the expression of LC 3B and Beclin- 1,as well as the phosphorylation of AMPK ,AKT and mTOR. RESULTS :Compared with blank control group ,32, 125,500 μg/mL ginseng root extract could increase the number of acidic autophagosomes in splenic lymphocytes of mice(P<0.05 or P<0.01);125,500 μg/mL Ginseng root extract could significantly enhance the survival rate of splenic lymphocytes,the levels of IL- 4,IL-6 and TNF-α and the transformation of LC3BⅠ to LC 3BⅡ,significantly increased the protein expression of Beclin- 1, quinacrine cationic liposomes for treating non-small cell @163.com lung cancer[J]. J Drug Target ,2015,23(3):232-243. the phosphorylation of AMPK protein ,and significantly reduced the phosphorylation level of AKT and mTOR proteins ,with statistical significance (P<0.05 or P<0.01). CONCLUSIONS :Ginseng root extract can induce autophagy of mice splenic lymphocytes,activate the activity of splenic lymphocytes ,regulate the secretion of cytokine by activating AMPK and inhibiting the activation of AKT which can inhibit the activity of mTOR ,so as to exert immune enhancement effect.
