ABSTRACT

Objective:To explore the application value of combined detection of p16 and human papillomavirus (HPV) typing in the diagnosis of cervical intraepithelial neoplasia (CIN).Methods:A total of 8 346 patients aged between 25 years old and 65 years old at Baoji Central Hospital of Shaanxi Province from February 2019 to February 2020 were selected. There were 2 882 patients with cervical lesions diagnosed by colposcopy biopsy. Patients were divided into the different groups based on the age range, and then the condition of HPV infection in all age groups was analyzed. Taking biopsy as the gold standard and according to the pathological results, the detection rate of p16 and HPV typing and the diagnostic value of the single and combined detection in CIN were also analyzed.Results:The age group with the highest positive rate of p16 and HPV was 31-40 years old [47.42% (1 014/2 427) and 36.84% (894/2 427), respectively], followed by 41-50 years old group [30.15% (907/2 942) and 28.11% (827/2 942)], and there were statistically significant differences in positive rate of p16 and HPV in all age groups (all P < 0.05). Among 2 882 patient with cervical lesions diagnosed by pathological examination, there were 2 572 cases (89.24%) of p16 positive, and 2 169 cases (75.26%) of HPV positive. With the disease progression of cervical lesions, the positive rate of p16 and HPV was gradually increased, and the positive rate of p16 of inflammation, CINⅠ, CINⅡ, CIN Ⅲ, cervical squamous cell carcinoma (SCC) was 11.68% (23/197), 94. 85% (1 105/1 165), 93.57% (771/824), 96.76% (538/556), 96.43% (135/140), respectively; the positive rate of HPV was 17.77% (35/197), 77.60% (904/1 165), 80.22% (661/824), 80.40% (447/556), 87.14% (122/140), respectively, and HPV infection was mostly HPV16/18 infection type with the disease progression of cervical lesions. The sensitivity, specificity, positive predictive value and negative predictive value in detecting CIN of HPV was 75.26%, 81.13%, 67.78% and 86.14%, respectively; the sensitivity, specificity, positive predictive value and negative predictive value in detecting CIN of p16 was 89.24%, 84.74%, 75.51% and 93.72%, respectively; the diagnostic efficacy of p16 was higher than that of HPV in detecting CIN, and the difference was statistically significant ( P < 0.05). The sensitivity, specificity, positive predictive value and negative predictive value of HPV combined with p16 in detecting CIN was 94.10%, 91.33%, 85.12%, 96.71%, which were higher compared with those of single detection (all P < 0.05). Conclusions:HPV infection mainly occurs in women aged 31-40 years old followed by 41-50 years old, and the infected population of CIN tends to be younger. p16 is superior to HPV in detecting the diagnostic efficacy of CIN; combined detection of p16 and HPV can increase the sensitivity and specificity, reduce the rate of misdiagnosis, and can play a key clinical value in early diagnosis and treatment of CIN.

ABSTRACT

OBJECTIVE To establish a platform of electrophysiology using vagal ganglion neurons (VGNs) isolated from adult canines. METHODS The VGNs were enzymatically isolated from adult canines of either gender and cultured under experimental conditions. Action potential (AP), repetitive firing, voltage-gated outward K+ currents (IK) and hyperpolarization-mediated inward currents (Ih) were recorded under current-and voltage-clamp configurations before and after treatment. RESULTS Analysis of AP waveform showed that ① inaddition to traditionally classified myelinated A- and unmyelinated C-types, myelinated Ah-types could also be identified in females rather than in males; ② step current depolarization evoked a stimulus intensity-dependent repetitive discharge, and to reach a similar firing frequency, the lowest stimulus intensity was required for A-types, a similar or slightly higher stimulus intensity was needed for Ah-types, and the highest stimulus intensity was required for C-types;③tetro?dotoxin significantly reduced the rate of depolarization and positively shifted the AP firing threshold of Ah-types, and iberiotoxin dramatically increased the neuroexcitability of Ah-types;④all tested neurons functionally expressed IK and Ih, and the current density for both channels on average was A-types>Ah-types>C-types; ⑤ although the distribution of afferent types of VGNs differed between males and females, the known difference in discharge profiles of A- and C-types isolated from male and female rats was not studied here. CONCLUSION The VGNs can be successfully isolated from adult canines, AP, IK and Ih can be recorded. The tight seal can be held for at least 30 min, which may be enough for pharmacological investigation.

ABSTRACT

Fenofibrate, an agonist for peroxisome proliferator-activated receptor alpha (PPAR-α), lowers blood pressure, but whether this action is mediated via baroreflex afferents has not been elucidated. In this study, the distribution of PPAR-α and PPAR-γ was assessed in the nodose ganglion (NG) and the nucleus of the solitary tract (NTS). Hypertension induced by drinking high fructose (HFD) was reduced, along with complete restoration of impaired baroreceptor sensitivity, by chronic treatment with fenofibrate. The molecular data also showed that both PPAR-α and PPAR-γ were dramatically up-regulated in the NG and NTS of the HFD group. Expression of the downstream signaling molecule of PPAR-α, the mitochondrial uncoupling protein 2 (UCP2), was up-regulated in the baroreflex afferent pathway under similar experimental conditions, along with amelioration of reduced superoxide dismutase activity and increased superoxide in HFD rats. These results suggest that chronic treatment with fenofibrate plays a crucial role in the neural control of blood pressure by improving baroreflex afferent function due at least partially to PPAR-mediated up-regulation of UCP2 expression and reduction of oxidative stress.

@#Objective To investigate the situation of family intimacy and adaptability, and social avoidance and distress in severe burn patients, and explore the correlation between them, so as to provide basis for intervention. Methods From January, 2013 to June, 2015, 289 patients with severe burn were surveyed with self-designed general condition questionnaire, Family Adaptability and Cohesion Evaluation Scale (FACES Ⅱ) and Social Avoidance and Distress Scale (SADS), and analyed with Pearson correlation analysis. Results The real family intimacy and adaptability were lower in the severe burn patients than in national norm (t>3.830, P<0.01), and the ideal family intimacy, dis-satisfied intimacy level, ideal adaptability and dissatisfied adaptability level were higher in the severe burn patients than in national norm (t>3.857, P<0.01). The SADS score was higher in the severe burn patients than in national norm. The real intimacy, real adaptability and the to-tal score of FACES II negatively correlated with the scores of SADS (P<0.05), while the levels of dissatisfied intimacy and adaptability posi-tively correlated (P<0.05). Conclusion The social avoidance and distress exist in severe burn patients, which may associated with the family intimacy and adaptability, that some interventions may target to.

ABSTRACT

Objective To investigate the application of alpha-fetoprotein (AFP),α-L-fucosidase(AFU),5'-nucleotidase(5'-NT) and γ-glutamyltransferase (GGT) in diagnosis of hepatocellular carcinoma(HCC),and to provide the basis for its early and rapid diagnosis.Methods The clinical data of 120 cases with initial diagnosis as hepatocellular carcinoma,150 cases with liver cirrhosis,200 cases with hepatitis were analyzed retrospectively from June 2013 to August 2014,meanwhile,100 healthy people were selected as control group.The serum AFP,AFU,5'-NT and GGT levels were measured by electrochemiluminescence immunoassay,rate method,peroxidase method and enzymatic colorimetric assay,respectively.The results were analyzed by receiver operating characteristic (ROC) curve,while the sensitivity,specificity,AUC were calculated at the same time.Results The levels of serum AFP,AFU,5'-NT and GGT were significantly different among HCC group,liver cirrhosis group,hepatitis group and control group (all P<0.05).When tested alone,the sensitivities of AFP,AFU,5'-NT and GGT were 58.33 %(70/120),82.50 %(99/120),85.00 %(102/120),78.33 %(94/120),the specificities were 88.33 %(106/120),73.33 %(88/120),88.33 %(106/120),71.67 %(86/120),and AUC were 0.655,0.702,0.814,0.754.When four indexes were performed with combined test,the sensitivity was 98.33 %(118/120),specificity was 96.67 %(116/120),AUC was 0.975,which were higher than the single detection (all P<0.05).Conclusion The combined detection of serum AFP,AFU,5'-NT and GGT can greatly increase the sensitivity and specificity for diagnosing HCC and will gain mutual complement advantages,which is of great clinical value for early and rapid diagnosis.

ABSTRACT

Objective] To evaluate effect of health literacy intervention in occupational groups , explore suitable ways and provide suggestions for workplace health promotion . [ Methods ] In application of convenience sampling , Minhang District , Hongkou District and Qingpu District each selected 2 enterprises respectively according to their type , scale and location , setting one as intervention enterprise and the other as control .Then a six-month comprehensive intervention was conducted among intervention groups .Two hundred employees in each enterprise were randomly investigated before and after intervention to evaluate the effect . [ Results ] The overall health literacy level of participants in the intervention group rose from 16.7% to 33.4%, with improvement on the literacy levels of scientific view , infectious diseases prevention and treatment , chronic disease control and prevention and the three aspects of health literacy content ( P<0 .05 ) , while such improvement was not reflected in the control group , and health skill literacy, safety and first aid literacy declined (P <0.05). [Conclusion] Comprehensive intervention based on health needs could improve the level of health literacy of occupational groups . Intervention in future should focus on improving the literacy of chronic disease control and prevention and basic medical care in occupational population .

ABSTRACT

Objective To explore the clinical application of single or combined detection of serum Cys-C,5'-NT and AFP in hepatocellular carcinoma (HCC) in order to provide the platform for early diagnosis and prognosis evaluation.Methods The serum levels of Cys-C,5'-NT and AFP from 148 cases of HCC patients (HCC group),135 cases of liver cirrhosis patients (LC group) and 155 cases of hepatitis (hepatitis group) and 100 healthy people (control group) were measured by latex-enhanced immunoturbidimetric assay (LEITD),peroxidase method and electrochemiluminescence immunoassay (ECLI),respectively.Then their differences had been compared.The sensitivity,specificity and Youden's index were calculated and the results were analyzed by receiver operating characteristic (ROC) curve.Results The levels of serum Cys-C,5'-NT and AFP were significantly different in 4 groups.The differences were statistically significant (F =12.35,42.25,58.12,P =0.000).The sensitivity of three indicators for diagnosing HCC was 100 %,which was higher than that of single or two combined detection (P < 0.05),and the specificity was 78.97 %.The area under ROC curves of combined detection of three indicators was 0.977,which was also higher than single or two combined detection (P < 0.05).Conclusions The combined detection of serum Cys-C,5'-NT and AFP can greatly increase the sensitivity and accuracy for diagnosing HCC,which has an important clinical value for early diagnosis.Therefore it is worth popularizing and applying on clinic.

ABSTRACT

Objective To investigated the effects of combined arsenic trioxide(ATO) and resveratrol(Res)on the viability of NB4 human leukemia cells. Methods NB4 human leukemia cell was used in this experiment.Cells were cultured in ATO (0,0.1875,0.3750,0.7500, 1.1250, 1.5000,2.2500,3.0000,5.0000 μmol/L) and Res (0, 1.5625,3.1250,6.2500, 12.5000, 18.7500,25.0000,37.5000,50.0000 μmol/L). Cell viabilities were measured by MTT in different treatment groups. Half inhibitory concentration(IC50) was calculated. The ratio of concentration of ATO and Res 1.5∶ 18,1.5∶ 25,1.5∶ 35 was added to cells, and the combination index(CI) was calculated. The level of ROS in control, ATO( 1.5000 μmol/L), Res(25.0000 μmol/L) and ATO(0.9000 μmol/L) + Res( 12.5000μmol/L) groups was measured by chemiluminescence assay. Results ①ATO( ≥0.7500 μmol/L) reduced the viability of NB4 cells in a concentration-dependent manner(P < 0.05 ), and IC50 was (1.78 ± 0.11 )μmol/L. ②)Res (≥18.7500 μ mol/L) dose-dependently decreased the viability of NB4 cells (P < 0.05 ), and IC50 was ( 18.71 ±0.18)μ mol/L. ③Combination of ATO and Res showed an antagonistic effect on NB4 cells viability. ④The ROS in Res group( 1670.55 ± 13.97) was significantly lower than that in control group(2345.88 ± 14.48,P < 0.05). The ROS in ATO group (3092.42 ± 94.84) was significantly higher than that in control group(P < 0.05). The ROS in ATO + Res group (1860.27 ± 15.99) was significantly lower than that in ATO group(P < 0.05). Conclusions NB4 cell survival rate can be decreased by ATO and Res. The combination of arsenic trioxide and Res presents an antagonistic effect on NB4 cell viability, in part by reducing intracellular ROS formation.

ABSTRACT

Objective To study the effect of Asi-antidiarrheal capsule on gastrointestinal goblet cell of thyroid hormone-induced diarrhea.Methods Total of 120 SD male rats aged about 8 weeks were randomly divided into 2 groups:control group(10 rats)and thyroid hormone-induced diarrheic group(110 rats).Rats in control group were lavaged with normal saline 1 ml/d.Thyroid tablets were partly desolved into normal saline forming a 40 mg/ml suspension.Rats in thyroid hormone-induced diarrheic group were given the thyroid suspension 1 ml/d to make thyroid hormone-induced diarrheic model.Serum FT3 and FT4 were tested.Fourty thyroid hormone-induced diarrheic rats were screened out according to serum FT3 and FT4 levels,body weight and wet stool.The fourty rats were randomly divided into 5 groups,8 rats in each group:positive control group,berberine group,low-dose,mediandose and high-dose groups.Normal saline of 1 ml/d was admnistered to diarrhea control group,1.94 g·kg-1·d-1 Berberine capsule was given to positive control group,and 0.63,1.26,2.52 g·kg-1·d-1 Asi-antidiarrheal capsule to low-dose,mediandose and high-dose groups,respectively.After sever days treatment,rats are executed.Duodenum,jejunum,ileum and colon were dissected,respectively.Histology observation and cell counting were carried out under light micmscopo on HE coloration.Cell counting unit was defined as:cell/high power field of vision (cells/hpf).Results In jejunum,the number of goblet cells in berberine group,mediandose group and high-dose group[(15.32±2.53),(20.24±1.24),(14.98±1.10)cells/hpf,respectively],were all lower than that of the diarrhea control group[(25.73±4.55)cells/hpf,all P<0.05]with an exception of low-dose group[(23.98±2.28)cells/hpf].The numbers of goblet cells in berberine control group,low-dose group,mediandose group and highdose group[(18.29±1.33),(20.61±2.12),(19.38±2.01),(16.34±1.55)cells/hpf,respectively]were all less than that of the control group[(23.36±3.10)cells/hpf,all P<0.05].The numbers of goblet cells of diarrhea control group and high-dose group were obviously lower than that of the low-dose group(all P<0.05)in jejunum and colon.The numbers of goblet cells of Duodenum and ileum were not significantly different between groups(F=2.81,2.67,all P>0.05).The numbers of goblet cells in the diarrhea control group increased markedly observed under microscope,but decreased following therapeutic treatment.Conclusions The numbers of goblet cells from jejunum and colon in thyroid hormone-induced diarrheic rats are increased significantly.Asi-antidiarrheal capsule can remarkably decrease the number of goblet cells in jejunum and colon,and reduce mucus secretion.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate and analyze the clinical therapeutic effect and characteristics of acupuncture combined with medicine on depression of different syndrome types.</p><p><b>METHODS</b>Eighty cases of depression were dividede into liver qi stagnation and spleen deficiency group (group A, 35 cases), deficiency of both heart and spleen group (group B, 21 cases) and yang deficiency of both spleen and kidney group (group C, 24 cases) according to the syndrome differentiation. They were all treated with acupuncture at Baihui (GV 20), Yintang (EX-HN 3), Sishen-cong (EX-HN 1), Neiguan (PC 6) etc. and oral administration of 5-HT selective serotonin reuptake inhibitors (SSRI) tablets. The Hamilton depression scale (HAMD) and self-made syndrome differentiation symptoms evaluation scale were adopted to evaluate the therapeutic effect.</p><p><b>RESULTS</b>After treatment of 4 weeks, the total effective rate of 91.4% in group A and of 90.5% in group B, both were superior to that of 87.5% in group C (P < 0.01,P<0. 05). There was a significant difference in comparison of HAMD scores between the group A and group C after treatment of 4 weeks (P < 0.05). The symptom scores of syndrome differentiation in all three groups were obviously decreased after 1 week, 2 weeks and 4 weeks treatment compared with those before treatment (all P < 0.001), the symptom scores of group C had significant differences compared with the group A and group B after treatment of 2 weeks (P < 0.01, P < 0.05). The reducing scores rate of the group A had singificant differences compared with the group B and the group C after treatment of 4 weeks (both P < 0.05).</p><p><b>CONCLUSION</b>Acupuncture combined with SSRI has better therapeutic effect on depression of liver qi stagnation and spleen deficiency type. The syndrome differentiation symptoms evaluation scale is more exact and sensitive to evaluate the clinical therapeutic effect of TCM treatment.</p>

ABSTRACT

Objective Investigate the Asi-antidiarrheal capsule's effect on gastrointestinal horrnones in blood plasma of thyroid hormone-induced diarrheic(Abbreviation:Hyperthyroid Diarrhea.,D)rats.Methods One hundred and twenty SD male rats about 8 weeks old were randomly divided according to their constitution into control group of 10 rats and thyroid hormone-induced diarrheic group of 110 rats.The control group rats welle hvaged with isotonic Na chloride 1 ml/d.Thyroid tablets were made with isotonic Na chloride into 40 g/L susnl. The such solution with 1 ml/d was intragastrically administered to each rat in thyroid hormone-induced diardleic group.After three weeks,blood was sampled from vena caudalis of each rat.FT4 were then detected in blood semm.Fourty-two thyroid hormone-induced diarrheic rats were screened based on FT3 and FT4 level in blood serum, wet stool and body weisht.Fourty thyroid hormone-induced diarrheic rats were stochastically re-divided into 5 groups with 8 in each.The physiological saline with 1 ml/d was given to blank group,1.94 g·-kg-1·d-1 Berberine capsule to positive control group,and 0.63,1.26,2.52 g·kg-1·d-1 to low-dose,moderate-dose and high-dose groups respectivelv. Intragastric administration of each group continued for 7 days.Venous blood was centrifuged before and after administration and underwent radioimmunoassay to observe the effect of Ashi-antidiarrheal capsule on motillty (MTL),gastrin(GAS),somatostatin(SS),vasoactive intestinal polypeptide(VlP)in blood plasma of thyroid homlone- induced diarrheic rats.Results①Weight of thyroid hormone-induced diarrheic rats decreased[(344.0±12.9)g], FT3[(4,58 ±0.70)mol/L]and FT4[(23.44±4.40)mol/L]increased,and weight of wet stool[(17.4±3.2)g] increased.Compared to control group[(386.0±1.8)g,(2.08±0.10)mol/L,(10.18±2.00)mol/L,(9.1±0.6)g], there was a statistical significance(t=6.85,9.80,7.66,7.18,P<0.01).②After treatment,high-dose Ashi-antidiarrheal group[(80.54 ±3.80)ng/L]and positive control group[(90.63 ±9.99)ng/L]blood plasma MTL, compared to pre-therapy[(204.27±17.69),(187.79±13.32)ng/L]was decreased,there wag a statistical significance (t=8.60,4.57,P<0.01)③GAS contentshad respectively decreased comparedtopre-therapy[(192.75±11.80), (193.09±3.81),(190.60±9.31),(196.33±18.13)ng/L]in positive control group[(56.06 ±6.36)ng/L],low- dose group[(90.88±4.18)ng/L],midst-dose group[(75.64±7.09)ng/L]and hish-dose group[(44.32±3.72) ng/L],except for blank group.There Wag a statistical significance(t=15.27,7.62,13.43,13.22,all P<0.01).The intm-group difference of MTL,GAS and VIP level had statistic signifieances before and after the treatment(F= 166.68,1503.53,216.68,P<0.01).Conclusion Ashi-antidiarrheal capsule Can significantly lower the level of MTL and GAS in blood plasma。And raise the level of VIP.

ABSTRACT

<p><b>BACKGROUND</b>Airway smooth muscle (ASM) is suspected to be a determining factor in the structural change of asthma. However, the role of protein kinase C alpha (PKCalpha) and cyclin D1 involved in the dysfunction of ASM leading to asthmatic symptoms is not clear. In this study, the central role of PKCalpha and cyclin D1 in ASM proliferation in asthmatic rats was explored.</p><p><b>METHODS</b>Thirty-six pathogen-free male Brown Norway (BN) rats were randomly divided into 2 groups: control groups (group N1, N2 and N3) and asthmatic groups (group A1, A2, and A3). Groups A1, A2 and A3 were challenged with ovalbumin (OA) for 2 weeks, 4 weeks and 8 weeks respectively. Control animals were exposed to an aerosolized sterile phosphate buffered saline (PBS). The ASM mass and nucleus numbers were studied to estimate the degree of airway remodeling by the hematoxylin-eosin staining method. PKCalpha and cyclin D1 expression in the ASM cells was detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry. The relation between PKCalpha and cyclin D1 was assessed by linear regression analysis. PKC agonist phorbol 12-myristate 13-acetate (PMA), PKC inhibitor Ro31-8220 and an antisense oligonucleotide against cyclin D1 (ASOND) were used to treat ASM cells (ASMCs) obtained from the 2 weeks asthmatic rats. The cyclin D1 protein expression level was detected by Western blotting.</p><p><b>RESULTS</b>Compared with the control group, the PKCalpha and cyclin D1 mRNA levels were increased in the asthmatic group. Similar to RT-PCR results, immunohistochemistry analysis for PKCalpha and cyclin D1 expression revealed an increased production in ASMCs after allergen treatment for 2, 4 and 8 weeks compared with the respective control groups. No difference in expression of PKCalpha and cyclin D1 in ASM were found in the 2, 4 or 8 weeks asthmatic rats. There were significant positive correlations between PKCalpha and cyclin D1 expression, both transcriptionally (r = 0.944, P < 0.01) and translationally (r = 0.826, P < 0.01), in ASM. The content of cyclin D1 in asthmatic ASMCs increased after being stimulated by PMA, and decreased when induced by Ro31-8220. ASOND targeting for cyclin D1 lowered the expression of cyclin D1 induced by PMA.</p><p><b>CONCLUSIONS</b>Increased expression of PKCalpha and cyclin D1 in ASM along with smooth muscle structure changes might implicate PKCalpha and cyclin D1 participation in the proliferation of ASM and contribute to the pathogenesis of asthma after repeated allergen exposure in rats. The results suggested that cyclin D1 might be downstream of PKC signal transduction pathway.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the role of Shenfu Injection (SFI) in rats with systemic inflammatory response syndrome (SIRS).</p><p><b>METHODS</b>The SIRS rat model was induced by the intravenous injection of lipopolysaccharide (LPS). Forty-five male Wistar rats were randomly divided into 3 groups, the sham operative control group (control group, n=5), the SIRS model group (model group, n=20) and the SFI treatment group (SFI group, n=20). LPS was injected through the external jugular vein (12 mg/kg, 6 mg/mL) to all rats except for those in the control group, and SFI (10 mL/kg) was given to those in the SF group only once through intraperitoneal injection, while the normal saline (10 mL/kg) was given to those in the model group. For those in the control group, normal saline was given through the external jugular vein (2 mL/kg) and intraperitoneal injection (10 mL/kg). Then, rats in the model group and SFI group were divided into 4 subgroups according to the time points, i.e., 1 h, 2 h, 4 h and 6 h subgroups, 5 rats in each group. The activity of nuclear factor of kappa B (NF-kappa B) of in blood mononuclear cells and the plasma levels of tumor necrosis factor-alpha (TNF-alpha) and interleukin 6-(IL-6) were determined using enzyme-linked immunoabsordent assay (ELISA) at 1 h, 2 h, 4 h and 6 h after modeling. Histopathologic changes of the lung and liver were observed under a light microscope.</p><p><b>RESULTS</b>Compared with the control group, the activity of NF-kappa B in mononuclear cells and the plasma level of TNF-alpha were obviously increased at each time points (all P<0.01), reaching the peaks at 2 h after modeling. The plasma level of IL-6 increased gradually as time went by in the model group (P<0.01). Pathological examination showed pulmonary alveoli hemorrhage, edema and inflammatory cell infiltration in the lung tissue, and angiotelectasis, congestion, and local necrosis in the liver tissue in the model group. Compared with the model group, the activity of NF-kappa B and the levels of TNF-alpha and IL-6 in plasma decreased significantly in the SFI group (P<0.01), and the pathological injury in the lungs and liver was significantly alleviated.</p><p><b>CONCLUSION</b>SFI plays a protective role by inhibiting the activity of NF-kappaB, and reducing the expressions of TNF-alpha and IL-6 in SIRS rats.</p>

ABSTRACT

This study is to investigate the expression of CyclinD1 in asthmatic rats and construct expression plasmids of sense and antisense CyclinD1 gene and transfect them to asthmatic airway smooth muscle cell to study the effects of CyclinD1 on the proliferation of airway smooth muscle cells in asthmatic rats. CyclinD1 cDNA was obtained by RT-PCR of total RNA extracted from the airway smooth muscle in asthmatic rats. The sequence was inserted into eukaryotic expression vector pcDNA3.1 (+) to recombinate the sense and antisense pcDNA3.1-CyclinD1 eukaryotic expression vector. The two recombinations and vector were then separately transfected into airway smooth muscle cell in asthmatic rats by using liposome. The expression level of CyclinD1 was certificated by Western blotting analysis. The proliferations of ASMCs isolated from asthmatic rats were examined with cell cycle analysis, MTT colorimetric assay and proliferating cell nuclear antigen (PCNA) immunocytochemical staining. Results showed (1) Compared with control group, the content of CyclinD1 was significantly increased; (2) It was comformed by restriction endonucleasa digestion and DNA sequence analysis that the expression plasmid of sense and antisense CyclinD1 were successfully recombinated. There was significant change of CyclinD1 expression between vector and sense CyclinD1 transfected cells, and the expression level of CyclinD1 in ASMC transfected with antisense CyclinD1 was lower than that in vector transfected cells (P <0.01); (3) In the asthmatic groups, compared with the vecter group, the percentage of S + G2M phase, absorbance A value of MTT and the expression rate of PCNA protein in ASMC transfected with pcDNA3. 1-CyclinD1 vector significantly increased. The values decreased remarkably in the pcDNA3,1-as CyclinD1 group. Statistical analysis revealed that there were significant differences in these indicators of cell proliferation in three groups (P <0.01). In the normal groups, statistical analysis revealed that there were significant differences in the percentage of S + G2M phase, a value of MTT and the expression rate of PCNA protein in three groups (P <0.01). Sense CyclinD1 eukaryotic expression vectors could have a positive effect on the proliferation of ASMC, however the antisence one have a negative effect, which implicated that CyclinD1 might contribute to the process of airway smooth muscle cell proliferation.
