ABSTRACT

Objective To investigate the mechanism of Guiqiyiyuan Ointment (GO) for preventing and treating 12C6+ beam radiation induced lung and kidney bystander effect to provide a new strategy for prevention and treatment of clinical radiation injury. Methods Sixty healthy male Wistar rats were randomly and equally divided into 3 groups: NC group, SR group (simple radiation 2ml/kg), GO group(GO 2ml/kg intragastric administration for 7 days). The right side of the lung was modeled by 12C6+beam radiation. After modeling, the rats were killed at 48h. The left lung, left and right kidney tissues were taken from the rats. The DNA methylation rate was detected by ELISA assay, pathological changes were observed by HE staining, and the expressions of Dnmt1, Dnmt3a and Dnmt3b were detected by immunohistochemistry. Results Compared with the NC group, the level of DNA methylation was decreased significantly (P<0.01), the left lung showed inflammation, no abnormal finding was seen in the left and right kidneys, and the expressions of Dnmt1, Dnmt3a and Dnmt3b were significantly increased in the SR group (P<0.01). Compared with the SR group, the level of DNA methylation was increased significantly (P<0.01), the left lung inflammation became better, and the expressions of Dnmt1, Dnmt3a and Dnmt3b were significantly decreased in the GO group (P<0.01). Dnmt1, Dnmt3a and Dnmt3b proteins were expressed in the cytoplasms of bronchial and renal tubular epithelial cells in all the groups. The NC group presented as light brown-brown staining, showing a weak positive expression, the SR group as brown-brown staining, showing astrong positive expression, and the GO group as light brown-brown staining, showing a moderate positive expression. Conclusion The GO can reduce the bystander effect caused by 12C6+ beam radiation, and its mechanism is related to improving the level of DNA methylation.

ABSTRACT

Objective Xanthelasma palpebrarum presents a therapeutic and aesthetic challenge because of its high visibility and frequent rate of recurrence with a wide variety of treatments.Many patients were disappointed by an initial unsuccessful treatment and fail to return for further therapy until the problem had become quite conspicuous.So a simple treatment with few side effects and good acceptance remains an important goal.Methods Eleven patients with xanthelasma lesions were treated with the 1540nm erbium:glass laser.We could see erythema and swell around the skin lesion immediately.It needed irradiate again after one month if there was residual lesion.The end point was the lesions completely removed.The therapeutic effect of 1540nm erbium laser,the adverse reaction and the satisfaction of patients were observed.Results All lesions were removed without hyperpigmentation or scarring.Conclusions The 1540 nm erbium:glass laser represents an effective means for treating xanthelasmas with few side effects and good acceptance.

ABSTRACT

Objective To investigate the assessments from family member and nurse for nursing care needs of critical patients based on the Maslow's hierarchy of needs. Methods Questionnaires regarding nursing care needs of critical patients were designed according to the Maslow′s hierarchy of needs; these questionnaires were performed to family members and nurses, and the results were analyzed. Results There was difference regarding the order of various needs of critical patients between family members and nurses; compared to nurse, family members had significantly higher scores on love and belongingness need [(4.08±0.72) points vs. (3.44±0.63) points, t=5.61, P<0.01], and had obviously lower scores on safety needs [(3.71±0.62) points vs. (3.92±0.69) points, t=2.18, P<0.05]. Conclusions The asymmetry in assessments from family member and nurse for nursing care needs of critical patients exist objectively;during clinical nursing, communication with family members should enhance and provide humanized services and management measures should be adopted to improve family members′satisfaction towards nursing service.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the anti-apoptotic effect of low molecular weight heparin (LMWH) in the placenta of rats with preeclampsia-like symptoms.</p><p><b>METHODS</b>Thirty pregnant Wistar rats were randomized equally into 3 groups and received subcutaneous saline injection (control group), 200 mg/kg L-NAME injection to induce preeclampsia symptoms (PE group), or L-NAME with 40 µl/kg LMWH injections (LMWH group). The blood pressure, urine protein, the number of pups and the weight of the fetuses and placenta were measured, and the placental apoptotic index was measured by TUNEL assay. The expression of cleaved caspase-3, Bcl-2 and Bax in the placenta were examined by Western blotting.</p><p><b>RESULTS</b>Compared with the control group, L-NAME injections caused significantly increased blood pressure and urine protein (P<0.05), which were significantly lowered by LWMH (P<0.05). The number and weight of normal pups were significantly lower in PE group than in the control group (P<0.05) and LMWH group (P<0.05); in LMWH group, the weight of pups was still lower than that of the control group (P<0.05) but the number of normal pups was comparable (P>0.05). The LMWH group showed a significantly lower placental apoptosis index than the PE group (P<0.05) with also significantly lower cleaved caspase-3 and Bax and higher Bcl-2 expressions (P<0.05). The apoptosis index and expressions of cleaved caspase-3, Bcl-2 and Bax protein were similar between LMWH group and normal pregnant group (P>0.05).</p><p><b>CONCLUSION</b>LMWH can alleviate preeclampsia-like symptoms and decrease the apoptosis in the placenta of rats possibly by enhancing Bcl-2 and suppressing Bax expressions.</p>

ABSTRACT

Objective To express the unique gene of herpes simplex virus type 2(HSV-2) gG-2(gG-2T) in E.coli BL21,and study preliminarily the lemologic characteristics of gG-2T protein. Methods We cloned the gene of gG-2T and had gG-2T fusion protein expressed in E.coli BL21,and identified its activity by Western blot and indirect ELISA.Results Glycoprotein G-2T fusion protein could combine with GST polyclonal antibody through gG-2T fusion protein expressed in prokaryotic system and SDS-PAGE had a specific binding strap in 46 ku nearby.The gG-2T fusion protein could react with HSV-2 patients' serum,but not with HSV-1 patients' serum and r-Globulin.Conclusion We obtained gG-2T fusion protein,which can combine with GST polyclonal antibody and react with HSV-2 patients' serum specifically.It will provide an experimental basis for diagnostic kit and vaccine of gG-2 protein target area.
