ABSTRACT

The aim of this research was to evaluate the effect of abscisic acid (ABA) on gas exchange and the activity of antioxidant enzymes of Ormosia arborea (Vell.) Harms seedlings under water deficit and its influence on the recovery potential of the seedlings. The experiment was conducted using four treatments, being daily irrigation or water restriction without and with 10 μM ABA. Seedlings under water deficit + ABA showed greater adjustment to drought, and when re-irrigated, they restored photosynthetic metabolism and water potential. ABA minimizes the reduction in the photosynthetic metabolism and water potential of the leaf, however, it does not increase the antioxidant activity of the O. arborea seedlings under water deficit. These results suggest that this species exhibits plasticity, which enables it to survive also in environments subjected to temporary water deficit regardless of the supplementation of ABA. We suggest that other doses of ABA be researched to expand the beneficial effect of ABA on this species.


O objetivo deste trabalho foi avaliar o efeito do ácido abscísico (ABA) nas trocas gasosas e na atividade de enzimas antioxidantes de mudas de Ormosia arborea (Vell.) Harms sob deficiência hídrica e sua influência no potencial de recuperação das mudas. Resumo O objetivo deste trabalho foi avaliar o efeito do ácido abscísico (ABA) nas trocas gasosas e na atividade de enzimas antioxidantes de mudas de Ormosia arborea (Vell.) Harms sob deficiência hídrica e sua influência no potencial de recuperação das mudas. O experimento foi conduzido com quatro tratamentos, sendo eles irrigação diária ou restrição hídrica sem e com 10 M ABA. ABSTRACT

RESUMO: A Artrogripose Múltipla Congênita (AMC) é uma síndrome rara, que representa um grupo de condições congênitas caracterizado por contraturas articulares em duas ou mais articulações, não progressivas, geralmente simétricas, de etiologia desconhecida e que pode estar associada a outras malformações, como viscerais e neurológicas. Apresenta-mos dois casos isolados com achados característicos de AMC, atendidos em ambulatório de pediatria de alto risco de uma cidade do Oeste do Paraná. Caso 1: lactente, sexo feminino, 14 dias de vida, com redução dos movimentos fetais referidos pela mãe, apresentando malformações envolvendo somente os membros, visualizadas logo após o nasci-mento. Caso 2: lactente, sexo masculino, quatro meses de vida, com diagnóstico de hipocinesia fetal ao ultrassom obstétrico de 23 semanas, ao nascimento apresenta contraturas dos membros e disfunção do sistema nervoso cen-tral. O diagnóstico da artrogripose é complexo e essencialmente clínico, sendo necessária uma anamnese meticulosa, contemplando uma avaliação da gestação, via de parto, história familiar e um exame físico minucioso, classificando quais os tipos de contraturas, membros envolvidos, presença de musculatura alterada e de alterações neurológicas, a fim de diferenciar a AMC das outras síndromes que também cursam com contraturas articulares. O seguimento e tratamento dos lactentes com artrogripose devem ocorrer em acompanhamento multidisciplinar devido às variadas manifestações que a doença pode apresentar e à característica de recidiva ao longo dos anos. ABSTRACTArthrogryposis Multiple Congenita (AMC) is a rare syndrome that represents a group of congenital conditions character-ized by joint contractures in two or more joints, non-progressive, usually symmetrical, of unknown etiology and which may be associated with other malformations, such as visceral and neurological. We present two isolated cases with characteristic features of AMC, seen in a high-risk pediatrics outpatient clinic in a city in western Paraná. Case 1: infant, female, 14 days old, with reduced fetal movements reported by the mother, presenting malformations involving only the limbs, seen immediately after birth. Case 2: infant, male, four months old, diagnosed with fetal hypokinesia on 23-week obstetric ultrasound, at birth, presents limb contractures and central nervous system dysfunction. The diagnosis of ar-throgryposis is complex and essentially clinical, requiring a meticulous anamnesis, including an assessment of pregnan-cy, delivery, family history and a thorough physical examination, classifying what types of contractures, involved limbs, presence of altered musculature and neurological changes to differentiate AMC from other syndromes that also have joint contractures. The aftercare and treatment of infants with arthrogryposis must occur in multidisciplinary follow-up due to the varied manifestations that the disease may present and the characteristic of recurrence over the years. ABSTRACT

Background: ADAMTS are metalloproteases with disintegrin and thrombospondin motifs. They are secreted proteases playing a role in biological processes such as inflammation, angiogenesis, and urogenital development. ADAMTS have specific substrates, such as the proteoglycans (PG) versican, aggrecan, and brevican. Despite data indicating a role of ADAMTS in tumor invasion and metastases, effects played by these molecules in cancer progression are still controversial. In ovarian cancer, the importance of ADAMTS gene mutations was recently described and related to chemotherapy outcome. Objective: To analyze protein levels of ADAMTS-1, -4, and -5, and TIMP-3 in human ovarian cancer classified as benign, borderline, or malignant. We also assessed the expression of the ADAMTS substrates aggrecan, brevican, and versican in these neoplasms. Correlations between overall survival and protein expression were performed. Methods: Tumors were classified according to the WHO Classification of Tumors of Female Reproductive Organs. Protein and PG expression was studied by immunohistochemistry. Differences in labeling were analyzed by percent measurements of stained areas. Results: ADAMTS-1, ADAMTS-5, and its tissue inhibitor TIMP-3 are increased in borderline and malignant tumors compared to benign neoplasms. Aggrecan and versican levels were increased in malignant subtypes compared to benign ovarian cancer. Higher ADAMTS-1, TIMP-3, and versican expression was associated with a shorter overall survival. Conclusions: Comparison of protease, TIMP-3, and substrate expression showed that in malignant tumors all ADAMTS and TIMP-3 expression levels were significantly raised compared to the substrates studied.

ABSTRACT

A Aloe vera (L.) Burm.f. tem sido utilizada há milhares de anos na medicina tradicional para o tratamento de diversos males. O intuito desse trabalho foi o levantamento bibliográfico de artigos que evidenciassem a atividade farmacológica da Aloe vera. A revisão contemplou livros e periódicos nacionais e internacionais indexados nas bases de dados MEDLINE, LILACS e SciElo, nos idiomas português, inglês e espanhol, utilizando as palavras-chave citadas. Após o levantamento bibliográfico, constatou-se que várias atividades biológicas são atribuídas a Aloe vera. Evidências sugerem eficácia no tratamento da psoríase, herpes genital, queimaduras e hiperglicemia. Além disto, também foram demonstradas atividades antineoplásica, antimicrobiana, anti-inflamatória e imunomodulatória por estudos in vitro e in vivo, entretanto, na cicatrização de feridas, os resultados foram conflitantes. No tratamento de dermatite por radiação e em queimaduras solares sua eficácia não foi comprovada e foram relatados casos de hepatite aguda devido ao consumo de preparações orais. Tendo em vista as várias atividades comprovadas e poucos relatos acerca de sua contra indicação, conclui-se que o uso desta espécie corrobora o vasto uso popular.


The Aloe vera (L.) Burm.f. has been used for thousands of years in traditional medicine to treat various ailments. The aim of this study was to carry out a bibliographical review on the pharmacological activity of Aloe vera. This review included books and national and international journals indexed to MEDLINE, LILACS and SciELO, in Portuguese, English and Spanish, using the key words mentioned. After the literature review, we found that several biological activities have been attributed to Aloe vera. Evidence suggests efficacy in the treatment of psoriasis, genital herpes, burns and hyperglycemia. Moreover, antineoplastic, antimicrobial, anti-inflammatory and immunomodulatory activities have also been demonstrated by in vitro and in vivo studies; however, in wound healing, the results were conflicting. In the treatment of radiation dermatitis and sunburn, its efficacy has not been proven, and cases of acute hepatitis from the consumption of oral preparations have been reported. Considering the various proven activities and the few reports about the contraindications of Aloe vera, we conclude that the use of this species confirms its wide popular usage.

ABSTRACT

Hepatocellular carcinoma (HCC) is the third highest cause of cancer death worldwide. In general, the disease is diagnosed at an advanced stage when potentially curative therapies are no longer feasible. For this reason, it is very important to develop new therapeutic approaches. Retinoic acid (RA) is a natural derivative of vitamin A that regulates important biological processes including cell proliferation and differentiation. In vitro studies have shown that RA is effective in inhibiting growth of HCC cells; however, responsiveness to treatment varies among different HCC cell lines. The objective of the present study was to determine if the combined use of RA (0.1 µM) and cAMP (1 mM), an important second messenger, improves the responsiveness of HCC cells to RA treatment. We evaluated the proliferative behavior of an HCC cell line (HTC) and the expression profile of genes related to cancer signaling pathway (ERK and GSK-3β) and liver differentiation (E-cadherin, connexin 26 (Cx26), and Cx32). RA and cAMP were effective in inhibiting the proliferation of HTC cells independently of combined use. However, when a mixture of RA and cAMP was used, the signals concerning the degree of cell differentiation were increased. As demonstrated by Western blot, the treatment increased E-cadherin, Cx26, Cx32 and Ser9-GSK-3β (inactive form) expression while the expression of Cx43, Tyr216-GSK-3β (active form) and phosphorylated ERK decreased. Furthermore, telomerase activity was inhibited along treatment. Taken together, the results showed that the combined use of RA and cAMP is more effective in inducing differentiation of HTC cells.

ABSTRACT

Milk production of transgenic does was evaluated by ultrasound measurements of the mammary gland. Two Canindé goats, which were nine months of age were used in the trial, one non-transgenic or other transgenic for hG-CSF. For hormone-induced lactation, animals were given estradiol (0.25mg/kg, IM), progesterone (0.75mg/kg, IM), and prednisolone (0.4mg/kg, IM). Ultrasonographic exams were carried out during milking, using a Falcon 100 ultrasound equipment with a 5MHz convex probe and were performed by the same operator. The results were expressed as mean±standard error. The maximum greater length and shorter length of the cistern were respectively 5.14cm and 1.36cm for the transgenic animal and 7.28cm and 2.25cm for non-transgenic, which is consistent with the maximum milk volume produced. The relationship between the average area of cisterns and milk yield was expressed as a linear correlation curve, with a correlation coefficient significantly positive for both transgenic (Y=-1.1314+10.8538*x; r=0.97) and non-transgenic (Y=-21.7551+18.3634*x; r=0.97) animals. In conclusion, the ultrasound is a practice and appropriate technique to evaluate the cisterns in ruminant udders in transgenic animal.

ABSTRACT

Eremanthus erythropappus, commonly known as “candeia”, is an abundant pioneer tree species, forming dense populations known as “candeial”, but it is also found in forests at middle stages of succession. Trees from forests are bigger and occur in lower density than in the “candeial”. The objectives of the present study were to investigate if the decrease in population density during successional process is accompanied by 1) changes in within-population genetic diversity, and 2) differentiation of populations. Eight populations, four of early successional stage (“candeial”) and four of middle successional stages (forest), were analyzed with RAPD markers. The genetic diversity found was high compared to other tree species analyzed with RAPD markers. AMOVA revealed that most of the genetic variations of E. erythropappus were found within populations (85.7%), suggesting that this species is predominantly outcrossing. The relatively low differentiation among the populations can be attributed to small distances among the populations analyzed (0.2 to 10.8 km). No indication that populations from middle successional habitats show lower genetic variation than populations from early successional stages was found. The percentage of polymorphic fragments (82.8 and 84.8%) and the Shannon indexes (0.442 and 0.455) were similar in “candeial” and forest, respectively. These results suggest that if an increase in selection intensity occurred during succession, it did not result in a decrease in genetic diversity or that the selection effect was balanced by other factors, such as gene flow. Higher significant differentiation among E. erythropappus populations from “candeial” in relation to that among populations from forest was also not detected.

Mammalian seminal plasma contains among others, proteins called spermadhesins, which are the major proteins of boar and stallion seminal plasma. These proteins appear to be involved in capacitation and sperm-egg interaction. Previously, we reported the presence of a protein related to spermadhesins in goat seminal plasma. In the present study, we have further characterized this protein, and we propose ion-exchange chromatography to isolate this seminal protein. Semen was obtained from four adult Saanen bucks. Seminal plasma was pooled, dialyzed against distilled water and freeze-dried. Lyophilized proteins were loaded onto an ion-exchange chromatography column. Dialyzed-lyophilized proteins from the main peak of DEAE-Sephacel were applied to a C2/C18 column coupled to an RP-HPLC system, and the eluted proteins were lyophilized for electrophoresis. The N-terminal was sequenced and amino acid sequence similarity was determined using CLUSTAL W. Additionally, proteins from DEAE-Sephacel chromatography step were dialyzed and submitted to a heparin-Sepharose high-performance liquid chromatography. Goat seminal plasma after ion-exchange chromatography yielded 6.47 +/- 0.63 mg (mean +/- SEM) of the major retained fraction. The protein was designated BSFP (buck seminal fluid protein). BSFP exhibited N-terminal sequence homology to boar, stallion and bull spermadhesins. BSFP showed no heparin-binding capabilities. These results together with our previous data indicate that goat seminal plasma contains a protein that is structurally related to proteins of the spermadhesin family. Finally, this protein can be efficiently isolated by ion-exchange and reverse-phase chromatography.

ABSTRACT

This pilot project was designed to determine if normal kids could be produced after microinjection in pronuclear embryos and subsequent transfer to recipients in a transgenic goat program in Brazil. Twelve donors of the Saanen breed and 17 recipients of an undefined breed were used. The estrus of both donors and recipients was synchronized by a standard progestagen treatment and superovulation obtained by six pFSH injections. Donors in estrus were mated with fertile Saanen bucks. Zygotes were recovered surgically by flushing oviducts. The recovered zygotes with visible pronuclei were microinjected with 500 to 1000 copies of the human G-CSF gene. Two or four embryos were surgically transferred into the oviducts of recipients. One recipient became pregnant and two kids were born. No transgenic goat was identified after PCR analysis. Even though transgenic goats were not obtained, this experiment establishes the basis of a synchronization and superovulation regimen for use in goats raised in Brazil, for the purpose of collecting and manipulating the pronuclear embryos. This project also showed that microinjected one-cell goat embryos can survive to produce live young following surgical transfer

ABSTRACT

Utilizou-se a sincronizaçäo do estro com esponjas vaginais de 30mg de acetato de fluorogestona durante 12 dias para avaliar o efeito de diferentes doses de eCG sobre o desempenho reprodutivo de ovelhas. Na retirada das esponjas as ovelhas foram divididas em três grupos para receberem 0 (n=26), 200 (n=30) ou 400UI (n=30) de eCG. O estro foi detectado a cada 12h utilizando-se um rufiäo. As fêmeas foram inseminadas por laparoscopia 60h após a retirada das esponjas. Realizaram-se colheitas de sangue aos 5 e 18 dias pós-inseminaçäo para dosagem de progesterona e determinaçäo do número de ovulaçöes e prenhezes, respectivamente. A fertilidade foi verificada por ecografia aos 60 dias e ao parto. Das 86 ovelhas 70,9 por cento apresentaram estro. Essa porcentagem foi maior (P<0,05) nas fêmeas tratadas com eCG: 96,7 por cento (400UI) e 76,7 por cento (200UI) versus 34,6 por cento (0 UI). O intervalo entre o final do tratamento e o início do estro foi maior (P<0,05) no grupo sem eCG, 54,7 +/- 6,3h versus 45,9 +/- 7,8h para 200UI e 40,4 +/- 10,3h para 400UI. Verificou-se menor (P<0,05) número de ovulaçöes e prenhezes no grupo sem eCG. A näo aplicaçäo de eCG influiu negativamente no desempenho reprodutivo de ovelhas deslanadas

ABSTRACT

The effect of peritoneal fluid (PF) on the human sperm acrosome reaction (AR) was tested. Sperm was pre-incubated with PF and the AR was induced by calcium ionophore A23187 and a neoglycoprotein bearing N-acetylglycosamine residues (NGP). The AR induced by calcium ionophore was inhibited 40 percent by PF from controls (PFc) and 50 percent by PF from the endometriosis (PFe) group, but not by PF from infertile patients without endometriosis (PFi). No significant differences were found in the spontaneous AR. When the AR was induced by NGP, pre-incubation with PFc reduced (60 percent) the percentage of AR, while PFe and PFi caused no significant differences. The average rates of acrosome reactions obtained in control, NGP- and ionophore-treated sperm showed that NGP-induced exocytosis differed significantly between the PFc (11 percent) and PFe/PFi groups (17 percent), and the ionophore-induced AR was higher for PFi (33 percent) than PFc/PFe (25 percent). The incidence of the NGP-induced AR was reduced in the first hour of pre-incubation with PFc and remained nearly constant throughout 4 h of incubation. ABSTRACT

No Setor de Reproduçäo Humana da Disciplina de Ginecologia da Escola Paulista de Medicina os AA. avaliam os resultados de microcirurgia tubária em 202 pacientes, com e sem uso de Dextran 70%. Nos 202 casos referem permeabilizaçäo tubária em 90% dos casos. As taxas de gravidez pós-cirúrgicas foram de 27% no grupo sem o uso de Dextran e de 33,9% naqueles em que essa soluçäo glicosada foi utilizada. Concluem afirmando ser o uso do Dextran 70 útil para reduzir as aderências tubárias pós-operatórias

ABSTRACT

Os AA avaliaram o resultado da microcirurgia tubária em 196 casos operados na Clínica Ginecológica da Escola Paulista de Medicina, no período de 10 anos; de 1978 a 1987. Separam os casos de acordo com o tipo de cirurgia em Salpingostomia: 52 vezes; lise de aderências (salpingolise): 48; anastomose ístmico ístmica: 62 e reimplante tubo-uterino: 34. Descrevem as técnicas e concluem expondo seus resultados em termos de permeabilizaçäo e de gravidez. Afirmam que cerca de 25% dos casos resultam em gestaçäo, na média de todos os casos e especificam as porcentagens de cada tipo de cirurgia realizada
