ABSTRACT

Abstract Although invasive infections and mortality caused by Candida species are increasing among compromised patients, resistance to common antifungal agents is also an increasing problem. We analyzed 60 yeasts isolated from patients with invasive candidiasis using a PCR/RFLP strategy based on the internal transcribed spacer (ITS2) region to identify different Candida pathogenic species. PCR analysis was performed from genomic DNA with a primer pair of the ITS2-5.8S rDNA region. PCR-positive samples were characterized by RFLP. Restriction resulted in 23 isolates identified as C. albicans using AlwI, 24 isolates as C. parapsilosis using RsaI, and 13 as C. tropicalis using XmaI. Then, a group of all isolates were evaluated for their susceptibility to a panel of previously described killer yeasts, resulting in 75% being susceptible to at least one killer yeast while the remaining were not inhibited by any strain. C. albicans was the most susceptible group while C. tropicalis had the fewest inhibitions. No species-specific pattern of inhibition was obtained with this panel of killer yeasts. Metschnikowia pulcherrima, Pichia kluyveri and Wickerhamomyces anomalus were the strains that inhibited the most isolates of Candida spp.


Resumo Embora as infecções invasivas e a mortalidade causada por espécies de Candida estejam aumentando entre pacientes comprometidos, a resistência a agentes antifúngicos comuns também é um problema crescente. Analisamos 60 leveduras isoladas de pacientes com candidíase invasiva utilizando como estratégia PCR/RFLP baseada na região espaçadora transcrita interna (ITS2) para identificar diferentes espécies patogênicas de Candida. A análise por PCR foi realizada a partir de ADN genómico com um par de iniciadores da região ITS2-5.8S rDNA. As amostras PCR-positivas foram caracterizadas por RFLP. A restrição resultou em 23 isolados identificados como C. albicans usando AlwI, 24 isolados como C. parapsilosis usando RsaI e 13 como C. tropicalis usando XmaI. Em seguida, avaliou-se o grupo de todos os isolados quanto à sua susceptibilidade a um painel de leveduras killer previamente descritas, resultando em 75% sendo suscetíveis a pelo menos uma levedura killer, enquanto que as restantes não foram inibidas por qualquer cepa. C. albicans foi o grupo mais suscetível enquanto C. tropicalis teve o menor número de inibições. Não se obteve um padrão de inibição específico da espécie com este painel de leveduras killer. Metschnikowia pulcherrima, Pichia kluyveri e Wickerhamomyces anomalus foram as cepas que inibiram a maioria dos isolados de Candida spp.

ABSTRACT

Abstract Melipona colimana Ayala is an endemic species inhabiting temperate forests of pine and oak of south of Jalisco in Mexico. During a year, it was recorded every 15 days foraging activity, environmental parameters and the development of colonies of M. colimana in its wild habitat. For five minutes every hour from 7:00 to 21:00, the bees that entered and left the hive and bringing pollen and resin were registered. Every hour the relative humidity, temperature, wind speed and light intensity was recorded and related to foraging activity. Additionally, the weight of the colonies recently transferred to wooden boxes, the number of brood combs, honey pots and pollen were registered. The time of beginning and ending of the foraging activity differs from the reports of stingless bees of tropical weather and the same happens with the pollen collection. The environmental parameters that affect other tropical stingless bees in the foraging activity also affect M. colimana in temperate climate. It was determined that the major activity season and the presence of more pollen pots in the colony is from November through February, for what it could be the best time of the year for the division and obtainance of new colonies, while the critical period of minor activity and pollen flow was during rainy season. These data may be useful for the future sustainable use of this species in temperate climate.


Resumo Melipona colimana Ayala é uma espécie endémica de clima temperado que habita nas florestas de pinho e de azinheira do sul de Jalisco, no México. Registrou-se durante um ano, todos os quinze dias, a atividade de forragem, os parâmetros ambientais e o desenvolvimento de colônias de M. colimana no seu habitat silvestre. Registrou-se durante cinco minutos, cada hora das 7:00 às 21:00, as abelhas que entraram e saíram da colmeia e as que traziam pólen e resinas. Registrou-se toda hora, a humidade relativa, temperatura, velocidade do vento e intensidade da luz e relacionaram-se à atividade de forragem. Além do mais, registrou-se o peso de colônias recém transferidas a gavetas de madeira e contabilizou-se o número de panais, potes de mel e pólen. A hora de início e de fim da atividade de forragem difere com os reportes das abelhas sem ferrão de clima tropical e o mesmo acontece com a recolecção de pólen. Os parâmetros ambientais que afetam outros meliponinos de clima tropical na forragem também afetam a M. colimana em clima temperado. Determinou-se que a temporada de maior atividade e a presença de mais potes de pólen na colônia é de Novembro a Fevereiro, razão pela qual pode ser a melhor temporada do ano para a divisão e obtenção de novas colônias, enquanto que o período crítico de menor atividade e fluxo de pólen foi durante a temporada de chuvas. Esses dados podem ser úteis para o futuro aproveitamento sustentável de esta espécie em clima temperado.

ABSTRACT

Phenotypic diversity has been described in the central repeated region of the circumsporozoite protein (CSP) from Plasmodium vivax. Two sequences VK210 (common) and VK247 (variant) have been found widely distributed in P. vivax isolates from several malaria endemic areas around the world. A third protein variant called P. vivax-like showing a sequence similar to the simian parasite P. simio-ovale has also been described. Here, using an immunofluorescent test and specific monoclonal antibodies, we assessed the presence of two of these protein variants (VK210 and VK247) in laboratory produced sporozoite. Both sequences were found in parasite isolates coming from different geographic regions of Colombia. Interestingly, sporozoites carrying the VK247 sequence were more frequently produced in Anopheles albimanus than sporozoites with the VK210 sequence. This difference in sporozoites production was statistically significant (p <0.05, Kruskal-Wallis); not correlation was found with parameters as the total number of parasites or gametocytes in blood from human donors used to feed mosquitoes. Previous studies in the same region have shown a higher prevalence of anti-VK210 antibodies which in theory may suggest their role in blocking the development of sporozoites carrying the CSP VK210 sequence

ABSTRACT

Se einforman seis casos de enfermedad de Waldenstrom estudiados en el orden clinico y humoral, y se destaca la diversidad de formas clinicas presentes. Se hizo evidente la presencia de manifestaciones hemorragicas e infecciones; esta ultima asociada frecuentemente a una inmunodeficiencia secundaria. Llama la atencion la presentacion de herpes zoster en dos enfermos La anemia estuvo presente en la totalidad de los pacientes con una persistente elevacion de las cifras de eritrosedimentacion. Es fundamental el estudio inmunologico como complemento del estudio hematologico clasico. Todos los pacientes sobre pasaron la edad de 50 anos

ABSTRACT

Entre el 26 de noviembre de 1982 y el 11 de julio de 1983, se han realizado 10 interposiciones viscerales con anastomosis mecanica EEA en el esofago cervical, 8 por cancer avanzado inextirpable y 2 luego de la reseccion del tumor. Se interpuso 6 veces el estomago y 4 el colon, usando preferentemente la via retroesternal. Se analizan las indicaciones, tecnica y resultados. No se han observado beneficios tales que justifiquen la suplantacion de la sutura manual
