ABSTRACT

Background Research on non-target organ damage of biological pesticides has attracted much attention. Rotenone exposure may be far beyond the occupational environment, and the exposureduring pregnancy may be increased through bioaccumulation, fruit or vegetable residues, and other forms of oral intake. At present, the effects of rotenone on placental development and its mechanism are still unknown. Objective To investigate the developmental damage of rat placenta and evaluate the expression levels of glycogen synthase kinase (GSK-3β) and beta catenin (β-catenin) followed by rotenone exposure through the placental barrier during pregnancy, as well as to propose possible associated mechanisms. Methods Eighteen sexually mature SD female infertile rats without specific pathogens were selected and divided into three groups: blank control group (0.9% saline), corn oil group, and rotenone group (corn oil + 2 mg·kg−1 rotenone) by random number method, six female animals in each group. Another six male rats were selected and mated to the female rats at night with a female to male ratio of 3:1 per cage. Pregnant rats were given 0.9% saline, corn oil, and 2 mg·kg−1 rotenone preparation by isovolumetric gavage once daily for the entire gestation period (19 d), and their conditions were observed after the last dose. The pregnant rats were anesthetized, and the size of the placenta and blood perfusion were detected by ultrasound the next day of the last dose of rotenone. Then, 3 pregnant rats in each group were sacrificed immediately and the placenta and umbilical cord tissues were dissected. The remaining 9 pregnant rats gave birth naturally, and the fetuses were observed for developmental evaluation and weighed. The histopathological changes of umbilical cord and placenta were observed by hematoxylin-eosin staining. The reactive oxygen species levels of placenta tissues were detected by flow cytometry. The Ca2+-ATPase activity of placenta tissues was detected by colorimetric method. The localization and levels of GSK-3β and β-catenin expression of placenta were detected by immunohistochemistry. The p-GSK-3β/GSK-3β and p-β-catenin/β-catenin protein expression in placental tissues were measured by Western blotting. Results No injury or death was recorded during the pregnant rats receiving rotennon administration. Adverse pregnancy outcomes such as fetal absorption and postpartum stillbirth were found in the rotenone group, and the weight of the fetal mice decreased (P<0.05). The B-ultrasound showed disc-shaped placenta with a thick middle and thin edge, smooth fetal surface, rough maternal surface, visible placental lobules, granular echotexture of the placenta with comma-like echogenic densities, and chorionic plate showing deep indentations, no calcification, degeneration, or necrosis in each group. Compared with the corn oil group, the fetal surface diameter of the placenta was reduced in the rotenone group (P<0.05). The Doppler color ultrasound showed that interplacental blood flow was reduced in the rotenone group, while interplacental blood flow was abundant in the blank control and the corn oil groups. The hematoxylin-eosin staining results showed that smooth muscle cells in the umbilical cord tissues of rats were loosely arranged, with fuzzy nuclei and inflammatory infiltration in the rotenone group. The placental trophoblast cells were small in size, disorderly arranged with nuclear fragmentation and cytoplasm turbidity. The tissue reactive oxygen species level in the rotenone group was higher than those in the other two groups (P<0.05). The Ca2+-ATPase activity of placental tissues was reduced in the rotenone group (P<0.05). The immunofluorescence low-magnification observation showed that GSK-3β and β-catenin were expressed in placental tissue, weak fluorescence expression in the decidua basalis, strong fluorescence expression in the labyrinthine layer structure. The labyrinthine layer under high magnification showed that compared with the blank control group and the corn oil group, the brightness of β-catenin fluorescence expression in the rotenone group decreased (P<0.05), and the brightness of GSK-3β expression increased (P<0.05). The Western blotting results showed that the expression of β-catenin and p-GSK-3β proteins decreased (P<0.01), and the expression of GSK-3β protein increased (P<0.01) in the rotenone group. No significant expression of p-β-catenin protein was detected in the placenta tissue of each group. Conclusion Rotenone exposure during pregnancy induces placental hypoperfusion, growth retardation, and oxidative stress in rats, as well as down-regulation of β-catenin and p-GSK-3β protein expression, and up-regulation of GSK-3β protein expression, which may further lead to abnormal pregnancy and fetal restricted growth.

ABSTRACT

Objective To investigate the influence of fast track surgery on clinical effect,anal dynamics and living quality in treatment of low retcal cancer anus saving patients.Methods Screening 100 cases of low retcal cancer anus saving patients as research subjects.According to a random number ta-ble method,100 cases were divided into twe group:study group(50 cases)and control group(50 cases). Patients in control group were given traditional perioperative treatment,and patients in study group were given FTS perioperative treatment,then the two groups were compared in terms of the clinical effect and a-nal dynamics.Results There was no statistically significant difference in operation time between the two groups(P>0.05).The Intraoperative blood loss of study group and control group were(221.4 ± 93.1)ml and(263.2 ± 76.4)ml(P<0.05),first exhaust time after operation of the two group were(3.7 ± 0.6)d and(5.6 ± 0.8)d(P<0.05),intraoperative infusion volume of the two group were(2261.5 ± 670.2)ml and(2574.2 ± 635.0)ml(P<0.05),hospital stay after operation of the two group were(8.5 ± 2.1)d and (13.2 ± 2.0)d(P<0.05),first feeding time of the two group were(4.1 ± 1.2)d and(6.6 ± 1.8)d(P<0.05).The postoperative complication rate in study group(18.0%)was lower than the control group (36.0%)(P<0.05);3 months after operation,the anal function good rate of the two group were 74.0%and 54.0%(P<0.05),anal maximum systolic blood pressure,anus maximum resting pressure,static vec-tor volume and contraction vector volume of study group were(245.2 ± 20.1)mmHg,(165.0 ± 17.3) mmHg,(32792.4 ± 3887.6)cm(mmHg)2and(68647.5 ± 2865.3)cm(mmHg)2),respectively.While in control group were(189.5 ± 18.5)mmHg、(88.9 ± 16.8)mmHg、(28986.6 ± 3946.8)cm(mmHg)2and(43462.9 ± 3013.0)cm(mmHg)2),respectively(P<0.05).The score of FACT-G of one year after op-eration in in the study group were higher than the control group(P<0.05).Conclusion The fast track surgery on low retcal cancer anus saving patients has a more significant advantage,it can improve clinical effect,reduce the postoperative complication rate and improve the anal dynamics,thus it is worthy of cini-cal promotion.

ABSTRACT

Objective To investigate the expression alteration and significance of long non-coding RNA (lncRNA)by GWAS(Genome-wide association study)between Esophageal squamous cell carcinoma(ESCC)and its adjacent normal esophageal tissues. Methods lncRNA and mRNA differential expression in 6 pairs of ESCC and matched non-cancerous tissues were screened by microarray assay. The target genes were predicted. Finally , GO(Gene Ontology)and Pathway analysis was used for the further research of lncRNA. Results A total of 680 lncRNA and 1472mRNA were differentially expressed at more than two-fold change(P ≤ 0.05,with 161lncRNA and 653mRNA up-regulated,519lncRNA and 819mRNA down-regulated between ESCC and its adjacent normal esophageal tissues. Gene ontology and pathway analysis results suggested that the differentially expressed genes were involved in 11 pathways.Theyare potentially associated with esophageal squamous cell carcinoma ,including post-translational protein modification ,mucin type O-Glycan biosynthesis and sphingolipid metabolism pathways , which mainly related to the changes of molecular function ,cellular components and biological processes. Through cis and trans analysis,a total of 15 differentially expressed lncRNA had cis-and/or trans-regulated target genes in the database,with 13 lncRNA had cis-regulated target genes,3 lncRNA had trans-regulated target genes,and 1 lncRNA had both cis- and trans-regulated target genes. Conclusion Compared with adjacent normal tissues ,a large number of lncRNA were expressed differentially in ESCC in Xinjiang Han people.Aberrantly expressed lncRNA may play important roles in ESCC development and progression through some signaling pathways ,which are of great significance for further search of new targets for the diagnosis and treatment of ESCC.

The implementation of online academic lectures for librarians by making use of QQ group was studied by analyzing the feasibility and advantages of giving online academic lectures for librarians in Guizhou Province , which showed that this method can solve the problems , such as a small number of librarians attending a lecture and high cost in organizing a lecture, thus opening a new way for continuing education and training of librarians.

ABSTRACT

<p><b>OBJECTIVE</b>To screen the microRNAs involved in colon cancer proliferation and to investigate the expression and regulating function of target miRNA in colon cancer.</p><p><b>METHODS</b>Mitochondrial transcription factor A(TFAM), which was proved to be an oncogene to colon cancer in prior study, was used as target gene. The microRNAs involved in colon cancer proliferation were screened with miRWalk 2.0 software. The expression of screened miRNAs was examined in 30 samples of colon cancer tissue, para-cancer tissue, normal colon cell strain, and 3 colon cancer strains (SW480, HT-29, and HCT116) by real-time PCR. MiR-204 presenting lowest expression was selected to further study in SW480 cells. Dual luciferase reporter assays was performed to examine the association of TFAM with miR-204. Anti-miR-204 lentivirus and miR-240 lentivirus were used to down-regulate and up-regulate miR-204 expression respectively. Change of TFAM protein expression in SW480 cells was detected by Western blotting, and change of SW480 cells proliferation was detected by MTT and BrdU assay after lentivirus transfection.</p><p><b>RESULTS</b>After screening, the candidate miRNAs were miR-204, miR-211, miR-214, miR-381 and miR-590-3p. Expressions of miR-204, miR-211, miR-214 and miR-381 were lower, but miR-590-3p expression was higher, in colon cancer tissues than those in para-cancer tissues(all P<0.05). Meanwhile expressions of above 4 miRNAs(miR-204, miR-211, miR-214 and miR-381) were also lower, but miR-590-3p expression was higher as well, in SW480, HT-29 and HCT116 cells compared to normal colon cells(all P<0.05). Among above 4 miRNAs, miR-204 showed the lowest expression in both colon cancer tissues and cell lines. Expression of miR-204 was negatively correlated with TFAM expression in colon cancer tissues(P<0.05). Dual luciferase reporter assays revealed TFAM could be integrated with miR-204 directly, suggesting TFAM as the direct target of miR-204. After up-regulating miR-204 by lentivirus, expression of TFAM decreased and proliferation increased in SW480 cells(all P<0.05). After down-regulating miR-204 by lentivirus, expression of TFAM increased and proliferation decreased in SW480 cells(all P<0.05).</p><p><b>CONCLUSION</b>MiR-204 inhibits TFAM expression and up-regulates the proliferation of colon cancer cells SW480.</p>

ABSTRACT

Objective To investigate the correlation between serum interleukin-8 (IL-8 )and immunoglobulin/complement 3A (IgA/C3)and immune imbalance in Henoch-Schonlein Purpura(HSP)in children,and provide evidence and guidance for clinical therapy of HSP.Methods Serum IL-8 was measured by Enzyme-linked immunosorbent assay(ELISA),and IgA and C3 were de-tected by automatic biochemistry analyzer,and calculate the ratio of IgA/C3.Results Serum IL-8 levels were (389±5 1.68)ng/L, significantly higher than that in healthy children (P < 0.05 ).Serum IgA levels were(306.2 ± 32.21 )mg/L,significantly higher (P <0.05).Serum C3 levels were (1 14.5 ±20.7)mg/L,of no statistical difference.IgA/C3 were (3.041 ±0.508 5),significantly higher(P <0.05).There was a positive correlation between IL-8 and IgA/C3 (r =0.534,P <0.05 ).Conclusion The children of HSP has cytokine and immunoglobulin disturbance,and presents a hyperreactivity in both humoral immunity and cellular immunity. Positive correlation between IL-8 and IgA/C3 was observed.

ABSTRACT

Objective To construct a recombinant Bacillus Calmette-Guerin ( BCG ) vaccine strain, rBCG::Rv3478-pMV261, expressing the Rv3478 protein of Mycobacterium tuberculosis and to inves-tigate its immunogenicity.Methods The gene fragments encoding Rv3478 antigen were amplified by PCR and then respectively cloned into pMV261 and pET-28a vectors to construct the recombinant expression plas-mids (Rv3478-pMV261 and Rv3478-pET-28a).The Rv3478-pMV261 plasmids were transformed into the BCG cells to construct the rBCG vaccine strains, while the Rv3478-pET-28a plasmids were transformed into Escherichia coli BL21 strains for the expression of Rv3478 protein.Polyclonal antibodies were induced in mice upon the immunization with Rv3478 protein.The rBCG vaccine strains overexpressing Rv3478 protein were screened out with Western blot assay.The C57BL/6 mice were divided into four groups including the PBS treated group, BCG treated group, rBCG::pMV261 ( R0) treated group and rBCG::Rv3478-pMV261 ( R3) treated group.All mice were sacrificed in 4 or 12 weeks after immunization.Enzyme-linked immunos-pot assay ( ELISPOT) , ELISA and flow cytometry analysis were performed to evaluate the induced humoral and cell-mediated immune responses in mice.Results The Rv3478 protein was successfully expressed and could induce polyclonal antibodies in mice.High levels of IFN-γand TNF-αwere detected in mice treated with R3, indicating that the immunization with R3 enhanced the cellular immunity.Moreover, the ratios of CD4+to CD8+T cells and the percentages of CD44+CD62L+T cells were increased in mice upon the immuni-zation with R3.Conclusion The recombinant BCG vaccine strain overexpressing Rv3478 protein could in-duce stronger cell-mediated immune responses in mice.It might be have a great significance as a new tuber-culosis( TB) vaccine strain against TB infection in the future.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the feasibility of monitoring clopidogrel resistance with flow cytometric analysis of platelet vasodilator stimulated phosphoprotein (VASP) phosphorylation.</p><p><b>METHODS</b>Twenty patients with coronary heart disease (CHD) and 17 healthy volunteers were examined for platelet aggregation rate and phosphorylation of VASP (calculated as platelet reactivity index, PRI) using traditional optical nephelometry and flow cytometry before and after concurrent therapy of clopidogrel and aspirin.</p><p><b>RESULTS</b>No significant differences were found in PRI between CHD group and healthy control group [(89.45∓5.22)% vs (86.58∓4.35)%] before treatment. The PRI in CHD group was significantly lowered after treatment to (67.66∓19.77)% (P<0.05). Clopidogrel resistance was found in 6 (30%) cases in CHD group by flow cytometric analysis, which showed a higher sensitivity than optical nephelometry (10%).</p><p><b>CONCLUSION</b>Flow cytometric analysis of VASP phosphorylation is a more reliable test to specifically evaluate clopidogrel resistance.</p>

ABSTRACT

OBJECTIVE@#Discussing the use of endoscopy in the operation of microvascular decompression of cranial nerves to treat trigeminal neuralgia (TN), hemifacial spasm (HFS), glosspharyngeal neuralgia (GN) and some tumors of the cerebellopontine angle (CPA).@*METHOD@#Since 2006, 973 cases (including 420 cases of TN, 487 cases of HFS, 66 cases of GN) had received the operation of nervous decompression by approach of postauricular suboccipital to the CPA. All cases who used the endoscopy and the material was described in detail.@*RESULT@#In 420 TN patients, 390 (92.9%) were found responsible blood vessels. Two (0.5%) were found no responsible blood vessels but arachnoid adhesion, 28 were found CPA tumor secondary trigeminal neuralgia. In 487 HFS patients, 486 were found responsible blood vessels. Between the vascular and trigeminal nerve, teflon was used to decompress and isolate the facial nerve. In 66 GN, the glossopharyngeal nerve were cut and vagus nerve were decompressed. The short-term cure rate was 100%. Cerebrospinal fluid rhinorrhea occurred in 3 cases, 1 case of cerebellar infarction, 2 patients of cerebellar hemorrhage, 2 cases of pneumatosis. All complications were cured at last. There were no deaths. All cases were followed-up for 1 to 5 years. Four cases of TN and 4 cases of HFS recurrent, and none of GN recurrence.@*CONCLUSION@#The microneurosurgery of nervous decompression endoscopy technology for the treatment of TN, HFS, GN and some tumors of the CPA is an ideally functional and etiotropic operation. It is helpful to detect the responsible blood vessel and to protect the brain tissue and nerve function with endoscope, which can improve the success rate of the operation and avoid the complications. This technology has clinical application value.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of voluntary exercise on the proliferation and differentiation of hypothalamus progenitor cells in adult rats.</p><p><b>METHODS</b>Male Wistar rats were divided into voluntary exercise (EX) and sedentary (SE) groups, both of which were further divided into 6 subgroups for observation on days 6, 13, 23, 33, 43 and 53. Bromodeoxyuridine (BrdU) was intraperitoneally injected daily for 5 consecutive days after commencing voluntary exercise, and at the specified time points during voluntary exercise, the rats' brains were removed to observe the numbers of BrdU-positive cells in the hypothalamus.</p><p><b>RESULTS</b>Immunohistochemical analysis showed that the numbers of BrdU-positive cells in the hypothalamus of EX subgroups were significantly greater than those of SE subgroups on days 23, 33, 43 and 53. In EX group, the number of BrdU-positive cells double-stained for a mature neuron marker increased after 43 days of voluntary exercise, which did not occur in SE group.</p><p><b>CONCLUSION</b>Long-term voluntary exercise can promote the proliferation of neuronal progenitor cells in the hypothalamus and their differentiation into neurons.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of acute heat stress on the day-night circadian gene Per2 mRNA expression in the liver of rats.</p><p><b>METHODS</b>Male Wistar rats were randomly divided into two groups and exposed to heat at 32 degrees celsius; or to a room temperature at 24 degrees celsius; (control). After 7 days of heat exposure, the body temperature was measured by telemetry. The relative weight of the pituitary and adrenal glands was determined after the exposure, and liver Per2 mRNA expression level was detected using RT-PCR.</p><p><b>RESULTS</b>Acute heat stress did not obviously affect body temperature or body weight of the rats. Seven days of heat exposure increased the relative weight of the pituitary and adrenal glands and significantly lowered Per2 mRNA expression level at night.</p><p><b>CONCLUSION</b>Acute heat stress can interfere with the day-night circadian gene Per2 mRNA expression in rats.</p>

ABSTRACT

Objective:To study the impact of rotary cell culture system(RCCS) on the quality and activity of fetal islets of pancreas after cryopreservation and resuscitation.Methods:The fetal islets of pancreas were assigned into three groups averagely,experiment group 1,2 and control group.In experiment group1 and 2,fetal islets of pancreas were cultivated in RCCS and common culture respectively.While in control group,fresh fetal islets of pancreas were cultivated in RCCS all the time.We resected the fetal pancreas and digested with Collagenase V and purified with ficoll solution.Then standard cryopreservation step was adopted,and after resuscitation,the islets of pancreas were cultivated continuously.And the quantity,quality and activity of the islets of pancreas,and insulin stimulation test results in all groups were detected.Results:After purification,islets got from one fetal pancreas reached 2,331.98-5,115.43 IEQ(average 3,551.27? 253.76 IEQ) .The survival rate,the insulin release and stimulation index in islets of pancreas of group 1 incubated in RCCS were higher than those in common culture.Conclusion:RCCS are good for the growth and proliferation of islets of pancreas,and the islets of pancreas cultivated in RCCS have better insulin secreting capacity.RCCS combined standard cryopreservation method may further improve the cryopreservation effect.

Objective To identify one umbilical blood sample with abnonnal gap-PCR products of three bands of α2,-α3.7 and-SEA.further family pedigree were analyzed for the source of genetic variations,Methods One fetal umbilical blood sample was drawn from a woman of 24-weeks pregnancy.Gap-PCR for α-thalassemia was routinely conducted and abnornlal three bands of α2.-α3.7 and-SEA were observed.which could not be interpreted according to the kit manual and suspected as rare variation.With informed consen,DNA samples from the parents and grandparents were obtained for further study.Singleplex andnested PCR techniques were utilized to analyze the molecular characteristics of DNA samples from this fetus and its parents and grand-parents.Results Hematological phenotype study showed that fetal Hb Ban's was 7.6%,and its mother and maternal grandfather were both with typical α-thalassemia.while its father,grandfather and grandmother and maternal grandmother are without abnormal hematological change.Molecular study showed that fetal blood DNA was a heterozygosity for HKaa and-SEA.its father and grandfather are both HKαα/αα,its mother and maternal grandfather are both-SEA/aa,its grandmother and maternal grandmother are with both normal alleles of αα/αα.Then after genetic counseling the fetas was saved and iS a she baby now.Conclusion ThroUgh careful molecular tests one case of prenatal heterozygosity of HKαα/-SEA was identified,and the fetus is kept successfully through careful clinical counseling.

OBJECTIVE@#To investigate the value of the endoscopy in the operation of microvascular decompression (MVD) for the hemifacial spasm by approach of postauricular suboccipital to the cerebellopontine angle (CPA) with posterior auricular small incision.@*METHOD@#Two hundred and eighty-six cases of hemifacial spasm had received the operation of MVD with endoscopy by approach of postauricular suboccipital to the CPA.@*RESULT@#Responsible blood vessels were found in 285 cases (99.7 percent), including 264 cases of anterior inferior cerebellar artery, 21 cases of basilar artery. The root entry zone of the facial nerve were completely decompressed with Teflon. There is no responsible blood vessels but adhesion in 1 cases (0.3 percent). After surgery, hemifacial spasm immediately disappeared. House-Brackmann(1985) grading system was used to evaluated the recovery of facial nerve function. After 1 week of operation 196 cases' facial nerve function are stage 1/6, 62 cases' are stage 2/6, 23 cases' are stage 3/6, 4 cases' are stage 4/6, 1 cases' is stage 5/6. And after Six months of operation, 274 cases' are stage 1/6, 10 cases' are stage 2/6, 2 cases' are stage 3/6. After 1 month of operation there is no significant change of hearing in 238,there are 35 cases of hearing loss less than 20 dB, 10 cases of hearing loss more than 20 dB, but less than 50 dB,3 cases of hearing loss more than 50 dB. Ear-nose cerebrospinal fluid leakage occurred in 2 cases are cured. During 1 year to 4 years following-up, only 3 (1.0 percent) preliminary suffered relapse,among which 1 case was cured by the secondary operation. The long term cure rate was 99.3 percent without occurrence of serious complications such as death.@*CONCLUSION@#The microneurosurgery of MVD for the treatment of hemifacial spasm is an ideally functional and etiotropic operation. It is useful not only to discover the responsible blood vessels which are regarded as those pressing the root entry zone of facial nerve,but also to protect the function of the brain tissue and nerves as well. It is a safe, minimally invasive and efficient operation. To avoid the complications, enough knowledge of the craniotopography and skilled technique of endoscopic operation are primary.

ABSTRACT

OBJECTIVE:To establish a HPLC-fluorescence detection method for the determiniation of the serum concentration of metoprolol.METHODS:The determination was performed on Symmetry C18 column,and the mobile phase consisted of acetonitrile-water-triethylamine -phosphoric acid(110∶390∶2.5∶1.6) at a flow rate of 0.8 mL?min-1.The excitation wavelength was 265 nm and the emission wavelength 298 nm,and the sample size was 20 ?L.RESULTS:Good linearity was obtained for metoprolol over the range of 2.0~100.0 ?g?L-1 with correlation coefficient r=0.999 6.At low,medium and high concentrations,the average recoveries of metoprolol were 99.73%,98.21% and 99.38%,respectively.The intra-day RSD were 2.89%,2.36% and 1.32%,respectively,and the inter-day RSD were 3.73%,3.03% and 2.25% respectively.The lowest detectable limit was 1.0 ?g?L-1.CONCLUSION:This method is precise,accurate,specific,simple yet with high recovery,and it is applicable for clinical monitoring of blood concentration and pharmacokinetic study.

ABSTRACT

Objective:Cholangioduodenal reflux is still an unresolved problem in interposed jejunal choledochoduodenostomy (IJC) yet.To settle this problem,we designed IJC with distal suspending (IJCDS).Methods:IJCDS is to insert and suspend the distal end of the 25 cm-ling interposed jejunum to the duodenal cavity,which is about 2.5 cm long.The pressure of the biliary tract and the biochemical composition or the bile were measured during and after the operation and the results were compared.Barium meal and cholangiography through T tube were performed after the operation.Follow-up surveys were also done.Results:The pressure or the biliary tract decreased distinctively after the operation.Barium meals found no cholangioduodenal reflux although the patients had changed many postures.Cholangiography through T tube showed parisaltic or the interposed jejunum and the contrast medium was continuously pushed into the duodenum.Noreflux cholangitis and stone recurrence were found in the follow-up survey in 14 patients.Conclusion:IJCDS has the powerful function on preventing cholangioduodenal reflux.On the other hand,it avods the shortcomings of other reflux-preventing operative pattern.So it had the good prospect to extend its clinical usage.
