ABSTRACT

Objective To investigate the effect of short-term complications after D2 radical gastrectomy on long-term survival rate of gastric cancer patients. Methods A retrospective case-control study was conducted on 421 patients with gastric cancer who underwent D2 radical gastrectomy. According to the short-term postoperative complications, they were divided into experimental group (complication group, n=76) and control group (without complication group, n=345). In order to reduce the selection bias, the long-term survival rate of the two groups was tested by Kaplan-Meier survival analysis method after balancing the variables by propensity score matching (PSM). Log rank method was used for univariate analysis and Cox multivariate analysis was used for prognostic factors. Results There was no significant difference in long-term survival rate between the experimental group and the control group (P > 0.05). Histological type, lymph node metastasis rate and pTNM stage were independent risk factors for long-term survival. Conclusion The short-term complications after D2 radical gastrectomy have no significant impact on the long-term survival rate of gastric cancer patients, but it has certain clinical significance to actively prevent and control complications.

ABSTRACT

Objective: To construct a lentiviral vector with overexpression of endoplasmic reticulum protein 29 ERp29), and to explore the effects of ERp29 overexpression on the proliferation, migration and invasion of gastric cancer MGC803 and SGC7901 cells. Methods: The lentiviral plasmid with ERp29 pCDH-ERp29) tagged with red fluorescentce protein and control plasmid pCDH-Vector) were constructed, and then the MGC803 and SGC9901 cells were infected with these lentivital vectors. Under fluorescence microscope, the infection of these cells were observed. CCK-8 assay and clone formation assay were performed to test the proliferation abilities of MGC803 and SGC7901 cells infected with ERp29. Transwell chamber assay was used to test the abilities of migration and invasion of MGC803 and SGC7901 cells infected with ERp29. Real-time PCR and Western blotting methods were used to detect the expression levels of ERp29 mRNA and protein in MGC803 and SGC7901 cells which were stably infected by ERp29, respectively. Results: The enzyme digestion identification result showed that the pCDH-ER29 overexpression vector was successfully constructed. The fluorescence microscope result showed the successful infection in the gastric cancer cells. The CCK-8 assay and clone formation assay results showed that compared with pCDH-Vector group, the proliferation ability of the cells in pCDH-ERp29 group had no marked changes (P > 0.05). Transwell chamber assay revealed that compared with pCDH-Vector group, the number of migration and invasion cells in the MGC803 and SGC7901 cells in pCDH-ERp29 group were significantly decreased (P < 0 . 05). The expression levels of ERp29 mRNA and protein in the cells in pCDH-ERp29 group were significantly increased compared with pCDH-Vector group (P < 0 . 05). Conclusion: The lentiviral vector with overexpression of ERp29 is constructed successfully, and the overexpression of ERp29 in MGC803 and SGC7901 cells can significantly inhibit their abilities of migration and invasion.

ABSTRACT

Objective: To analyze the expressions of myeloid differentiation factor 88 (MyD88) in cancer tissue and adjacent tissue of the patients with colorectal cancer and to explore the relationships between its expression and the clinicopathological parameters and the prognosis of the patients with colorectal cancer, and to clarify the clinical significance of MyD88 expression. Methods: A total of 90 cases of tumor tissue and adjacent tissue of the patients with colorectal cancer were collected and their clinical pathological materials were analyzed. The expressions of MyD88 in tumor tissue and adjacent tissue of the colorectal cancer patients were detected by immunohistochemistry. The expression levels of MyD88 in different tissues were analyzed by Image-Pro Plus 6. 0 software. Kaplan-Meier method was used for survival analysis in the 90 patients with colorectal cancer. Results: The expression level of MyD88 in colorectal cancer tissue was significantly higher than that in adjacent colorectal cancer tissue (P<0. 001). The MyD88 expression level had no correlations with the age, gender, tumor size and histologreal differentiation of the patients with colorectal cancer (P<0. 05). The MyD88 expression level had correlations with the clinical stage, T stage, M stage and lymph node metastasis (P<0. 05). The survival rate of the patients with colorectal cancer with higher expression of MyD88 was significantly lower than that of the patients with colorectal cancer with low expression of MyD88 (P<0. 05). Conclusion: MyD88 is highly expressed in colorectal cancer tissue and it is associated with the clinical stage, T stage, M stage, and the situation of lymph node metastasis of the patients with colorectal cancer.

Objective To study the clinical efficacy of Bikini incision minimally invasive total hip arthroplasty on elder patients with femoral head necrosis and its effect on bleeding.Methods Seventy-six elder patients with femoral head necrosis in Sanmenxia Central Hospital from January 2015 to December 2016 were randomly divided into the study group(38 cases)and the control group(38 cases).The patients in the study group were treated with Bikini incision minimally invasive total hip arthroplasty by anterior approach,while the control group underwent total hip arthroplasty by anterolateral approach.Harris score and WOMAC score were used to evaluate the clinical function of hip joint.The operation time,length of incision,intraoperative blood loss, blood transfusion,hospitalization time,hemoglobin level at 72 h after surgery,the first ambulation time,the lower limb length discrepancy,postoperative complication were recorded and compared between two groups.Results The Harris scores at 1,2,4 months after operation in the two groups were significantly improved(P<0.05), while WOMAC scores of patients at 1,2,4 months after operation were significantly lower than those before treatment(P<0.05).However,there were no significant differences at different time between the groups(P>0.05).The hospitalization time and the first ambulation time in the study group were significantly shorter than those in the control group((6.1±2.8)d vs.(7.9±3.2)d,P=0.011;(5.3±1.5)d vs.(6.4±1.7)d,P=0.004).After 4 months of treatment,the lower limb length discrepancy in the study group was less than that of the control group((6.5 ± 2.3)mm vs.(10.4 ± 3.5)mm,P<0.01).In addition,there were no significant differences between two groups in the operation time,length of incision,complication occurrence,intraoperative blood loss and blood transfusion(P>0.05).Conclusion Treating femoral head necrosis with Bikini incision minimally invasive total hip arthroplasty can achieve favorable effectiveness,with no increase in intraoperative blood loss and rapid recovery of postoperative joint function,but further follow-up is needed.

ABSTRACT

Objective To discuss whether acupuncture at Neiguan (PC 6) inhibits myocardial hypertrophy via monoamine neurotransmitters.Method Sixty female Sprague-Dawley rats were randomized into a control group, a model group, a sham electroacupuncture (EA) group, and an EA group, 15 cases in each group. The rat models of myocardial hypertrophy were developed by subcutaneous injection of Isoprenaline hydrochloride (ISO) for 14 d. The EA group was intervened by EA at Neiguan (PC 6), and the sham EA group was by EA at a sham acupoint [5 mm away lateral to Neiguan (PC 6). The Electrocardiogram (ECG) of each group was recorded, electron microscope was used to observe the microstructure of the rats’ myocardial tissues, and high performance liquid chromatography (HPLC) was adopted to detect the contents of catecholamines in myocardial tissues.Result The heart rate, R-R interval, ST-elevation, and the contents of catecholamines in rats’ myocardial tissues (adrenaline, noradrenaline, and dopamine) in the model group and sham EA group were significantly different from that in the control group (P<0.05). The heart rate, R-R interval, ST-elevation, and the contents of catecholamines in rats’ myocardial tissues (adrenaline, noradrenaline, and dopamine) of the EA group were significantly different from that of the model group (P<0.05).Conclusion EA at Neiguan (PC 6) can improve myocardial hypertrophy, which is related to the regulation of the topical neurotransmitters; it has provided laboratory evidences for the relationship between Neiguan (PC 6) and heart, i.e. the relationship between acupoints and internal organs.
