ABSTRACT

@#Hepatitis B virus(HBV)-infected hepatitis is one of the most common infectious disease worldwide. To find novel effective anti-HBV agents, a series of Matijin-Su(MTS)derivatives with aromatic heterocycles were synthesized and evaluated for their anti- HBV activities in HepG2 2. 2. 15 cells. Among them, compounds 7a(IC50=2. 94 μmol/L)and 9a(IC50=2. 21 μmol/L)exhibited more potent inhibitory activity against the replication of HBV DNA in HepG2 2. 2. 15 cells than that of lead compound MTS(IC50=11. 16 μmol/L). Notably, both 7a and 9a displayed a high selective index(SI)of 146. 39 and > 250, respectively, which were also much higher than that of MTS(SI=10. 78). Therefore, compounds 7a and 9a may be promising anti-HBV agents with safety profile for HBV infection.

ABSTRACT

Object To study the suitable conditions for the pac king of volatile oil in Rukang Capsule, which will inhibit the loss of volatile oil. Methods With the packing rate as index, even design method wa s applied to investigate the optimal packing conditions. Results The optimal packing conditions were found to be: volatile oil volume (mL)-cy c lodextrin (g)=1∶6, the packing process should be carried out for 120 min under 40 ℃. Conclusion The repetition rate is very good in the repeti tion experiments.

ABSTRACT

Object To compare the contents of chlorogenic acid in the different processed crude drugs of Lonicera fulvotomentosa Hsu et S.C. Cheng. Methods The contents of chlorogenic acid were deter mined by HPLC. Chromatographic conditions included Hypersil C 18 column and the mobile phase consisting of a mixture of acetonitrile 0 4% phosphoric acid (13∶87). Chlorogenic acid was detected at 327 nm wavelength. Results The contents of chlorogenic acid were 0 54%, 0 81%, 0 57% in the sun cured flower bud, white bloom, gold flower in L. fulvotomentosa and 2 14%, 1 65%, 2 04% in the steaming sun cured crude drugs, respectively. Conclusion The contents of chlorogenic acid in the steaming sun cured crude drugs are higher than those in the sun cured crude drugs.

ABSTRACT

Object To determine the content of glycyrrihizic acid obtained when each individual ingredient in MAHUANG DECOCTION was decocted separately and then mixing the extracts with boiling water in comparison with that obtained by decocting the total composition together as a whole in the traditional way. Methods The contents of glycyrrhizic acid was determined by HPLC. Hypersil C 18 column was used, with acetonitrile∶0.1% acetic acid (33∶67) as the mobile phase and detected at the wavelength of 254 nm. Results The average recovery of glycyrrhizic acid when separately decocted was 102.43%, RSD=2.65%, while that of decoction in whole was 99.41%, RSD=3.11%. Conclusion The method was simple and accurate and was not interfered by other constituents in the prescription.

ABSTRACT

AIM: To determine the contents of ephedrine、laetrile、glycyrrhizic acid and liquiritin in Huagai Powder traditional decoction and granule decoction(Herba Ephedrae,Semen Armeniacae Amarum,Radix et Rhizoma Glycyrrhizae,etc.) by HPLC. METHODS: An Agilent SB-C_(18) column was used for ephedrine,the mobile phase was acetonitrile-0.1% phosphoric acid(4∶96),detection wavelength at 207 nm;An Agilent XDB-C_(18) column was used for laetrile,the mobile phase was methanol-water(23∶77),detection wavelength at 215 nm;An Agilent XDB-C_(18) column was used for glycyrrhizic acid,the mobile phase was methanol-0.2 mol/L ammonium-acid(67∶33∶1),detection wavelength at 250 nm; An Agilent XDB-C_(18) column was used for liquiritin,the mobile phase was acetonitrile-0.5% acetic acid(20∶80),detection wavelength at 276 nm. RESULTS: There were discrepancies between traditional decoction and granules in the contents of four ingredients,the average contents in granules were more than those in the traditional decoction. CONCLUSION: The peaks of the four ingredients are(segregated)(effectively),and the method is simple,conveient,exact and has good reproducibility,the other ingredients do not have interferences for the determination.

ABSTRACT

Objective: To compare the contents of ferulic acid in single medicine decoction (preparing the refined granules of single traditional Chinese herb from the different kinds of medicinal materials separately then mixing the granules with boiling water) and mixed medicine decoction (decocting the all kinds of the medicinal materials together as the traditional method) of Tangkuei Blood Supplementing Decoction (composed of Radix Angelicae Sinensis and Radix Astragali). Methods: HPLC conditions: Hypersil C 18 column methanol -0.05% acetic acid solution (45∶55) as mobile phases, detection wavelength at 324 nm. Results: The average recovery of single medicine decoction was 100.86% with RSD =2.33%( n =6), and that of mixed medicine decoction was 101.37% with RSD =1.52%( n =6).Conclusion: The content of ferulic acid mixed medicine decoction was higher than that in single medicine decoction.

ABSTRACT

Objective:To study the protective effect of polysaccharide from Rose roxburghii Tratt (RRTP-1) on neural stem cells damaged by glutamic acid (Glu). Method: RRTP-1 was obtained by ion exchange column chromatography and gel chromatography. The neural stem cells were cultured from the striata of fetal mice and damaged by glutamic acid to simulate hypoxic-ischemic brain injury. The cultures were divided into control group, glutamic acid group and test groups in which different doses of RRTP-1 were added with glutamic acid. The death rate of neural stem cells damaged by Glu and LDH leakage were observed. Results:The neural stem cells damage of test groups were alleviated,and the death rate of neural stem cells and LDH leakage of high dose test group were significantly lower than glutamic acid group. Conclusion:RRTP-1 has remarkable protective effect on neural stem cells damage.
