ABSTRACT

Venlafaxine (VEN) is a new antidepressant drug that can effectively antagonize the reuptake of serotonin (5-HT) and norepinephrine (NE), compared with other antidepressants, venlafaxine pharmacokinetics / pharmacodynamics (PK-PD) is more regular and has the characteristics of less toxic side effects, fast oral absorption, and high bio-availability. This article reviews the PK-PD model-ling of venlafaxine and its quantitative relationship, as well as the factors affecting the process in vivo of venlafaxine, including sex, body weight, individual genotype, liver and kidney function impairment, drug-drug interaction and other related factors.

ABSTRACT

OBJECTIVE To optimize the extraction technology fo r phenylethanol glycosides from Cistanche deserticola by natural deep eutectic solvents (NADESs),and to provide reference for the development and utilization of C. deserticola . METHODS The optimal NADESs was selected using total extraction efficiency of echinacoside ,acteoside and isoacteoside as indexes. Based on single factor test ,response surface methodology was used to select the optimal NADESs molar ratio ,the optimal NADESs water content ,the optimal liquid-solid ratio ;and the results were optimized by genetic algorithm . Using vitamin C (VC) as positive control ,the extraction effects of NADESs and traditional solvent (50% methanol)were compared in respects of extraction efficiency and antioxidant activities. RESULTS The optimal extraction solution was NADES- 11 composed of 1, 4-butanediol and malonic acid. The optimal extraction technology was as follows as the molar ratio of 1,4-butanediol-malonic acid was 1 ∶ 2.5,water content of NADES- 11 was 18%,liquid-solid ratio was 30 mL/g,extraction time was 30 min and extraction temperature was 30 ℃. The extraction efficiency of NADES- 11 was significantly higher than that of 50% methanol(P<0.05). IC 50 values of NADES- 11 extract(261.17 and 744.34 µg/mL)to 1,1-diphenyl-2-trinitrophenylhydrazine radical and hydroxyl radical were all lower than those of 50% methanol extract (420.97 and 1 175.12 μg/mL). ABSTRACT

Objective To explore the expressions of N-cadherin,E-cadherin,and P63 protein in hepatocellular carcinoma (HCC) tissues,and the relationship between their expressions and clinicopathologic factors.Methods Immuohistochemistry was used to detect the expressions of N-cadherin,E-cadherin,and P63 protein in 60 normal liver tissues,60 HCC tissues and their peficancerous tissues,and then the relationship between their expressions and clinicopathologic factors were analyzed.Results The positive expression rates of N-cadherin in normal liver tissue,peficancerous tissues and HCC tissue were gradually decreased,the difference was statistically significant (P < 0.05).And The positive expression rates of E-cadherin in normal liver tissue,peficanceroustissues and HCC tissue were gradually increased,the difference was statistically significant (P < 0.05).The expression of P63 in normal liver tissue was significantly higher than that in peficancerous tissues and HCC tissue,and the differences wcrc all significant(P < 0.05).The positive rates of expression of N-cadherin were related with clinical stage,portal vein invasion,metastasis and pathological grading.The positive rates of expression of E-cadherin were related with clinical stage,portal vein invasion and pathological grading.The positive rates of expression of P63 were related with clinical stage,tumor sizeand pathological grading.Spearman rank correlation analysis showed that the expression of P63was positively correlated with the expression of E-cadherin,but negatively correlated with the expression of N-cadherin.Conclusion The high expression of N-cadherin and low expression of P63 protein,E-cadherin of HCC tissues may play a role in tumor metastasis and carcinogenesis.

ABSTRACT

Objective To evaluate endoscopic sphincterotomy(EST)for the management of choledocholithiasis. Methods Between July 1987 and March 2007,991 cases of choledocholithiasis treated with EST were reviewed,and 710 cases were followed-up. Results All cases were treated with EST.The common bile duct stones were removed in 909 cases(91.7%,909/991).The rate of complications after EST was 6.0%(59/991)including pancreatitls in 29 cases(2.9%),hemorrhage in 18 cases(1.8%),cholangitis in 11 cases(1.1%),and duodenal perforation in 1 case(0.1%).There was no mortality in this group.Follow-up found reflux cholangitis in 51 cases(7.2%)and recurrent choledocholithiasis in 42 cases(5.9%). Conclusion Therapeutic endoscopy for choledocholithiasis is safe and effective.

ABSTRACT

The appliance of bilingual teaching of Medical Immunology offers an effective means of mastery and communication of the medical essential subject.This study has tried the bilingual teaching of medical immunology for clinical,dentistry and phylaxiology specialty students,discussed the model of bilingual teaching,investigated the teaching effect,analyzed and summarized the teaching experience.After the practice of bilingual teaching,we found that the feasible method of bilingual teaching to apply Chinese as the primary language and pervade English into the medical immunology course.

ABSTRACT

Objective To map the specific gene responsible for primary erythromelalgia and identify gene mutations in a Chinese family and one sporadic patient with primary erythromelalgia. Methods Geno-mic DNA was extracted from peripheral lymphocytes of the family members of the pedigree and the sporadic patient. Scanning the genes on chromosome 2q that had been identified was performed by using 6 microsatellite markers for the family members with primary erythromelalgia. Then linkage analysis and haplotype analysis were conducted. All exons of SCN9A gene were analyzed by PCR-DNA sequencing. The mutation identification was also confirmed by restriction fragment length polymorphism(RFLP). Results A maximum 2-point LOD score of 2.11 was found at a recombination fraction (? = 0.00) with markers D2S2370 and D2S2330. Recombination events were detected by markers D2S1353 and D2S2345 in this family by the haplotype analysis. There were two missense heterozygous point mutations in the 15th exon of SCN9A gene both in the family(T2573A) and the sporadic patient(T2543C), leading to the substitution of the amino acid leucine to histidine(L858H) and isoleucine to threonine(I848T), respectively. The above mutations were not found in 400 normal alleles. Conclusion It is proved that primary erythromelalgia is caused by mutations in SCN9A gene.
