ABSTRACT

The regulator does not encourage the use of fetal bovine serum (FBS) as a cell culture supplement to reduce the risk of zoonosis and heterogeneous immune responses.In addition,FBS production is subject to strict examination because of animal welfare principles.Mesenchymal stem cells (MSC) in the platelet lysate (PL) showed a faster growth rate,while maintaining good osteogenic differentiation,PL also have the key factors of MSC attachment,biological safety and immune regulation ability after MSC amplification.PL was proposed as an altemative to MSC in vitro expansion of FBS.This paper briefly introduces the latest research on the field of PL amplification in vitro.

ABSTRACT

Bone regeneration strategy to repair bone defects shows great potential.Although the skeleton has a strong self-healing and repair capabilities, for huge bone defect caused by trauma, congenital malformations,infection and surgery,the skeleton is difficult to complete the self-repairing process.In order to promote bone regeneration,bone growth must be induced by biologically active implants,cell and intracellular molecular signaling pathways.Mesenchymal stem cells(MSC)play an important role in bone repair and remodeling.Understanding the role of MSC and signaling pathways in bone repair and regeneration is critical for the study and development of bone repair materials.

ABSTRACT

Objective:To study the diagnosis value of Transcranial Doppler ultrasound for intracranial artery stenosis in patients with cerebral infarction.Methods:A total of 140 patients with acute cerebral infarction,transient ischemic attack and posterior circulation ischemic attack in Department of Neurology,Xinjiang cardiovascular and cerebrovascular disease hospital from October 2014 to October 2016 were selected as research object,CT angiography (CTA) and TCD detection were performed in all patients.Used CTA examination results as the gold standard,the detection results of intracranial artery stenosis in two groups were compared,the diagnostic value of TCD and the diagnostic results of TCD to the degree of bilateral middle cerebral artery (MCA) stenosis were analyzed.Results:CTA diagnosis showed that 140 patients had a total of 105 patients with intracranial artery stenosis,in the anterior and posterior circulation vessel of 1155 intracranial segments,CTA detection showed that 249 vessels were narrow,TCD detection showed that 236 vessels were narrow.Com-pared with CTA,TCD was better in the diagnosis of patients (Kappa value>0.75).The diagnostic sensitivity and positive predictive value of TCD for MCA were the highest,which were 91.26% and 93.07%,the consistency was the best (Kappa value =0.917).CTA detection showed that 210 MCA vessels had 103 stenoses,mild stenosis 17,moderate stenosis 41,severe stenosis 45,TCD detection showed that the stenosis was 101,mild stenosis 16,moderate stenosis 40,severe stenosis 45.The Kappa test showed that the diagnostic results of TCD to the degree ofMCA stenosis was better consistency compared with CTA (Kappa value=0.884.Conclusion:TCD has a high diagnostic value for cerebral artery stenosis in patients with cerebral infarction,and it is consistent with the diagnosis of CTA.

ABSTRACT

Objective To discuss effects of three different-intensity exercise on oxidative stress and adiponectin/adiponectin receptor1/2 of the liver in obese rats.Methods Thirty-two Sprague-Dawley rats were fed high-lipid fodder and became the obesity model after 16 weeks.Then they were randomly divided into a high-lipid control group (HF),a low-intensity exercise group (HS),a moderate-intensity exercise group (HM) and an exercise group with progressive intensity (HI),each of 8.Another 8 rats were chosen into a normal control group (NC).The speed of treadmill running of group HS and HM was 10m/min and 15m/min respectively,while that of group HI was10m/min for 20 min,followed by another 20min at and still another 20min at 28~30m/min,five days a week for 6 weeks.Six weeks later,the weight,alanine transaminase (ALT),aspartate transaminase (AST),superoxide dismutase (SOD),malondialdehyde (MDA),tumor necrosis factor alpha (TNF-α),interleukin-6 (IL-6),adiponectin receptors1/2,adiponectin,triglyceride (TG),cholesterol (TC),high-density lipoproteincholesterol (HDL-C),low-density lipoproteincholesterol (LDL-C) and free fatty acid (FFA) were tested.Results Compared with group NC,significant increase was observed in the average weight,TG,TC,LDL-C,FFA,ALT,AST,MDA,TNF-α and IL-6 (P<0.05 or P<0.01),while significant decrease was found in the average HDL-C,SOD,adiponectin and adiponectin receptors1/2 of group HF (P<0.05 or P<0.01).Compared with group HF,there was significant decrease in the average weight,TG,LDL-C,FFA,ALT,MDA,TNF-α and IL-6 (P<0.05 or P<0.01),but significant increase in the average SOD,adiponectin,AR1 and AR2 of group HS,HM and HI (P<0.05 or P<0.01),and only HDL-C of group HM increased significantly(P<0.05).Compared with group HS and HI,significant decrease was found in the average FFA and MDA (P<0.05),while significant increase was found in the average adiponectin and AR2 of group HM (P<0.05).Conclusion Aerobic exercise can improve abnormal hepatic lipid metabolism and oxidative stress,strengthen free radical scavenging capacity,prevent liver cell inflammation injury and ameliorate liver metabolism disorder of obese rats induced by high-lipid fodder.Moreover,the effect of moderate-intensity exercise is superior to low-intensity exercise or exercise with progressive intensity,whose mechanism may be related to further activating liver adiponectin receptor 2 through increasing serum adiponectin.

ABSTRACT

The purpose of this study was to compare Heat Stress Protein70 (HSP70) in human leucocytes, VO2max and Blood Lactate (BLa) during incremental exercise under a thermoneutral condition without taking a sauna with those during the same workload and under same thermoneutral condition after a sauna and after intermittent hypoxia stimuli. Ten unacclimated men performed an incremental test to exhaustion on a treadmill under a thermoneutral condition without taking a sauna (N25℃, relative humidity 65%) and during the same workload and under same thermoneutral condition after a sauna (D25℃, relative humidity 65%), and after intermittent hypoxia stimuli (LN25℃, relative humidity 65%) .
