ABSTRACT

【Objective】 To investigate the prevalence of occult hepatitis C virus infection (OCI) in Shaanxi blood donors and characterize the occult hepatitis C virus in this cohort. 【Methods】 Between July and September of 2019, 112, 117, 46, and 75 blood samples were obtained from eligible donors, alanine aminotransferase (ALT) elevated donors(ALT>50 U/L), hepatitis B virus surface antigen (HBsAg) positive donors, and anti-hepatitis C virus (anti-HCV) positive donors, respectively. Peripheral blood lymphocytes (PBMCs) were isolated from all the samples. HCV 5′UTR was amplified from total RNA of PBMCs using reverse transcription nested PCR (RT-Nested PCR) to detect the infection status of OCI in blood donors. HCV RNA core/E2 region was amplified from HCV RNA positive samples with RT-Nested PCR to determine HCV genotypes and subtypes via sequencing. 【Results】 Two (2.67%, 2/75) cases of OCI were identified in anti-HCV positive samples, 1(0.85%, 1/117) case of OCI was identified in abnormal ALT samples, and all OCI were genotyped as 1b. OCI was not found in HBsAg positive and eligible blood donors. 【Conclusions】 Our findings suggest that OCI does exist among Shaanxi blood donors and that ALT may be useful in diagnosing OCI.

ABSTRACT

Objective:Mineral trioxide aggregate (MTA),a wildly used pulp capping material, could affect the proliferation and differentiation of dental pulp cells. The aim of this study is to study the roles of Notch signalling and autophagy in MTA induced human dental pulp cells (hDPCs) proliferation promotions. Methods:Healthy human third molars were collected and hDPCs were isolated by a combined digestion of collagenase Ⅰ and dispase Ⅱ.MTA extracts of different concentrations (0.5,1.0,2.0,5.0,10.0 mg/mL) were used to test the cytotoxicity by cells counting kit (CCK-8) assays and to select the optimum concentration for hDPCs survival..Expressions of Notch1,Hes1,LC3Ⅱ / Ⅰ and p62 in wild type and MTA treated hDPCs were detected by western blotting. Results:MTA extracts in a concentration of 1.0 mg/mL exerted most profoundly promotion effects on the proliferation of hDPCs among all concentrations tested.MTA of high concentration (10 mg/mL) was toxically to cells.Compared with that of wild type hDPCs,the expressions of Notch1 and Hes1(P<0.05),or p62 and LC3Ⅱ/Ⅰ (P<0.01) in MTA treated hDPCs were significantly increased.Much lower expression of Notch1 was detected in hDPCs when autophagy was induced by Earle's balanced salt solution (EBSS) starvation for 24 h.Conclusions:MTA could up-regulated hDPCs proliferation with highly relevant in stimulating Notch1-Hes1 signalling and inhibition of autophagy. The study is supposed to provide new insight in unrevealing the mechanisms of MTA mediated dental pulp cells proliferation.

ABSTRACT

Using the theory and method of bibliometrics, this paper made statistical analysis on the research papers of 16 main woody medical plants published in the period of 1998-2007, from aspects of years distribution, journals distribution, themes distribution, authors distribution and research institutes distribution, and so on. The results showed the studies, status quto and development potential of woody medical plants.

ABSTRACT

Objective Studies on the changes of gastrin and SS immunoreactive cells in pancreatic islets during experimental gastric ulcer. Methods The immunohistochemical ABC technique was used. Results The gastrin immunoreactive cells were located in most of the pancreatic islet. The mumber of G cells in experimental gastric ulcer group were higher than that of control group on the 4th and 10th day after operation. The D cells raised on the 10th day. Conclusion The present work provides the evidence that the G and D cells of pancreatic islets might be involed in the self-healing process of the experimental gastric ulcer by endocrine or paracrine regulation.
