ABSTRACT

BACKGROUND@#Hair follicles are easily accessible and contain stem cells with different developmental origins, including mesenchymal stem cells (MSCs), that consequently reveal the potential of human hair follicle (hHF)-derived MSCs in repair and regeneration. However, the role of hHF-MSCs in Achilles tendinopathy (AT) remains unclear. The present study investigated the effects of hHF-MSCs on Achilles tendon repair in rabbits.@*METHODS@#First, we extracted and characterized hHF-MSCs. Then, a rabbit tendinopathy model was constructed to analyze the ability of hHF-MSCs to promote repair in vivo . Anatomical observation and pathological and biomechanical analyses were performed to determine the effect of hHF-MSCs on AT, and quantitative real-time polymerase chain reaction, enzyme-linked immunosorbent assay, and immunohistochemical staining were performed to explore the molecular mechanisms through which hHF-MSCs affects AT. Furthermore, statistical analyses were performed using independent sample t test, one-way analysis of variance (ANOVA), and one-way repeated measures multivariate ANOVA as appropriate.@*RESULTS@#Flow cytometry, a trilineage-induced differentiation test, confirmed that hHF-derived stem cells were derived from MSCs. The effect of hHF-MSCs on AT revealed that the Achilles tendon was anatomically healthy, as well as the maximum load carried by the Achilles tendon and hydroxyproline proteomic levels were increased. Moreover, collagen I and III were upregulated in rabbit AT treated with hHF-MSCs (compared with AT group; P  < 0.05). Analysis of the molecular mechanisms revealed that hHF-MSCs promoted collagen fiber regeneration, possibly through Tenascin-C (TNC) upregulation and matrix metalloproteinase (MMP)-9 downregulation.@*CONCLUSIONS@#hHF-MSCs can be a treatment modality to promote AT repair in rabbits by upregulating collagen I and III. Further analysis revealed that treatment of AT using hHF-MSCs promoted the regeneration of collagen fiber, possibly because of upregulation of TNC and downregulation of MMP-9, thus suggesting that hHF-MSCs are more promising for AT.

ABSTRACT

Objective To evaluate the quality coherence of Trillium tschonoskii Maxim.from different producing areas by HPLC fingerprint and PCA; To provide a method for quality control. Methods Samples were separated by Hibar C18 (4.6 mm × 250 mm, 5 μm) with acetonitrile-water as gradient mobile phase at the flow rate of 1.0 mL/min. The wavelength was 203 nm and the temperature was 30 ℃. Chromatographic Fingerprint Similarity Evaluation System and PCA were used to analyze the data. Results The results of method validation of HPLC fingerprint met technical standards.15 common peaks was verified and the similarities of 14 batches of Trillium tschonoskii Maxim. from different producing areas were among 0.389–0.979. 3 principal components with the characteristic root cumulative contribution rate reaching 87.674% were screened out by PCA results. The composite score of S2 was the highest (4.926), and the quality was the best. Conclusion The application of HPLC combined with PCA can objectively and effectively evaluate the quality difference of Trillium tschonoskii Maxim.from different producing areas.

ABSTRACT

<p><b>OBJECTIVE</b>To study the effects of RhoA down-regulation by RNA interference on the invasion of tongue carcinoma Tca8113 and SCC-4.</p><p><b>METHODS</b>Determination of the human RhoA sequence as well as the design and constructionof a short specific small interfering RNAs (siRNA) were performed. The siRNA of RhoA gene was transfected into humantongue squamous cell carcinoma Tca8113 and SCC-4 cells line by Lipofectamine 2000. Quantitative real-time polymerasechain reaction was used to examine the mRNA expressionlevels of RhoA. Protein expressions of mRNA, galectin-3,and matrix metalloproteinase (MMP)-9 were evaluated byWestern blot. Transwell invasion assay was performed toassess the invasion ability of tongue carcinoma.</p><p><b>RESULTS</b>RhoA expressions in Tca8113 and SCC-4 cells were reducedsignificantly after transfection of RhoA-siRNA. Protein levels f galectin-3 and MVP-9 were also down-regulated significantly. Invasion ability was inhibited as well.</p><p><b>CONCLUSION</b>RhoA-siRNA can effectively inhibit RhoA expression in Tca8113 and SCC-4 cells. The invasion ability of tongue carcinoma cells decreased with down-regulation of the protein expressions of galectin-3 and MMP-9, indicating that RhoA-siRNA can inhibit invasion of tongue carcinoma. Results show that RhoA may play an important role in the processes of invasion and metastasis of tongue carcinoma.</p>

ABSTRACT

Objective To investigate the infection status of hepatitis B virus(HBV) and hepatitis C virus(HCV) in healthy adult in Xi′an ,in order to provide references for hepatitis prevention and health education .Methods A total of 1 052 healthy adults from 10 communities in Xi′an were collected by using multistage stratified random sampling method .HBV serum markers and anti‐HCV antibody were detected ,and characteristics of distribution of HBV and HCV infection were analysed .Results A total of 37 adults were observed with positive hepatitis B surface antigen(HBsAg) ,the total positive rate was 3 .52% ,and the positive rate of male and female was 4 .08% and 3 .07% respectively ,no statistically significant difference was found between male and female(P>0 .05) .Apart from male adults aged 30 - 0 .05) .A total of 31 adults were observed with HBsAg ,hepatitis B e antibody (HBeAg)and hepatitis B core antibody(HBcAb) positive ,the positive rate was 2 .95% .The positive rate of hepatitis B surface antibody(HBsAb) was 54 .18% .Serological patterns between different gender had no statistically signifi‐cant differences(P>0 .05) .A total of 9 adults(0 .86% ) were observed with positive anti‐HCV antibody ,and the positive rate of male and female was 0 .64% and 1 .02% respectively ,no statistically significant difference was found between male and female(P>0 .05) .Conclusion Healthy adults in Xi′an have relatively low infection rates of HBV and HCV ,while for the control of hepatitis B and hepatitis C ,further strengthening the health education ,improving awareness of prevention and routine monitoring infectious dis‐eases and vaccination may still be necessary .

ABSTRACT

Objective To investigate the serum levels of tumor markers CEA,CA724,PGⅠ,PGⅡ,PGI/PGII and SF in gas-tric cancer.Methods The serum levels of CEA,CA724,PGⅠ,PGⅡ,PGI/PGII and SF were detected in 46 healthy con-trols,45 atrophic gastritis patients and 39 gastric cancer patients.Serum levels of CEA,CA724 were measured by ELC meth-od,PGⅠ,PGⅡ by time resolved fluorescence immunoassay (TRFIA)and SF by immunoturbidimetry(ITM).Results ①Compared to the healthy controls,the statistical contrast of serum levels of CEA,CA724,PGⅠ,PGⅡ and PGI/PGII except SF was significant in gastric cancer patients(P<0.05),only PGⅠ,PGⅡ,SF had significant difference in atrophic gastritis patients (P<0.05).Compared to atrophic gastritis patients,the serum levels of CEA,CA724 were significantly higher,but PGⅠwas lower in gastric cancer patients(P<0.05).②When CEA,CA724,PGⅠ,PGⅡ,PGI/PGII and SF were used to di-agnose gastric cancer individually,the sequence of the area under ROC curve was CEA,SF,CA724,PGI/PGII,PGⅡ and PGⅠ.Only the areas of PGⅠ,PGⅡunder ROC curve had significant statistical difference (P<0.05).③The sensitivity,speci-ficity,PPV and NPV were different when these indexes were used to diagnize gastric cancer individually or incorporatedly. Conclusion The serum levels of tumor markers CEA,CA724,PGⅠ,PGⅡ,PGI/PGII and SF had important reference value for the diagnosis of gastric cancer although the diagnostic value was different individually or incorporatedly;the content of serum PG and the ratio of PGI/PGII were closely related to the gastric mucosa.

ABSTRACT

A case of methamphetamine-related cerebral infarction was reported and the relevant databases of CBMdisc and Medline were searched.Among 14 cases of ischemic stroke , the ratio of male:female was 9∶5 and the age range 19-45 years.There were anterior circulation lesions ( n=13,92.8%) and 10 of them (71.3%) had cerebral vascular occlusion or stenosis.And the predominant manifestation was hemiplegia (85.7%);among 34 cases of hemorrhagic stroke ,the ratio of male: female was 12∶5 and the age range 16-60 years.And 28 cases (82%) were≤45 years.Cerebral hemorrhage was the most common (85%).The major manifestations were headache ( n =26, 76%) and disturbance of consciousness (n=21,62%).The methamphetamine-related stroke occurs frequently in young males.And ischemic stroke tends to involve anterior circulation and cause hemiplegia.Yet hemorrhagic stroke has extensive attack sites and headache and disturbance of consciousness are quite common.

ABSTRACT

Patients with late-stage cancer commonly have distant lymph node metastasis; however, poor health often contraindicates surgical treatment. Although the quality of life and overall survival for these patients are low, there is neither a consensus nor a guide for treatment. Ablation technique and surrounding tissue damage are two possible reasons for limited study of radiofrequency ablation in patients with superficial distant lymph node metastasis. Here, we report two patients treated successfully with ultrasound-guided radiofrequency ablation for superficial distant lymph node metastasis. In these patients, deionized water was injected to the surrounding tissues of the lymph node to decrease heat injury. Results from these patients suggest that radiofrequency ablation may play an important role in the treatment of patients with distant lymph node metastasis.

ABSTRACT

Stem cell marker LIN28, related closely with SOX2 and OCT4, has been studied as a biomarker for the maintainance of pluripotent cells in several malignancies. Our previous study showed that SOX2 and OCT4 were negative predictors for hepatocellular carcinoma (HCC). However, the predictive value of LIN28 in HCC outcome is still undetermined. We hypothesized that LIN28 may also play a role as a biomarker for HCC. To test this hypothesis, we examined the expression of LIN28 in 129 radically resected HCC tissues using reverse transcription-polymerase chain reaction and analyzed the association of LIN28 expression with clinicopathologic features and prognosis. Our study showed that LIN28 was expressed at a higher frequency in tumor tissues than in non-HCC tissues (45.0% vs. 21.7%, P = 0.020). Moreover, LIN28 expression was significantly increased in cases with large tumor size (P = 0.010). Univariate analysis did not reveal a significant correlation between LIN28 expression and overall survival or recurrence-free survival. For HCC patients who met the Milan criteria, stratified analysis revealed shorter overall survival (P = 0.007) and recurrence-free survival (P < 0.001) in those with detectable LIN28 expression compared to those with no detectable LIN28 expression. Furthermore, multivariate analysis revealed that LIN28 was a negative independent predictor for both overall survival (hazard ratio= 7.093, P = 0.017) and recurrence-free survival (hazard ratio=5.518, P = 0.004) in patients who met the Milan criteria. Taken together, our results suggest that LIN28 identifies low-risk and high-risk subsets of HCC patients meeting the Milan criteria who undergo hepatectomy.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the relation ship of estrogen receptor 2 gene (ESR2) polymorphism associated with intrahepatic cholestasis of pregnancy (ICP) in Chengdu of China.</p><p><b>METHODS</b>By polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method, the Rsa I polymorphism in exon 5 and the Alu I polymorphism in exon 8 of ESR2 were detected in 100 pregnant women with ICP (ICP group) and 100 normal pregnant women (control group) in Chengdu.</p><p><b>RESULTS</b>(1) The frequency of the allele A of Alu I polymorphism in exon 8 was significantly higher in ICP group than in control group (P=0.031, OR=1.975), so did the frequency of the Aa+AA genotypes (P=0.028, OR=2.144). (2) The genotype distributions (rr, Rr and RR) and allele frequencies (r and R) of Rsa I polymorphism in exon 5 were not significantly different between the two groups (P>0.05).</p><p><b>CONCLUSION</b>The Alu I polymorphism in exon 8 of ESR2 may be associated with the susceptibility of ICP in Chengdu. The Aa+AA genotype significantly elevated the risk suffering from the ICP. The Rsa I polymorphism in exon 5 of ESR2 is not associated with the risk getting the ICP in Chengdu.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the relationship between human leukocyte antigen-DQA1 (HLA-DQA1) allele gene polymorphism and intrahepatic cholestasis of pregnancy (ICP).</p><p><b>METHODS</b>Forty-five patients with ICP, eighteen ICP families, forty-five normal pregnant women and eighteen normal control families were tested for HLA-DQA1 allele gene polymorphism by polymerase chain reaction with sequence-specific primer (PCR-SSP) method.</p><p><b>RESULTS</b>The frequency of HLA-DQA1*0301 in normal pregnant women was markedly higher than that in the ICP group (P>0.05). No significant differences were observed between the frequencies of other detected HLA-DQA1 alleles in both groups. The analysis of feto-maternal or couples sharing of the HLA-DQA1 alleles showed that no significant differences were observed between the two groups.</p><p><b>CONCLUSION</b>The above findings suggest that there is no significant association between the genetic polymorphisms in HLA-DQA1 and ICP in Chengdu district; HLA-DQA1*0301 may be a protective gene against ICP. It may prevent the development of ICP.</p>

ABSTRACT

OBJECTIVE@#To evaluate the application of sequential noninvasive following invasive mechanical ventilation in chronic obstructive pulmonary disease (COPD) patients with severe respiratory failure by investigating the appearance of pulmonary-infection-control-window.@*METHODS@#From November 2001 to October 2004, 76 case of COPD patients with severe respiratory failure due to pulmonary infection were intubated and recruited in the study. When the pulmonary infection was significantly controlled (the time of pulmonary infection control was called PIC window) by the antibiotic and comprehensive therapy, all cases were randomized into noninvasive veatiation group (NIV) and control group. The early extubation was conducted and followed by noninvasive mechanical ventilation via facial mask with bilevel positive airway pressure mode immediately in the NIV group. Conventional invasive synchronized intermittent mandatory ventilation (SIMV) plus pressure support ventilation (PSV) was used as the weaning technique in the control group.@*RESULTS@#Thirty eight cases among 76 patients were in the NIV group, and the rest in the control group. The NIV group and the control group had similar age, sex, APACHE scores, RR, HR, MAP, PaO2 and PaCO2 at the time of commencement and PIC window (P > 0.05). The time of PIC window was (7.5 +/- 1.9) d in the NIV group, and (8.0 +/- 2.5) d in the control group (P > 0.05). In the NIV group, the durations of invasive mechanical ventilation (MV) and total MV were (7.5 +/- 1.9) d and (12.5 +/- 4.0) d respectively, while the durations were (23.5 +/- 9.5) d in the control group (P < 0.05). The durations of RICU stay and hospital stay were shorter than that in the control group. The incidence of ventilation associated pneumonia (VAP) was 18.4% (7/38) in the NIV group, 39.5% (15/38) in the control group respectively (P < 0.05). The incidence of reintubation was 13.2% (5/38) in the NIV group, 34.2% (13/38) in the control group respectively (P < 0.05). Hospital mortality was 7.9% (3/38) in the NIV group, and 28.9% (11/38) in the control group (P < 0.05).@*CONCLUSION@#In those COPD patients requiring intubation and mechanical ventilantion who have severe respiratory failure due to pulmonary infection, sequential noninvasive following invasive mechanical ventilation at the appearance of PIC window can significantly reduce the MV duration, the length of RICU stay and hospital stay, and decrease the occurrence of VAP, reintubation and hospital mortality as well. So it is an efficient strategy to be generalized.

0.05).Conclusions The reduced expression of HLA-G on placenta in ICP patients may alter the maternal-fetal immune response and thus be involved in the pathogenesis of this disorder. Dexamethasone can upregulate the expression of HLA-G on placenta.The 14 bp deletion polymorphism in exon 8 of HLA-G gene might not have a significant influence on the development of ICP.
