ABSTRACT

Aim: Reactive oxygen species and high concentrations of proinflammatory cytokines are related to diseases that are often triggered during the aging process. This study aimed to investigate the effect of moderate-intensity physical exercise, twice a week, on oxidative stress and inflammation biomarkers. Methods: Participants were older women aged 60-70 years, engaged in a moderate-intensity exercise program carried out in 60 minutes sessions, twice a week, for 24 weeks. Exercise sessions consisted of step, resistance and stretch exercises. Thiobarbituric acid reactive substances (TBARS), serum iron and cytokines IL-6, IL-1β and IL-1ra were determined using specific kits. Physical fitness was assessed using tests provided by AAPHERD. A paired t test was performed. Results: Moderate-intensity exercise program provided a reduction in lipid peroxidation (27%) and in the serum release of prooxidant iron ions (40%). Cytokine levels were reduced by 37% for IL-6, 16% for IL-1β and 32% for IL-1ra. Improvements in cardiorespiratory fitness (13%), muscular endurance (11%) and flexibility (12%) were also observed. Conclusion: Diminished redox state and inflammation were obtained using a twice-weekly exercise program. These results have important implications for older adults who are unable or unwilling to attend exercise programs more than twice a week. Moreover, these results could re-establish the minimum exercise activity necessary for obtaining health benefits in the elderly population.(AU)

ABSTRACT

Two oil blends (sunflower/canola oils 85/15 (BL1) and canola/linseed oils 70/30 (BL2)), were prepared and enzymatically interesterified to be applied to surgically-induced wounds in rats. Following surgery, the animals were submitted to the Treatment with Physiological Saline (TPS) (control group), Blends (TBL), and Structured Lipids (TSL). The control group (TPS) received physiological saline solution for 15 days. In TBL, BL1 was administered during the inflammation phase (days 0-3) and BL2 in the tissue formation and remodeling phase (days 4-15). In TSL, Structured Lipid 1 (SL1) and Structured Lipid 2 (SL2) were used instead of BL1 and BL2, respectively. The aim of this study was to compare wound closure evolution among rats treated with the blends or structured lipids versus control rats treated with physiological saline. The wound healing process was evaluated by measuring the wound areas along the treatments and the concentrations of cytokines. An increase in the areas of wounds treated with the blends and structured lipids in the inflammatory phase was observed, followed by a steeper closure curve compared to wounds treated with physiological saline. Duas misturas de óleos vegetais (girassol/canola 85/15 (BL1) e canola/linhaça, 70/30 (BL2) foram preparadas e interesterificadas por via enzimática para serem aplicadas em feridas induzidas cirurgicamente em ratos. Após a cirurgia, os animais foram submetidos ao tratamento com soro fisiológico (TPS) (grupo controle), tratamento com as misturas (TBL) e tratamento com os lipídios estruturados (TSL). O grupo controle (TPS) recebeu soro fisiológico por 15 dias. Em TBL, BL1 foi administrada durante a fase de inflamação (dias 0-3) e BL2 na fase de formação de tecido e remodelação (dias 4-15). Em TSL, os lipídios estruturados SL1 e SL2 foram usados em vez de BL1 e BL2, respectivamente. O objetivo deste estudo foi avaliar a evolução do fechamento das feridas dos grupos de ratos tratados com as misturas ou lipídios estruturados em comparação com os ratos do grupo controle, tratados com soro fisiológico. O processo de cicatrização das feridas foi avaliado através da medição das áreas das feridas ao longo dos tratamentos e pela determinação das concentrações de citocinas. Observou-se aumento das áreas das feridas tratadas com as misturas e os lipídios estruturados na fase inflamatória, seguida por um fechamento acentuado de feridas comparado com o tratamento com solução salina. ABSTRACT

El Síndrome Metabólico corresponde a una serie de trastornos relacionados con obesidad e inactividad física. Poco se conoce respecto de la falta de ejercicio, en estadios tempranos del desarrollo, en la susceptibilidad a un fenotipo insulinoresistente inducido por una dieta alta en grasas. Akt juega un rol clave en la síntesis de proteínas y el transporte de glucosa en el músculo esquelético y ha mostrado ser regulada por la actividad muscular. El objetivo del presente estudio fue determinar el efecto de la inactividad física temprana sobre el crecimiento muscular y la susceptibilidad de adquirir un fenotipo diabético y evaluar su relación con la expresión de Akt. Cuarenta ratas Wistar fueron distribuidas en 2 grupos (Grupos Control, Std) y Restricción de movimiento (RM). Entre los días postnatal 23 y 70 los animales del grupo RM fueron alojados en pequeñas jaulas que no permitían una actividad motora relevante. A partir del día postnatal 71 y hasta el día 102, 10 ratas de cada grupo fueron alimentadas con una Dieta Alta en Grasas (RM-DAG y Std-DAG). No se observaron diferencias en el peso corporal total pero DAG generó un significativo incremento en la grasa epididimal. RM generó una disminución significativa en el peso de los músculos sóleo. La captación de glucosa estimulada por insulina fue menor en el grupo RM-DAG. Los niveles de proteína Akt fueron menores en los grupos RM. El análisis de PCR a tiempo real mostró que la restricción de movimiento disminuyó los niveles de ARNm de AKT1 en el músculo sóleo, independiente de la dieta administrada. Estos hallazgos sugieren que la inactividad física temprana limita el crecimiento muscular y contribuye en la instauración un fenotipo insulino resistente, lo cual puede ser en parte explicado por una desregulación en la expresión de Akt.


Metabolic Syndrome is a group of conditions related to obesity and physical inactivity. Little is known about the role of physical inactivity, in early stages of development, in the susceptibility to insulin resistant phenotype induced by high fat diet. Akt plays a key role in protein synthesis and glucose transport in skeletal muscle and has been regulated by muscle activity. The objective of present study was to determine the effect of early physical inactivity on muscle growth and susceptibility to acquire a diabetic phenotype and to assess its relationship with Akt expression. Forty Wistar male rats were distributed in two groups (standard group, Std) and movement restriction (RM). Between days 23 and 70 after birth, RM group was kept in small cages that did not allow them to perform relevant motor activity. From day 71 to 102 after birth, 10 rats of each group were fed with hyperlipidic diet (groups Std-DAG and RM-DAG). No differences were observed in total body weight although DAG increased epididymal fat pad weight. RM decreased significantly the soleus weight. Insulin-mediated glucose uptake was lower in RM-DAG group. Akt protein levels were lower in RM groups. Real time RT-PCR analysis showed that movement restriction decreased mRNA levels of AKT1 in soleus muscle, regardless of supplied diet. These findings suggest that early physical inactivity limits muscle's growth and contributes to instauration of insulin resistant phenotype, which can be partly explained by dysregulation of Akt expression.

ABSTRACT

A proteína de fase aguda, amilóide sérica A (SAA), exerce função importante na resposta inflamatória, estimulando a expressão e a liberação de TNF-`alfa´-IL-8 e IL-8 e IL-1ß em neutrófilos e células mononucleares (1, 2, 3, 4). Visando obter mais informação sobre o processo, estudamos as vias de sinalização envolvidas na liberação dessas citocinas, após as células serem estimuladas com SAA. Para tal, avaliamos o efeito de alguns inibidores da cascata de sinalização e evidenciamos indiretamente a participação das proteínas quinases ativadas por mitógenos (MAPK) e da fosfatidil inositol 3 quinase (PI3K) (2, 3). Com a utilização de inibidores específicos, observamos que há inibição da liberação de IL-8 quando utilizamos um antagonista da proteína Gi e observamos também a participação do fator de transcrição, NF-`capa´B na liberação de IL-8 e TNF-`alfa´ promovida por SAA...
