ABSTRACT

Objective@#Cytoreductive surgery followed by adjuvant chemotherapy is a standard frontline treatment for epithelial ovarian cancer (EOC). We aimed to develop an ovarian cancer risk score (OVRS) based on the expression of 10 ovarian-cancer-related genes to predict the chemoresistance, and outcomes of EOC patients. @*Methods@#We designed a case-control study with total 149 EOC women including 75 chemosensitives and 74 chemoresistants. Gene expression was measured using the quantitative real-time polymerase chain reaction. We tested for correlation between the OVRS and chemosensitivity or chemoresistance, disease-free survival (DFS), and overall survival (OS), and validated the OVRS by analyzing patients from the TCGA database. @*Results@#The chemosensitive group had lower OVRS than the chemoresistant group (5 vs.15, p≤0.001, Mann-Whitney U test). Patients with disease relapse (13 vs. 5, p60 months) of patients with OVRS ≥10 were significantly shorter than those of patients with OVRS <9). The high OVRS group also had significantly shorter median OS than the low OVRS group in 255 patients in the TCGA database (39 vs. 49 months, p=0.046). @*Conclusions@#Specific genes panel can be clinically applied in predicting the chemoresistance and outcome, and decision-making of epithelial ovarian cancer.

ABSTRACT

OBJECTIVE: To provide scientific evidence for establishing perfect rationality evaluation indicator system of antibiotics. METHODS: On the basis of literature research, Delphi method was used to conduct several rounds of consultations on 30 experts in related fields with E-mail and questionnaire field investigation. After several rounds of consultations, rationality evaluation index system for antibiotics was determined. The effective recovery rate of expert consultation questionnaire was used to express the positive coefficient of experts; the authoritative coefficient was used to express the authoritative degree of experts; the index importance assignment and the full score ratio were used to reflect the concentration degree of experts’ opinions; the coefficient of variation and the coefficient of coordination were used to express the coordination degree of experts’ opinions. RESULTS & CONCLUSIONS: After two rounds of consultations, response rates of the questionnaire in two rounds of surveys were 100% and 96.67%, indicating experts were highly motivated. The authoritative coefficients were 0.91 and 0.88, indicating experts had a high degree of authority. Finally, an evaluation index system for rationality of antibiotics was established, which included four first-level indicators (indications, drug selection, medication process, management indicators) and 35 second-level indicators. Among them, the mean value of importance scoring of first-level indicators ranged from 4.28 to 5.00, the full score ratio from 0.93 to 1.00, the coefficient of variation from 0.00 to 0.15 and the coefficient of coordination was 0.446 (P<0.001). While, the mean value of importance scoring of second-level indicators ranged from 3.83 to 4.79; the full score ratio from 0.67 to 1.00; variation coefficient from 0.10 to 0.26 and the coefficient of coordination was 0.115 (P<0.001), which indicating the system was reliable.

ABSTRACT

Objective To compare the efficacy of bismuth-containing quadruple therapy on H.pylori eradication for 10 days versus 14 days in a randomized controlled trial.Methods A total of 240 patients diagnosed with H.pylori infection were randomly assigned to two groups:treated with esomeprazole,bismuth,clarithromycin and amoxicillin for 10 days(n =117) and for 14 days (n =123).Eradication was defined by rapid urease test or 13C-urea breath test at 4 weeks after drug withdrawal.Results A total of 219 patients were complete the trial.Intention-to-treat(ITT) analysis showed that the eradication rate of 10 and 14 days were 77.8% (91/117)and 76.4% (94/123)respectively.Per protocol(PP) analysis indicated that the eradication rate of 10 and 14 days were 85.8% (91/106) and 83.2% (94/113) respectively.There was no statistically significant differences between the two groups(P > 0.05).The incidence of adverse events were 7.7% (9/117)and 9.8% (12/123),respectively.There was no significant difference (P > 0.05).Conclusion The eradication rate of bismuth-containing quadruple therapy for 10 days and 14 days were essentially identical,bismuth-containing quadruple therapy for 10 days has a higher potency ratio.

ABSTRACT

Objective To study the impact of different low concentrations of cerium oxide for hepatocellular carcinoma cell prolifera-tion.Methods Three different types of hepatoma cells (Huh7, HepG2,7721) were cultured,and added different concentrations of cerium oxide (0.005,0.01,0.05,0.1,1 μg/mL),of which the cell proliferation was detected by CCK8.The apoptosis-related genes was detected by qRT-PCR technology.The cell cycle was analyzed by flow cytometry.And the effect of low concentration cerium oxide on hepatocellular carci-noma cells tumorigenicity was confirmed by the nude mice experiments.Results CCK8 experiment showed that low concentrations of cerium oxide could promote proliferation of hepatocellular carcinoma cell, especially in concentration of 0.01μg/mL.The qRT-PCR showed that low concentration of cerium oxide could inhibit the apoptosis of hepatocellular carcinoma cell.The flow cytometry analysis had not found any effect of cerium oxide on cell cycle.The tumorigenicity experiments confirmed that low concentrations of cerium oxide could enhance the tumorigenic ability of hepatocellular carcinoma cell.Conclusion Low concentration of cerium oxide can significantly improve the proliferation of liver cancer cells.

ABSTRACT

Objective ToevaluatetherelationshipbetweentypeIIdiabetes,hyperlipidlevelandcarotidartery arthrosclerosisbyultrasonography.Methods Atotalof150retiredofficerswereselected,ofwhich63diabeticpatients were divided into case group while 87 subjects free of diabetes were divided into control group.Blood laboratory examination wasconductedandthemanifestationofcarotidarterywasexaminedbyultrasonography.Results TCH,TG,LDLand ApoB of case group were significantly higher than those in control group (P<0.05 ).The intimamedia thickness (IMT)of carotid artery in case group was increased significantly (P<0.05 ).Moreover,arteries narrowing and plaque were more than those in control group (both P <0.05 ).T peak velocity in carotid artery and internal carotid were of statistical significance between two groups (P<0.01 ).Furthermore,multiple linear regression analysis considering the degree of carotid artery arthrosclerosis and serum lipid showed that multiple correlation coefficient of case group was 0.870 (P<0.01)and0.902(P<0.01)ofcontrolgroup.Conclusion Hyperlipidemiamayhavethepromoteeffectontheformation of arteriosclerosis plaque and ultrasonography could be one of the effective methods to detect early vessel lesion of diabetic patients.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the anti-tumor immunity in vitro induced by prostate cancer cell vaccine transfected with recombinant adenovirus encoding 4-1BBL in mice.</p><p><b>METHODS</b>The replication-deficient adenovirus AdEasy-1 system was used to construct recombinant adenovirus Ad-m4-1BBL and Ad-eGFP. The prostate cancer cell RM-1 of mice was transfected with Ad-m4-1BBL and Ad-eGFP, and treated with mitomycin (MMC) to produce TCV, TCV-Ad-eGFP and TCV-Ad-m4-1BBL, followed by co-culture with syngeneic murine spleen cells. Then the cytotoxic activity of the lymphocytes against RM-1 cells was analyzed with CCK-8 solution, and IL-2 and INF-gamma were detected by ELISA.</p><p><b>RESULTS</b>The 4-1BBL protein was highly expressed in the TCV-Ad-m4-1BBL of the 4-1BBL-transfected mice. TCV-Ad-m4-1BBL significantly increased the expressions of IL-2 ([180.24 +/- 2.22] pg/ml) and INF-gamma ([1512.46 +/- 23.64] pg/ml) as compared with TCV and TCV-Ad-eGFP (P < 0.05), and induced higher RM-1 cell specific cytotoxicity ([34.24 +/- 2.64]%) than the latter two ([9.82 +/- 1.48]%) and ([14.65 +/- 3. 21]%), (P < 0.05). But none of them exhibited significant cytotoxicity against hepatocellular carcinoma Hepal-6.</p><p><b>CONCLUSION</b>The m4-1BBL-expressing prostate cancer cell vaccine can effectively induce anti-tumor immune responses.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To discuss the role of calcium-overloading in initiation and maintenance of atrial fibrillation (AF).</p><p><b>METHODS</b>The right atrial appendages were obtained from 14 patients with AF and 12 patients with sinus rhythm. The mRNA expression of proteins influencing the calcium homeostasis was measured by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and normalized to the mRNA level of glyceraldehyde-3- phosphate dehydrogenase. The left atrial diameter (LAD), mitral valvular area (MVOA), and systolic pulmonary arterial pressure were obtained by echocardiography before surgery.</p><p><b>RESULTS</b>Compared to sinus rhythm group, the mRNA levels of L-type calcium channel alc, sarcoplasmic reticulum (SR), calcium adenosine triphosphatase (Ca2+ -ATPase), and ryanodine receptor type-2 (R(Y) R2) were significantly decreased (P < 0.01); the mRNA level of inositol triphosphate receptor type-1 (IP3R1) was significantly increased (P < 0.05). No changes in the mRNA expression of phospholamban and calsequestrin were observed between two groups (P > 0.05). Correlations were found between MVOA and mRNA levels of LVDC-Calc, SR Ca2+ -ATPase (r = 0.719, P = 0.004; r = 0.625, P = 0.017). The mRNA level of SR Ca2+ -ATPase was negatively correlated with LAD (r = -0.573, P = 0.032).</p><p><b>CONCLUSIONS</b>Calcium loading may be responsible for the occurrence and maintenance of AF, and abnormal regulation in the mRNA expression may be the molecular mechanism of intracellular Ca2+ overload. The progressive nature of AF involves structural change.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the association of polymorphisms in the apolipoprotein B gene (APOB) 3'variable number of tandem repeat with natural longevity in the Xinjiang Uighur nationality people.</p><p><b>METHODS</b>Totally 191 healthy individuals over 90 years and 53 individuals aged 65-70 years were recruited among Xinjiang Uighur population, the nationality, gender and living area were matched. Genotyping was performed using polymerase chain reaction-sequence specific primer(PCR-SSP) and PCR-sequencing.</p><p><b>RESULTS</b>Fourteen alleles were found in the Xinjiang Uighur nationality population. The frequency of HVE36 and HVE42 in the natural longevity group were significantly higher than that in the control group (both P<0.05) and HVE44, HVE46, HVE48 and HVE58 were only found in the natural longevity group. However, the frequency of HVE26, HVE30 and HVE34 were markedly lower in the natural longevity group compared to the control group. Logistic regression analyses revealed that allele L and the genotypes LL were positively associated with age, whereas the allele S and genotype SS were negatively associated with age (both P<0.05). Each allele consists of 15 bp tandem repeats with rich-AT by PCR-sequencing.</p><p><b>CONCLUSION</b>These results indicate that the S allele, and SS genotype are frail factors in China Uighur natural longevity people, whereas allele L and genotypes LL are protective factors.</p>

ABSTRACT

Objective To observe the induction of tolerogenic dendritic cells (DCs) by curcumin (Cur) and its mechanism.Methods After immature DCs from bone marrow cells of Wistar rats were treated with different concentrations of Cur (0,10,20 and 30?mol/L) respectively,and then the DCs were tested by flow cytometry for the surface molecules expression.After the immature DCs were treated by 30?mol/L Cur with or without stimulation of LPS,endocytosis of DCs to dextran was tested by flow cytometry.The production of IL-12 in DC culture supernatant was determined by ELISA.The levels of NF-?B p65 and RelB translocation to the nucleus were investigated by Western- blot.The activity of NF-?B was detected by NF-?B-binding ELISA and luciferase reporter gene analy- sis.The ability of DCs to stimulate the proliferation of T cells from Lewis rats were analyzed by mixed leukocyte reactions (MLR).Results Cur suppressed LPS-indueed cell-surface expression of costimu- latory molecules (CD80,CD86 and CD40) in a dose-dependent manner.When Cur was used at a con- centration of 30?mol/L,there was no marked difference in the surface molecules expression of LPS- inducing DCs as compared with immature DCs.After DCs were induced by LPS (LPS group),the positive rate of FITC-Dextran uptake was (36.6?7.2)%,and the secretory amounts of IL-12 were (415.9?42.7) pg/ml.In DCs of LPS group,the intranuclear RelB and p65 were highly expressed and their DNA binding activity was 0.65?0.08 and 0.74?0.07 respectively.The luciferase activity of reporter gene in LPS group DCs was remarkably increased to 435% as compared with that in the controls.DCs in LPS group showed strong capacity to stimulate T cells proliferation.When DCs were treated with 30?mol/L Cur followed by induction with LPS (Cur+LPS group),the positive rate of Dextran uptake was (78.6?14.2)% and remarkably higher than in LPS group (P

ABSTRACT

<p><b>OBJECTIVE</b>To explore the distribution of polymorphisms of apolipoprotein E(apoE) gene and its relation to natural longevity in the Xinjiang Uighur people.</p><p><b>METHODS</b>Using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP), the authors obtained data from centenarians(n=42), 90-year-old people (n=102), 65-70-year-old people (n=70) and controls(n=53).</p><p><b>RESULTS</b>The frequencies of apoE genotypes epsilon 3/4, epsilon 3/3 and epsilon 2/3 were 2.4%, 69.0% and 23.8% respectively in the centenarian group, and the frequencies of their epsilon 4, epsilon 3 and epsilon 2 alleles were 1.2%, 82.1% and 16.7%. The frequencies of genotype epsilon 3/4 and epsilon 4, epsilon 3 allele in the centenarian group were significantly lower than those in controls(P< 0.01), whereas the frequencies of genotype epsilon 2/3 and epsilon 2 allele in the centenarian group were significantly higher than those in controls(P< 0.01). Longevity was noted to be positively correlated with epsilon 2 and negatively correlated with epsilon 4.</p><p><b>CONCLUSION</b>This relationship reflects a genetic influence on differential survival and may point to pleiotropic age-dependent effects on longevity.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate whether the insertion/deletion(I/D) polymorphism in the angiotensin converting enzyme(ACE) gene is associated with essential hypertension in Xinjiang Kazakh isolated population.</p><p><b>METHODS</b>The study covered 201 hypertensives and 151 normotensive controls in Xinjiang Barlikun Kazakh population. The I/D polymorphism of ACE gene was determined by polymerase chain reaction.</p><p><b>RESULTS</b>The frequencies of D and I in the hypertensive group (0.44 and 0.56, respectively) were not significantly different from the controls(0.39 and 0.61, respectively, P=0.16). The frequencies of ACE genotypes of DD, ID, and II were 0.18, 0.52, 0.30 in hypertensives respectively and 0.17, 0.43, 0.40 in control group respectively. There was no significant difference in genotypes between hypertensive group and normotensive group (P=0.14).</p><p><b>CONCLUSION</b>The results suggested that the I/D polymorphism of ACE gene might not be associated with hypertension in the Kazakh population of Xinjiang Barlikun area.</p>

ABSTRACT

Electrocardiac signal is one of the most important signals which is used to trigger ventricular assist device (VAD), and the delay time of VAD assistance is very important to get a satisfied result. Proper delay will give VAD relatively enough time to assist, avoiding left heart failure caused by the collision of the heart and VAD during systolic phase. This becomes much more important when the left atrium drainage is insufficient. The aim of our study is to set up an equation to calculate the delay time by RR interval. We try to set up an equation about RR and R-Ao like: R-Ao = A x (RR)n + B(A and B are constant). RR represents the RR interval and R-Ao represents the duration of the period between the peak point of QRS and the point of aortic valve closing; First, calculate RR according to weighting average method, and then, calculate the anticipant R-Ao according to the before-mentioned equation. After adjustment, R-Ao will be used as assistance delay time. R-R interval was measured in 457 selected pediatric patients who were undergiong left heart catheterization and who did not have arrhythmias. From the ECG recording during catheterization, R-R interval was measured while R-Ao was obtained from aortic pressure wave chart; Plot graphs with R-Ao as dependent variable and (RR)n as independent variable; find out correlating model and calculate the arguments A and B of R-Ao = A x (RR)n + B. The results showed that the relation between (RR)1/3 and R-Ao is the most significant, the relation coefficient is 0.733, the regress coefficient is -0.182 (P < 0.001) and the interception is 1.070. This means that R-Ao = (-0.182) (RR) 1/3 + 1.070. The likelyhood degrees of different sections differ markedly. When heart rate is less than 120 beats per min. The relation argument is about 0.733 while 0.45 when heart rate is more than 120 beats per min, Therefore, we can use the equation R-Ao = (-0.182) (RR)1/3 + 1.070 to calculate R-Ao when heart rate is less than 120 beats per min.
