ABSTRACT

BACKGROUND:A pure chitosan (CS) scaffold has poor mechanical properties,and cannot play a load-bearing role in the bone defect region.Therefore,many scholars attempt to develop multi-walled carbon nanotube (MWCNT)/chitosan (MWCNT/CS) bone tissue scaffolds.OBJECTIVE:To develop the functionalized MWCNT/CS composite scaffolds carrying zinc (f-MWCNT/CS-Zn) and to test its mechanical properties,antibacterial activity and biocompatibility.METHODS:(1) F-MWCNT composite scaffolds were prepared by the acid mixture method,and f-MWCNT/CS-Zn composite scaffolds were prepared through mixing CS,f-MWCNT (0.5%) with different concentrations of ZnSO4·7H2O (the concentrations of Zn in the composite scaffolds were 0.2%,1%,2%,respectively).The surface hardness and tensile strength of the pure CS scaffold and the f-MWCNT/CS-Zn composite scaffolds were measured.(2) The inhibitory effect on Staphylococcus aureus or Escherichia coii of the pure CS scaffold and f-MWCNT/CS-Zn scaffolds was tested by inhibition zone test.(3) The osteoblasts MC3T3-El were cultured with the leaching solution of f-MWCNT/CS-Zn composite scaffolds.The cell proliferation was detected by MTT assay at 1,2,3 days after culture,then the relative proliferation rate was calculated and the cytotoxicity was evaluated.RESULTS AND CONCLUSION:(1) The surface hardness and tensile strength of the f-MWCNT/CS-Zn composite scaffolds were higher than those of the pure CS scaffold (P < 0.05),and there was no significant difference in the surface hardness and tensile strength between the different zinc-containing composite scaffolds.(2) The antibacterial properties of the three f-MWCNT/CS-Zn composite scaffolds against Staphylococcus aureus and Escherichia coii were higher than those of the pure CS scaffold (P < 0.05),and the antibacterial activity was enhanced with the increase of zinc mass fraction.(3) The f-MWCNT/CS-0.2%Zn and f-MWCNT/CS-1%Zn composite scaffolds had no cytotoxicity.To conclude,the f-MWCNT/CS-1%Zn composite scaffold has good mechanical properties,antibacterial activity and biocompatibility.

ABSTRACT

@#To determine the effects of celastrol on the proliferation and apoptosis of cell lines HL-60 and Jurkat in human leukemia. Human leukemia cell lines were treated with celastrol at different concentrations. The inhibitory rate of cell proliferation was detected by MTT assay; the apoptosis rate was detected by AnnexinV-FITC/PI double staining; cell cycle was observed by PI staining; cell morphology was observed by transmission electron microscope(TEM). Cell proliferation was inhibited by celastrol, with IC50 of(0. 46±1. 05)μmol/L and(0. 88±1. 13)μmol/L at 24 h. Celastrol induced cell apoptosis in a dose-dependent manner. The cell cycle distribution of G1 phase rate increased, S phase rate decreased(P< 0. 05)with typical cell apoptosis-induced morphological changes. Results showed that celastrol could significantly inhibit cell proliferation and induce apoptosis in human leukemia cell lines of HL-60 and Jurkat.

ABSTRACT

<p><b>OBJECTIVE</b>To compare the efficacy difference on the treatment of supraspinal ligament injury in deficiency syndrome between only-puncturing at Dazhong (KI 4) and routine acupuncture therapy and then explore an optimum process.</p><p><b>METHODS</b>One hundred and twenty-six cases were randomly divided into a Dazhong group (63 cases) and a routine acupuncture group (63 cases). Based on the same treatment including TDP irradiation, cupping at Shenshu (BL 23), Dachangshu (BL 25), Mingmen (GV 4), Yaoyangguan (GV 3) and Ashi points, the patients in the Dazhong group received only-puncturing Dazhong (KI 4) with respiratory reinforcing method once daily or every other day, three times of which made a course of the treatment, with on interval of one day or without between different courses. For the routine acupuncture group, the same reinforcing method was applied at shenshu (BL 23) pointect, and the other requirements such as courses were also the same.</p><p><b>RESULTS</b>The cured and markedly effective rate was 88.9% (56/63) in the Dazhong group, which was significantly superior to that of 54.0% (34/63) in the routine acupuncture group (P < 0.01). Compared before and after the treatment, the score of Numerical Rating Scale (NRS) and the clinical symptom score of the two groups were significantly decreased (all PF < 0.01). Compared between the two groups, there were significant differences in NRS and clinical symptom score the Dazhong group was better than the routine acupuncture group (both P < 0.01).</p><p><b>CONCLUSION</b>The only-puncturing Dazhong (KI 4) method combined with cupping and TDP irradiation are simple and effective for supraspinal ligament injury in deficiency syndrome.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effects of di-butyl phthalate (DBP) on the reproductive system of adolescent male rats.</p><p><b>METHODS</b>Sprague-Dawley (SD) rats aged 5 weeks were assigned to receive corn oil (vehicle control) or DBP orally at 10, 100 and 500 mg/(kg x d) for 30 days. After the exposure, the testis, epididymis, liver and pituitary of the rats were weighted and their ratios to the body weight obtained. Histopathological changes of the testis and epididymis were examined by Hematoxylin-eosin staining, the levels of testosterone (T), luteinizing hormone (LH) and follicle stimulating hormone (FSH) in the serum were measured by radioimmunoassay, and the relative mRNA expressions of the steroidogenesis acute regulatory protein (StAR), proliferating cell nuclear antigen (PCNA), cytochrome P450 cholesterol side chain cleavage enzyme (P450scc) and scavenger receptor (SR) were detected by real-time quantitative RT-PCR.</p><p><b>RESULTS</b>DBP induced significant histopathological changes in the testicular tissue at 100 and 500 mg/(kg x d), and decreased the testicular and epididymal weights, inhibited the mRNA expressions of StAR and PCNA, reduced the levels of T and LH, and elevated the level of FSH at 500 mg/(kg x d). At the dose of 10 mg/(kg x d), DBP increased serum LH and FSH and the mRNA expression of P450scc. While the SR mRNA expression showed no significant changes in all the groups.</p><p><b>CONCLUSION</b>High level of DBP has apparent toxic effect on reproductive system of male rats. Low - dose DBP can increase the level of serum gonadotropin LH and affect the mRNA expression of P450scc in the testis.</p>
