ABSTRACT

Objective:The homogeneity regularity of "imprinting templates" of compound preparation of traditional Chinese medicine(TCM) was revealed, which provided the basis for effectively controlling the quality of compound preparation of TCM, by clarifying the dynamic changeability in number and content of volatile ingredients in different batches of Houttuyniae Herba. Method:The volatile oil of Houttuyniae Herba was extracted by steam distillation.The volatile components in 11 batches of Houttuyniae Herba were determined by GC-MS(electron ionization, ion source temperature of 230℃, detection range of m/z 40-500) combined with literature and ChemicalBook database.The relative content of each component was calculated by peak area normalization method and analyzed by the total quantum statistical moments of fingerprint. Result:There were only 15 common ingredients in the volatile oil from 11 batches of Houttuyniae Herba, accounting for 3.35%of total number and 73.94%of total content, while the number of non-common ingredients was 433, accounting for 96.65%of total number.However, the RSD of total quantum first moment of GC-MS fingerprint was 6.8%, the RSD of total quantum second moment was 14.8%(RSD of standard deviation was 7.4%), and the RSD of similarity of total quantum statistical moment was 3.8%.The type and content of volatile ingredients in Houttuyniae Herba were heterogeneous, but their chromatographic behavior of GC-MS fingerprint was uniform, the whole "imprinting templates" was similar. Conclusion:The total quantum statistical moment(similarity) of TCM fingerprint is applicable to quality control of dynamic Chinese medicines, which will play a breakthrough role in the study of homogeneity and stability of material reference and compound preparation for ancient classical formulas.

ABSTRACT

Objective: To explore the effect of Scutellariae Radix on the diversity of intestinal flora in rats under physiological conditions, in order to determine the property of Scutellariae Radix property. Method: 16S rRNA high-throughput gene sequencing technique was used to detect the cecum solutes of rats treated with Scutellariae Radix (10 g·kg-1). The number, richness and diversity index of the intestinal flora taxon (OTUs) and the differential phylum and genus were comprehensively analyzed. The network visualization was used to find the correlation between differential phylum and genus. Result: Based on the Illumina Miseq platform, compared with the blank group, the number of OTUs and the index of richness and diversity of the intestinal flora of the rats treated with Scutellariae Radix decreased. Bacteroidetes and Spirochaetes were significantly up-regulated(PPPPPRuminococcus, Paraprevotella, Prevotella and Oscillospira. Conclusion: Scutellariae Radix can reduce the diversity of intestinal flora and inhibit the metabolism of the body, so its property is cold.

ABSTRACT

In this paper,immune fingerprint was used to screen the allergenic components of Shuanghuanglian Injection(SHLI) by enzyme-linked immuno sorbent assay(ELISA) combined with HPLC/MS method. ELISA-embedded anti-IgE antibody could successfully adsorb allergens in SHLI and its plasma samples containing drugs through different routes of administration,suggesting that SHLI can induce type I hypersensitivity in rats. HPLC fingerprints and MS map of SHLI and drug-containing plasma samples from different routes of administration before and after anti-IgE antibody adsorption were established. According to the similarity evaluation of HPLC fingerprints and analysis results MS map,the sensitization of traditional Chinese medicine injections can be changed by different administration methods. There were 22 kinds of components that can be adsorbed by specific anti-Ig E antibodies in Shuanghuanglian Injection and its drug-containing plasma,most of them were acids and nitrogen compounds. Based on supramolecular theory,it was inferred that these compounds came from SHLI or body,and may form supramolecular hapten,which results in immunotoxicity and allergic reaction when being used as injection instead of oral liquid. Immune fingerprint is not only used to screen out single component allergen,but also more comprehensive,sensitive and easy to operate. It can provide reference for the future research methods of allergic reaction of traditional Chinese medicine injections.

ABSTRACT

Objective To observe the clinical efficacy of acupuncture plus intradermal needle in treating mild-to-moderate post-stroke depression (PSD). Method Ninty patients with mild-to-moderate PSD were randomized into group A, B and C, with 30 cases in each group. All the groups were intervened by selecting Baihui (GV20), Shenting (GV24) and Yintang (GV29). Group A received acupuncture plus intradermal needle; group B, intradermal needle alone; group C, acupuncture plus sham acupuncture. Hamilton depression rating scale (HDRS) score and Barthel index (BI) score in the three groups were observed before and at the end of the treatment, and the clinical efficacies of the three groups were compared. Result The HDRS score and BI score showed a significant change after the treatment in the three groups (P<0.01). After the treatment, the HDRS score and BI score in the group A and group B was significantly different from those in group C (P<0.05). The total effective rate was 90.0% in group A versus 86.7%in group B, and 73.3% in group C; there was a statistically significant difference between group C and group A or B (P<0.01, P<0.05). Conclusion Both acupuncture plus intradermal needle and intradermal needle alone are effective approaches in treating mild-to-moderate PSD. The treatment efficacy of these two methods is similar.

ABSTRACT

<p><b>OBJECTIVE</b>To construct CD19-specific artificial antigen-presenting cells (aAPCs) for in vitro activation and expansion of CD19 chimeric antigen receptor (CAR)-modified T cells (CD19-CAR-T) and investigate their cytotoxic effect.</p><p><b>METHODS</b>CD19-specific aAPCs (NIH3T3-CD19/86, NIH3T3-CD19/86/137L) expressing costimulatory molecules CD86 and/or CD137L were prepared on the basis of NIH3T3 backbone cells by lentivirus-mediated gene transfer. Irradiated CD19-specific aAPCs were co-cultured with CD19-CAR-T cells to activate and amplify CD19-CAR-T cells. The growth curve of CD19-CAR-T cells was determined by trypan blue exclusion assay, and CD19CAR expression and phenotype on CD19-CAR-T cells were detected by flow cytometry. The in vitro cytotoxicity of CD19-CAR-T cells against the target cells was evaluated by bioluminescence-based cytotoxicity assay.</p><p><b>RESULTS</b>Flow cytometry showed that NIH3T3-CD19/86 and NIH3T3-CD19/86/137L expressed high levels of CD19, CD86 and/or CD137L. Both NIH3T3-CD19/86 and NIH3T3-CD19/86/137L cells could amplify CD19-CAR-T cells efficiently, but NIH3T3-CD19/86/137L cells had better amplification effect. After 14 days of co-culture with NIH3T3-CD19/86/137L cells, the number of CD19-CAR-T cells was significantly greater than that of NIH3T3-CD19/86 cells (P<0.05), and the proportion of CD19-CAR-T cells in the total T cells increased significantly (P<0.05). CD19-CAR-T cells amplified by CD19-specific aAPCs produced target-specific cytotoxicity and were able to specifically kill CD19-positive target cells. About 20% central memory T cells were present in the final products expanded by NIH3T3-CD19/86/137L.</p><p><b>CONCLUSION</b>We successfully prepared CD19-specific aAPCs that can specifically amplify functional CD19-CART cells in vitro, which facilitates the acquisition of clinical-scale high-quality CD19-CART cells.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the blood test patterns for blood donors after nucleic acid detection in blood center.</p><p><b>METHODS</b>The collected blood samples after voluntary blood donors first were detected by conventional ELISA, then 31981 negative samples were detected via HBV/HCV/HIV combined nucleic acid test of 6 mixed samples(22716 cases) or single samples(9265 cases) by means of Roche cobas s201 instrument. The combined detection method as follows: the blood samples were assayed by conventional nucleic acid test of 6 mixed samples, at same time, 6 mixed samples were treated with polyethylene glycol precipitation method to concentrate the virus, then the nucleic acid test of blood samples was performed; the single detection method as follows: firstly the conventional nucleic acid test of single sample was performed, then the positive reactive samples after re-examination were 6-fold diluted to simulate the nucleic acid test of 6-mixed samples. The positive rate of positive samples detected by combined nucleic acid test, positive samples detected by nucleic acid test of mixed virus concentration and positive samples detected by single nucleic acid test was statistically analyzed. In addition, for HBVpersons the serological test yet should be performed.</p><p><b>RESULTS</b>In 22 716 samples detected by nucleic acid test of 6 mixed samples (MP-6-NAT) , 9 cases were HBV(0.40‰, 9/22716); at same time, the detection of same samples by nucleic acid test of mixed sample virus concentration showed 29 cases of HBV(1.28‰, 29/22716). In 9265 samples detected by single nucleic acid test(ID-NAT) 12 cases showed HBV(1.30‰, 12/9265), meanwhile the detection of these 12 samples with HBVby 6-fold dilution for virus concentration found only 4 samples with HBV. In serological qualified samples, ID-NAT unqualified rate was 1.28‰, which was higher than that of MP-6-NAT(0.4‰) (χ=8.11, P<0.05); but there was no statistical difference between unqualified rate of ID-NAT and MP-6-NAT(1.3‰ vs 1.28‰)(χ=0.00, P>0.05). In 41 samples with HBsAgHBV DNAdetected by ELISA, 36 samples were confirmed to be occult HBV infective(OBI) by HBsAb, HBcAb test of ELISA; out of these 41 samples, 33 samples showed HBcAb(91.66% of OBI), 5 might be HBV "window period" infective, moreover the HCV RNA and HIV RNA positive samples were not found.</p><p><b>CONCLUSION</b>To avoid the missdiagnosis of donors with low level of virus, the nucleic acid test must be carried out after virus concentration of mixed samples when the blood test pattern of donors is nucleic acid test of mixed samples, otherwise the single nucleic acid test must be performed to obtain more high detected rate of virus nucleic acid. The HBcAb serologic test and physical examination of donors before blood donation must be enhanced on basis of serological test of HBsAg; for high risk people, the persuading no blood donation is simplest pattern.</p>

ABSTRACT

Objective To fulfill the precise control of the liquid flow by a multifunctional medical flowmeter. Methods The flowmeter had a separated structure, and was composed of a liquid pipeline unit and a flow control unit. The flow control unit apprehended a velocity of liquid level changes in the sterile pipeline by ultrasonic sensing technique, and regulated fluid speed by a SCM-controlled electromagnetic valve. Results The requirements for sterile environment were fulfilled by the sterile liquid pipeline, and the flow control unit realized precision control of flow rate and achieved the expected effect. Conclusion The flowmeter has simple structure, low cost, convenient operation and use, can greatly reduce the workload of the nursing staff and the possible health risks, and thus is worthy promoting practically.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the clinical value ofF-FDG PET/CT for patients with B cell lymphoma-associated hemophagocytic syndrome.</p><p><b>METHODS</b>The clinical characteristics, laboratory parameters andF-FDG PET/CT data of 23 newly diagnosed patients sufferred from B cell lymphoma-associated hemophagocytic syndrome were retrospectively analyzed. The correlation between PET and laboratory parameters were determined using Spearman correlation test. The prognostic factors were analyzed by the Kaplan-Meier method.</p><p><b>RESULTS</b>23 patients were all examined byF-FDG PET/CT before chemotherapy, theF-FDG uptake of spleen positively correlated with neutrophil count and hemoglobin content (r=0.588, P=0.035;r=0.699, P=0.008), respectively, and theF-FDG uptake of bone marrow positively correlated with neutrophil count only (r=0.691, P=0.009). Among all the clinical or laboratorial parameters andF-FDG PET/CT, only PET parameter was poor factor affecting prognosis of patients with B cell lymphoma-associated hemophagocytic syndrome. Out of 6 patients received PET/CT scans after 6 cycles of treatment, the 5 patients with negative PET/CT survived, and one patient with positive result died.</p><p><b>CONCLUSION</b>BaselineF-FDG PET/CT may provide prognostic information for the management of patients with B cell lymphoma-associated hemophagocytic syndrome, and the data ofF-FDG PET/CT before chemotherapy may predict the pregnosis of the patients with negative results, and the negative PET/CT results after chemotherapy betokens a better prognosis of patients.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the value of pretherapeuticF-FDG PET/CT in the focus detection, staging and prognosis evaluation of adult Burkitt's lymphoma(BL).</p><p><b>METHODS</b>The clinical data and detection results ofF-FDG PET/CT scan of 18 patients with BL before the treatment from December 2008 to February 2015 were analyzed retrospectively. The lesions distribution were observed, the maximal standard uptake value(SUV) ofF-FDG by lesions and the maximal diameter of lymph nodes were measured. One-way analysis of variance was used to analyze the SUV, and the Spearman correlation was used to analyze the relationship between SUVand the maximum diameter of lymph nodes. The prognostic factors were analyzed by using Kaplan-Meier survival analysis.</p><p><b>RESULTS</b> F-FDG PET/CT showed 100% positive detection rate for 18 adults with BL, lymph nodal lesions were found in all the patients, and the most common area was lymphonodi coeliaci (9/18, 50%), 8 cases showed extranodal soft tissue involvement, 6 cases showed gastrointestinal tract, bone marrow involvement was detected in 7 cases, out of which the mutiple-focal involvement was found in 4 cases. The median SUVof lymph nodes, extranodal soft tissue and bone marrow was 11.7 (range of 3.4 to 28.5), 9.85 (range of 6.7 to 21.9) and 11.8(range of 5.6 to 23.8), respectively, there were no significant differences among these 3 types of lesions (F=1.013, P=0.369). And the median SUVof all lesions was 16.5 (range of 8.8 to 28.5). The clinical staging of 2 patients (2/8, 11.1%) was changed byF-FDG PET/CT imaging. The univariate analysis revealed that only the IPI (international prognostic index) could be the prognostic factor (χ=6.602, P=0.010), but the prognosis was not different between the patients below and above the median SUVof all lesions and lymph node (all P>0.05).</p><p><b>CONCLUSION</b>BL in adult is an intenselyF-FDG-avid tumor, theF-FDG PET/CT is an effective imaging tool for the detection and staging of this disease. In our study, the prognostic value of pretherapeuticF-FDG PET/CT still is unclear, that is not recommended for the prognosis of the adult BL patients.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the PET-CT manifestation and clinical features of patients with Waldenstrom macroglobulinemia(WM) .</p><p><b>METHODS</b>The clinical features, laboratorial examination results and PET-CT manifestation of 12 patients with WM were analyzed retrospectively.</p><p><b>RESULTS</b>The average age of 12 patients with WM was 63.5 years old, the most common incipient symptoms were fatigue and hyperviscosity syndrome. The median SUVof bone marrow was 4.9 (range of 2.1 to 21.9), with diffusely increasedF-FDG uptake in all the patients. In 6 patients, the median SUVof splenomegaly and lymphadenectasis was 3.2 (range of 2.3 to 5.2) and 5.2 (range of 3.6 to 11.2), respectively. The SUVof bone marrow, splenomegaly and lymphadenectasis did not related with white blood cell count, hemoglobin level, blood platelet count, β2- microglobulin level and serum monoclonal IgM of such patients, respectively (both P>0.5). In univariate analysis, serum monoclonal IgM and the SUVof lymphadenectasis were found to be the prognostic factors (both P<0.5).</p><p><b>CONCLUSION</b>TheF-FDG PET-CT feature of Waldenstrom macroglobulinemia has some characteristics which are helpful for setting up the clinical therapeutic schedule.</p>

ABSTRACT

Chuangxinmycin (CM) from Actinoplanes tsinanensis was an antibiotic discovered by Chinese scientists about 40 years ago. It contains a new heterocyclic system of indole fused with dihydrothiopyran, whose biosynthetic mechanism remains unclear. CM is used as an oral medicine in the treatment of bacterial infections in China. The simple structure makes CM as an attractive candidate of structure modification for improvement of antibacterial activity. Recently, we analyzed the secondary metabolites of Actinoplanes tsinanensis CPCC 200056, a CM producing strain, as a natural CM analogue. We discovered the first natural CM analogue 3-demethylchuangxinmycin (DCM) as a new natural product. Compared to CM, DCM exhibited a much weaker activity in the inhibition of the bacterial strains tested. The finding provides valuable information for the structure-activity relationship in the biosynthesis of CM.

ABSTRACT

To reveal the characterization of interaction between Chinese and western medicinal injections, isothermal titration calorimetry (ITC) was applied to evaluating the interaction of Yiqi Fumai injection (YQFM, as mode drug) with epinephrine hydrochloride injection (YS) and 5% glucose injection (5% GS). The diversification of Gibbs free energy (ΔG), enthalpy (ΔH), and entropy (ΔS) were determined to judge the reaction types of colliquefaction procedures of different injections. Meanwhile, the fingerprints of YQFM before and after combined with the various injections were compared to validate the results. This work demonstrated that during the titration procedure of YQFM and YS, [ΔH] > T [ΔS] , that was to say the reaction was enthalpy-driving. And the reactive profile indicated that a great deal of heat gave out during the procedure. Obviously, chemical reactions happened and the internal component changed. On the other side, the reaction of YQFM combined with 5% GS was entropy-driving, because [ΔH] < T [ΔS]. The reactive profile showed there was only a little heat released. So non-chemical reactions happened and the major ingredients did not change. ITC could be applied to the evaluation on compatibility of other kinds of Chinese and western medicinal injection combination.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the value of PET/CT after chemotherapy for evaluating therapeutic efficacy of patients with diffuse large B-cell lymphoma (DLBCL).</p><p><b>METHODS</b>(18)F-FDG PET/CT was performed before and after 6-8 cycles of chemotherapy in 73 newly diagnosed patients with DLBCL from September 2005 to January 2012. The results of pre-treatment PET/CT was compared with results of post-treatment PET/CT. These patients were divided into 3 groups: complete response group, partial response group and no response group. The post-treatment PET/CT results was used to assess its ability to predict progression-free survival (PFS) and overall survival (OS).</p><p><b>RESULTS</b>The comparison of PET/CT results before and after chemotherapy showed that 2-year PFS rates of complete response group, partial response group and no response group were 82% (41/50), 45.5% (5/11) and 8.3% (1/12), respectively; and the 3-year OS rates of 3 groups were 88% (44/50), 54.5% (6/11) and 8.3% (1/12) respectively (both P < 0.01). The 2-year PFS rate and the 3-year OS rate of the complete response group and partial response group were significantly higher than those of no response group (both P < 0.05), and there was also significant statistical difference between partial response group and complete response group in 2-year PFS rate and 3-year OS rate (both P < 0.05).</p><p><b>CONCLUSION</b>The post-treatment PET/CT can be accurately used to evaluate the curative efficacy of chemotherapy and prognosis of patients with DLBCL.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the relationship between nitric oxide (NO) in central nervous system and exercise-induced fatigue stress and to study the effect of L-arginine (L-Arg), as a substrate of nitric oxide, on the exercise capacity and NO content in the exhausted rat brain and blood.</p><p><b>METHODS</b>Through an implanted cannula, the normal saline or L-Arg was microinjected into rat's intracerebroventrical for consecutive four days. Then an acute exhaustive model (on the speed of 18 m/min, an inclination of 5 degrees) was established with animal treadmill. The time of exercise till exhaustion was recorded, and the total workload was calculated that represented the exercise capacity. Nitrate and nitrite (NO3/NO2-, NOx-) levels in blood, hypothalamus and hippocampus were assayed.</p><p><b>RESULTS</b>Both the time of exercise till exhaustion and total workload in the LArg group increased respectively by 51.8% and 50.08% (P < 0.05), compared with those in the control. The NOx- content in hypothalamus in the L-Arg group (8.93 +/- 1.83) micromol/g pro was larger than that in the control (4.25 +/- 0.79) micromol/g pro, (P < 0.01). There was no significant difference in NOx- content in brain and hippocampus between the two groups. The total workload was positively correlated with NOx- concentration in hypothalamus (P < 0.01). However, there was no correlation between workload and changes in hippocampus NOx- content at fatigue.</p><p><b>CONCLUSION</b>Intracerebroventricular microinjection of L-Arg may enhance the exercise capacity and lead to up-regulation of NO by means of L-Arg-NO signal path in the hypothalamus. Hypothalamus may be a key site in brain in the modulation of physiological exercise.</p>

ObjectiveTo evaluate the functional changes of stunned myocardium before and after coronary artery bypass graft(CABG) treatment,and clear the meaning of revascularization which CABG has brought to patients with diffused vascular changes.MethodsA total of 36 patients with 99% diffused coronary artery stenosis in left anterior descending branch underwent non-pump CABG treatment in the Department of Cardiothoracic Surgery the First affiliated hospital of Harbin Medical University.Real-time three-dimensional echocardiography (RT3DE) was repeatedly performed 1 week before operation and 10 days,1 month,and 3 months after CABG.Regional diastolic volumes,systolic volumes,ejection fractions,regional stroke volume to global diastolic volume and the values of abnormal segments before and after CABG were studied.ResultsOne week before operation and 10 days,1 month and 3 months after CABG,the differences of volumes between groups in the last phases of diastole and systolic were statistically significant in anterior wall basement segment,anterior septal basement segment,anterior wall intercalary segment,anterior septal intercalary segment,anterior wall of apex cordis and septation of apex cordis(F =3.51,3.55,4.08,4.05,2.98,3.01,all P < 0.05; F =4.51,4.55,4.08,3.00,2.96,2.99,all P < 0.05).The values of the six segments mentioned above,3 months after operation[(6.74 ± 1.23),(6.64 ± 1.21),(6.02 ± 1.10),(5.95 ± 1.09),(5.82 ± 1.06),(5.10 ± 0.93)ml; (2.74 ± 0.50),(2.69 ± 0.49),(2.51 ± 0.46),(2.32 ± 0.42),(2.36 ± 0.43),(2.03 ± 0.37)ml] were compared with those of 1 week before operation[(8.33 ± 1.52),(8.20 ± 1.50),(7.43 ± 1.36),(7.36 ± 1.34),(7.19 ± 1.31),(6.29 ± 1.15)ml; (4.94 ± 0.90),(4.85 ± 0.88),(4.53 ± 0.83),(4.18 ± 0.76),(4.25 ± 0.78 ),(3.65 ± 0.67)ml],the differences were statistically significant (all P < 0.05); the differences between groups in regional ejection fractions,regional-global ejection fractions were statistically significant(F =4.56,4.88,4.28,3.15,2.93,2.88,P < 0.01 or < 0.05; F =5.56,5.28,4.98,5.15,3.03,2.78,P < 0.01 or < 0.05).Compared with 1 week before the operation, 1 month after the operation in regional ejection fractions,10 days,1 month in regionalglobal ejection fractions after the operation,4 segments of them were significantly improved(all P < 0.05) and 3 months after operation,all the 6 segments had been improved significantly(all P < 0.05).The maximum volume of the sum of group difference of the 6 segments and the 4 segments in the last phase of diastole was statistically significant(F =2.58,5.81,P < 0.05 or < 0.01 ),and the summation began to decrease 10 days after the operation.The values of 3 months after operation[ (36.27 ± 1.10),(25.35 ± 1.16)ml] were compared with that of 1 week before operation[ (44.80 ± 1.36),(31.32 ± 1.43)ml ] the difference was statistically significant (all P< 0.05).The maximum volume summafion comparisons of 6 segments and 4 segments in the last phase of systolic had statistical significance(F =5.77,5.57,all P < 0.01 ),and 10 days after the operation,the summation began to decrease.The values of 1 month[(16.4 0 ± 0.48),(11.58 ±0.51 )ml],and 3 months after operation[ (14.65 ± 0.45),(10.26 ± 0.46)ml],were compared with those of 1 week before operation[ (26.40 ± 0.80),(18.50 ± 0.84)ml],the differences were statistically significant (all P < 0.05).ConclusionsStunned myocardium can be improved through CABG in myocardium systolic,diastole function and ejection fractions of the relevant segments and all of this have proved that patients undergoing CABG revascularization can improve the heart function of the ischemic area.

ABSTRACT

The present study was to determine the effect of c-SRC on the viability of human cervical cancer HeLa cells and the expression of phosphorylated signal transducer and activator of transcription-3 (p-STAT3) of the cell. Post-transfection of c-SRC RNA interference vector, RT-PCR and Western blot were utilized to observe the contents of c-SRC mRNA and protein, respectively, in HeLa cells. The MTT was used to observe the viability of the cells. Cell cycle was observed by flow cytometry. The content of p-STAT3 in the cells was also investigated after knockdown of c-SRC. Knockdown of c-SRC significantly decreased the contents of c-SRC mRNA and protein in the cells. The viability of the cells decreased by 23.1%, 29.3%, 38.6% and 45.0% (all P < 0.05), respectively, after the cells were transfected with c-SRC RNA interference vector for 24, 48, 72, and 96 h. The number of S-phase cells decreased by 5.6%, 10.0%, 15.2% and 19.9% (all P < 0.05), respectively, after transfection of c-SRC RNA interference vector for 24, 48, 72, and 96 h. The content of p-STAT3 also decreased when c-SRC was knockdowned. Compared with the control group, after treatment of HeLa cells with STAT3 inhibitor Piceatannol for 24, 48, 72, and 96 h, the cell viability decreased by 23.8%, 29.7%, 37.3% and 45.4% (all P < 0.05), respectively, while increase of c-SRC content could not reverse the inhibitory effect. These results suggest that the inhibited viability of HeLa cells caused by knockdown of c-SRC is associated with the decreased content of p-STAT3 protein.

ABSTRACT

<p><b>BACKGROUND</b>Celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, is a non-steroidal anti-inflammatory drug used as an adjuvant to sensitize cancer cells to apoptosis. However, in rats suffering from acute rejection, celecoxib reduced apoptosis of myocardial cells. We hypothesize that celecoxib reduces myocardial apoptosis either by inducing apoptosis in peripheral blood lymphocytes (PBLs) or by altering the percentage of CD4(+) and CD8(+) lymphocytes.</p><p><b>METHODS</b>After cardiac transplantation, rats were administered intragastrically with celecoxib (50 mg/kg per day) for 3, 5 or 7 days, at which time the graft was excised and evaluated for organ rejection. In addition, PBLs were isolated from the blood to determine PBLs apoptosis, and the percentage of CD4(+) and CD8(+) lymphocytes.</p><p><b>RESULTS</b>Celecoxib induced PBLs apoptosis in 3 days, but protected the cells from apoptosis at 5 and 7 days. Also, the percentage of CD4(+) lymphocytes decreased only at 3 days, but a reduction in the percentage of CD8(+) lymphocytes was not seen until 7 days after the transplant surgery. Celecoxib only decreased acute rejection at 5 days, with no discernible difference in rejection after 3 and 7 days.</p><p><b>CONCLUSIONS</b>The results suggested that celecoxib displayed a multiple physiological function in a time-dependent manner.</p>

ABSTRACT

<p><b>BACKGROUND</b>Pachymic acid (PA), a natural triterpenoid, is known to significantly reduce cell proliferation and induce apoptosis in vitro through initiation of mitochondria dysfunction. However, its effect on immune cells and anti-rejection following organ transplantation remains unknown.</p><p><b>METHODS</b>In this study, we investigated PA as a treatment to control acute rejection occurred in rats which had accepted cardiac transplantation. We measured apoptosis of peripheral blood lymphocyte (PBLs), and CD4(+) lymphocyte, as well as the number of CD4(+) and CD8(+) lymphocytes and the effect of PA on acute rejection in rats 7 days after cardiac transplantation.</p><p><b>RESULTS</b>PA treatment might decrease allograft rejection, protect PBLs from apoptosis, and reduce the percentage of CD8(+) lymphocyte. PA neither regulated the number nor the apoptosis rate of CD4(+) lymphocyte.</p><p><b>CONCLUSIONS</b>Our findings indicated that PA has an anti-apoptotic effect acting on PBLs through a novel mechanism involving stabilization of the PBLs mitochondrial transmembrane potential, an anti-rejection effect in rats after cardiac transplantation and an inhibiting effect to CD8(+) lymphocyte.</p>

ABSTRACT

<p><b>BACKGROUND</b>A living fetus within the maternal uterus provides an example of allogene tolerance in mammals. Poria cocos Wolf is the main component of many Chinese medicinal combination drugs that have therapeutic effects on recurrent spontaneous abortion and that can maintain pregnancy until delivery. It was hypothesized that this herbal medicine can also prolong allograft survival after organ transplantation. Here, in an in vivo study, we report the anti-rejection effect of the ethanol extract of Poria cocos Wolf (EEPCW) in rats after cardiac allograft implantation.</p><p><b>METHODS</b>Ten normal rats were healthy controls. Eighty rats receiving homologous heart transplants were divided into 4 groups of 20 rats each based on type of treatment: olive oil 8 ml.kg(-1).d(-1), EEPCW 25 mg.kg(-1).d(-1), EEPCW 50 mg.kg(-1).d(-1) or cyclosporin A 5 mg.kg(-1).d(-1). Allograft survival was observed in 10 rats from each group. On the seventh day post transplantation, pathological lesions and percentages of CD3+, CD4+, and CD8+ lymphocytes and the CD4+/CD8+ ratio in peripheral blood were assessed in another 10 rats from each group and in 10 normal rats.</p><p><b>RESULTS</b>The survival time of donor hearts in the two EEPCW groups was significantly prolonged, to (15.9 +/- 2.4) days and (30.0 +/- 0.0) days, respectively, compared with (6.7 +/- 0.8) days in the control group. Pathological lesions in the two EEPCW groups were also less severe, and the percentages of CD3+, CD4+, and CD8+ lymphocytes and CD4+/CD8+ ratio were significantly lower in the EEPCW groups.</p><p><b>CONCLUSIONS</b>Acute rejection of heart transplants and cellular immune reaction can be effectively suppressed using the EEPCW. Taking advantage of novel immunosuppressants derived from Chinese medicinal herbs used to treat abnormal pregnancy provides a hopeful road for future research and treatment in organ transplantation.</p>
