ABSTRACT

AIM: To illuminate the effect of SWI/SNF-related, matrix-associated, actin-dependent regulator of chromatin, subfamily b, member 1 (SMARCB1) in early diagnosis and prognosis of hepatocellular carcinoma (HCC) by determining the clinical expression of SMARCB1 in HCC tissue and benign liver tissue.METHODS: The specific target gene SMARCB1 was selected from these genes by using The Cancer Genome Atlas (TCGA).SMARCB1 expression in HCC tissue and benign liver tissue was measured by immunohistochemistry.Further statistical analysis of TCGA was performed to illuminate the role of SMARCB1 on HCC occurrence and progression.RESULTS: Compared with the benign liver tissue, immunohistochemical staining showed that SMARCB1 expression was significantly up-regulated in the HCC tissue (P<0.01).In addition, SMARCB1 expression was significantly associated with advanced tumor stage (P<0.05).The relation between SMARCB1 expression at mRNA level and clinical prognosis was analyzed.The results indicated that high SMARCB1 expression was an independent prognostic factor for HCC (P<0.05).CONCLUSION: SMARCB1 may play a part as a carcinogenic gene in tumorigenesis.We can distinguish primary HCC samples from non-malignant samples according to its different clinical expression.High SMARCB1 expression probably predicts poor outcome in HCC patients.

ABSTRACT

Objective To analyze expression of Bmi-1 gene in thyroid tissues and the relationship between Bmi-1 gene and metastasis of papillary thyroid carcinoma.Methods RT-PCR was used to detect expression of Bmi-1 in normal thyrroid tissues(8 eases),nodular goiters(18 cases)and papillary thyroid carcinomas (24 cases).The result was analyzed by Fisher exact propability test.Results Bmi-1 expression was detected only in 1 case(12.5%)in normal thyroid tissues group,3 cases(16.7%)in nodular goiters group,and 19 cases(79.2%)in papillary thyroid carcinoma group.The difference of Bmi-1 expression between the 3 groups had statistical significance(P<0.05).There were 16 cases of lymph node metastasis(66.67%)in the 24 cases of papillary thyroid carcinoma.The difference of Bmi-1 expression between cases with cervical lymph node metastasis and those without metastasis had statistical significance(P<0.05).Among the 19 thyroid carcinoma cases in which Bmi-1 expression was positive,12 eases(63.2%)were over45 years old,and 7 cases(36.8%)were under 45 years.The difference had statistical significance(P<0.05).Conclusions Expression of Bmi-1 gene in papillary thyroid carcinoma tissue is significantly higher than that in nodular goiters and normal thyroid tissues.Bmi-1 expression is closely relevant to lymph node metastasis and age,implying that Bmi-1 expression has clinical significance of distinguishing thyroid malignant tumors from benign diseases and it may provide clues to unravel the mechanism and prognosis of papillary thyroid carcinoma metastasis.

ABSTRACT

In this work, growth, cellular chemical composition and production of fusaricidin type antimicrobial compounds by P. polymyxa SQR-21 were compared in tryptone broth supplemented with four concentrations of iron (25, 50, 100 and 200 µM). The data revealed that the growth of P. polymyxa SQR-21 was increased by 3-8 percent with the increase in concentration of ferric ion (Fe3+). The production of fusaricidin type compounds was increased by 33-49 percent only up to 50 µM Fe3+ and the highest level of Fe3+ was inhibitory. Increase in the liquid culture Fe3+concentration increased the intracellular protein (2 percent), intracellular carbohydrate (14 percent), extracellular protein (7 percent) and polysaccharide contents (18 percent) while the intracellular lipid contents were increased (11 percent) only up to 50 µM Fe3+. In addition, the regulatory effects of Fe3+ were also reflected by the increase in total RNA contents and relative expression of the fusaricidin synthetase gene (FusA) by 3-13 and 35-56 percent, respectively, up to 50 µM Fe3+, after that a continuous decrease was observed.


Tipo compostos do fusaricidin do produto das tensões do polymyxa de Paenibacillus que é ativo de encontro a uma variedade larga das bactérias e de fungos gram-positive. O crescimento, a composição química celular e a produção do fusaricidin datilografam compostos antimicrobial pelo P. o polymyxa SQR-21 foi comparado no caldo de carne do tryptone suplementado com as quatro concentrações (25, µM 50, 100 e 200) do ferro. Os dados revelaram que o crescimento do P. o polymyxa foi aumentado por 3-8 por cento com o aumento na concentração do íon férrico (Fe3+) e o tipo produção do fusaricidin dos compostos foi aumentado 33-49 por cento somente até 50 pelo µM Fe3+ quando o nível o mais elevado de Fe3+ era inhibitory. O aumento na concentração de Fe3+ na cultura líquida aumentou a proteína intracellular (2 por cento) e os índices de hidrato de carbono (14 por cento) e a proteína extracellular (7 por cento) e os índices do polysaccharide (18 por cento) quando os índices intracellular do lipid eram (11 por cento) somente até 50 o µM aumentado Fe3+. Além, os efeitos regulatory de Fe3+ foram refletidos também pelo aumento em índices totais do RNA e na expressão relativa do gene do synthetase do fusaricidin (FusA) por 3-13 e por 35-56 por cento respectivamente, até 50 o µM Fe3+, em seguida que uma diminuição contínua estêve observada.
