ABSTRACT

Objective To study the repairment of pancreas mesenchymal stem cells(MSCs) on injured pancreas of rats,and find a new source of cells for treatment of diabetes. Methods ① Pancreases were taken out of three-day-old Wistar rats under bioclean condition,and cells were obtained by V-collagenase digestion. Generations were passed by conventional culture,cells were purified by adherence screening method,cell morphous was observed by Giemsa staining. The expressions of surface marker CD34 and CD44 were determined by FCM,and compared with bone marrow MSCs(BM-MSCs),the differences in character and function were observed. ②The experimental rats were divided into two groups randomly. Pancreas ischemic necrosis model was made by deligation,then the purified pancreas MSCs were marked with DAPI and then were transplantated partially. After two weeks,the survival rate was measured and histopathological detection was performed. Results ① The cells had concordant morphous gradually with vigorous generation. There was no significant difference in morphology and surface antigen compared with BM-MSCs.② The survival rate in experimental group was 75% ,the necrotic tissue had basi-rebounded. ABSTRACT

@# ObjectiveTo explore the effect of bone marrow mesenchymal stem cells transplantation on the deep partial thickness derma burn injury in guinea pigs.MethodsSuspension cells of MSCs were cultured in vitro and transplanted into wounds of the female guinea pig of the deep partial thickness derma burn injuring model with the concentration of 2×106cell/ml(group A),2×107cell/ ml(group B). The rate of wound healing was observed, factor Ⅷ was detected using immunohistochemistry. Y chromosome on the wound in guinea pig was detected using PCR 15,30,50 days after MSCs had been transplanted. ResultsThe rate of wound healing of groups A and B was better than that in the control group, but there was not significantly different between groups A and B. Microvessel density of wound in groups A and B was better than those in the control group on the 15th and 30th days. There was not significantly different between the group A and B. PCR results showed that Y chromosome gene was expressed on some recoveried wound of female guinea pig. ConclusionBone marrow mesenchymal stem cells transplantation is effective in deep partial thickness derma burn injury in guinea pigs.

ABSTRACT

Objective: To adjust the distance between SD sequence and ATG in the same expressive plasmid pLX1 to enhance expression of heterologous bFGF gene in E. coli. Methods: Adjusting the different distance between SD sequence and ATG by Klenow and Mung Bean Nuclease. SDS PAGE and Western blot showed the expressed protein bFGF in E.coli. bFGF proteins were purified by HPHIC, HPGFC and HAC. Biological activity was examined by MTT. Results: Recombinant plasmids pLX2, pLX3 were obtained and the expressive levels were 8.03%, 9.9% respectively. Also the purified bFGF was obtained by HPHIC, HPGFC, HAC and its ED 50 was 2.29 ng/ml. Conclusion: Increasing the bFGF gene dosage by adjusting the distance between SD sequence and ATG could increase the expression level of a desired protein.
