ABSTRACT

Objective:To explore the molecular mechanism of Wendan Decoction in the treatment of Hypertension by the network pharmacolog. Methods:The potential active components and targets of Wendan Decoction were screened in TCMSP and TCMIP database. The TTD, genecards and OMIM databases were used to collect hypertension targets. Then the protein interaction was analyzed by using String database. The software of Cytoscape v3.7.1 was used to construct the network diagram of Wendan Decoction in the treatment of Hypertension and using the software of ClueGO to analyze GeneOntology and KEGG pathway of the potential targets. Finally, we used molecular docking technology for the appropriate of molecules and targets. Results:A total of 141 potential active components and 108 potential therapeutic targets were selected from Wendan Decoction. And 20 of core targets selected by MCC in CytoHubba. The GeneOntology analysis contained 164 GO terms, 133 of biological processes, 24 of molecular functions, and 7 of cell components. Seven key pathways were selected by enrichment analysis of KEGG pathway. Molecular docking results showed good docking between key components and targets.Conclusion:Wendan Decoction can treat hypertension through multiple ways and targets. This study predicts the possible mechanism of Wendan Decoction and provides data for its clinical application.

Traditional Chinese medicine treatment is the key to restore the balance of Yin and Yang through gasification. So it is important to discuss the mode of gasificaiton of Qi and Chinese medicine and their relationship in the machanism of the Chinese medicine for diseases. Life is a mode of Qi and Qi movement and gasificaiton make people alive. The Chinese medicine works on the Qi gasification to balance the Yin and Yang to keep the peace inner and outsides.

ABSTRACT

Objective To investigate the protective effects and possible mechanism of pigment epithelium derived faetor (PEDF) on myocardial cells H9C2 under hypoxia and serum-free condition.Methods H9C2 cells were culture in vitro and performed the hypoxia and serum-free processing.The cells were divided into the control group (H9C2),hypoxia group (hypoxia + H9C2),PEDF group(hypoxia+H9C2 +PEDF) and mitochondrial fission inhibitor(Mdivi-1) group(hypoxia+h9C2+Mivi-1).The apoptotic rate was detected by TUNNEL staining.The proteins levels of dynamin related peptide1 (Drp1) and cleaved-caspase 3 were measured by Western blot.Electron Microscopy and MitoTracker Red were used to detect the mitochondria morphology,the mitochondrial membrane potential was evaluated by cationic dye JC-1.MitoSOXTM was used to detect mitochondrial reactive oxygen species (ROS).Results Hypoxia induced mitochondrial fission(P<0.05).The hypoxia group (6 h) and control group had statistical difference(P<0.05).PEDF reduces mitochondrial fission under hypoxia condition(P<0.05),which had statistical difference between the PEDF group and hypoxia group (6 h)(P<0.05).PEDF and Mdivi-1 could decrease cell apoptosis under hypoxia condition(24 h),compared with the hypoxia group,the difference was statistically significant(P<0.05).Conclusion PEDF decrease cell apoptosis by inhibiting H9C2 cells mitochondrial fission under hypoxia condition.

ABSTRACT

Objective To explore the effects of Ginkgolide B on the expression of hypoxia inducible factor 1 α (HIF-1α) and P I3K/Akt pathway in the hippocampus of developing rats after pentylenetetrazol(PTZ)-induced status epilepticus,and to investigate the correlation between HIF-1α expression and PI3K/Akt pathway.Methods Ninety-six SD rats aged 21 days were randomly divided into normal saline group(group NS),status epilepticus group (group P),GKB treatment groups (group G+P),GKB +wortmannin treated group (group G+P+W),wortmannin treated group(group P+W).The brain tissue were harvested from the rats at 4 and 8 hours after the inducement,but in the group G+P at 1 h,4 h,8 h,24 h.Immunohistochemistry and Western blot were used respectively to detect HIF-1α and p-Akt protein expression.Results (1) For the group G+P,there were statistical differences in the expression levels of p-Akt protein between 1 h,4 h,8 h and 24 h(P<0.01),The p-Akt protein reached the peak level at 4 hours (0.85±0.03),there were statistical differences in the expression levels of HIF-1α protein between 1 h,4 h,8 h and 24 h(P<0.01),the HIF-1α expression reached the peak level at 8 hours(1.00±0.13).(2) The expression of HIF-1α in all the groups at 8 hours time point:the expression levels of HIF-1α in the group P and group G+P were significantly higher than those in the group NS (P<0.01) and the expression levels of HIF1α in the group G+P were higher than those in the group P(P<0.01).Using wortmannin,the PI3K/Akt specific inhibitor,HIF-1α protein expression in the group G+P+W and P+W was significantly decreased when compared with the group G+P and P (P<0.01).(3)The expression of p-Akt in all the groups at 4 hours time point:the expression levels of p-Akt in the group P and group G+P were significantly higher than those in the group NS (P<0.01) and the expression levels of p-Akt in the group G+P were higher than those in the group P (P< 0.01).Using wortmannin,p-Akt protein expression in the group G+P+W and P+W was significantly decreased when compared with the group G+P and P (P<0.01).Conclusion GKB can activate PI3K/Akt signaling pathway,and the pathway is involved in regulating the expression of HIF-1α.

ABSTRACT

Objective To study the effect of hesperidin on the locomotor activity of mice with infective brain injury induced by li-popolysaccharide (LPS)in the dark field environment.Methods 45 ICR mice were randomly divided into 3 groups:Saline group, LPS group and LPS+Hesperidin group,15 mice in each group.The moving distance and the number of crossing line in the dark field were recorded using the ZH-ZFT locomotor activity video analysis system.ResultsThe LPS group had significantly shorter moving distance as well as lower frequency of the number of crossing line than those of the Saline group(P<0.05);compared with the LPS group,the Hesp group moved longer and crossed line more frequently,the differences between the two groups had statisti-cal significance (P<0.05).Conclusion Hesperidin could significantly improve the capacity for action in the dark field of mice with infective brain inj ury induced by LPS.

ABSTRACT

Objective To investigate the therapeutic efficacy of combining Kangfuxin solution with compound Bingpengsan on patients with diabetes mellitus complicated with pressure sores. Methods Fifty-two diabetes mellitus patients complicated with pressure sores admitted in Department of Emergency in the PLA 155th Central Hospital were divided into observation group and control group by lot method,26 cases in each group. Observation group was treated by combining Kangfuxin solution with compound Bingpengsan,the control group was given Mepliex application therapy,they all had change of dressing once a day. After treatment for 20 days,the therapeutic efficacy of pressure sore,healing time and frequency of changing dressing were observed. Results The total effective rate in observation group was significantly higher than that of control group〔96.1%(25/26)vs. 80.6%(21/26),P<0.05〕, in the observation group,the pressure sore healing time was significantly shorter than that of the control group(day:Ⅱstage:9.5±1.7 vs. 13.0±2.1,Ⅲstage:13.1±3.1 vs. 18.1±5.1,Ⅳstage:15.3±3.7 vs. 19.6±5.9,all P<0.05)and the number of times of changing dressing was significantly reduced compared with that of control group (times:Ⅱ stage:16.39±1.89 vs. 19.32±2.26,Ⅲ stage:19.56±2.52 vs. 22.36±2.69,Ⅳ stage:23.54±2.86 vs. 26.47±3.96,all P<0.05). Conclusion The Kangfuxin solution combined with compound Bingpengsan for treatment of deep pressure ulcers in patients with diabetes mellitus has significant effect,its cure rate is relatively high,the pressure sore healing time is reduced and the patients' suffering is alleviated.

ABSTRACT

OBJECTIVE To investigate the nasopharynx carriage status of opportunistic pathogens in non-infectious old people.METHODS The totally of 592 pharyngeal samples from old persons were collected.RESULTS The isolated rates of main opportunistic pathogens were Haemophilus parainfluenzae,Stomatococcus mucilaginosus,Streptococcus pneumoniae and Klebsiella pneumoniaa.The isolated rate of single opportunistic pathogen was 45.8% and that of multiple opportunistic pathogens was 3.9%.The total carriage rate of opportunistic pathogen was 49.7%.The carriage rate of G+ or G-bacteria was 13.2% or 86.8%,respectively.The isolated rates of 2 multiple pathogens,S.pneumoniae and K.pneumoniae were increased with aging.The isolated rate of K.pneumoniae was higher in persons with more than 3 kinds of underlying diseases.CONCLUSIONS Aging,pharyngeal carriaging of S.pneumoniae,H.parainfluenzae,K.pneumoniae,et al and with multiple underlying diseases might be the risk factors for elder people to suffer from infectious respiratory disease.Isolating pharyngeal opportunistic pathogen among non-infection people is necessary and significant for prevention and treatment of infectious respiratory disease.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the feasibility of endothelialization of bioprosthesis by transfer of vascular endothelial growth factor (VEGF) gene.</p><p><b>METHODS</b>Bovine pericardium treated with glutaraldehyde and L-glutamic acid was positioned into the pig right atrium. pcD(2)/hVEGF(121) gene (1 mg) was transferred into the right ventricular myocardium using surgical sutures Reverse transcri ption polymerase chain reaction (RT PCR) was employed to evaluate the expression of myocardial VEGF mRNA. The determination of concentrations of VEGF protein in blood from both the right atrium and peripheral vein, and histological and ultrastructural analysis of implanted bovine pericardium were completed simultaneously.</p><p><b>RESULTS</b>The concentration of VEGF derived from the right atrium in pcD(2)/hVEGF(121) group was significantly higher than that in the pcD(2) group 10 days after VEGF gene transfer (P < 0.01). The expression of myocardial VEGF mRNA in pcD(2)/hVEGF(121) group was much higher in comparison with that in the pcD(2) group. The morphological analysis demonstrated that the coverage rate of host endothelium in the pcD(2)/hVEGF(121) group was 2.6 times as fast as that in the pcD(2) group at 16 days after VEGF(121) gene transfer (P < 0.01). Entire endothelialization occurred at 30 days after VEGF gene transfer. In addition, higher expression of myocardial VEGF mRNA was still available.</p><p><b>CONCLUSIONS</b>VEGF gene transfer by surgical suture can remarkably accelerate endothelialization of bioprosthesis, which may provide a new approach for inhibiting biological valve calcification and improve biocompatibility and long-term durability of the bioprosthesis.</p>

ABSTRACT

ObjectiveTo investigate the clinical manifestati on and pathogen of hospital acquired low respiratory tract infection in elderly patients with diabetes.MethodsTo analyze retrospectively th e clinical data, the pathogens isolated from cultured sputum and the antibiotic sensitivity of bacteria in 85 elderly patients with diabetes.Results Bacilli in sputum were common in these patients(73 9%), including Pse udomonas aeruginosa (18 2%), Klebsiella pneumoniae(15 9%), Escherichia co li(14.8%), 14% complicated with 2 different bacilli, 6.8% with fungus. The bac illi sensitive rate to antibiotics was as high as 72.3% to Ceftazidime, 70.8% to Ofloxacin, 63 1% to Ceftriaxone, 61 5% to Cefoperazone or Cefoperazone, 60 .0% to Cefazolin, 53.8% to Piperacillin respectively. Cocci sensitive rate wa s 100% to Vancomycin. Uncontrolled hyperglycemia, severe basic diseases and dia betic complications were involved in the pathogenesis of hospital acquired low respiratory tract infection and the poor prognosis.Conclusion Hospital acquired low respiratory tract infection is common in elderly patient s with diabetes, and should be treated based on the antibiotic sensitivity of ba cteria and on metabolic control.

ABSTRACT

AIM: To investigate biological effect of endothelin-1 (ET-1) on differentiation of cardiomyogenic cells (CGCs) induced from rabbit bone mesenchymal stem cell (BMSCs) in vitro. METHODS: The BMSCs from shaft of femur of Japan rabbits were isolated and propagated for cell culture. Cultured BMSCs were randomly divided into six groups: Group Ⅰ: the control group (uninduced group); Group Ⅱ: the ET-1 group, added ET-1 (30 nmol/L) into culture medium; Group Ⅲ: 5-aza group, added 5-aza (10 ?mol/L) into culture medium; Group Ⅳ: 5-aza+ET-1 group, after inducing with 5-aza for 3 weeks, ET-1 was added into culture medium. This group was divided into three parts-Ⅳ 1、Ⅳ 2、Ⅳ 3, and separated to add 10、 30、 50 nmol/L ET-1. During the cell culture, growing and differentiation of BMSCs were observed. After inducing for 4 weeks, the differentiation rate and the diameter of cardiomyogenic cells were calculated; Western-blot was employed to analyze the expressions of GATA-4 protein and phosphorylation level. The expression of ?-MHC mRNA was assessed by RT-PCR. Immunohistochemistry staining of Troponin-I and ultrastructure observation of induced cardiomyogenic cells were also completed simultaneously. RESULTS: The cell diameters of CGCs in group Ⅳ 2 were enlarged significantly (P0.05). ABSTRACT

Objective To determine the cellular localization and expression pattern of AR and FSHR in adult rat testis must be helpful to understand the action site and mechanisms that T and FSH regulate spermatogenesis. Methods We applied in situ Hybridization to detect the expression of AR and FSHR on adult testis, in which Dig labeled cRNA probe was used to carry out the experiment on frozen sections; at the same time, following the technique of transillumination assisted microdissection we separated seminiferous epithelium into four stages(Ⅱ Ⅵ,Ⅶ Ⅷ,Ⅸ Ⅻ and ⅩⅢ Ⅰ), extracted total RNA and carried out dot hybridization, using ? 32 P labeled cDNA probe, in order to test qualitatively and quantitatively the location of AR and FSHR mRNA and their expression pattern in adult rat testis. Results Our results showed that the positive signal of AR mRNA was located in Sertoli cells and Leydig cells. The signal in Sertoli cells began to appear in Ⅱ Ⅵ stages, strongest in Ⅶ Ⅷ stages and weakest in Ⅸ Ⅰ stages ( P
