ABSTRACT

Objective To investigate the effect of berberine (BBR) extracted from Chinese Goldthread Rhizome on the expression of claudin-1 in ulcerative colitis mice. Methods UC model was established by dextran sulphate sodium (DSS) . Sixty BALB/c mice were randomly divided into a blank control group, a model control group, a low dose BBR group, a high dose BBR group and a salazosulfapyridine (SASP) positive control group. Then daily gavage administration was given for 7 days. During the course of experiment, the general conditions of mice were observed, and disease activity index (DAI) were assessed. At the end of treatment, colon tissue were dissected, its macroscopic damage index (CMDI) and tissue damage index (TDI) were evaluated, and the contents of claudin-1 protein and mRNA were detected by immunohistochemical method and real-time PCR separately. Results Compared with model control group, the clinical and pathological symptoms of colitis in BBR groups and SASP group were improved to various degrees, the DAI, CMDI and TDI in high dose BBR group and SASP group were all reduced (P<0.01) . The expression levels of claudin-1 mRNA in five groups were 1.00±0.03、 0.12±0.02、 0.21±0.02、 0.63±0.03 and 0.81±0.03, respectively. The expression levels of claudin-1 and its mRNA of colonic tissue in UC mice were all markedly lower than normal mice (P<0.01), which were all increased after BBR or SASP treatment when comparing with blank control group, whose difference was statistically significant. Conclusion Berberine can effectively relieve the colitis in UC mice by raising the expression of claudin-1, but the therapeutic effect was a little weaker than SASP.

Objective To explore the standardized management of high-value medical consumables during medical insurance management.Methods The definition of high-value medical consumables was introduced,and the problems of high-value medical consumables were analyzed in medical insurance management.The standardized management of highvalue medical consumables was discussed from the aspects of admission and purchase management,approval of high-value medical consumables,training on medical insurance policies,construction of high-value medical consumables examination and evaluation.Results Standardized management of high-value medical consumables contributed to slowing down the increase of per capita medical consumables,controlling the amount of medical consumables during hospitalization and increasing the ratio of domestic consumables.Conclusion All the medical facilities have to avoid irrational increase of highvalue medical consumables to utilize medical insurance fund reasonably with involvement of information platform,management regulation,approval flow as well as examination and evaluation system.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate four candidate variable number tandem repeat (VNTR) loci for genotyping Mycobacterium tuberculosis complex strains.</p><p><b>METHODS</b>Genomic sequences for two M. tuberculosis strains (CCDC5079 and CCDC5180) were generated, and using published sequence data, four candidate VNTR loci were identified. The VNTRs were used to genotype 225 Chinese clinical M. tuberculosis complex strains. The discriminatory power of the VNTRs was evaluated using BioNumerics 5.0 software.</p><p><b>RESULTS</b>The Hunter-Gaston Index (HGI) for BJ1, BJ2, BJ3, and BJ4 loci was 0.634, 0.917, 0.697, and 0.910, respectively. Combining all four loci gave an HGI value of 0.995, thus confirming that the genotyping had good discriminatory power. The HGI values for BJ1, BJ2, BJ3, and BJ4, obtained from Beijing family strain genotyping, were 0.447, 0.878, 0.315, and 0.850, respectively. Combining all four loci produced an HGI value of 0.988 for genotyping the Beijing family strains. We observed unique patterns for M. bovis and M. africanum strains from the four loci.</p><p><b>CONCLUSION</b>We have shown that the four VNTR loci can be successfully used for genotyping M. tuberculosis complex strains. Notably, these new loci may provide additional information about Chinese M. tuberculosis isolates than that currently afforded by established VNTR loci typing.</p>
