ABSTRACT

Objective To evaluate the effects of adenovirus-thediated PTEN gene transfer on growth and invasion of H22 cells in vitro. Methods H22 cells were tnmsfected with Ad-PTEN and Ad-PTENG-129R it vitro, The mRNA expressions of PTEN were detected with RT-PCR. Cell viability was determined by MTT assay. Invasion of H22 in vitro was observed using Boyden chamber. Results Gene and protein expressions of PTEN were shown to be up-regulated when H22 was transfected with Ad-PTEN. MTT assay showed that the group transfected with Ad-PTEN had much lower cell viability than other groups (P<0.05) . The number of 1422 invasion cells in the group of Ad-PTEN was (15.64±3.27) %, which was lower than the number in other groups (P<0.05). Conclusion These results imply that the number of H22 invasion cells was significantly decreased after infected with Ad-PTEN in vitro.

ABSTRACT

To investigate the effects of dendritic cells (DCs) transfected with full length wild type p53 and modified by gastric cancer lysates on immune response, the wild type P53 was transducted to DCs with adenovirus, and the DCs were modified by gastric cancer lysates (Lywt-P53DC). The concentration of the surface molecules (B7-1, B7-2, MHC-I , MHC-II) of all DCs was determined by FACS, and the ability of the DCs to induce efficient and specific immunological response in anti-51Cr-labeled target cells studied. BALB/c mice model infected with DCs and Mk28 was established. CTL response in mice immunized with Lywt-p53DC and the effectiveness of Lywt-p53DC in the treatment of tumor-bearing mice was assayed. FACS revealed that the surface molecules of Ly-wt-P53 DC had a high expression: for B7-1 86.70% +/- 0.07%, B7-2 18.77% +/- 0.08%, MHC-I 87.20% +/- 0.05%, MHC-II 56.70%+/-0.07%; The T lymphocytes had a specific CTL lysing ability induced by Lywt-P53DC with the CTL lysis rate being 81%. The immune protective effect of Lywt-p53DC group was more obvious than any other groups (P<0.05). The tumor diameter in Lywt-p53DC group was 3.10+/-0.31 mm, 2.73+/-0.23 mm, 3.70+/-0.07 mm on the day 13, 16 and 19, smaller than DC, wtp53DC and LyDC groups (P<0.05). On the other hand, the growth rate of tumor in Lywt-p53DC group was slower than any other groups (P<0.05). It was suggested that DCs transfected with wild type P53 and modified by gastric cancer lysates had specific CTL killing capability.

ABSTRACT

Objective To determine the effective prognostic parameter and the best prognostic index of gastiric carcinoma. Methods The prognostic relevance of clinical and pathological variables were evaluated in 83 patients with histologically proved stomach carcinoma (including the expression of nm23, c erbB2, microbloodvessel and PTNM) by the multivariate analysis of COX regression.Result PTNM staging was the only parameter to enter the COX model. Conclusion The PTNM staging is the most important, reliable and best varible factor in predicting the clinical outcome of stomach cancer.

ABSTRACT

Objective To investigate the expression of Notch1 and its ligand DLL4 in human gastric carcinoma tissues and its correlation with tumor angiogenic metastasis. Methods Immunohistochemical SP method was used to detect the expression of Notch1,DLL4 and VEGF in 45 gastric carcinoma tissues and paired adjacent normal gastric mucosa, and the relationship between them and clinico-pathological parameters were analyzed. Results The positive expression rate of Notch1,DLL4 and VEGF in gastric carcinoma were higher than that in normal gastric mucosa(P

ABSTRACT

Objective To study the effects of 5-aza-2-deoxycytidine (ZdCyd), a methylation inhibitor, on the growth of bile duct cancer cell line QBC939 in vivo and in vitro. MethodMTT method and flow cytometry were used to detect the growth and apoptosis of QBC939 after being treated with different dosage of ZdCyd singly or in combination with other chemotherapy drugs. Nude mouse model was used to test the effect of ZdCyd. Results5-aza-2-deoxycytidine blocks cell cycle at G1 phase increasing apoptosis rate. These effects within certain extent are dose and time dependent.5-aza-2-deoxycytidine also coordinates with other chemotherapy drugs in the anti-tumor effects. In nude mouse model the tumor occurrence rate decreased in 5-aza-2-deoxycytidine pro-treated cells of QBC939, and ZdCyd also impedes the growth of transplanted tumor. Conclusion 5-aza-2-deoxycytidine inhibits the growth of QBC939 in vivo and in vitro by inducing cell apoptosis and enhances the antitumor effect of chemotherapy drugs.
