ABSTRACT

Objective:To identify the morphological alterations in the Golgi apparatus of skin fibroblasts in spinocerebellar ataxia type 3 (SCA3) patients.Methods:In this study, 3 SCA3 patients and 3 healthy volunteers were obtained in the First Affiliated Hospital of Zhengzhou University from 2016 to 2020. The cytosine, adenine, and guanine repeats of 3 SCA3 patients were 14/76, 20/80 and 21/82, respectively. Tissue mass culture was used to amplify skin fibroblasts derived from SCA3 patients and healthy volunteers. Cell viability and apoptosis were detected using cell counting kit-8 assay and flow cytometry, respectively. Western blotting and immunofluorescence assay were used to detect the protein expression of ataxin-3, Golgi reassembly stacking protein 2 (GORASP2), and Golgi matrix protein 130 (GM130) in the skin fibroblasts. The morphology of the Golgi apparatus in skin fibroblasts was detected using transmission electron microscopy.Results:Tissue culture of skin fibroblasts of both SCA3 patients and healthy volunteers was successfully established. The patient-derived dermal fibroblasts expressing mutant ataxin-3 protein exhibited reduced cell viability ( t=5.06,P=0.007), increased apoptosis ( t=3.77, P=0.020), fragmentation of the Golgi apparatus, increased expression of GM130 ( t=5.23, P=0.006), and decreased expression of GORASP2 ( t=4.35, P=0.012). Transmission electron microscopy revealed that the Golgi apparatus was disorganized in skin fibroblasts. Conclusion:Fragmentation of the Golgi apparatus occurs in the skin fibroblasts of SCA3 patients, and abnormal morphology and structure of the Golgi apparatus may be involved in the pathogenesis of SCA3.

ABSTRACT

This study was aimed to observe the level of T helper 17 [Th17] cells and CD4[+] CD25[+] Foxp[3+] regulatory T cells [Tregs] and their related factors in carotid atherosclerosis [AS] patients and AS model rats to explore the influence of Th17 on the pathological process of AS and its specific mechanism. 60 cases with AS in our hospital from July 2012 to July 2014 were recruited for this study as the observation group, and 40 healthy people who came to the hospital for a physical examination were the control group. Flow cytometry [FCM] was used to detect the Th17 and Treg cells in the peripheral blood of the two groups, ELISA was used to detect IL-17 and transforming growth factor-beta [TGF-beta], and RTPCR was used to test the ROR gamma T mRNA and Foxp3 mRNA expression levels. An AS model was created in rats using high-fat+ VD[3] to explore the mechanism of Th17 on AS. The Th17 count, serum level of IL-17, and ROR gamma T mRNA level of the observation group were significantly higher than those of the control group [P<0.001]. The Tregs count, serum TGF-beta level, and Foxp3 mRNA level were significantly lower in the observation group than in the control group [P<0.001]. In addition, the findings of the AS model in rats showed that the Th17 cell count and serum level of IL-17 in high-fat rats were significantly higher than in the normal rats [P<0.05]. The Treg count and TGF-beta levels of the observation rats were significantly lower than in the normal rats [P<0.05]. The IL-17 level, serum total cholesterol, and triglyceride in the high-fat-feed rats decreased after being injected with the IL-17 neutralizing antibody, but TGF-beta levels increased, and the difference was significant [P<0.05]. Th17 cells and their related factors can be involved in promoting the pathological process of AS, while Tregs and its related factors can be involved in the inhibition of AS. Blocking IL-17 can be one potential method of treating AS

ABSTRACT

To evaluate the diagnosis significance of single high-frequency ultrasonography and mammography and combination therapy of both on breast cancer. 352 cases of female breast cancer patients were selected from The First Affiliated Hospital of Zhengzhou University from January 2012 to December 2014. Among the 352 patients, 124 patients had only performed high-frequency ultrasonography detection, 102 cases of patients were only conducted mammography, and 126 patients had applied the combination detection of high-frequency ultrasonography and mammography. The coincidence rate of single mammography detection was 79.4%, the misdiagnosis rate was 10.8%, and the missed diagnosis rate was 9.8%; the coincidence rate of single high frequency ultrasonography detection was 83.9%, the misdiagnosis rate was 11.5%, the missed diagnosis rate was 4.6%; the coincidence rate of combination of high frequency ultrasonography detection was 89.7%, the misdiagnosis rate was 6.3%, the missed diagnosis rate was 4.0%. The detection rate and missed diagnosis rate of combination diagnosis had statistical difference with single high frequency ultrasonography and single mammography. There was no statistical difference on misdiagnosis rate. mammography and high frequency ultrasonography respectively had their own advantages. The combination application of both had better diagnosis complementary, and could significantly improved the detection rate and accuracy rate on breast cancer, and decreased the misdiagnosis rate and missed diagnosis rate

ABSTRACT

Objective To observe the influence of individual behavior intervention on emotion and quality of life of elderly females with stress urinary incontinence and provide guidance for health intervention.Methods 124 elderly females with stress urinary incontinence were selected.Individual behavior intervention was used,and after 1,3,6 months,the effect was evaluated by anxiety,depression,ICIQ -FLUTS,UDI -7,and SCL -90.Results After the individual behavior intervention,anxiety and depression scores became lower[anxiety scores:before intervention (59.16 ±6.12)points,1month (52.68 ±5.62)points,3months (48.28 ±5.32)points,6months (39.86 ± 4.27)points,there were statistically significant differences between the two groups(F =298.69,P =0.000).Depres-sion scores:before intervention (59.16 ±6.12)points,1 month (52.68 ±5.62)points,3 months (48.28 ± 5.32)points,6 months (39.86 ±4.27)points,there were statistically significant differences between the two groups (F =252.10,P =0.000)].ICIQ -FLUTS and UDI -7 scores became less[ICIQ -FLUTS scores:before interven-tion (31.79 ±3.74)points,1 month (29.62 ±3.18)points,3 months (27.54 ±2.67)points,6 months (23.32 ± 2.16)points,there were statistically significant differences between the two groups(F =161.15,P =0.000).UDI -7 scores:before intervention (19.29 ±1.76)points,1 month (16.09 ±1.48)points,3 months (14.32 ±1.37)points, 6 months (11.57 ±1.26)points,there were statistically significant differences between the two groups(F =531.23, P =0.000)].There were significant differences in somatization,obsession,sensitivity of interpersonal relationship, depression,anxiety,hostility,fear,crankiness and psychotic diseases(somatization scores:F =18.933,P =0.000;obsession:F =5.153,P =0.000;sensitivity of interpersonal relationship:F =3.314,P =0.020;depression:F =9.306,P =0.000;anxiety:F =4.902,P =0.002;hostility:F =6.852,P =0.000;fear:F =7.782,P =0.000;cranki-ness:F =11.917,P =0.000;psychotic diseases:F =8.560,P =0.000).Conclusion After the individual behavior intervention,anxiety,depression,ICIQ -FLUTS,UDI -7,and scl -90 scores become lower.Individual behavior inter-vention has a positive effect on emotional states and quality of life,which is worthy of clinical promotion.

ABSTRACT

Objective To observe the cognitive function and its related factors in patients with essential epilepsy.Methods The cognitive function of 70 essential epilepsy patients (epilepsy group) and 40 healthy controls(control group) were evaluated by means of Wechsler Intelligence Scale for Adult-Chinese (WAIS-RC).The relation between the cognitive dysfunction and its related factors were analyzed.Results The scores of performance intelligence quotient,verbal intelligence quotient,full intelligence quotient in epilepsy group were significantly lower than those in control group [(98.06 ± 15.24) scores vs.(113.80 ± 12.14) scores,(98.09 ± 16.06) scores vs.(120.65 ± 11.28) scores,(98.06 ± 15.80) scores vs.(119.42 ± 11.85) scores] (P < 0.01).The scores of 11 numbers of rating scales were significantly lower than those in control group (P < 0.01).The factors related to the cognitive function were education level,age numbers,duration of the disease,frequency of seizures attack before medication,the duration of seizures and the quantity of antiepileptics.Conclusions Many of the essential epilepsy patients have cognitive function deficit.The cognitive condition of essential epilepsy patients should be pay more attention and reduce the dangerous factors in order to improve the life quality.

ABSTRACT

Objective To study the protein expressions of Fas-associated death domain protein (FADD) and caspase-8 in rats with intracerebral hemorrhage ,and the effects of erythropoietin tp reveal the mechanism of neu-m-protection by EPO. Methods 126 male SD rats were randomly divided into three groups: Sham-operated group, intracerebral hemorrhage group, and EPO group. Each group was divided into seven subgroups according to the differ-ent time points (3,6,12,24,48,72 h and 7 d). The model of intracerebral hemorrage was established in rats by in-tracerebral injection of autogenous blood. The protein expressions of FADD and caspas-8 in rats tissue around the hemorrhagic and the normal brain tissue were detected by immunohistochemistry. Results The protein expressions of FADD and caspase-8 were increased [(4.66±0.46 ) and ( 15.89±1.81)] at 3 h after intracerebral hemorrhage, and peaked at 48 h [ (35.88±4.24 ) and (45.04±3.99)], the expressions of FADD and caspas-8 in the region around hematoma in EPO group significantly decreased compared with model group[ (3.92±0.64) and (28.24±1.90), (13.32±2.01 ) and (35.08±2.82)] at 3 h and 48 h. Conclusion The protein expressions of FADD and easpase-8 are markedly increased after intracerebral hemorrhage. EPO can protect the neurons by signifi-cantly reducing the expressions of FADD and caspase-8.

ABSTRACT

@#ObjectiveTo explore the related factors influencing effect of community-based rehabilitation (CBR) on activities of daily living (ADL) in stroke patients in.Methods202 stroke patients were randomly divided into the CBR group (n=103) and control group (n=99). The patients of the CBR group treated with rehabilitation and following-up including risk factors controlled with drug, rehabilitation, health education and mental treatment, those of the control group only treated by following-up. Before and five months after treatment, patients of two groups were assessed with Barthel Index (BI), clinical never function limitation score, cognitive item of Functional Comprehensive Assessment (FCA). Multielement regression analysis was applied, the ADL score of last assessment was taken as dependent variable, group information, location, smoking, sex, age, course of diseases, education, drinking, sleeping, the first scores of FCA and BI were taken as independent variable.ResultsThe improvement of ADL of stroke patients was related with group information, drinking, course of diseases, the first scores of FCA, BI, and etc.ConclusionEarly CBR can significantly improve the ADL of the stroke patients. Cognitive deficit also has influence on ADL.
