ABSTRACT

@#The chemical constituents of solid rice culture of the endophytic fungus Aspergillus sp.Dq-25 from barnacle were isolated and purified by silica gel, Sephadex LH-20, C18 reversed silica gel column chromatography and recrystallization.Their structures were identified by the physical and chemical properties, and by various spectroscopic methods.Six compounds were isolated and identified as: demethyldihydropenicillic acid (1), dihydropenicillic acid (2), penicillic acid (3), fortisterol (4), 22E, 24R-3P, 5a-dihydroxyerogosta-7, 22-diene-6-one (5), and (22E, 24R)-ergosterol-7, 22-diene-3β, 5α, 9α-triol-6-one (6).Compound 1 was a new butyrolactone.MTT method was used to analyze cytotoxicity, and the result showed that compound 3 exhibited inhibitory activity on five cell lines, including K562, HeLa, SGC-7901, A542 and BEL-7402, with IC50 values of 38.0 ~ 105.0 μmol/L.

ABSTRACT

Objective:To investigate the safety and efficacy of nimotuzumab combined with IMRT radiotherapy in the treatment of elderly patients with locally advanced cervical cancer (LACC).Methods:A retrospective analysis was conducted for 34 elderly LACC patients treated with nimotuzumab combined with IMRT radiotherapy or concurrent chemoradiotherapy in the Zhangzhou Affiliated Hospital of Fujian Medical University from June 2020 to December 2021. The efficacy and side effects were evaluated one and two years after treatment.Results:Median follow-up time was 13.3 months (6.1-24.3 months). A total of 24 cases of complete response (CR) and eight cases of partial response (PR) were achieved after treatment, with an objective response rate (ORR) of 94.1% (32/34). The tumor diameters were (49.56 ± 19.22) mm before treatment and (19.61 ± 14.59) mm after treatment, with a tumor regression rate (TRR) of 59.22%.The 1- and 2- year progression-free survival (PFS) rates were 84.9% and 84.9%, overall survival (OS) rates 91.8% and 87.2%, respectively, disease-free survival (DFS) rates 91.8% and 87.2%, respectively, and the cancer-specific survival (CSS) rates 95.7% and 90.9%, respectively. The main adverse events included radiation enteritis, leukopenia, hypoproteinemia and anemia.Conclusions:Nimotuzumab combined with IMRT radiotherapy or concurrent chemoradiotherapy is safe and effective in the treatment of LACC.

ABSTRACT

Objective:To explore the effect of a Chinese medicinal composition ( Xiadanqi) on the prevention of radon exposure induced injuries of lung in vitro and in vivo. Methods:Mice were randomly divided into three groups of blank control group, radon-exposed group alone and radon-exposed group intervened with Chinese medicinal composition. The pathological changes of lung tissues in each group after 120 WLM were observed by HE and Masson staining, and the expressions of α-SMA protein and Vimentin protein in lung tissues were detected by immunohistochemistry staining. The levels of oxidative stress in lung tissue of each group were detected with SOD and MDA kits. At the same time, a radon exposed cell model and a radon exposure + Xiadanqi intervention cell model were constructed using an ecological radon chamber. The cell adhesion abilities of different groups were detected by an adhesion kit. The cell migration ability of each group was determined by the transwell migration experiment. The expression of E-cadherin and Vimentin protein was detected by Western blot. Results:Compared with the radon exposure group, the concentration of MDA was decreased ( t=4.43, P<0.05), the activity of SOD was increased ( t=3.22, P<0.05), and α-SMA and Vimentin protein expressions were decreased ( t=3.08, 7.57, P<0.05) in lung tissue of mice intervened with 2 mg/g Xiadanqi. In vitro, compared with radon exposure group, the migration ability was reduced ( t=4.78, 13.01, P<0.05), the cell adhesion property was enhanced ( t=3.41, 12.55, P<0.05), the expression of E-cadherin protein was increased ( t=2.96, 19.57, P<0.05), and the expression of Vimentin protein was obviously reduced ( t=21.00, 33.32, P<0.05) in radon-exposed cells with the treatment of Chinese medicine (150 μg/ml and 200 μg/ml). Conclusions:The Chinese medicinal composition ( Xiadanqi) has a certain radioprotective effect on radon exposure induced injury by reducing oxidative stress, attenuating EMT and fibrosis, and thus it may be applied as a protective agent for radon induced injury.

ABSTRACT

OBJECTIVE:To compare the chemical compo sition difference in methanol and petroleum ether fraction from Curcuma longa of different habitats. METHODS :The ultrasonic method was used to extract C. longa from 7 defferent producingareas(S1-S7),and methanol and petroleum ether fraction were obtained and calculated yield. The curcumin compounds in methanol fraction were determined by LC-MS ;The chemical components in petroleum ether fraction were analyzed by GC-MS , and the relative percentage content was determined by peak area normalization method after determining its structure by comparing NIST 2005 standard mass spectra and Wiley 275 standard mass spectra. SPSS 25.0 software was used for principle component analysis(PCA)and cluster analysis of relative percentage content of common components in petroleum ether fraction from C. longa of different habitats. At the same time ,the influence of latitude of the habitats on the content of total tumerone (by tumerone and ar-tumerone )was analyzed. RESULTS :The yield of methanol fraction were 1.35%-8.90% from C. longa of 7 habitats;the yield of petroleum ether fraction were 0.81%-4.90%,which were the highest in C. longa from Longyan of Fujian Province. There was no significant difference in the relative content of curcumin compounds(reference peak area )from S 1,S3-S7,which was in descending order as follows as curcumin >desmethoxycurcumin>bisdemethoxycurcumin. There was slightly different in curcumin compounds of C. longa from S 2,mainly manifesting as the content of bisdemethoxycurcumin was higher than that from other producing areas. Totally 48 chemical compositions were identified from petroleum ether fraction in C. longa from different habitats , mainly being sesquiterpenoids and monoterpenoids. 23,10,15,18,11,14,15 chemical compositions were identified from S1-S7,accounting for 94.49%,96.09%,95.66%,98.98%,99.24%,89.05% and 97.27%. There were 4 common compositions in C. longa from different habitats ,which were tumerone (17.90%-43.07%),ar-tumerone(6.97%-33.66%),(6R,7R)-bisabolone (1.60%-4.28%),curlone(6.80%-20.63%). PCA analysis showed that accumulative contribution rate of former 6 principle components was 100%. Cluster analysis showed that S1,S2, S6 was clustered into a category ,respecrively;and others intoa category. Total content of total tumerone decreased first and then increased as the increase of latitude ,which was the highest in Mianyang of Sichuan province (64.28%)and the lowest in Zhangzhou of Fujian province (26.92%). CONCLUSIONS : There are difference in composition and content of methanol and petroleum ether fractions in C. longa from different habitats.

ABSTRACT

OBJECTIVE: To study on the effects of prophylactic administration of Ramulus mori polysaccharides (RMP) on inflammatory response of renal ischemia reperfusion injury (RIRI) model mice and to explore its possible mechanism. METHODS: Totally 60 C57BL/6 mice were randomly divided to sham operation group, model group, atorvastatin group (positive control, 15 mg/kg), RMP low-dose, medium-dose and high-dose groups (300, 600, 1 200 mg/kg). Except for sham operation group, RIRI model was induced in other 5 groups. 24 h before surgery, they were given relevant medicine intragastrically, once a day, for consecutive one week. 24 h after reperfusion, the mice were sacrificed. The serum levels of Scr and BUN were detected. The morphological changes of renal tissue were observed under optical microscope. The serum levels of IL-1β, IL-6, IL-10 and TNF-α were determined by ELISA. Western blot assay was used to determine the protein expressions of Toll-like receptor 4 (TLR4), p38 mitogen-activation protein kinase (p38MAPK) and p-p38MAPK. RESULTS: Compared with sham operation group, the serum levels of Scr and BUN were significantly elevated in model group  (P<0.01). RIRI led to typical inflammatory response of renal tissue, widespread renal tubular epithelial cell degeneration and necrosis, and inflammatory cells infiltration. Serum levels of IL-1β, IL-6, IL-10 and TNF-α were increased significantly (P<0.01). The protein expressions of TLR4, p38MAPK and p-p38MAPK were increased significantly in renal cortex (P<0.01). Compared with model group, serum levels of Scr and BUN were decreased significantly in administration groups (P<0.05 or P<0.01). The pathological damage of renal tissue was improved in varying degrees, especially in the RMP medium-dose and high-dose groups. Serum levels of IL-1β and IL-6 were decreased significantly in administration groups (P<0.05 or P<0.01). Serum levels of IL-10 were further increased in atorvastatin group and RMP high-dose group (P<0.01), and serum level of TNF-α was decreased significantly in atorvastatin group and RMP medium-dose and high-dose groups (P<0.05 or P<0.01). The protein expressions of TLR4 and p-p38MAPK in renal cortex were decreased significantly in administration groups (P<0.05 or P<0.01). CONCLUSIONS: RMP prophylactic administration can improve RIRI of mice, the mechanism of which may be associated with relieving the inflammatory response through inhibition of TLR4/p38MAPK signaling pathway.

ABSTRACT

OBJECTIVE: To isolate and identify the coumarins from the seeds of Clausena lansium, and to study their inhibitory activity of α-glucosidase and nematicidal activity against Panagrellus redivivus. METHODS: Column chromatography, reversed phase silica gel column chromatography and HPLC method were used to separate and purify the coumarins from the seeds of C. lansium. The structures of compounds were identified according to physicochemical properties, 1H-NMR and 13C-NMR spectral data. Using acarbose and avermectin as positive control, PNPG and Berman funnel methods were used to investigate the α-glucosidase inhibitory activity and nematicidal activity against P. redivivus, respectively. RESULTS: Seven coumarins compounds were isolated from the seeds of C. lansium, and were identified as 7-hydroxy-1-benzopiran-2-one (Ⅰ), Wampetin (Ⅱ), Lansiumarin-C (Ⅲ), Claucoumarin A (Ⅳ), Clausenalansimin A (Ⅴ), (E,E)-8-(7-hydroxy-3,7-dimethylocta-2,5-dienyloxy) psoralen (Ⅵ), Dihydroindicolactone (Ⅶ). Under 0.25 mg/mL, the α-glucosidase inhibitory rates of compounds Ⅰ, Ⅲ, Ⅴ were (32.4±1.9)%,(37.1±6.0)%, (39.5±1.1)%, respectively. Under 2.5 mg/mL, corrected mortality of compounds Ⅰ, Ⅳwere 50.5% and 47.9%. CONCLUSIONS: Compounds Ⅰ, Ⅲ, Ⅴ show α-glucosidase inhibitory activity, and compounds Ⅰ,Ⅳ display nematicidal activity against P. redivivus.  α-Glucosidase inhibitory activity of compounds Ⅲ, Ⅴ, and nematicidal activity of compound Ⅳ are found for the first time.

ABSTRACT

OBJECTIVE: To analyze the difference of volatile oil components from the leaves of Clausena lansium and Clausena excavata. METHODS: The volatile oil was extracted from the leaves of C. lansium and C. excavata by steam distillation. GC-MS method was adopted to analyze volatile oil to obtain TIC. After mass spectra scanning of the chromatographic peaks in the TIC diagram by HPMSD chemical workstation, chemical components of volatile oil in 2 kinds of samples were identified by retrieving and comparing mass spectrum database NIST Version 1.7. The peak area normalization method was used to calculate the relative mass fraction of each component. RESULTS: A total of 43 and 31 kinds of components were identified in volatile oil from the leaves of C. lansium and C. excavata; total relative mass fractions were 97.59% and 98.57%. Relative mass fractions of 19 and 18 components in volatile oil from the leaves of C. lansium and C. excavata were more than 1%, mainly being sesquiterpenoids. Relative mass fractions of 7 and 5 components in volatile oil from the leaves of C. lansium and C. excavata were more than 5%; the volatile components in volatile oil from the leaves of C. lansium were mainly (-)-spatol (12.35%) and (E)-5-{(1R,3R,6S)-2,3-dimethyltricyclic [2.2.1.02,6] heptane-3-yl}-2-methyl pentane-2-enol (14.70%); those from the leaves of C. excavata were mainly (E)-sesquihydrated betuline (24.94%) and 1-(1, 5-dimethy-4-hexenyl)-4-methyl-benzene (16.15%). A total of 4 components were found in volatile oil from the leaves of C. lansium and C. excavata, mainly being α-humulene, (E)-5-{(1R,3R,6S)-2,3-dimethyltricyclic [2.2.1.02,6] heptane-3-yl}-2-methylpentaeryl-2-enol, caryophyllene oxide and (-)-spatol; the content differences of them were not significant. CONCLUSIONS: The components of volatile oils from the leaves of C. lansium and C. excavata are basically similar However, the composition and comtent of specific components are quiet different and can not substituted for each other.

ABSTRACT

OBJECTIVE:To study the protective effect of fingolimod on renal ischemia reperfusion injury (RIRI) model mice and its mechanism.METHODS:A total of 60 mice were randomly divided into sham operation group,model group,fingolimod group (1 mg/kg) and fingolimod+wortmannin group [fingolimod 1 mg/kg+phosphatidylinositol 3-kinase (PI3K) specific blocker wortmarmin 1.4 mg/kg],with 15 mice in each group.Except for sham operation group,RIRI model was induced in other 3 groups,and those model mice were given relevant medicine via caudal vein at once 24 h before surgery.Serum of mice were collected in each group after 24 h perfusion.Serum levels of Scr and BUN were measured by automatic biochemical analyzer.The pathological changes of renal tissue were observed under light microscope.The protein expression of intercellular cell adhesion molecule-1 (ICAM-1),monocyte chemoattractant protein-1 (MCP-1) and phosphorylated protein kinase B (p-Akt) in renal tissue were measured by Western blot assay.RESULTS:Compared with sham operation group,the serum levels of Scr and BUN in model group were increased significantly (P<0.01).Pathological changes were found in the kidney,and RIRI led to widespread renal tubular epithelial cell injury,apoptosis and inflammatory cells infiltration.The protein expression of ICAM-1 and MCP-1 in renal tissue were increased significantly (P<0.01),the protein expression of p-Akt was increased slightly (P>0.05).Compared with model group,other indexes of fingolimod group were improved significantly (P<0.01) except that the protein expression of p-Akt in renal tissue was increased significantly (P<0.01).Compared with fingolimod group,above indexes of fingolimod+wortmannin group were reversed (P<0.05 or P<0.01).CONCLUSIONS:Fingolimod can obviously ameliorate renal injury induced by RIRI in mice,the mechanism of which may be associated with the activation of PI3K/Akt signaling pathway.

ABSTRACT

Objective@#To investigate the potential clinical role of nitric oxide (NO) and endothelial nitric oxide synthase (eNOS) in infants suffering from infantile cytomegalovirus hepatitis (ICH).@*Methods@#NO and eNOS were measured in 56 ICH infants and 50 healthy controls by using ELISAmethod .@*Results@#NO and eNOS were significantly increased in ICH group as compared with the controls (P<0.01) accompanied by increased ALT in infants of ICH group (P<0.05). Both of NO and eNOS levels were significantly positively correlated with the level of ALT(r=0.682, r=0.746, P<0.05). However, neither NO level nor eNOS level was significantly correlated with the number of copies of HCMV-DNA (r=-0.087, r=-0.134, P>0.05).@*Conclusions@#The expressions of NO and eNOS were significantly increased in infants with human cytomegalovirus hepatitis, and were closely related to the severity of liver lesion.

ABSTRACT

Background and purpose:Currently, subjective questionaire is the most frequently used methods to evaluate swallowing dysfunctions after radiotherapy in nasopharyngeal carcinoma patients, while lacking of effective objective examinations. This study aimed to explore effective methods to evaluate swallowing dysfunctions after radiotherapy in nasopharyngeal carcinoma patients, and gain knowledge of the incidence and severity of swallowing dysfunctions. Methods: From Oct. 2013 to Dec. 2013, 128 consecutive outpatients with previously treated nasopharyngeal carcinoma received esophageal barium lfuoroscopy examination at there regularly follow-ups to evaluate swallowing function. Among these patients, 89 were primary treated with intensity modulated radiation therapy (IMRT) and 39 with conventional radiotherapy (CRT). In this study, each patient received esophageal barium lfuoroscopy examination for 3 times with thin, thick and pasty barium and were dynamically observed using X-ray fluoroscopy from front and lateral direction. Swallowing dysfunctions were defined as follows:①The bolus could not be swallowed and blocked in the mouth;②The dilute barium diverted to the glottis or trachea;③Residual barium delayed in the pyriform sinus and vallecula;④The movement of the hyoid bone or epiglottis were restricted;⑤Bolus prolong through the pharynx;⑥Barium slowed down when went though the esophageal entrance. Results:Of the 128 patients, incidence of dysphagia was 60.2%for the entire cohort, 52.8%for IMRT group and 76.9%for CRT group. Incidence of dysphagia for IMRT group was signiifcantly lower than CRT group (P=0.018). Dysphagia incidence within 1 year, 1 to 2 years and more than 2 years after RT were 63.1%, 33.3%and 69.0%, respectively (P=0.019). Conclusion:There was a high incidence of swallowing dysfunction for the nasopharyngeal carcinoma patients treated with radiotherapy and dysphagia incidence decreased when treated with IMRT. Esophageal barium lfuoroscopy examination is objective method to evaluate the incidence and severity of the swallowing dysfunction.

ABSTRACT

Objective: To explore the effects of monocrotaline (MCT) on right ventricular function and expression of cardiac canonical transient receptor potential channels (TRPC) subfamily in experimental rats. Methods: The SD male rats were randomly divided into 2 groups:Control group, the rats were normally fed and MCT group, the rats received a single dose injection of MCT 60 mg/kg to induce myocardial hypertrophy. n=10 in each group and all animals were treated for 3 weeks. The right ventricular hemodynamics parameters and right ventricular hypertrophy index (RVHI) were measured, right ventricular myocardium tissue section was observed by HE staining, the mRNA and protein expressions of TRPC subfamily were examined by RT-PCR and Western blot analysis. Results: Compared with Control group, MCT group had increased RVSP, RVHI, RV+dp/dtmax and decreased RV-dp/dtmax, all P Conclusion:Right ventricular hypertrophy could be induced by 3 weeks MCT treatment, it up-regulating the mRNA and protein expressions of TRPC6 which might be involved in the occurrence and development of cardiac hypertrophy in experimental rats.

ABSTRACT

Objective To explore the effect of Fas, Fas ligand (FasL), soluble Fas (sFas) and their clinical significance in KD. Methods The expression of Fas, FasL in peripheral blood lymphocytes (PBLC) were detected with flow cytometery at acute and remission stages in patients with KD; and the serums Fas was detected by double antibody sandwich ELISA in the patients with KD at acute and remission stage, meanwhile erythrocyte sedimentation rate (ESR), C-reactive protein (CRP) were also tested. Results The expression of Fas, FasL in PBLC in patients with KD at acute stage was (14.2±0.5)% and (1.61±0.09)% respectively , which were significantly lower than those at remission stage [(15.7±0.5)%, (1.95±0.09)% respectively (P<0.05 and P<0.01)]. The expression of Fas in PBLC in the patients with KD at acute and remission stage was both significantly lower than that in normal control group (20.8±0.5)% (P<0.01 both);The expression of FasL in PBLC in patients with KD at acute and remission stage was both significantly lower than that in normal control group (20.8±0.5)% (P<0.01 both); the serum sFas in patients with KD at acute and remission stage was (1906±55)μg/L and (1622±52)μg/L respectively , which was significantly higher than that in normal control group (1151±51)μg/L (P<0.01 both); the serum sFas at acute stage was obviously higher than that at remission stage (P<0.01); there was positive correlation between sFas and ESR, CRP (P<0.01 both). Conclusion There are abnormal expressions of Fas/FasL in PBLC and sFas in patients with KD. Fas/FasL is lower and sFas is higher than that of the controls. The abnormal expression of Fas/ FasL in lymphocytes and the apoptosis triggered by sFas are probably involved in the immunological aberrance and pathogenesis of KD. sFas may be used as a marker to evaluate the disease activity and therapeutic efficacy.

ABSTRACT

Objective To investigate the serum amyloid A (SAA) and interleukin-18(IL-18)concentration in the pathogenesis of type 2 diabetes mellitus (T2DM) and its macrovascular complications, and study the relation between them. Methods ELISA was used to assay serum SAA and IL-18 levels in 65 T2DM patients (including 31 cases with macrovascular complications) and 30 healthy controls. Results Serum SAA and IL-18 levels [(3.09±0.96)mg/L, (98.8±36.4)ng/L]were significantly elevated in patients with T2DM as compared with those in control subjects [(1.06±0.45)mg/L, (58.9±15.6)ng/L](P<0.05). There was significant difference of SAA and IL-18 levels between T2DM patients with [(6.34±1.52) mg/L,(141.2±48.3)ng/L]and without macrovascukar complications [(2.65±0.39)mg/L, (80.2±20.1)ng/L](P < 0.05).Univariate linear regression analysis showed significant positive correlations between serum IL-18 with SAA (r =0.615, P<0.05), SAA, IL-18 and fasting blood glucose (FBG) had mutual positive correlations (r=0.312, 0.428, P< 0.05, respectively). Conclusions In patients with T2DM, serum SAA and IL-18 concentration is greater than in non-diabetic subjects. SAA and IL-18 play important roles in the initiation and development of T2DM. The study suggests that SAA and IL-18 might be an important independent risk factor.

ABSTRACT

Objective To study the characteristics of child acute lymphocytic leukemia (ALL) immuno-phenotype and evaluate its diagnosis value. Methods Direct immunofluorescence staining and CD45/Side Scatter (SSC) gating of flow cytometry were used for immunophenotyping in 111 cases of child ALL. The relation of mor-phology and immunology classification was analyzed. Results Three categories could be identified,including 81 ca-ses (73.0%) of B lineage ALL, 16 cases (14.4%) of T lineage ALL and 14 cases (12.6% ) of B/T lineage ALL. There were 25 cases (22.5% ) of ALL expressing myeloid-associated antigens. According to the FAB Morphology classification,59 cases (53.2%) of L1 type and 47 cases (42.3%) of L2 type were diagnosed. The two cases (1.8%) of L3 type were classified as one case of null-ALL and one case of B-ALL. One case (0.9%)of acute my-eloblastic leukemia (AML-M2a) was identified as null-ALL. The two cases that could not be diagnosed by FAB clas-sification were c-ALL. Conclusion The immunophenotyping helps to identify the character of leukemia with an im-portant value in diagnosis of child ALL.

Objective To explore the changes of sTfR and Tf in serum in children of acute infection with iron deficiency,and their significance in diagnosing. Methods To detect the level of sTfR, Tf and SF in serum by velocity scatter immune turbidimetry, volunteers were divided into groups as IDAI before and after treatment,IDA and the healthy controlled.While iron in bone marrow was observed by iron staining. Results The sTfR and Tf levels in serum in IDAI and IDA groups were significantly higher than that in the controlled group(P0.05).After the treatment,Hb in IDAI group,though was still abnormal comparing with the controlled group(P0.05) .Conclusions sTfR and Tf in serum were specific and sensitive in diagnosing the iron deficiency, and the results would not change if infected.So they are reliable indexes in diagnosing iron deficiency and monitoring the treating effect in the children of anemia with infection.Therefore the two indexes should be higher valued than SF or Iron in the bone marrow.

ABSTRACT

AIM:To explore the supportive and expansive effects of aorta-gonad-mesonephros(AGM) region derived stromal cells on hematopoietic stem cells(HSC) in vitro.METHODS:The murine stromal cells were separated and cultured from AGM region of a 11 day postcoitum(dpc) mouse embryo and 6 week mouse.After identification by Wright's staining and flow cytometry,the stromal cells were co-cultured with the embryonic stem cell(ESC)-derived,cytokine-induced HSCs,and the maintenance and expansion of HSCs were evaluated by detecting CD34+,CD34+Sca-1+cells with flow cytometry.Blast colony-forming cell(BL-CFC) and high proliferative potential colony-forming cells(HPP-CFC) were determined by semi-solid medium clonal culture.RESULTS:AGM-derived and bone marrow(BM)-derived stromal cells were similar in morphology and phenotype,and had common character of stromal cells.Supported by AGM stromal cells or by BM stromal cells,more primitive progenitor cells HPP-CFC were expanded,but BL-CFC expansion was only detected in AGM-derived stromal cells.In the supporting of BM stromal cells CD34+ hematopoietic stem/progenitor cells were expanded 3-4 times,but no significant expansion in CD34+Sca-1+ cells was observed.While in the supporting of AGM stromal cells,both CD34+ hematopoietic stem/progenitor cells and CD34+Sca-1+ cells were expanded significantly from 4 to 5 times,respectively(P

ABSTRACT

AIM:To direct embryonic stem cells(ESCs)into hematopoietic stem cells(HSCs)in vitro by simulating the hematogenic microenvironment in human early embryonic aorta-gonad-mesonephero(AGM)region.METHODS:Murine E14 embronic stem cell line was used for two-step differentiation.In the first step of primary differentiation,E14 ESCs were seeded into semisolid methylcellulose-based medium containing bone morphogenesis protein 4(BMP4)and vascular endothelial growth factor(VEGF)for embryoid body(EB)formation.On days 3,6,9,12 and 15,single EB cells were analyzed for Flk-1+ cells amount through flow cytometry.In the second step,single cell from EB containing most Flk-1+ cells was further co-cultured with human AGM stromal cells in non-contact system.On co-culture days of 3,6,9 and 12 days,cells were collected for cell count,flow cytometry for Sca-1+c-kit+ cells analysis,and colony forming cell assay.RESULTS:During the EB formation,BMP4+VEGF promoted Flk-1+ cell genesis on day 9 at peak pencentage value of 27.53%?2.84%,which was statistically higher than that in control group as 8.77?1.10(P
