ABSTRACT

Platinum-based chemotherapy resistance is a key factor of poor prognosis and recurrence in hepatocellular carcinoma (HCC). Herein, RNAseq analysis revealed that elevated tubulin folding cofactor E (TBCE) expression is associated with platinum-based chemotherapy resistance. High expression of TBCE contributes to worse prognoses and earlier recurrence among liver cancer patients. Mechanistically, TBCE silencing significantly affects cytoskeleton rearrangement, which in turn increases cisplatin-induced cycle arrest and apoptosis. To develop these findings into potential therapeutic drugs, endosomal pH-responsive nanoparticles (NPs) were developed to simultaneously encapsulate TBCE siRNA and cisplatin (DDP) to reverse this phenomena. NPs (siTBCE + DDP) concurrently silenced TBCE expression, increased cell sensitivity to platinum treatment, and subsequently resulted in superior anti-tumor effects both in vitro and in vivo in orthotopic and patient-derived xenograft (PDX) models. Taken together, NP-mediated delivery and the co-treatment of siTBCE + DDP proved to be effective in reversing chemotherapy resistance of DDP in multiple tumor models.

ABSTRACT

Chronic obstructive pulmonary disease (COPD) has a high global morbidity and mortality and a severe disease burden, yet progress in treatment and prevention has been slow in recent decades. Early COPD has few symptoms and is severely underdiagnosed and undertreated; it is crucial to search for effective clues of early COPD and provide management interventions. By reviewing the definition, risk factors, diagnosis and management interventions, this study explores the disease evolution of early-stage COPD, which can help clinical practice to develop more effective preventive and therapeutic strategies for stopping or slowing down the natural progression of the disease, improving the long-term prognosis, and reducing the disease burden.

ABSTRACT

Objective To understand the influencing factors of oppositional defiant disorder(ODD)in junior middle school students and their interrelationships,and to provide theoretical basis for formulating ODD intervention programs.Methods Cluster random sampling was used to investigate 670 freshmen of two junior middle schools in Wuhan.ODD scale,parenting style scale,bullying scale,adolescent mental resilience scale and mental sub-health scale were used for questionnaire sur-vey.Structural equation model was constructed to explore the effects of parenting style,school bullying,mental resilience and mental sub-health on ODD.Results The prevalence of ODD among junior middle school students in Wuhan city was 9.55%.Rural household registration,instability of fathers'career,and poor parental relationship were latent risk factors of ODD among junior middle school students.ODD was positively associated with negative parenting style,school bullying and mental sub-health,and negatively correlated with positive parenting style and psychological resilience.The parenting style models construc-ted in both fathers and mothers had satisfying fitting effects,and each fitting index had reached the fitting standard.The total effect value of each factor in the father's parenting style model on ODD was father's punishment(0.191),psychological sub-health(0.171),psychological resilience(-0.097),bullying(0.071),father's refusal and denial(0.010).The total effect value of each factor in the mother's parenting style model on ODD was mother's punishment(0.215),psychological sub-health(0.163),psychological resilience(-0.092),bullying(0.067),mother's emotional warmth and understanding(-0.012),mother's rejec-tion and denial(0.010).Conclusion Parenting style can directly affect ODD,and it can also indirectly affect ODD through psy-chological resilience,school bullying and mental sub-health.ODD can be prevented by improving parenting style,reducing school bullying and improving personal psychological quality.

ABSTRACT

The oral and maxillofacial region contains oral organs and facial soft tissues. Due to the complexity of the structures and functions of this region, the repair of related defects is complicated. Different degrees of defects require different repair methods, which involve a great combination of medicine and art, and the material requirements are extremely high. Hence, clinicians are plagued by contemporary oral repair materials due to the limitations of bone harvesting, immune rejection, low osteogenic activity and other problems. Decellularized extracellular matrix has attracted much attention as a bioactive scaffold material because of its nonimmunogenic properties, good osteogenic properties, slow release of growth factors, promotion of seed cell adhesion and maintenance of stem cell characteristics. This article reviews the sources, preparation methods, application and research progress of extracellular matrix materials in the repair of oral and maxillofacial defects to provide an overview for fundamental research and clinical development.

ABSTRACT

Bronchial asthma (asthma) is a complex heterogeneous disease, with a high rate of missed diagnosis and misdiagnosis. Repeated attacks of bronchial asthma can cause complications such as chronic obstructive pulmonary disease, emphysema, and pulmonary heart disease. In recent years, mass-spectrometry-based metabolomics has developed rapidly. It can sensitively identify metabolic fluctuations and pathological changes in patients with asthma. By analyzing the molecules produced by various metabolic pathways, it can help us to find relevant biomarkers and provide a better method for early diagnosis and severity assessment of asthma. We reviewed and analyzed the literature of metabolomics technology in disease progression, early diagnosis and severity assessment, so as to provide reference for asthma research.
